Abstract
【解決手段】本発明は、動物、特にヒト又は他の哺乳動物の免疫応答を調節、特に抑制するための組成物の調製における樹状細胞の表面上のC型レクチンに結合する化合物の使用に関する。該組成物は、特に、樹状細胞とT細胞、具体的には、樹状細胞の表面上のC型レクチンとT細胞の表面上のICAMとの相互作用を調節する。組成物は、特異的な抗原に対する免疫応答を阻止/阻害するために、免疫療法のために、免疫抑制のために、自己免疫疾患の治療、アレルギーの治療、及び/又はHIV感染を阻害するために使用できる。C型レクチンに結合する化合物は、好ましくは、マンノース、フコース、植物レクチン、抗生物質、糖、タンパク質、又はC型レクチンに対する抗体である。本発明は、このような抗体にも関し、このような抗体を用いて樹状細胞を単離する方法にも関する。

Description
(項目1) 動物、特にヒト又は他の哺乳動物の免疫応答を調節、特に抑制するための組成物の調製における樹状細胞の表面上のC型レクチン(但し、該C型レクチンは、DEC−205受容体ではない)に結合する化合物の使用。
(項目2) 樹状細胞とT細胞間の一以上の相互作用を調節するための、特に抑制するための組成物の調製における項目1に記載の使用。
(項目3) 樹状細胞とT細胞間の接着、特に樹状細胞の表面上のC型レクチンとT細胞の表面上のICAM受容体、特にT細胞の表面上のICAM−2又はICAM−3受容体間の接着を調節するための、特に抑制するための組成物の調製における項目1及び/又は2に記載の使用。
(項目4) 特異的な抗原に対する免疫応答を阻止し、又は阻害するための、免疫寛容を誘導するための、免疫療法のための、免疫抑制のための、自己免疫疾患の治療のための、及び/又はアレルギーの治療のための組成物の調製における項目1〜3の何れか1項に記載の使用。
(項目5) 樹状細胞のHIV感染を阻害するための、特に樹状細胞の表面へのHIV表面タンパク質(すなわち、gp120)の接着を阻害することによって、前記樹状細胞内へのHIVの侵入を阻害するための組成物の調製における樹状細胞の表面上のC型レクチンを結合する、又はC型レクチンに結合することができる化合物の使用。
(項目6) 感染した樹状細胞から感染していないT細胞へのHIVの転移を阻害するための組成物の調製における、樹状細胞の表面上のC型レクチンを結合する、又はC型レクチンに結合することができる化合物の使用。
(項目7) 動物、特にヒト又は他の哺乳動物の前記抗原に対する免疫応答を調節するための、特に生じさせるための、増加するための、及び/又は促進するための組成物の調製における1)樹状細胞の表面上のC型レクチンに結合し、これに付着した化合物:2)抗原又はその断片若しくは一部の組み合わせの使用。
(項目8) 前記抗原が、C型レクチンに結合することができる化合物に共有結合し、又は融合している項目7に記載の使用。
(項目9) 抗原が、前記抗原、又は感染症(に対するワクチンで使用される)抗原を含有、又は発現する腫瘍細胞に対して免疫応答を生じせしめるために使用することができる癌抗原から選択される項目7又は8に記載の使用。
(項目10) 樹状細胞の表面上のC型レクチンに結合できる化合物が、マンナン及びD−マンノースのようなマンノース炭水化物;L−フコースのようなフコース炭水化物;コンカナバリンAのような植物レクチン;プラジミシンAのような抗生物質;N−アセチル−D−グルコサミン及びガラクトースのような糖;gp120及びその類縁体又はそれらの断片のようなタンパク質;及び樹状細胞の表面上に発現されたC型レクチンに対して誘導された抗体又はその一部、断片、若しくはエピトープからなる群から選択される項目1〜9の何れか1項に記載の使用。
(項目11) 樹状細胞の表面上のC型レクチンが、配列番号1のアミノ酸配列を有するタンパク質、又はその天然の変異形、又はその均等物である項目1〜9の何れか1項に記載の使用。
(項目12) 樹状細胞の表面上のC型レクチンに結合することができる化合物が、モノクローナル抗体、好ましくは、配列番号1のアミノ酸配列を有するC型レクチンに対して誘導されたモノクローナル抗体、又はその天然の変異形若しくは均等物;及び/又はその一部、断片、若しくはエピトープである項目10又は11に記載の使用。
(項目13) 配列番号1又は図9のアミノ酸配列を有するC型レクチンに対して誘導された抗体、好ましくはモノクローナル抗体、又はその天然の変異形若しくは均等物;及び/又はその一部、断片、若しくはエピトープ。
(項目14) AZN−D1又はAZN−D2の何れかである項目13に記載の抗体。

