Abstract

Description
図1は、TetRと、VP16に由来する最小の酸性活性化ドメインとの融合物の概略図である。ドメインのアミノ酸配列を右側に示す:[F](これはまた配列番号1として示される)は、442位にフェニルアラニンを含有するVP16の436位〜447位の間の野生型配列を表す。変異した最小のドメイン[G](これはまた配列番号2として示される)またはドメイン[Y](これはまた配列番号3として示される)において、Phe442は、それぞれ、グリシンまたはチロシンによって置換されている。最小のドメインの様々な組合せをTetRに融合させて、左側に示される一群の融合タンパク質が得られた。

A.発現ベクター
トランス活性化因子融合タンパク質をコードする本発明の核酸は、上記のように、宿主細胞における融合タンパク質の発現に適切な形態で組換え発現ベクターに組み込むことができる。用語「宿主細胞における融合タンパク質の発現に適切な形態で」は、組換え発現ベクターが、核酸のmRNAへの転写およびmRNAの融合タンパク質への翻訳を可能にする方法で融合タンパク質をコードする核酸に機能的に連結された1つまたは複数の調節配列を含むことを意味するものとする。用語「調節配列」はこの分野で認識されており、プロモーター、エンハンサーおよび他の発現制御エレメント(例えば、ポリアデニル化シグナル)を含むものとする。そのような調節配列は、当業者に知られており、GoeddelのGene Expression Technology:Methods in Enzymology 185、Academic Press、San Diego、CA(1990)に記載されている。発現ベクターの設計は、トランスフェクションされ得る宿主細胞の選択および/または発現され得る融合タンパク質の量のような要因に依存し得ることを理解しなければならない。

トランス活性化因子融合タンパク質をコードする核酸は、本発明の融合タンパク質が1つまたは複数の細胞タイプで発現するトランスジェニック動物を作製するために、ヒト以外の動物の受精した卵母細胞に入れることができる。トランスジェニック動物は、出生前の段階(例えば、胚の段階)で動物または動物の祖先に導入されているトランスジーンを含有する細胞を有する動物である。トランスジーンは、トランスジェニック動物が発生する細胞のゲノムに組み込まれ、そして成熟した動物のゲノムに残留し、それによって、コードされた遺伝子産物の発現をトランスジェニック動物の1つまたは複数の細胞タイプまたは組織でもたらすDNAである。1つの実施形態において、ヒト以外の動物はマウスであるが、本発明はマウスに限定されない。他の実施形態において、トランスジェニック動物は、ヤギ、ヒツジ、ブタ、ウシまたは他の家畜である。そのようなトランスジェニック動物は、タンパク質の大規模な生産(いわゆる、「遺伝子農場」)に有用である。

tetシステムまたは逆tetシステムに基づく本発明のトランス活性化因子融合タンパク質をコードする核酸、ならびにtetオペレーター配列に機能的に連結されたヌクレオチド配列(すなわち、転写され得る目的の遺伝子)を有する宿主細胞において、tetオペレーター配列に機能的に連結されたヌクレオチド配列の高レベルの転写は、(tetシステムまたは逆tetシステムが使用されているかどうかに依存して)テトラサイクリンの有無に依存する。宿主細胞での転写を誘導するために、宿主細胞は、(tetシステムに関しては)Tcの非存在下で培養されるか、あるいは(逆tetシステムに関しては)テトラサイクリンまたはテトラサイクリンアナログと接触させられる。従って、本発明の別の局面は、本発明のトランス活性化因子融合タンパク質を発現する宿主細胞または動物において、tetオペレーター配列に機能的に連結されたヌクレオチド配列の転写を調節するための方法に関する。この方法は、細胞をテトラサイクリンまたはテトラサイクリンアナログと接触させること、あるいはテトラサイクリンまたはテトラサイクリンアナログを、そのような細胞を含有する被験体に投与することを含む。
