界面活性剤等の皮膚刺激緩和剤として、トレハロースモノ脂肪酸エステル(特許文献1:特開平10−45560号公報)、パーフルオロアルキル基およびポリオキシアルキレン基を側鎖として持つシリコーン(特許文献2:特開平6−199629号公報)、アルコール刺激緩和剤として、フェニルエチル−α−グルコシド、フェニルエチル−β−グルコシドのようなグルコース誘導体(特許文献3:特開平8−283121号公報)、脂溶性薬剤の刺激緩和剤としてポリアルキレングリコール(特許文献4:特開2002−212024号公報)、経皮外用剤の刺激緩和剤としてクエン酸トリアルキル、多価アルコール、水(特許文献5:特開平5−255118号公報)が開示されている。しかしながら、化粧品の基剤として大きな配合比を占める油剤に関しては、安全性に関しては研究されているが(非特許文献1: 杉山、太田、「化粧品油性原料の皮膚刺激性評価」、日皮協ジャーナル、日本産業皮膚衛生協会、1999年2月、第41巻、p.136−142)、刺激緩和作用については研究されていない。

クレンジング化粧料は、油性のメークアップ化粧料を除去するために用いられる化粧料であり、界面活性剤の水溶液を増粘剤で増粘させた水性ゲル状クレンジング化粧料や水中油型乳化のクレンジングクリームや油剤、多価アルコール、界面活性剤でゲル構造を形成した多価アルコール性ゲル状クレンジング化粧料や油剤に界面活性剤を溶解した油性クレンジング化粧料等が用いられている。
水性ゲル状クレンジング化粧料は油性メークアップ化粧料となじみにくく、クレンジング力を高めるためには界面活性剤の濃度を高める必要があり、界面活性剤の刺激が問題とされている。水中油型乳化のクレンジングクリームや多価アルコール性ゲル状クレンジング化粧料についても油性メークアップ化粧料となじませるために、油相を高含有させる必要があり、油剤自体の溶剤としての刺激が生じる。油性クレンジング化粧料はそもそも油剤を主成分としており、油剤自体の溶剤としての刺激が生じると共に、洗い流しのために多量の界面活性剤を配合することによる刺激も問題となる。
クレンジング化粧料の刺激を低減することは重要な課題であり、数多くの試みがなされてきた。例えば、特許文献7(特開2005−104892号公報)には、特定のアルキレンオキシド誘導体を配合することにより刺激を低減したクレンジング化粧料が示されている。特許文献8(特開2005−194290号公報)には、抗炎症剤とジグリセリンテトラオレートを配合した刺激を低減したクレンジング化粧料が示されている。特許文献9(特開2006−232717号公報)には特定のグリセリン誘導体を配合した目への刺激が少ないクレンジング化粧料が示されている。特許文献10(特開2007−217302号公報)には界面活性剤を低減し、目や皮膚への刺激を低減したクレンジング化粧料が示されている。しかしながら、機能性と安全性を両立させたクレンジング化粧料を実現するためにさらなる技術開発が望まれている。

ームを各種濃度で細胞に暴露したとき、培地中のクリームの濃度を0.156%から10%まで増大させても、各例とも細胞生存率は約80%から約70%にほぼ同様に低下する程度であった。
しかしながら、培地中のクリームの濃度を20%にすると0.1%の濃度でラウリル硫酸ナトリウムを含有させた試験例1のクリームは54%の細胞生存率を維持したが、0.1%の濃度でラウリル硫酸ナトリウムを含有させた参考比較例1及び2のクリームでは細胞が殆ど死滅した(細胞生存率1%)。
従って、試験例1のクリームはラウリル硫酸ナトリウムの刺激を緩和する効果が特に優れている。

DEG−DA5およびDEG−DA5を配合したクリーム3種、参考比較例として化粧品用油剤として一般的に使用される流動パラフィンとスクワランを配合したクリーム4種及び皮膚科で処方される保湿剤の代表としてワセリンを前処理被験品とした。無処理を対照とした。

