Abstract
【解決手段】 本発明は、マイクロテスラ磁場において磁束トランスを介して高臨界温度(高Tc)超電導量子干渉素子(SQUID)磁束計を用いた高分解能プロトン核磁気共鳴およびイメージング(NMR/MRI)の方法及びその装置を提供する。SQUIDと入力コイルの両方が、環境雑音をシールドし、SQUIDを安定した動作状態にする超伝導容器内に設置される。本発明はまた、試料がSQUID検出器から遠くにあってもNMR信号を維持できるという利点を提供する。

(SQUID)を利用しており、これは液体ヘリウムでの冷却が必要な低臨界温度(低Tc)SQUIDを使用するよりも安価で作業が容易である。
10mT以上の範囲の前磁場を発生できる。最良の実施例において、前記前磁場は
45mTである。高Tc SQUIDベースのNMRスペクトロメータは電磁シールド室内に設置される。一様磁場を発生するために、3組のコイルペアから成る平面コイルシステムが設置され、これは笹田らによる「Planar coil system consisting of three coil pairs for producing a uniform magnetic field(一様磁場を発生するための3組のコイルペアから成る平面コイルシステム)」Jounal of Applied Physics99、08D904(2006)が開示されている。これら3組のコイルペアは順に接続され、64cm3の試料に101μTで104分の1より優れた均一度を形成する。101μTの静磁場B0は高Tc SQUID磁束計の平面に平行なz軸に沿ってアクティブである。45mTの前分極磁場BpがやはりSQUID磁束計の平面に平行なy軸 に沿って印加される。前記前分極磁場の強度は前記測定静磁場の強度よりずっと高いため、水の核スピン磁化の方向は前記前分極磁場のy軸に沿ってほぼ揃えられる。継続時間Tbpの間前分極磁場を印加した後、前記前分極磁場が3ms後にクエンチされる。核磁化の歳差運動はBoの方向に沿っている。プロトンスピンの自由誘導減衰(FID)信号が検出コイルによって検出される。検出コイルのノーマルはz軸方向に沿っており、超伝導容器内にシールドされた高Tc SQUIDに結合され、前記SQUIDの出力は増幅・積分された後、前記超伝導容器内のSQUIDの入力コイル近くに配置された帰還コイルに帰還される。FIDのNMR信号はバンドパスフィルタによってフィルタされる。その後、高速フーリエ変換(FFT)を通してNMRスペクトルを取得でき、かつ検出されたNMR信号からイメージを形成することで試料の磁気共鳴イメージング(MRI)を実行することもできる。
300Kで4.5Ω、入力コイルの抵抗Rinputは77.4Kで1Ωである。このため、熱雑音<V2>1/2 Johnson=2.91×10−10V/Hz1/2 が磁束トランスで生じる。前記熱雑音は187fT/Hz1/2の環境磁場雑音をSQUIDに結合する。この雑音は、101μTのプロトン共鳴周波数で動作する高Tc SQUID磁束計における雑音より小さい。さらに雑音を低減するため、グラジオメータ構成で高Tc超伝導線材または冷却された極低温磁束トランスを使用することができる。
図1に示すように、高Tc SQUIDベースのNMR/MRI検出システムは電磁シールド室に設置される。スピンは、キャパシタとインダクタンスに接続されて
NMR共振回路を形成する検出コイルと入力コイルを含む磁束トランスを介して
SQUID磁束計に誘導結合される。また、前記SQUIDと前記入力コイルは共に、環境雑音を遮蔽し、SQUIDをより安定した動作状態にするBi2Sr2Ca2Cu3Oyから成る超伝導容器内に設置される。
4.3kHzで280fT/Hz−1/2の磁性感度を示している。
cmで4回、第3コイルペアは半径38.9cmで140回巻装されている。これら3組のコイルペアは順に接続され、64cm3の試料に101μTで104分の1より優れた均一度を形成する。
図4にNMR/MRI測定に用いられるシーケンスを示す。101μTの静測定磁場B0 はSQUID磁束計の平面に平行なz軸方向に沿ってアクティブであり、45mTの前分極磁場Bp がSQUID磁束計の平面に平行なy軸に沿って印加される。前分極磁場の強度が静測定磁場の強度よりずっと大きいため、プロトンの核スピン磁化の方向は、ほぼy軸に沿った状態となる。Tbpの短時間分極磁場を印加した後、前分極磁場は3ミリ秒でオフにされる。核磁化は測定磁場B0の方向に沿って進行する。MRIに24.6 μT/mの勾配磁場を発生する勾配コイルがx、y、z方向に沿って設けられる。MRIは、印加された勾配磁場を各イメージングシーケンスについて15°回転させ、逆投影法で撮られている。
図5に純水10mlの典型的なNMR信号およびそのフーリエ変換を示す。SNRは45、線幅は0.9Hzである。101μTおよび5×103分の1の磁場でシングルショット測定されている。
24.6μT/mの勾配磁場で逆投影法を使った方向に沿って撮られている。フィルタ補正逆投影MRIははっきりとした画像コントラストを示している。画像内に記された被検ファントムの形状を示す。
MRIにおいて空間分解能はそれぞれΔx=1および3mmが見込まれる。この分析は、図7(b)に示された画像は分解できないが、図7(c)に示された画像ははっきりと分解が可能であることを説明している。
リン酸塩(31P)核とプロトン(1H)核間のJ結合を示すリン酸トリメチルのNMRスペクトル((CH3O)3PO)において、磁場B0におけるプロトンのスピンのハミルトニアンは次のように表すことができる。

