(試験例1)ヒト表皮角化細胞におけるヒアルロン酸量測定1
ビタミン類は、2−プロパノールで1000倍濃度に調製した後、培地中に0.1%添加した。すなわち、96穴プレートに正常ヒト表皮角化細胞(クラボウ)をHuMedia KG-2培地(クラボウ)にて10,000cells/wellの密度で播種し、1日後、ビタミンB6のみ、VA+VC+VD+VEおよびVA+VB6+VC+VD+VEを表5に記載の濃度及びそれらの10倍希釈液及び100倍希釈液を添加したHuMedia KB-2培地(クラボウ)と交換した。なお、コントロールは媒体の0.1%2−プロパノールとした。被験物質添加より2日後に培地を採取し、培地中に分泌されたヒアルロン酸濃度を、ヒアルロン酸測定キット(生化学バイオビジネス株式会社)を用いて定量した。結果を表6及び図1に示した。なお、データは各群4例の平均値である。
