この試料混合物を、ガラスキャピラリーの尖端に取り、素子1の収容部18内の基板表面に付着させた。脂質二分子膜の自発展開の様子を調べるために、レーザー共焦点顕微鏡を用いて、以下の手順で観察を行った。
顕微鏡の対物レンズと試料との間に、トリス(ヒドロキシメチル)アミノメタン10mM、NaCl100mMの濃度で塩酸によりpH7.6に調製されたバッファー溶液を満たすことによって、脂質二分子膜の自発展開を開始させた。
図2にその結果を示す。図2は、上記の条件で脂質二分子膜を自発展開させた時の流路部17の経時変化を示したものであり、脂質二分子膜がナノギャップ電極に到達した時刻をt=t0(秒)とし、図中上側から、t0(秒)に対して−300秒、−100秒、0秒、+20秒、+100秒、+200秒および+400秒における流路部17の観察結果をそれぞれ示している。
図2中、流路部17の左側から右側へと経時的に帯状に伸びて行く領域は、試料混合物中に含まれるフルオレセイン−DHPEのフルオレセイン色素からの蛍光を示しており、この結果から、脂質二分子膜の単一膜が、図中左側の収容部から流路部17に入り、流路部17に沿って左から右へと成長していく様子が確認できた。また、脂質二分子膜が、ナノギャップ電極に到達した後、そのギャップを通って、さらにもう一端の収容部へ向かって成長していったことが確認できた。
また、フルオレセイン色素からの蛍光の観察結果から、このとき、試料中のフルオレセイン−DHPEがバッファー溶液中に溶け出したり、ナノギャップ電極の金属部分や有機レジスト部分上に成長していくことがなかったことが確認できた。すなわち、L−α−PCと共に輸送されてきたフルオレセイン−DHPEは、ナノギャップ電極のギャップのみを通って輸送されていた。
なお、脂質二分子膜の自発展開を利用して試料を輸送する代わりに、予め、ベシクルフュージョン法により脂質二分子膜を収容部18、19および流路部17の表面に張っておき、収容部18にフルオレセイン−DHPEを注入したところ、フルオレセイン−DHPEが、脂質分子の拡散によって、上記と同様にギャップ10を通過したことが確認できた。

図3(a)には、本実施例で脂質二分子膜31の自発展開により試料(分析対象分子)32を輸送した際の素子の流路部36の状態を示す。流路部36には、実施例1と同様、1対のナノギャップ電極33、34が設けられており、該ナノギャップ電極33、34には電気測定装置35が接続されている。この素子は、実施例1において、ナノギャップ電極間のギャップを25nmとした以外は実施例1と同様にして作製した。
本実施例では、試料32として、表面を脂質分子で覆われている平均粒径20nmの金微粒子を用いた。該試料32には、可視化のために蛍光標識(NBD色素)が付けてある。該試料32と脂質分子との混合物を調製し、これを、上流側(図中、左側)の収容部(図示せず)に付着させ、実施例1と同様にして自発展開を開始させた。このとき、試料32の濃度は、試料間の距離が充分離れた状況となるよう、脂質分子に対して10−7mol%と充分に希薄な濃度とした。
脂質二分子膜31の成長と共に試料32がナノギャップ電極33、34間を通過することを確認するために、ナノギャップ電極33、34間に流れる電流をモニターした。このとき、該電流をモニターするために、ナノギャップ電極33、34間に50mV、100Hzの交流電圧を印加した。その結果、図3(b)に示すように、経時的にスパイク信号が得られ、ナノギャップ電極を通過した試料32を分子(粒子)レベルで計数することが確認できた。
また、ちょうど試料32がナノギャップ電極33、34間のギャップを通過したところで、交流印加電圧を500mVに上げたところ、ギャップ部分に試料32を捕捉できた。この試料32について、直流で電圧−電流特性を測定した。その結果、図3(c)に示すような結果が得られた。このように、試料32について、分子(粒子)レベルでの電気特性計測を実施できることが示された。

