本明細書中に議論するとおり、本発明の実施形態は、1種以上の治療薬をコードする第1配列及び生体応答調節物質(BRM)の発現に干渉する1種以上の薬剤が発現される第2配列を含む、新しい種類のベクターを提供する。好ましい実施形態において、干渉剤は、RNAi分子のこともある。単一のベクターから2種以上のタンパク質の発現を指示する核酸ベクターは、バイシストロン性又はマルチシストロン性ベクターとして当分野に公知である。シストロンは、単一のポリペプチドをコードする遺伝学的単位として定義される。本明細書でのシストロンは、哺乳動物宿主中で活性であり、その産物が、免疫治療又は遺伝子療法に直接関与する。いくつかの実施形態において、治療薬は、免疫療法で使用するための、1種以上の免疫原性剤のことがある。1種以上の免疫原性剤は、例えば、腫瘍関連抗原などの抗原でありうるが、これに限定されない。例えば、いくつかの実施形態において、治療薬は、遺伝子治療に使用するための、1種以上の遺伝子治療薬のことがある。

本明細書に開示された組成物のいずれも、キットにまとめることができる。より詳細には、本発明のバイシストロン性ベクター実施形態を設計又は構築するための成分の全て又はサブセットを、キット中に共に包装することができる。1種以上の治療薬及び生体応答調節物質の発現に干渉する1種以上の同時発現する薬剤は、別にでも、共にでも包装できる。いくつかの実施形態において、プラスミドを、1種以上の治療薬又は生体応答調節物質の発現に干渉する1種以上の同時発現する薬剤と共に包装することが好ましい。本発明の実施形態において、治療タンパク質、ペプチド、ポリペプチド、エピトープ又はそのようなものをコードする核酸は、共にでも、単一の分子としても、分子のセットとしても包装できる。いくつかの実施形態において、生体応答調節物質の発現に干渉する同時発現する1種以上の薬剤を、共にでも、単一の分子としても、分子のセットとしても包装できる。いくつかの実施形態において、1種以上の治療分子及び生体応答調節物質の発現に干渉する同時発現する1種以上の薬剤をキット中に共に包装できる。或いは、本明細書に開示される組成物は、治療薬としての使用のために、互いに組み合わせて使用し本明細書で開示されるバイシストロン性ベクターを設計及び構築する方法を記載する、印刷形態又は機械読み取り媒体での説明書と共に、個別に包装及び販売される。
pSEMプラスミド(pMA2Mとしても知られる)の構造及び構築は、すでに開示されている(米国特許出願公開第20030228634号及び国際公開第03/063770号)。簡単に述べると、pSEMプラスミドは、Melan−A26-35 A27L由来のHLA A2特異的CTLエピトープELAGIGILTV(配列番号1)及びアミノ酸31〜48及び56〜69にエピトープクラスターを含むMelan−A(配列番号2)の一部(アミノ酸31〜96)を持つ1ポリペプチドをコードする。これらのクラスターは、参照によりその全体が本明細書に取り込まれている、「エピトープクラスター」という名称の2000年4月28日出願の米国特許出願第09/561,571号にすでに開示された。ヒトチロシナーゼから誘導された短いアミノ酸配列(配列番号3及び4)が、定義されたMelan−ACTLエピトープに横付けし、免疫プロテアソームによるプロセシングによるMelan−4ハウスキーピングエピトープの遊離を促進する。さらに、これらのアミノ酸配列は、潜在的なCTLエピトープ自体を表す。プラスミド中のポリペプチドのcDNA配列は、サイトメガロウイルス由来のプロモーター/エンハンサー(CMVp)の制御下にあり、APCによる取り込みの際ポリペプチドのメッセンジャーを有効に転写する。コード配列の3’末端のウシ成長ホルモンポリアデニル化シグナル(BGHpolyA)は、安定性増大のためのメッセンジャーのポリアデニル化のための、並びに翻訳のための核から細胞質への移送のためのシグナルを与える。取り込み後の核へのプラスミド輸送を容易にするため、サルウイルス40(SV40)由来の核内移行配列(NIS)をプラスミド骨格に挿入した。プラスミドは、CpG免疫刺激性モチーフの2コピーを、1つはNIS配列に、1つはプラスミド骨格に保持する。最後に、プラスミド中の2つの原核生物遺伝因子は、大腸菌、カナマイシン耐性遺伝子(Kan R)及びpMB1細菌複製起点中の増幅に責任がある。
