lacZ発現の解析によって、マウスNaV2は4つの脳周囲器官及び中枢神経系のいくつかの神経核において発現することや、マウスNaV2発現細胞は組織分布のみならず細胞タイプにおいても多様性があることが判明したことから、NaV2チャネルの機能、特性の決定が困難となったが、4つの脳周囲組織が体液ホメオスタシスに関与していると考えられていることから(FASEB J 7,678-686(1993)、Annu.Rev.Physiol.59,601-619(1997)、Physiol.Rev.78,583-686(1997)、Physiol.Rev.58,582-603(1978)、Ann.NY Acad.Sci,877,258-280(1999))、マウスNaV2チャネルが体液容量オスモル濃度の感覚経路において機能していると仮定すれば、これらの器官におけるそのチャネルの活性と遺伝子発現はマウスNaV2変異体マウスにおいて影響を受けると想定された。そこで、マウス及びラットにおける細胞外液バランスに応じた神経活動の変化のマーカーであるFos核タンパクの中枢性発現に対して、水分飢餓がどのように影響するかを以下のように調べた。

次いでマウスNaV2チャネル欠損がマウスの水分及び食塩摂取に及ぼす影響を調べた。行動解析を行うにあたって変異体マウスを雄のC57BL/6Jと戻し交配させた。その結果、F1マウスとN4マウスで同様の結果を得た。ホモ接合型、ヘテロ接合型、野生型の同腹子における嗜好−嫌悪行動を、48時間2瓶嗜好性試験によって測定した。マウスは蒸留水と味溶液の選択を48時間自己のケージで行った。全ての行動解析には12−24週令の雄のマウスが使われた。マウスは一定の室温、湿度及び12/12時間明−暗サイクルにおいて各別にケージに入れられた。2つの瓶の位置は24時間毎に代えてサイド嗜好性の影響を回避した。動物毎の総摂取量を測定し、式;嗜好率=味溶液の量(ml)/味溶液及び水の総摂取量(ml)により嗜好性を計算した。結果を図5に示す。図5aは段階的濃度の食塩水、図5bは濃度固定の3種類の基本的な味物質についての結果を示し、ホモ接合型(-/-)、ヘテロ接合型(+/-)、野生型(+/+)とも5匹のマウスを用いた。図5aから、水分及び食塩充足条件下では食塩に対する濃度感受性は3つのグループにおいて変わらず、全てのグループにおいて0.1MのNaClを最も好み、0.3M又はそれ以上の濃度に対し嫌悪を示し、ヌル変異体が水分及び食塩が充足した条件下では各種の味物質に対して正常な嗜好を示すことがわかった。また図5bから、甘味(0.5Mの蔗糖)、酸味(0.01MのHCl)、苦味(0.02Mのキニン塩酸塩)に対する嗜好性に差が無いことがわかる。

ヌル変異体の味反応の正常性を調べるため、NaCl味をつかさどる鼓索神経に関する電気生理学的分析を行った。12−24週令の雄マウス(正常条件のために野生型4匹とホモ接合型マウス5匹;急性食塩欲求条件のための上記マウス3匹及び5匹)にペントバルビタールナトリウム(60mg/kg)を腹腔内投与して深く麻酔した後に各マウスを気管切開し、頭保持具で固定した。鼓索神経を露出させて周囲の器官から隔離させ、ブラ(bulla)の入口部位で切断した。神経の全束を切断し白金ワイヤー記録電極上(0.1mm直径)に持ち上げた。中性電極を近傍の組織に付けた。神経活動は増幅され、オシロスコープに表示され、オーディオアンプリファイアーでモニターされた。増幅された信号は時間定数0.3秒で積分機に通され、スリップチャート記録計上に表示された。

To examine the normality of the null mutant taste response, an electrophysiological analysis of the chorda tympani nerve responsible for the NaCl taste was performed. Nav2チャネルがナトリウム濃度依存性ナトリウムチャンネルであることを以下の実験により確認した。まず、抗Nav2抗体を次のようにして作製した。ラットNav2チャンネルのインタードメイン2−3に存在するアミノ酸配列(SVSETVPIASGESDIK;配列番号8)からなるペプチドにヘモシアニンを結合させ、このヘモシアニン結合ペプチドを免疫原として白色ウサギを免疫し、常法により抗ラットNav2ウサギ抗血清を作製した。作製した抗血清を、免疫原に用いたペプチドが固定化されたカラムによって精製し、抗Nav2抗体を得た。この抗Nav2抗体の特異性はウエスタンブロットと免疫組織化学的解析によって確認した。精製した抗Nav2抗体を使用すると、遺伝子欠損マウスから作製した脳、肺、脊髄後根神経節、舌の切片には非特異的な陽性シグナルは認められなかった。