そのうち、γHとSHはそれぞれ磁気回転比とプロトンの核スピンを表し、Jはスカラー結合定数、そしてmPはリン酸塩核スピンのz成分である。これは、1H核に対する31P核の作用が、プロトン核の位置で2πJmPi/γHの大きさを持つ追加磁場のようであることを意味する。この磁場は31P核による電子雲により誘起された磁場に対応する。
μTで低Tc SQUIDを用いたリン酸トリメチルのスカラー結合はJ3[P,H]=
(10.94 ± 0.6)であることが報告されている。NMRスペクトルは100の過渡信号の平均であり、McDermott Rらの「Liquid−State
NMR and Scalar Couplings in Microtesla
Magnetic Fields(マイクロテスラ磁場における液体状態NMRおよびスカラー結合)」、Science 295、2247、(2002)を参照する。図13で印加された磁場B0は、電源からの出力電流の違いのため図5で用いたものより若干高くなっている。リン酸トリメチルの分子量は140.1(密度=1.2g/ml)であり、純水の分子量(密度=1)は18である。実験における同量のリン酸トリメチルと純水において、水に比べてリン酸トリメチル中のより少量のプロトンがNMR信号に貢献した。このため、リン酸トリメチルで検出されたNMR信号は、純水に比べて小さいSNRを示している。本発明において、本発明の装置を用いることによりワンショットで高感度のJ結合が示された。
図14(a)は水のNMR強度、図14(b)、(c)、(d)、(e)、(f)はそれぞれ0.0003、0.0006、0.0009、0.0012、0.0015(単位emu g-1)磁性濃度の関数として磁性流体のNMR強度を示す。磁性流体は、水中に均一に分散されたデキストランコートを施した磁性ナノ粒子を含む。前記磁性ナノ粒子のサイズは直径30nmである。NMRスペクトルの線幅は、SNR45でのシングルショットで1Hzである。プロトンNMR信号の強度は磁性流体の磁性濃度(Ms)が増加するにつれて低下する。磁性濃度MsはNMR信号の強度を低下させ、線
幅の広がりを引き起こす。図14(g)はNMRスペクトルの線幅および磁性流体の濃度の関数としての強度を示す。磁性流体の磁性は磁場の均一度を低下させ、プロトン核スピンの位相の分散と、スピン‐スピンおよびスピン‐格子緩和の優勢を引き起こす。このため、磁性流体のMsが増加すると、プロトンNMRスペクトルの広がりと緩和率の上昇が認められる。

そのうち、M(t)は瞬間的時間tでの試料の磁化であり、M0=M(t=0)、T* 2は全緩和時間である。T * 2 は次の式で磁場の不均一性変数Γinhomogeneityに関連する。

そのうち、Γinhomogeneityは実効緩和率であり、測定磁場の不均一性によって引き起こされる。磁性流体のMsが0(純水)から0.0015emu g-1に増加されたとき、T* 2は0.43から0.07に減少した。磁性流体は次の式により緩和時間
T* 2に影響する。