HEK 293T細胞を、Melan−A発現プラスミドpcDNA−Melan―Aのみによりトランスフェクトし、或いは、それぞれ、pSEM−U6−Melan−A、pSEM−U6−GFP、Melan−AのsiRNA、又はコントロールsiRNAとコトランスフェクトした。トランスフェクトの48時間後、細胞を採取し、細胞溶解物を調製し、SDS−PAGE及び免疫ブロットにかけた。種々のsiRNA及びバイシストロン性プラスミドのノックダウン効果を評価した(図2)。Melan−Aに特異的なsiRNAのコトランスフェクションは、トランスフェクトされた細胞中でのMelan−A発現のレベルを著しく低下させ、ノックダウン効果は90%を超えた。pcDNA−Melan―A及びpSEM−U6−Melan−Aとコトランスフェクトされた細胞では、Melan−A発現のノックダウン効果は、80〜90%と見積もられる。Melan−A発現プラスミド及びpSEM−U6−GFP、又はコントロールsiRNAとそれぞれコトランスフェクトされた細胞のサンプルでは、Melan−A発現レベルのわずかな低下が観察された。
5群のHHDトランスジェニックマウス(n=10/群)を、第1日及び第4日に、各リンパ節に25μlのPBS中25μgを鼠径リンパ節に直接注入し、プラスミド(pSEM、pSEM−U6−GFP、pSEM−U6−Melan−A)で免疫した。10日後、第11日及び第14日に、マウスに、第2クラスターのDNA注入を行い、第34日及び第37日にMelan−A26-35A27Lペプチド(1mg/ml)を注入した(図3)。後眼窩静脈叢からの採血により個々のマウスから末梢血液を単離し、密度遠心分離(Lympholyte Mammal、Cedarlane Labs)を行った後、単核細胞を赤血球から分離した。免疫動物中の特異性CTL応答を、HLA−A2.1 MART−126-35(ELAGIGILTV)−APC及びFITCコンジュゲートラット抗マウスCD8a(Ly−2)モノクローナル抗体(BD Biosciences)により、40℃で1時間、単核細胞を同時染色して定量した。FACS Caliburフローサイトメーター(BD Biosciences)によりデータを集め、リンパ球集団にゲーティングし、CD8+集団内のテトラマー+細胞のパーセントを計算して、CellQuestソフトウェアにより分析した。値は各群内のテトラマー平均+/−SEMを表し、ナイーブの同腹仔のコントロールと比較した(図4)。
図4に示されるとおり、親プラスミド、pSEMによる免疫は、プラスミドのみの免疫後の7%のMelan−A26-35特異性CD8+T細胞の存在により、マウス中の検出可能な応答をもたらした。そのような細胞のパーセンテージは、Melan−Aペプチドの注入によるブーストの後、全CD8細胞の40%超までマウス中で著しく増加した。対照的に、ベースラインテトラマー陽性CD8細胞は、プラスミド、pSEM−U6−Melan−A、プレ及びポストペプチドブーストにより免疫されたマウス中に検出可能であった。これは、Melan−Aの発現が、pSEM−U6−Melan−Aを取り込んだ抗原提示細胞中で阻害され、そのようなプラスミド促進抗原発現が、プライムブーストレジームにおける免疫応答の誘起に必須であることを示している。pSEM−U6−GFPにより免疫されたマウスでは、おそらくdsRNAに関連するMAK/インターフェロンα経路の活性化により、pSEMで免疫されたマウスに比べ免疫応答の低下が見られた。しかし、ペプチドブースト後のこれらのマウスからの著しい応答(20%テトラマー陽性CD8細胞)は、プライミング事象の間のプラスミドからの抗原発現の重要性をさらに実証する。
細胞傷害性の測定の代わりに、エリスポットアッセイにおいて特異的なエフェクター細胞によるIFN−γ産生を測定して、CD8+CTL応答を評価できる。このアッセイでは、抗原提示細胞(APC)は、マイクロタイターウェルのプラスチック表面上に固体化されており、エフェクター細胞は種々のエフェクター:標的比で加えられる。抗原特異性エフェクター細胞によるAPCの結合により、エフェクター細胞によるIFN−γを含むサイトカインの産生が活性化される。細胞を染色して細胞内IFN−γの存在を検出でき、陽性に染色されている増殖巣(スポット)を顕微鏡下で計測する。