本発明は、多面発現作用または細胞傷害性を引き起こすことなく迅速かつ効率的に制御された方法で、遺伝子発現の開始および停止を行うことができ、あるいは遺伝子発現のレベルを調節できることが望ましい様々な状況に広く適用することができる。従って、本発明のシステムは、真核生物細胞、植物および動物における細胞発達および細胞分化の研究に幅広く適用することができる。例えば、ガン遺伝子の発現は、その機能を研究するために、細胞において制御された方法で調節することができる。さらに、このシステムを使用して、CREまたはFLPなどの部位特異的なリコンビナーゼの発現を調節することができ、それによってトランスジェニック生物の遺伝子型の不可逆的な改変が、特定の発達段階において制御された条件下で可能になる。例えば、トランスジェニック植物のゲノムに挿入され、特定のトランスジェニック植物の選択を可能にする薬物耐性マーカーは、Tcにより調節される部位特異的なリコンビナーゼによって不可逆的に取り除くことができる。本発明の調節システムの他の適用を下記に示す:
本発明は、遺伝子的疾患または後天的疾患のいずれかの処置における遺伝子治療目的に特に有用であり得る。遺伝子治療の一般的な方法には、機能的な活性の回復または増強のために、導入される遺伝物質によってコードされる1つまたは複数の遺伝子産物が細胞内で産生されるように核酸を細胞に導入することが含まれる。遺伝子治療法の総説に関しては、Anderson,W.F.(1992)Science 256:808〜813;Miller,A.D.(1992)Nature 357:455〜460;Friedmann,T.(1989) Science 244:1275〜1281;およびCournoyer,D.他(1990)Curr.Opin.Biotech.1:196〜208を参照のこと。しかし、現在の遺伝子治療ベクターは、典型的には、内在性の転写因子に応答し得る構成的な調節エレメントを利用している。このようなベクターシステムでは、被験体における遺伝子の発現レベルを調節し得ることは考慮されていない。これに対して、本発明の誘導性の調節エレメントはこのような調節能を提供する。
目的とするタンパク質の大規模な生産は、1)細胞におけるトランス活性化因子の発現に適する形態で本発明のトランス活性化因子融合タンパク質をコードする核酸、および2)tetオペレーター配列に機能的に連結された目的タンパク質をコードする遺伝子を含有するように改変された細胞のインビトロ培養物を使用して行うことができる。例えば、哺乳動物細胞、酵母細胞または真菌細胞を改変して、本明細書中に記載されているこのような核酸成分を含有させることができる。次いで、改変された哺乳動物細胞、酵母細胞または真菌細胞は、遺伝子発現を誘導させて目的タンパク質を産生させるために、Tcまたはそのアナログの存在下で標準的な発酵技術によって培養することができる。従って、本発明により、目的のタンパク質を単離するための製造プロセスが提供される。このプロセスにおいて、本発明のトランス活性化因子融合タンパク質をコードする核酸と、少なくとも1つのtetオペレーター配列に機能的に連結された目的タンパク質をコードする核酸との両方が導入された宿主細胞(例えば、酵母または真菌)を、テトラサイクリンまたはテトラサイクリンアナログが存在する培養培地において生産規模で増殖させて、目的のタンパク質をコードするヌクレオチド配列(すなわち、tetオペレーター配列に機能的に連結されたヌクレオチド配列)の転写を刺激し、そして目的とするタンパク質を、集めた宿主細胞または培養培地から単離する。標準的なタンパク質精製技術を使用して、目的とするタンパク質を培地または集めた細胞から単離することができる。
本発明により、トランスジェニック農場動物などの動物において目的とするタンパク質の大規模な生産もまた行われる。トランスジェニック技術での進歩により、ウシ、ヤギ、ブタおよびヒツジなどのトランスジェニック家畜の作製が可能になっている(Wall,R.J.他(1992)J.Cell.Biochem.49:113〜120;およびClark,A.J.他(1987)Trends in Biotechnology 5:20〜24における総説を参照のこと)。従って、本発明の誘導性調節システムの成分をそのゲノムに有するトランスジェニック家畜を作製することができる。この場合、目的とするタンパク質をコードする遺伝子は、少なくとも1つのtetオペレーター配列に機能的に連結されている。遺伝子の発現、従ってタンパク質の生産は、Tc(またはそのアナログ)をトランスジェニック動物に投与することによって誘導される。タンパク質の生産は、トランス活性化因子の発現をある種の細胞に限定する適切な組織特異的調節エレメントに連結することによって、トランス活性化因子融合タンパク質をコードする核酸を特定の組織に標的化することができる。例えば、乳清プロモーター(米国特許第4,873,316号および欧州特許出願公開第264,166号)などの乳腺に特異的な調節エレメントをトランス活性化因子トランスジーンに連結して、トランス活性化因子の発現を乳腺組織に限定することができる。従って、Tc(またはアナログ)の存在下において、目的のタンパク質は、トランスジェニック動物の乳腺組織で産生される。タンパク質は、トランスジェニック動物の乳汁に分泌させるように設計することができ、そして所望する場合には、次いで、タンパク質を乳汁から単離することができる。