DCは、非活性化T細胞と相互作用し、活性化する能力がユニークである。しかしながら、本発明の以前には、DC−T細胞接触がどのように開始され、調節されるかについては殆ど不明であった。本明細書では、DC−T細胞相互作用におけるICAM−3の役割が調べられている。DCは、ICAM−3に強く接着するが、この接着は、LFA−1、αD、又は他の全てのインテグリンによっては媒介されないことが実証されている。DC上の該新規ICAM−3受容体を検索する上で、DCが多く発現しているC型レクチン受容体がクローニングされ、DC−SIGNと名付けられた。DC−T細胞のクラスタリング及びT細胞応答の開始におけるその顕著な役割に加えて、我々は、DC−SIGNが、DCの感染、及びそれに続くT細胞へのHIV−1伝達に関与する主要なHIV−1受容体であることを発見した。このため、HIVと非活性化T細胞は、DCと相互作用するために、類似の高度に発現された受容体を利用する。
ここで、本発明は、以下に示された実験の部、及び図によってさらに説明されるであろう。
樹状細胞(DC)は、抗原を捕捉し、それらがナイーブT細胞に抗原を提示する場所である二次リンパ組織に移動する。HIV−1は、CD4+T細胞に接近するために、このユニークな能力を破壊する。本発明では、ICAM−2及び/又はICAM−3に高い親和性で結合するのみならず、HIV−1にも結合することができるDC特異的なC型レクチンがクローニングされ、DS−SIGNと名付けられた。また、一過性のDC−T細胞相互作用とDC誘導性のT細胞の増殖を阻害するのみならず、DCのHIV−1感染も効率的に阻害する抗DC−SIGN抗体が開発された。これらの発見は、CD4非依存性のHIVのDC中への侵入に対する理解に重要な結果を有するのみならず、一次免疫応答の開始におけるDC−SIGNの役割に新たな光を投じる。
最初のDC−T細胞接触におけるICAM−3を介した接着の役割を調べた。ICAM−3−Fcキメラをコートした蛍光ビーズ(Geijtenbeek et al.)を含む新規なフローサイトメトリー接着アッセイを利用して、DC、非活性化末梢血リンパ球(PBL)、及び単球がこのインテグリンリガンドに結合する能力をテストした。IL−4及びGM−CSFの存在下で、7日間単球を培養した後に得られた、未成熟なDCは、予めβ2インテグリンの活性化がなくても、強固にICAM−3を結合する(72%、図1A)。図1は、DCのICAM−3への接着が、Ca2+依存性であり、インテグリン非依存性であることを実証している:図1A、B、及びCには、少なくとも3つの実験のうち代表的な実験が1つ示されている(SD<5%)。
DC−SIGNをより詳細に調べるために、ICAM−3結合受容体に対する抗体を産生せしめた。DCで免疫したマウスの脾臓をSP2/0ミエローマ細胞と融合し、DC特異的なICAM−3への接着を阻害することができる抗体の存在について、得られたハイブリドーマの上清をスクリーニングした。2つのハイブリドーマを選択し、クローニングし、得られた抗体をAZN−D1及びAZN−D2と名付けた。精製された両抗体は、DCのICAM−3への接着を強く阻害するが、LFA−1を介した単球のICAM−3への接着には影響を与えない(図2A)。図2は、抗体AZN−D1とAZN−D2が、DCのICAM−3への接着を阻害し、DCによって特異的に発現されている抗原を認識することを実証している。
DC−SIGNを同定するために、抗DC−SIGN抗体AZN−D1を用いたDC溶解物からの予備的免疫沈降を行い、SDS−PAGEゲルから44kDaタンパク質を切り出した。トリプシン消化後、生じたペプチドをゲルから抽出し、予備的HPLCを用いて精製した。続いて、エドマン分解操作を用いて2つのペプチドのアミノ酸配列(0.5〜1pmol)を決定した。両ペプチドは、11アミノ酸残基からなり(図3B)、該ペプチド配列は、公知のタンパク質との相同性をEMBLデータベースでスクリーニングするために使用された。両ペプチドは、ヒトHIVgp120結合C型レクチンの推定アミノ酸配列(Curtis et al.,1992)と100%同一であることが証明された。該タンパク質は、これまでは、ヒト免疫不全ウイルス(HIV)エンベロープ糖タンパク質gp120に対するCD4非依存性の受容体として、専ら胎盤だけに同定されていた。
AZN−D1/D2抗体を用いた多岐にわたる造血細胞群のフローサイトメトリー分析は、抗原が、DCによって優先的に発現されていることを実証している(表1)。テストした全ての造血細胞は、DCを除いて、DC−SIGNの発現が陰性であった。さらに、RT−PCR分析は、DC−SIGNをコードするmRNAが、DC中で特異的に転写されているのを確認し、これは、DC−SIGNタンパク質の発現パターンと一致している(表1)。