前処理剤の安全性試験
前処理剤の安全性確認試験のため24時間クローズドパッチテストを実施した。貼付した4被験物質[DEG−DA5配合20%クリーム(試験例1)、流動パラフィン20%配合クリーム(参考比較例1)、スクワラン20%配合クリーム(参考比較例2)、ワセリン]の目視評価結果を表8に示した。DEG−DA5配合クリームや流動パラフィン配合クリームが各判定時間で全陰性の結果に対し、スクワラン配合クリームでスコア3を示した被験者が24h判定で1名、ワセリンではスコア1を示した被験者が24hで1名認めた。しかし、48h、7日目では刺激反応が低減しており、アレルギー症状ではないことが確認できた。以上の結果より、4被験物質は、24h閉塞貼付結果、明らかな一次皮膚刺激性はないことが確認できた。

3.パッチテスト被験品
皮膚一次刺激の緩和効果を評価するため、皮膚刺激物質として0.5%ラウリル硫酸ナトリウム(関東化学)水溶液(以下、SLSと呼ぶ)をパッチテスト被験品とした。各塗布部には、対照としてそれぞれ滅菌水を同時に貼付した。

4.変法クローズドパッチテスト試験方法
被験者の背部にプラスチック枠を用いて10mm×30mmの領域を10箇所設定し、DEG−DA5、ワセリン並びに試験例1〜5、参考比較例1〜4のクリームの一定量(約15μL)を微量分注器で量り採り、1箇所に1種類の前処理被験品を塗布した。前処理被験品塗布領域は9箇所であり、残りの1箇所は対照として無処理区とした。
その後、皮膚刺激物質である0.5%ラウリル硫酸ナトリウム水溶液(SLS)および対照の滅菌水をパッチテスト被験品として24時間クローズドパッチテストを実施した。
パッチテストには大正製薬のフィンチャンバーを使用した。
塗布部は背骨を挟んで左右5箇所を選択した。塗布部は試験担当医師が被験者の背部を所見し、紅斑等の炎症部位は避けて前処理被験品塗布、パッチ絆貼付を行った。
4). 6.評価方法
6.1 皮膚所見
目視判定は皮膚刺激判定基準(表9)に準じて試験担当医師が行った。判定結果は評点にかえ平均値を算出した。得られた値を無塗布部、ワセリン塗布部と比較して有効性を判断した。

7.皮膚科医による目視判定
貼付24時間後、48時間後、7日目に皮膚刺激基準に従って試験担当医師が目視判定した。貼付24時間後の目視判定結果を表10に、貼付48時間後の目視判定結果を表11に示す。図3、図4に各前処理剤における20名の0.5%ラウリル硫酸ナトリウム水溶液(SLS)皮膚刺激スコアのうち、皮膚刺激基準スコア3、4だけを抜粋し、24時間後、48時間後の総和人数を示した。皮膚刺激基準スコアが4を超える被験者はいなかった。
24時間判定では、無塗布部位と比較してDEG−DA5塗布部位、DEG−DA5 20%配合クリーム(試験例1)塗布部位で明らかに目視評価スコア3,4以上を示した被験者が少なく、次いでDEG−DA5 10%配合クリーム(試験例2)、DEG−DA5 5%配合クリーム(試験例3)であった。ワセリンは0.5%ラウリル硫酸ナトリウム水溶液(SLS)貼付による目視評価スコア3,4以上を示した被験者が5人と無塗布部位の12人と比較すると、少ない結果となったが、DEG−DA5およびDEG−DA5配合クリームと比較するとその人数は多かった。DEG−DA5配合はスコア1以上の合計点数として見ると全体的に刺激が緩和されていることが認められ、さらに、前述のように刺激が強いスコア3以上も少ないことが認められる。
Wilcoxon符号付順位和検定の結果、DEG−DA5塗布部位(DEG−DA5)、DEG−DA5 20%配合クリーム(試験例1)塗布部位、DEG−DA5 10%クリーム(試験例2)塗布部位でワセリン塗布部位と比較して有為にスコアが低い結果となった(p<0.05)。DEG−DA5 5%配合クリーム(試験例3)では抑制傾向はみられたものの有為差はなかった。また、DEG−DA5 20%配合クリーム(試験例1)は、流動パラフィン20%配合クリーム(参考比較例1)、スクワラン20%配合クリーム(参考比較例2))と比較して有意に刺激スコアが抑制されていた。さらに、DEG−DA5 10%配合クリーム(試験例2)は、流動パラフィン10%配合クリーム(参考比較例3)、スクワラン10%配合クリーム(参考比較例4)と比較して有意に刺激スコアが抑制されていた。