試料として、蛍光色素で標識した2種の脂質分子(図5(a)に示すテキサスレッド標識脂質分子および図5(b)に示すNBD標識脂質分子)の混合物を使用し、実施例1と同様にして試料を含む脂質二分子膜を自発展開で成長させた。所定時間経過後、流路部17表面をレーザー共焦点顕微鏡により観察し、各標識脂質分子からの蛍光強度を測定した。その結果を図4に示す。図4(a)はテキサスレッド標識脂質分子についての測定結果を示すグラフであり、図4(b)はNBD標識脂質分子についての測定結果を示すグラフである。図4に示すグラフの横軸は、ナノギャップ電極の位置を基準(0)とした、流路部に沿った距離を示している。縦軸は、各位置での試料からの蛍光の相対強度である。
テキサスレッド標識脂質分子については、ナノギャップ電極の位置よりも下流側で、30%程度の蛍光強度の落ち込みがあるのに対して、NBD標識脂質分子については、蛍光強度の落ち込みがほとんど見られなかった。これは、テキサスレッド標識脂質分子は、蛍光標識部位のテキサスレッドの大きさが、脂質分子の親水部に比べて立体構造的に大きいため、試料分子同士の立体障害およびナノギャップ電極通過に対する立体障害によってナノギャップ電極を通過しにくいこと、その一方、NBD標識脂質分子は、蛍光標識部位の大きさが脂質分子の親水部とほとんど同じ大きさであるために、脂質二分子膜を構成する脂質分子(標識のない脂質分子)と通過しやすさにほとんど差が出ないこと等によるものと考えられる。
この結果から、ナノギャップ電極、試料のふるいに用いることが可能であることが示された。すなわち、試料の大きさが、脂質二分子膜を構成する脂質分子と比べて有意に大きい場合、試料はナノギャップ構造を通過する際に該脂質分子よりも通りにくくなる。そのため、試料の大きさに合わせてナノギャップ電極のギャップ幅を調節すれば、ナノギャップ電極のギャップの通過前後での試料の濃度を変えることも可能である。

試料62として、表面を脂質分子で保護された平均粒径10nmの金微粒子を使用し、実施例1と同様にして試料62を含む脂質二分子膜61を自発展開で成長させた。
このとき、ナノギャップ電極63、64間の印加電圧が10mV(周波数100Hz)の時点では、図6(a)に示すように、試料がギャップに捕捉されずに通過していった。その後、印加電圧を500mV(周波数100Hz)にすると、次第に試料がギャップに捕捉されていき、やがて、図6(b)に示すように、ギャップが完全に塞がった。その後、印加電圧を10mV(周波数100Hz)に戻すと、図6(c)に示すように、捕捉された試料が放出され、再び試料が電極間を通過するようになった。
この結果から、ナノギャップ電極への印加電圧を変えるだけで試料の流れをコントロールできることが示された。
このように、ナノギャップ電極に印加する電圧を変えるだけで、電極の大きさを変えることなく、ナノギャップ電極のギャップの通過しやすさを制御でき、分子の流れをコントロールできることが示された。

本実施例では、流路部79に、それぞれギャップの幅が異なる3つのナノギャップ電極対71〜73を備える素子を実施例1と同様の手順で作製した。このとき、ナノギャップ電極対71、72、73のギャップの幅は、それぞれ、35nm、25nm、15nmとし、試料の進行方向にかけて次第に小さくなるように設定した。
試料74〜77としては、平均粒径がそれぞれ40nm、30nm、20nm、10nmの、脂質分子で表面保護された金微粒子の混合物を使用した。
該試料74〜77を含む脂質二分子膜を実施例1と同様にして試料を自発展開で成長させると、試料74〜77は、図7に示すような挙動を示す。
すなわち、まず、分子74〜77を含む脂質二分子膜78がナノギャップ電極71に到達すると、ナノギャップ電極71のギャップ幅よりも分子サイズが大きい分子74はそれよりも先へ進めず、ナノギャップ電極71よりも上流側にとどまる。一方、ナノギャップ電極71のギャップ幅よりも分子サイズが小さい分子75〜77はナノギャップ電極71を通過し、ナノギャップ電極72に到達する。すると、ナノギャップ電極72のギャップ幅よりも分子サイズが大きい分子75はそれよりも先へ進めず、ナノギャップ電極71とナノギャップ電極72との間にとどまる。一方、ナノギャップ電極72のギャップ幅よりも分子サイズが小さい分子76〜77はナノギャップ電極72を通過し、ナノギャップ電極73に到達する。すると、ナノギャップ電極73のギャップ幅よりも分子サイズが大きい分子76はそれよりも先へ進めず、ナノギャップ電極72とナノギャップ電極73との間にとどまる。一方、ナノギャップ電極73のギャップ幅よりも分子サイズが小さい分子77はナノギャップ電極73を通過し、結果、流路79内において、各ナノギャップ電極71〜73で分離された各区分に分子の分布が生じる。

このように、それぞれギャップの幅が異なる複数のナノギャップ電極を設けると、各ナノギャップ電極が分子ふるいの役割を果たし、試料中の複数の分子を分別することができる。
なお、本実施例では、複数のナノギャップ電極それぞれのギャップの幅を変えて試料中の各分子を分別したが、本発明はこれに限定されず、たとえば各ギャップの大きさを一定にして(例えば上記の例ではギャップの幅をナノギャップ電極対71のギャップ幅に統一して)、各ナノギャップ電極対71〜73に印加する印加電圧の大きさをそれぞれ変えるだけでも、実施形態Cと同様に、各分子の分別を行うことができる。この方法では、印加電圧を変更するだけでよいため、ギャップの幅を変えるよりも汎用性が高い。
