Abstract
【解決手段】ネムノキ属(Albizia julibrissin)ネムノキを極性溶剤或いは非極性溶剤にて抽出して得られるネムノキ抽出物によって、効果に優れたメラニン産生抑制剤、及びコラーゲン産生促進剤が得られる。

Description
〔抽出物の調製〕
ネムノキの樹皮を乾燥後粉砕したもの15gに90mLの精製水を加え、60℃にて一時間加熱抽出した。抽出液をろ過し、40℃で減圧乾燥した残留物を得た。乾燥物が0.01%となるように水溶液を調製し、試料溶液とした。
メラニン産生量の評価を行った。マウスメラノーマ細胞のトリプシン処理を行ない、5%牛胎児血清を含有するDMEM(Invitrogen社)培地で細胞を分散させ、12well plateに5×104/wellを播種し、1日間、37℃で培養を行なう。培地量は各wellあたり1mlになるように添加し培養する。
細胞内に産生されたメラニン量の測定は培養後、細胞を2N-NaOHに溶解し405nmの吸光度を測定した。また、細胞増殖度は2N-NaOHに溶解した細胞溶解液の一部を BCA法を利用したタンパク測定法により540nmの吸光度で測定し、タンパク量に換算した。メラニン産生度は、単位タンパク量あたりのメラニン量の割合で計算した。また、美白効果の陽性対照物質としてβ-アルブチンを用いた。
〔細胞の培養〕
コラーゲン産生量の評価を行った。ヒト線維芽細胞のトリプシン処理を行ない、15%牛胎児血清を含有するDMEM(Invitrogen社)培地で細胞を分散させ、24well plateに2.5×104/wellを播種し、1日間、37℃で培養を行なう。培地量は各wellあたり0.5mlになるように添加し培養する。
〔評価方法〕
細胞内に産生されたコラーゲン量の測定は培養後、細胞をPBSで洗浄し、0.1% SiriusRed酢酸溶液を0.3mL添加し室温で1時間放置したあと、10mM HClで5回洗浄した。その後、0.1M NaOHを0.5mL添加して5分間放置することで細胞を溶解し540nmの吸光度を測定した。また、細胞増殖度は溶解した細胞溶解液の一部を BCA法を利用したタンパク測定法により540nmの吸光度で測定し、タンパク量に換算した。コラーゲン産生度は、単位タンパク量あたりのコラーゲン量の割合で計算した。また、陽性対照物質としては、ビタミンC誘導体であるリン酸マグネシウム塩を用いた。