8.機器測定による経表皮水分蒸散量
表13、図5、図6に各被験物質の経表皮水分蒸散量値の平均値を示す。貼付24h後では、SLS貼付部位でその刺激性によるTEWL値抑制傾向が認められた箇所はDEG−DA5、DEG−DA5 20%配合クリーム(試験例1)、DEG−DA5 10%配合クリーム(試験例2)であった。DEG−DA5 20%クリーム(試験例1)、DEG−DA5 10%クリーム(試験例2)は無塗布部位に対して、またDEG−DA5 20%クリーム(試験例1)はワセリン塗布部位に対しても有為に刺激抑制効果が認められた(p<0.05)。貼付48h後では、すべての前処理剤において無塗部位と比較するとSLS貼付によるTEWL値上昇が有為に抑制されていた(p<0.05)。特にDEG−DA5、DEG−DA5 20%配合クリーム(試験例1)ではワセリン塗布部位と比較しても有為に抑制された(p<0.05)。

13.DEG−DAに関する被験者評価試験まとめ
本試験では、DEG−DA5およびDEG−DA5を配合したクリームの刺激成分浸透抑制効果をアトピー性皮膚炎患者の方、皮脂欠乏症の方20名を対象にして検証した。その結果、DEG−DA5およびDEG−DA5配合クリームが濃度依存的にSLSの刺激を抑制する傾向が認められ、ヒトに用いる際に効果を発揮するおおよその濃度が示唆された。
また、DEG−DA5およびDEG−DA5配合クリームのアトピー性皮膚炎疾患者、皮脂欠乏症疾患者に対する24時間クローズドパッチテストによる安全性確認ができた。
以上の結果より、DEG−DA5を配合した化粧料は、バリア機能が低下したアトピー性皮膚炎疾患者、皮脂欠乏症者の使用に適している。
13. 眼刺激性については、化学物質を能率よく、かつ経済的に安全性評価するために、また、動物愛護の必要性から、動物を用いない代替試験法が多く開発されてきた。欧米や日本でも多くの研究がなされ、日本国内で実施された多施設バリデーションの結果、ウサギ角膜由来の正常細胞(SIRC細胞)やチャイニーズハムスター肺由来線維芽細胞(CHL細胞)を用いた細胞毒性試験法の結果が、多くの水溶性物質について、ドレイズ試験の結果ときわめて良く対応することが明らかになった。特に培養液と混和しうるものであれば、再現性がよかった(大野泰雄ら、フレグランスジャーナル誌 1999年7月号, p.21-26, Ohno Y., et al.,Toxicology In Vitro, 13, 73-98(1999)、最新 動物実験代替法〜日米欧関連法規への対応/各種試験の手順〜 技術情報協会 2007年 p.87-96)。
眼に入りやすいシャンプーや洗顔料、クレンジング剤などの商品は眼刺激性が過去の文献からも低いものはなく、これらの処方には、界面活性剤が多く含まれている。