DCが、DC−SIGN依存性の態様で、ICAM−3を発現しているトランスフェクタントに結合することを実証するために、ICAM−3をコードするcDNAをトランスフェクトした白血病細胞株K562(K562−ICAM−3)がDCに結合する能力を調べた。図6Aに示されているように、相互作用が抗DC−SIGN抗体によってブロックできるので、DCは、DC−SIGN依存性の態様で、K562−ICAM−3とクラスターを形成する。DCとK562との間ではクラスタリングが全く観察されなかったことは、ICAM−3がDC−SIGNに対するリガンドであることを実証している。図6は、DC−SIGNを介したDCのICAM−3への接着が、DC−Tリンパ球相互作用に関与していることを示す。
DC−SIGNのICAM−3への結合は、DCのT細胞とのクラスタリングに重要なので、DC誘導性のT細胞増殖におけるDS−SIGNの役割も調べた。遮断抗DC−SIGN抗体の存在下、又は非存在下で、同種異系DCを用いて非活性化Tリンパ球を刺激した。図6Cに示されているように、抗DC−SIGN抗体AZN−D1とAZN−D2は、ともにTリンパ球の増殖を75%超、阻害した。同様に、T細胞上のCD2に結合し、T細胞増殖を媒介することも知られている共同刺激分子LFA−3に対する抗体は、T細胞の増殖を阻害する。抗LFA−3と抗DC−SIGN抗体の組み合わせは、T細胞の増殖を完全に阻害する(図6C)。
上記のごとく、DC−SIGNは、胎盤HIVgp120結合レクチンと同一であり、DCによって豊富に発現されていることが実証されたので、DC−SIGNは、HIV−1のDC中への侵入を促進し得るかもしれない。これを調べるために、DCにHIV−1をパルスし、DC−T細胞共培養中の増殖性感染を測定した。IL−4とGM−CSFの存在下で、7日間培養した後に採集したDCは、低レベルのCD4(Blauvelt et al.,1997;Granelli−Piperno et al., J Exp Med 184(6),2433−8,1996)と高レベルのDC−SIGN(図7)を発現した。図7に示されているように、DCに、2時間、HIV−1をパルスし、洗浄し、活性化されたPBMC細胞の存在下で培養すると強力な増殖性感染が起こる。DC−T細胞の共培養の3日目までに、HIV−1複製の指標であるp24Gagタンパク質が、培地中に蓄積し始め(図7)、他者によって示されたのと同様に(Blauvelt et al.,1997;Granelli−Piperno et al.,1998;Granelli−Piperno et al., Curr Biol 9(1),21−29,1999)、HIV−1が共培養中で効率的に複製されることを実証している。しかしながら、HIV−1パルスに先立って、DCを抗DC−SIGN抗体とプレインキュベートし、活性化されたPBMCとインキュベートすると、DC−T細胞の共培養の3及び5日目に示されているように、HIV−1の複製が75%超、阻害される(図7)。HIV−1でパルスした後、抗DC−SIGN抗体とDCをインキュベートすると、DC−T細胞の共培養中に、効率的なHIV−1複製が、なお観察された(図7)。これらの発見は、DCからT細胞へのHIV−1の伝達ではなく、HIV−1のDCへの結合の阻害を通じて、抗DC−SIGN抗体がHIV−1の感染をブロックすることを実証しており、DC−SIGNが、DC上の主要なHIV−1受容体として作用することを示している。このように、DC−SIGNは、DC上に高度に発現されており、ICAM−3とHIV−1の両者に対するDC特異的な受容体として機能する。
例9A:抗体
以下の抗体:KIM185(抗β2インテグリン、(Andrew et al.,Eur.J.Immunol.23,2217−2222,1993)、AZN−L19(抗β2インテグリン、)、NKI−L15(抗αL、(Keizer et al.,Eur.J.Immunol.15,1142−1147,1985))、AIIB2(抗β1インテグリン、(Da Silva et al.,J.Immunol.143,617−622,1989))、CBR−IC3/1及びCBR−IC3/2(抗ICAM−3(de Fougerolles et al.,J.Exp.Med.177,1187−1192,1993))、CD14(WT14())、CD4(wt4())を使用した。抗DC−SIGN抗体AZN−D1及びAZN−D2は、BALB/cマウスをDCで免疫し、続いて、ハイブリドーマ上清が、蛍光性ビーズの接着アッセイによて測定したDCのICAM−3への接着をブロックする能力をスクリーニングすることによって得られた。
