Abstract
【解決手段】哺乳動物の体性幹細胞から骨細胞への分化誘導培地は、哺乳動物細胞培養用基本培地と、哺乳動物の体性幹細胞から骨細胞への分化誘導剤と、内皮細胞分化遺伝子(Edg)ファミリーレセプターに対するリガンドと、セレンとを含み、無血清又は低血清である。体性幹細胞から骨細胞への分化誘導方法は、上記の培地中で、骨細胞へ分化し得る体性幹細胞を培養することを含む。

R−O−CH2CH(OH)CH2PO4H2 (I)
(式中、Rは、炭素数10〜30のアルキル基、炭素数10〜30のアルケニル基又は炭素数10〜30のアシル基である)
で表される化合物である。なお、上記の式(I)のR基についてのアシル基の炭素数は、カルボニル基の炭素数を含まない。
実施例1同様にグルタミン酸を含んだ基礎培地(DMEM)に5μMのLPA、1mMのアスコルビン酸2−リン酸、60nMのセレンおよび90μMのグルタミン酸を添加して無血清コントロール培地(比較例10)を製造した。
Claims (12)
- 哺乳動物細胞培養用基本培地と、哺乳動物の体性幹細胞から骨細胞への分化誘導剤と、内皮細胞分化遺伝子(Edg)ファミリーレセプターに対するリガンドと、セレンとを含み、無血清又は低血清である、哺乳動物の体性幹細胞から骨細胞への分化誘導培地。
- 内皮細胞分化遺伝子ファミリーレセプターに対する前記リガンドの培地中の濃度が0.01μM〜50μM、セレン濃度が1nM〜1μMである請求項1記載の培地。
- 内皮細胞分化遺伝子ファミリーレセプターに対する前記リガンドがリゾホスファチジン酸(LPA)及びその塩、スフィンゴシン1リン酸(S1P)並びに内皮細胞分化遺伝子(Edg)ファミリーレセプターのアゴニストから成る群より選択される少なくとも1種である請求項1ないし3のいずれか1項に記載の培地。
- メラトニン、ビタミンA、ビタミンD、ビタミンKおよび亜鉛から選択される少なくとも1種をさらに含む請求項1ないし4のいずれか1項に記載の培地。
- グルタミン酸をさらに含む請求項1ないし5のいずれか1項に記載の培地。
- 前記分化誘導剤が、β−グリセロリン酸、デキサメタゾン及びビタミンCである請求項1ないし6のいずれか1項に記載の培地。
- 前記基本培地が、DMEM、MEMα、MEM、Ham’s F−12、RPMI−1640、DMEM/F12、Williams培地E、MCDB培地、199培地、Fisher培地、Iscove改変ダルベッコ培地(IMDM)、McCoy改変培地から成る群より選ばれる請求項1ないし7のいずれか1項に記載の培地。
- 無血清培地である請求項1ないし8のいずれか1項に記載の培地。
- 内皮細胞分化遺伝子(Edg)ファミリーレセプターに対するリガンドと、セレンとを含む、哺乳動物の体性幹細胞から骨細胞への分化誘導培地添加剤。
- 体性幹細胞から骨細胞への分化誘導剤をさらに含む請求項10記載の添加剤。
- 水又は基本培地に溶解することにより請求項1ないし12のいずれか1項に記載の培地を与える組成を有する請求項10又は11記載の添加剤。
- 請求項1ないし9のいずれか1項に記載の培地中で、骨細胞へ分化し得る体性幹細胞を培養することを含む、体性幹細胞から骨細胞への分化誘導方法。
