(2)表皮細胞毒性試験
正常表皮角化細胞(クラボウ)を96ウエルプレートに2×104cells/wellの濃度で播種し、HuMedia-KG2培地(クラボウ)で72hr培養した。界面活性剤入り培地に交換し、48hr後、1mg/mL MTT〔3-(4,5-ジメチルチアゾール-2-イル)2,5-ジフェニルテトラゾリンブロマイド〕入り培地に交換した。3hr処理し、イソプロパノールで色素であるホルマザンを抽出し、570nmの吸光度を測定した。細胞残存率は下式で求めた。