そのうち、ΓMFは磁性流体により引き起こされた磁化による緩和率である。式(3)は磁場の不均一性の緩和時間T2に対する効果を表す。磁場均一度の効果に加え、式(4)は磁性流体の磁化が緩和時間T2.に追加的な効果をもたらすことを示す。
NMR測定において、磁場B0は101μTに保たれている。従って、Γinhomogeneityは同一値のままである。式(2)から(4)で、ΓMFはMsの関数として測定されたT* 2から導くことができる。図10(b)はMsの関数として導かれた値
ΓMF = 1/TMF をemu g-1単位で示しており、そのうちΓMFは磁性流体の実効緩和時間(秒単位)である。Msが、水に対して、0から0.0015emu g-1に増加したとき、ΓMFが0から13.5s-1 に増加したことが認められた。磁性流体のMsと磁場の不均一性の両方がプロトン核スピンの位相拡散を引き起こし、スピン‐スピン緩和時間を減少させている。縦緩和時間T1の調査は、Liao S Hらによる「

そのうち、S0は飽和でのNMR強度、tBpはNMR測定における前分極時間である。同じ方法を用い、TBpの関数としてNMR信号が測定され、Ms=0.0006
emu g-1の磁性流体についてT1=140ミリ秒が導かれた。磁性流体のMsの値が0から0.0006emu g-1に増加すると、T1が大きく減少することが分かった。減少したT1磁気双極子‐双極子相互作用によるものである。強磁性粒子とキャリア液体の分子間の不規則な衝突による熱雑音の十分な衝撃は揺らぎを誘起する。

- 試料の核磁気共鳴(NMR)を検出する方法であって、
(a) ミリテスラ磁場で試料の核スピンを前分極させる、
(b) 高臨界温度(高Tc)超電導量子干渉素子(SQUID)磁束計で検出コイルと入力コイルを含む磁束トランスを介しマイクロテスラ磁場で前記試料から核磁気共鳴(NMR)信号を検出する、
という手順を含み、そのうち、前記SQUIDと前記入力コイルが超伝導容器内に設置されることを特徴とする、試料の核磁気共鳴(NMR)を検出する方法。 - 請求項1に記載の試料の核磁気共鳴(NMR)を検出する方法であって、そのうち、前記検出磁場が約200μTから約1μTの範囲にあることを特徴とする、試料の

- 請求項1に記載の試料の核磁気共鳴(NMR)を検出する方法であって、さらにNMR信号からJ結合情報を取得する、という手順を含むことを特徴とする、試料の核磁気共鳴(NMR)を検出する方法。
- 試料の核磁気共鳴(NMR)を検出する装置であって、
(a)試料の核スピンを前分極させるためのミリテスラ磁場を提供する前分極コイル、
(b)検出コイルと入力コイルを含み、そのうち前記検出コイルが前分極コイルに嵌装された磁束トランス、
(c)前記試料から核磁気共鳴(NMR)信号を検出するための高臨界温度(Tc)超電導量子干渉素子(SQUID)磁束計を含み、
そのうち、前記SQUIDと前記入力コイルが超伝導容器内に設置されたことを特徴とする、試料の核磁気共鳴(NMR)を検出する装置。 - 請求項10に記載の試料の核磁気共鳴(NMR)を検出する装置であって、そのうち、前記検出磁場が約200μTから約1μTの範囲にあり、前記前分極磁場が約10mTまたは10mT以上の範囲にあることを特徴とする、試料の核磁気共鳴(
NMR)を検出する装置。 - 請求項10に記載の試料の核磁気共鳴(NMR)を検出する装置であって、そのうち、前記超伝導容器が超伝導Bi2Sr2Ca2Cu3Oyから成ることを特徴とする、試料の核磁気共鳴(NMR)を検出する装置。
- 請求項10に記載の試料の核磁気共鳴(NMR)を検出する装置であって、そのうち、前記高Tc SQUIDベースのNMR磁束計が電磁シールド室内に設置されたことを特徴とする、試料の核磁気共鳴(NMR)を検出する装置。
- 請求項10に記載の試料の核磁気共鳴(NMR)を検出する装置であって、さらに検出されたNMR信号からイメージを形成することにより試料の磁気共鳴イメージング(MRI)を実行する手段を含むことを特徴とする、試料の核磁気共鳴(NMR)を検出する装置。
- 請求項10に記載の試料の核磁気共鳴(NMR)を検出する装置であって、さらにNMR信号からJ結合情報を取得する手段を含むことを特徴とする、試料の核磁気共鳴(NMR)を検出する装置。