本発明の転写活性化因子タンパク質を使用して、ヒトの疾患の病理学を模倣するために動物の特定遺伝子の発現を刺激し、それによってヒト疾患の動物モデルを作製することができる。例えば、宿主動物において疾患に関与していると考えられる目的の遺伝子を、(例えば、本明細書中に記載されている相同組換えによって)1つまたは複数のtetオペレーター配列の転写制御下に置くことができる。そのような動物は、トランス活性化因子融合タンパク質のトランスジーンを有する第2の動物と交配して、テトラサイクリンにより調節される融合タンパク質遺伝子およびtetにより調節される標的配列の両方を有する子孫を作製することができる。このような子孫における目的とする遺伝子の発現は、テトラサイクリン(またはアナログ)を使用して調節することができる。

本研究の最小の活性化ドメインはVP16から得られた。これは、Seipel,K.、Georgiev,O.およびSchaffner,W.(1992)EMBO J 11、4961〜4968による436位〜447位を含む。このドメインおよびその変化体をコードする合成オリゴヌクレオチドを、それぞれ、[F]、[GF]、[FG]、[GG]および[Y]と名付けた:この場合、文字は、442位のアミノ酸を示す(下線を付したトリプレット)。コード鎖の配列を(442位のコドンに対応するトリプレットに下線を付けて)下記に示す。オリゴヌクレオチドは、括弧内の文字によって示されるように1つまたは複数の最小ドメインをコードする。

ColE1型プラスミドのpUHD141−1(Kistner,A.(1992)学位論文、ハイデルベルグ大学)は、翻訳の効率的な開始のために5′末端で最適化されているTetRのコード配列を含有する(Kozak,M.(1983)Microbiol Rev 47、1〜45)。tetR遺伝子の転写は、ヒトサイトメガロウイルスのIEプロモーター(Boshart,M.他(1985)Cell 41、521〜530)によって制御されている。XmaI切断によってtetRのオープンリーディングフレームの3′末端との読み枠を合わせてDNAを挿入するために、pUHD141−1を、tetRの停止コドンと重なっているAflIIで線状化した。突出した5′DNA末端をマングビーンヌクレアーゼによって除き、そして合成オリゴヌクレオチド5′−CCCGGGTAACTAAGTAA−3′(配列番号14)を、標準的なクローニング手順を使用してベクターに連結した。XmaI切断部位をtetR遺伝子のすぐ3′末端に含有する得られたプラスミドpUHD141−1/Xを配列分析によって確認した。
ルシフェラーゼのレポーター構築物pUHC13−3が染色体に組み込まれたHeLaX1/6細胞(Gossen,M.(1993)Ph.D.論文、ハイデルベルグ大学)およびHeLa(wt)細胞を、37℃および5%CO2で、10%ウシ胎児血清を補充したアール改変イーグル培地(GIBCOから得られるE−MEM)において維持した。リン酸カルシウム共沈澱によるトランスフェクションを、下記の改変を加えた標準的なプロトコルに従って行った:HeLaX1/6細胞を35mm培養ディッシュで50%〜60%のコンフルエンスに増殖させた。トランスフェクションする1時間前に、培養培地を新しい培地に変えて、細胞を37℃および6%CO2でインキュベーションした。リン酸カルシウム/DNAの沈澱物は1.5μgのプラスミドDNAを含有する(これは、0.5μgのトランス活性化因子構築物、トランスフェクション効率を正規化するために含められた0.4μgのlacZ発現ベクター(pUHD16−1)、および非特異的なキャリアDNAとして0.6μgのpUC18からなる)。沈澱物(培養ディッシュあたり100μl)をX1/6細胞に加え、次いで細胞を37℃および6%CO2で30時間さらにインキュベーションした。同時に行われている培養物のインシチウβ−ガラクトシダーゼ染色によって測定されたトランスフェクション効率は、60%〜80%の間であった。