化粧品成分が組み合わされ処方を構成すると考えると、化粧品処方における代替法バリデーションが完了しているわけではないが、過去の研究結果より細胞毒性試験法が処方の眼刺激性を予測するために有効な手段であると考えられ、すでに取り入れられている(Gordon V.C、フレグランスジャーナル誌 1992年8月号 p.15-21)。
皮膚刺激性についても、ヒト皮膚の刺激性強度を予想するin vitro代替法の開発が各国で進んでいる。界面活性剤の皮膚刺激性の予測において細胞毒性や炎症性サイトカインが有効な指標となることが示されている(林俊克、日本油化学会誌 45, 1179-1188(1996)、足利太可雄 フレグランスジャーナル誌 1999年7月号 p.35-40、Eun H. C. Current Problems in Dermatology, 55,,231-236, (1995)、Ikarahi, Y., J. Toxicol..,12(1),15-24, (1993))。また、新しい原料開発等の場面でも細胞毒性を用いて従来の成分と比較して皮膚刺激性が低い成分の開発がされている(Sanchez L, et al. , Pharm Res. 21(9):1637-1641, (2004))。一方、化粧品成分だけでなく、市販前化粧品処方の安全性評価法の1つとしてマウス由来線維芽細胞のニュートラルレッド取り込み試験法を採用している研究機関もある。D Jirovaらによると数多くの被験物質を評価した結果より、シャンプーやシャワージェルは他のカテゴリーの化粧品と比較して細胞毒性が高く、最も消費者が日常的に使用する化粧品の中で刺激を起こす可能性を持つ製品群であると報告している(Jirova D,, Toxicol . In Vitro. ;17(5-6):791-796, (2003))。
以上の背景より、細胞毒性試験は化粧品原料および処方の眼刺激性、皮膚刺激性についての安全性評価法として有用であり、特に界面活性剤を多量に用いた処方系の評価に適している。なお、細胞毒性試験結果は50%細胞死滅濃度(EC50値)で刺激性標準物質であるSDS、Triton X-100と比較するとクレンジング剤に多く用いられるポリオキシエチレンオクチルドデシルエーテル(25E.O.)は強刺激性に分類、モノラウリン酸ポリエチレングリコール(12E.O.)は強刺激〜中程度刺激性物質に分類される。従って、細胞毒性試験法でこれらの界面活性剤の刺激性は十分に予測することが出来ると考えられる。これらの界面活性剤を含有するクレンジング化粧料にDEG−DA5配合を配合することにより、細胞毒性が緩和されることから、眼刺激性、皮膚刺激性が緩和されたと考えられる。参考のため、定法により測定した50%細胞死滅濃度(EC50値)を表18に示す。