免疫シナプスの形成により、T細胞レセプターは、APC表面上の抗原とMHCの複合体を認識する。T細胞活性化には、T細胞とAPC上のB7ファミリー遺伝子との相互作用を含む共刺激シグナルも要する。さらに、新しく定義されたシグナル3サイトカイン(IL−12又はIL−1b)もT細胞のエフェクター機能に有用でありうる。
バイシストロン性ベクターを設計し、サイトメガロウイルス由来のプロモーター/エンハンサー配列(CMVp)の制御下におかれたMelan−A26-35をコードする核酸配列を含む。さらに、バイシストロン性ベクターは、U6プロモーターの制御下におかれた、B7分子に対して向けられるsiRNAに対応する配列を含む。

Claims (41)
- 少なくとも2つのシストロンを含むベクターであって、第1シストロンが、第1プロモーターと、1種以上の治療薬をコードする第1核酸配列とを含み、第2シストロンが、第2プロモーターと、生体応答調節物質又は前記治療薬の発現に干渉する1種以上のRNA分子とをコードする第2核酸配列を含み、第1配列の発現が第1プロモーターの制御下にあり、第2配列の発現が第2プロモーターの制御下にあるベクター。
- 前記ベクターがプラスミドベクター又はウイルスベクターである、請求項1に記載のベクター。
- 前記第1プロモーターが、機能的に連結されたプロモーター/エンハンサー配列である、請求項1又は2に記載のベクター。
- 前記プロモーター/エンハンサー配列が、CMVプロモーター/エンハンサー配列である、請求項3に記載のベクター。
- 生体応答調節物質の発現に干渉する1種以上のRNA分子が、RNAi、siRNA、又はshRNAである、請求項1から4のいずれかに記載のベクター。
- 前記第2プロモーターがU6プロモーター配列である、請求項1から5のいずれかに記載のベクター。
- 前記生体応答調節物質が、免疫応答の制御又は調節、抗原プロセシング及び提示、又は遺伝子サイレンシングに関与している、請求項1から6のいずれかに記載のベクター。
- 免疫応答の制御又は調節に関与している前記生体応答調節物質が、サイトカイン、ケモカイン、共刺激分子、チェックポイントタンパク質、転写因子、及びシグナル伝達分子からなる群から選択される、請求項7に記載のベクター。
- 抗原プロセシング及び提示に関与している前記生体応答調節物質が、TAPタンパク質、免疫プロテアソーム、標準プロテアソーム、β2マイクログロブリン、MHCクラスI分子及びMHCクラスII分子からなる群から選択される、請求項7に記載のベクター。
- 遺伝子サイレンシングに関与している前記生体応答調節物質が、DNAメチル化剤、クロマチン制御分子及びRNA調節分子からなる群から選択される、請求項7に記載のベクター。
- 前記転写因子が、T−bet、STAT−1、STAT−4、又はSTAT−6である、請求項8に記載のベクター。
- 前記サイトカインが、IFN−α、IFN−γ、IL−10、IL−18m、IL−12、又はTGF−βである、請求項8に記載のベクター。
- 前記共刺激因子が、CD40、B7.1、又はB7.2である、請求項8に記載のベクター。

- 前記生体応答調節物質が、LAG−3リガンドである、請求項1から17のいずれかに記載のベクター。
- 前記生体応答調節物質が、樹状細胞活性化抑制剤SOCS1である、請求項1から18のいずれかに記載のベクター。
- 前記DNAメチル化剤が、DMNT1である、請求項10に記載のベクター。
- 前記1種以上の治療薬が、免疫原を含む、請求項1から20のいずれかに記載のベクター。
- 前記免疫原が、腫瘍関連抗原、腫瘍特異性抗原、分化抗原、胎児抗原、癌精巣抗原、発癌遺伝子の抗原、変異した腫瘍抑制遺伝子、染色体転座から生じる独特な腫瘍抗原、ウイルス抗原、及びこれらの断片からなる群から選択される、請求項21に記載のベクター。

- 生体応答調節物質の発現に干渉する前記1種以上のRNA分子がMelan−A−siRNAである、請求項27に記載のベクター。