DCは、記載に従って単球から培養した(Sallusto and Lanzavecchia,J.Exp.Med.179,1109−1118,1994;Romani et al.,J.Exp.Med.180,82−93,1994)。簡潔に述べれば、接着工程によって、新鮮なPBMCから単球を単離した。IL−4(Schering−Plough,Brussels,Belgium;500U/mL)及びGM−CSF(Schering−Plough,Brussels,Belgium;1000U/mL)の存在下で、単球を7日間培養した。4日目に、新鮮なサイトカインを添加した。7日目に、MHCクラスI及びII、CD1a、p150,95、及びCD80の発現のフローサイトメトリー分析によって、培養されたDCの表現型を確認した。ICAM−3を発現している安定なK562トランスフェクタント(K562−ICAM−3)は、記載のごとく(Lub et al.,Mol.Biol Cell 8,719−728,1997)エレクトロポレーションによって、10μgのPCRII ICAM−3 R1プラスミド(D.Simmons博士から入手)と2μgのPGK−hygベクター(te Riele et al 1990)でK562をトランスフェクションすることによって作成した。非活性化T細胞(>90% CD3 陽性)は、記載のごとく(Figdor et al.,J.Immunol.Methods 68,73−87,1984)、健康なドナーの骨髄からPBMCの遠心による水力分級によって得た。
ラクトペルオキシダーゼ法(Pink and Ziegler,1979,in:Research Methods in Immunology,L.Lefkovits and B Pernis,eds.(New York:Academic pres),pp.169−180)により、細胞をNa125I(Amersham,Buckinghamshire、UK)で表面標識した。4℃で1時間、溶解用緩衝液(1% NP−40、50mMトリエタノールアミン(pH7.8)、150mM NaCl、1mM CaCl2、1mM MgCl2、1mM PMSF、0.02mg/mLロイペプチン)の中でDCを溶解した。4℃で15分間、13,000gでの遠心によって、核の破砕物を溶離液から除去した。タンパク質A−セファロースCL−4Bビーズに共有結合したmAbとともに、予め清澄にした溶離液を3時間インキュベートした(Pharmacia Fine Chemicals、Piscataway、NJ)。免疫沈降物を溶解用緩衝液で徹底的に洗浄し、Laemmliの操作(Laemmli,Nature 227,680685,1970)変法に従って、SDS−PAGEによって分析した。切り出したタンパク質をトリプシン消化し、生じたペプチドを精製し、Eurosequence BV(Groningen, The Netherlands)によって配列分析を行った。
酸性のグアニジニウムイソチオシアネート−フェノール−クロロホルム操作によって全RNAを単離した(Chomczyski and Sacchi,Anal Biochem 162(1),156−9,1987)。胎盤gp120結合C型レクチンをコードするcDNAは、全RNAに対するRT−PCRによって、DCから増幅した。PCRプライマーは、胎盤gp120結合C型レクチンのヌクレオチド配列(受付番号M98457、(Curtis et al.,1992))に基づいており、ヌクレオチド配列(5’から3’)は、以下のとおりである:XF29、AGAGTGGGGTGACATGAGTG;XR1265,GAAGTTCTGCTACGCAGGAGである。PCR断片をpGEM−Tベクター(Promega,Madison WI)中にクローニングし、配列を決定した。クローニングされたcDNAのヌクレオチド配列は、胎盤gp120結合C型レクチンの配列と同じであった(Curtis et al.,1992)。続いて、真核細胞発現ベクターpRc/CMV中にcDNAをクローニングし(pRc/CMV−DC−SIGN))、DEAEデキストラン法(Seed and Aruffo,Proc.Natl.Acad.Sci.U.S.A.84,3365−3369,1987)を用いて、pRc/CMV−DC−SIGNでCOS7細胞を一過性にトランスフェクトした。