The reproducibility was particularly good if it was miscible with the culture solution (Yasuo Ohno et al., Fragrance Journal, July 1999, p.21-26, Ohno Y., et al., Toxicology In Vitro, 13, 73-98 (1999), the latest alternative to animal experiments-compliance with Japanese, US and European laws / procedures for various tests-Technical Information Association 2007, p.87-96).
Products such as shampoos, facial cleansers, and cleansing agents that easily enter the eye have no low eye irritation from the past literature, and these formulations contain a large amount of surfactant.
Considering that cosmetic ingredients are combined to form a prescription, alternative method validation in cosmetic prescriptions has not been completed, but past studies have shown that cytotoxicity testing methods are effective in predicting eye irritation of the prescription. It is considered a tool and has already been adopted (Gordon VC, Fragrance Journal, August 1992, pages 15-21).
Regarding skin irritation, in vitro alternative methods for predicting the irritation intensity of human skin are being developed in various countries. Cytotoxicity and inflammatory cytokines have been shown to be effective indicators in predicting skin irritation of surfactants (Toshikatsu Hayashi, Journal of the Japan Oil Chemists' Society 45, 1179-1188 (1996), Kazuo Ashikaga Fragrance Journal Journal July 1999 p.35-40, Eun HC Current Problems in Dermatology, 55,, 231-236, (1995), Ikarahi, Y., J. Toxicol .., 12 (1), 15-24 , (1993)). In addition, in the field of new raw material development etc., a component having low skin irritation compared to conventional components has been developed using cytotoxicity (Sanchez L, et al., Pharm Res. 21 (9): 1637 -1641, (2004)). On the other hand, not only cosmetic ingredients but also some research institutions adopt the neutral red uptake test method of mouse-derived fibroblasts as one of the safety evaluation methods of pre-commercial cosmetic formulations. According to D Jirova et al., Shampoos and shower gels are more cytotoxic than other categories of cosmetics, and are most irritating among the daily consumer cosmetics, according to the results of a number of test substance evaluations It is reported to be a product group with potential (Jirova D ,, Toxicol. In Vitro.; 17 (5-6): 791-796, (2003)).
In view of the above background, the cytotoxicity test is useful as a safety evaluation method for eye irritation and skin irritation of cosmetic raw materials and formulations, and is particularly suitable for evaluation of a formulation system using a large amount of a surfactant. The cytotoxicity test results show that polyoxyethylene octyldodecyl ether (25E.O.), which is more frequently used as a cleansing agent compared to SDS and Triton X-100, which are stimulatory standards, at 50% cell killing concentration (EC50 value). Is classified as a strong irritant, and polyethylene glycol monolaurate (12E.O.) is classified as a strong to moderately irritating substance. Therefore, it is considered that the irritancy of these surfactants can be sufficiently predicted by the cytotoxicity test method. It is considered that eye irritation and skin irritation were alleviated by blending DEG-DA5 with cleansing cosmetics containing these surfactants, since the cytotoxicity was alleviated. For reference, Table 18 shows 50% cell killing concentration (EC50 value) measured by a conventional method.

Cell viability was evaluated when a commercial cleansing cosmetic was exposed to a three-dimensional model of the cornea (OCL-200).
1.試験方法
1.1 OCL−200キット
倉敷紡績(株)から市販されている、正常ヒト表皮角化細胞から無血清培地にて培養・構築されたヒト角膜構造に類似した三次元構造を有する「OCL−200」キットを使用した。
OCL−200は入手後直ちに4℃で保管し、48時間以内に使用することとした。試験前に、OCL−200を1時間、37℃、5%CO2、加湿状態で培養した。

1.4 MTTアッセイ法
被験物質の曝露後、OCL−200をPBS(−)で3回以上洗浄し、キット付属のアッセイ培地で10分間培養した後1mg/mLの濃度でMTT(和光純薬)をキット付属のアッセイ培地で希釈したMTT含有培地中で3時間培養した。培養後PBS(−)で軽く洗浄し抽出液(イソプロパノール)中にOCL−200を浸漬させ、抽出液が蒸発しないようにプレートをパラフィルムで覆い、暗所・室温で一晩静置した。抽出終了後、抽出液を各well200μLずつ96wellプレートへ移し、570nmと655nmの吸光度を測定した。570nmの吸光度から655nmの吸光度を差し引いたものを生存細胞数の指標値とし、精製水24時間曝露の指標値を100%として、細胞生存率を算出した。尚、精製水曝露2時間後の細胞生存率は、細胞毒性が生じないことが明らかなので測定しなかった。

After completion of extraction, the extract was transferred to a 96-well plate by 200 μL of each well, and absorbance at 570 nm and 655 nm was measured. The cell viability was calculated by subtracting the absorbance at 655 nm from the absorbance at 570 nm as an index value for the number of viable cells and the index value for exposure to purified water for 24 hours as 100%. The cell viability after 2 hours of exposure to purified water was not measured because it was clear that no cytotoxicity occurred.

被験物質を24時間暴露したときの細胞生存率は比較例3(33%)と比べて、実施例13(58%)は約2倍であり、DA5は顕著な刺激緩和効果を有する。