以前記載したとおりに(Geijtenbeek et al.,1999 投稿された)、ICAM−1FcとICAM−3を用いて、カルボキシレートで修飾されたTransFluorSpheres(488/645nm、1.0μm;Molecular Probes,Eugene,OR)をコートした。簡潔に述べれば、20μLのストレプトアビジン(50mM MES緩衝液中5mg/mL)を50μLのTransFluorSpheresに添加した。30μLのEDAC(1.33mg/mL)を添加し、混合液を室温で2時間インキュベートした。反応は、100mMの最終濃度になるようにグリシンを添加することによって停止された。ストレプトアビジンでコートされたビーズをPBS(50mM リン酸、0.9% NaCl pH7.4)で3回洗浄し、150μLのPBS、0.5%BSA(w/v)中に再懸濁した。0.5mLのPBA中、37℃で2時間、ビオチン化されたヤギ−抗ヒト抗FcFab2断片(6μg/mL)とともに、ストレプトアビジンでコートされたビーズ(15μL)をインキュベートした。PBS、0.5%BSAで一度ビーズを洗浄し、0.5mL中、ヒトIgG1Fc融合リガンド(ICAM−1 Fc、VCAM−1 Fc;250ng/mL)とともに、4℃で一晩インキュベートした。リガンドをコートしたビーズを洗浄し、100μLのPBS、0.5%BSA中に再懸濁し、4℃で保存した。ICAM−1 FcとICAM−3 Fcは、ヒトIgG1 Fc断片(D.Simmons博士が提供)に融合されたタンパク質の細胞外部分からなる。蛍光性ビーズの接着アッセイは、Geijtenbeekら(投稿された)によって記載されたように行った。簡潔に述べれば、Tris−Sodium−BSA緩衝液(20mM Trsi−HCl pH8.0、150mM NaCl、1mM CaCl2、2mM MgCl2、0.5% BSA;5×106細胞/mL)中に再懸濁した。遮断mAb(20μg/mL)あり/なしで、10分間室温で、96ウェルのV字底プレート中、50.000細胞をプレインキュベートした。リガンドをコートされた蛍光性ビーズ(20ビーズ/細胞)と様々な刺激/阻害剤を添加し、37℃で30分間、懸濁物をインキュベートした。細胞を洗浄し、100μLのTSA中に再懸濁した。FACScan(Becton and Dickinson & Co.,Oxnard,CA)を用いたフローサイトメトリーによって、結合した蛍光性ビーズを有する、細胞のパーセントを測定することによって接着を決定した。
DC、及びICAM−3を発現している細胞間のクラスタリングをフローサイトメトリーによって評価した。DC、及びICAM−3を発現している細胞(2×106細胞/mL)を、それぞれ、スルフォフルオレセイン(Molecular Probes,Eugene,OR;50μg/mL)とハイドロエチジン(Molecular Probes,Eugene,OR;40μg/mL)により、37℃で1時間標識した。洗浄後、DC、及びICAM−3を発現している細胞を混合し(それぞれ50×103細胞)、37℃でインキュベートした。異なる時点で、細胞をパラホルムアルデヒド(0.5%)で固定し、FACScan(Becton and Dickinson & Co.,Oxnard,CA)を用いたフローサイトメトリーによって、異種細胞クラスタリングを測定した。
遮断抗体(20 g/ml)の存在下で、同種異系の反応性Tリンパ球(100×103)をDC−刺激因子(1.5×103)に加えた。該細胞を4日間培養した。4日目に、細胞に、16時間、[3H]メチル−チミジン(1.52TBq/mmol/mmol、0.5μCi/ウェル;Amersham、Buckinghamshire、UK)をパルスし、取り込み量を定量した。
単球由来のマクロファージ(MDM)中で、HIVBa−Lを高力価になるまで増殖させた。MDMに対するウイルス株の滴定から7日後、p24抗原ELISA((Dignostics Pasteur,Marnes la Coquette,France)を用いてTCID50を決定し、104/mLと推定した。室温で20分間、抗体(50μg/mL)とともにプレインキュベートしたDC(50×103)を2時間、(105細胞当たり103の感染単位という多数の感染で)野生型HIVBa−Lをパルスし、洗浄し、PHA/IL−2活性化されたPBMC(50×103)とともに共培養した。DC−T細胞の共培養から3日及び5日後に、上清を集めて、p24抗原ELISAによって、p24抗原レベルを測定した(Diagnostics Pasteur、Marnes la Coquette,France)。PBMCは、IL−2(10U/mL)とPHA(10μg/mL)の存在下でそれらを培養することによって活性化した。
扁桃とリンパ節の凍結切片(8μm)を100%のアセトンで固定化し(10分)、PBSで洗浄し、第一の抗体(10μg/mL)とともに、室温で60分間インキュベートした。洗浄後、製造者のプロトコールに従って、ABC−APベクタスタインキット(Vector Laboratories,Burlingame,CA)を用いて最終染色を行った。核の染色は、ヘマテインを用いて行った()。