トランスフェクションされたX1/6細胞を含有する35mm培養ディッシュを3mlのリン酸緩衝化生理食塩水(PBS)で洗浄し、25mMTris−リン酸塩(pH7.8)、2mMジチオスレイトール、2mMジアミノシクロヘキサン四酢酸、10%グリセロールおよび1%TritonX−100を含有する125μlの溶解緩衝液において室温で10分間溶解した。溶解物を培養ディッシュから掻き取り、Eppendorf遠心分離器で10秒間遠心分離した。これらの抽出物におけるルシフェラーゼ活性を、Gossen,M.およびBujard,H.(1992)Proc.Natl.Acad.Sci.USA 89、5547〜5551の記載に従って測定した。ルシフェラーゼの値は、標準的な液体のO−ニトロフェニルβ−ガラクトピラノシドアッセイを行うことによってβ−ガラクトシダーゼ活性に正規化した(Miller,J.H.(1972)Experiments in Molecular Genetics、Cold Spring Harbor Laboratory Press、Cold Spring Harbor)。
HeLa細胞を、10cm培養ディッシュで50%〜60%のコンフルエンスに増殖させ、そして、様々なtTAをコードするプラスミドDNAの20μgを用いるリン酸カルシウム手順によりトランスフェクションした。トランスフェクションの30時間後に全細胞抽出物を下記のように調製した:細胞(約2×106)をPBSで洗浄し、遠心分離して、10mMHEPES、1.5mMMgCl2、10mMKCl、0.5mMジチオスレイトールおよび1mMフェニルメチルスルホニルフルオリドを含有する緩衝液の250μlに再懸濁し、そして0℃で20分間インキュベーションし、その後、素早く凍結および融解を行った。NaClを250mMの最終濃度に加え、そして0℃で20分間のインキュベーションを行った後に、サンプルをBeckmanTL−100超遠心分離器において435000gおよび0℃で10分間遠心分離した。抽出部の一部(10μl)を、10μlの結合緩衝液(20mMMgCl2、20mMTris(pH7.5)、10%グリセロール、2mg/mlニシン精子DNA、および1mg/mlウシ血清アルブミン)、およびpUHC13−3(Gossen,M.およびBujard,H.(1992)Proc.Natl.Acad.Sci.USA 89、5547〜5551)から42塩基対のTaqIフラグメントとして単離して[α−32P]dCTPの存在下でT4−DNAポリメラーゼによって突出末端を埋めた後の2fmolの32P標識したtetO DNAと混合した。25分後、反応混合物を、5%グリセロールを含有する5%ポリアクリルアミド/0.13%ビスアクリルアミドのゲルに負荷した。電気泳動を、45mMTris塩基、45mMホウ酸および1mMEDTAにおいて7V/cmで行った。
HeLaX1/6細胞を35mm培養ディッシュで増殖させ、そして上記のように、2μgの線状化プラスミドDNAを用いてトランスフェクションした。トランスフェクション混合物は、ハイグロマイシン遺伝子を有するプラスミドpHMR272(Bernard,H.U.他(1985)Exp.Cell.Res.158、237〜243)、およびpUHD15−1、pUHD19−1またはpUHD26−1(これらはtTA遺伝子の上流にKozak配列を含有する)をそれぞれ含有する。検討したプラスミドと選択マーカーとのモル比は40:1であった。24時間後、細胞を10cm培養ディッシュに移し、300μg/mlのハイグロマイシンを含有する培地で維持した。耐性クローンを単離し、個々に拡大培養して、ルシフェラーゼ活性について分析した(Gossen,M.およびBujard,H.(1992)Proc.Natl.Acad.Sci.USA 89、5547〜5551)。耐性クローンのルシフェラーゼ遺伝子のtTAに依存した活性化をさらに調べるために、細胞を、10000細胞/35mm培養ディッシュの密度で播種し、Tc(1μg/ml)の存在下または非存在下で増殖させた。5日後、細胞抽出物を上記のように調製して、ルシフェラーゼ活性を測定した。溶解物のタンパク質含有量を、Bradford(Bradford,M.M.(1976)Anal Biochem 72、248〜254)に従って測定した。
プラスミドpUHD15−1、pUHD19−1、pUHD20−1およびpUHD26−1を、選択マーカー遺伝子を挿入することによって改変した。それぞれの場合において、neo遺伝子を含有する発現カセットを、PhCMv(1)の上流に位置するXhoI部位に挿入した。得られたプラスミドを、それぞれ、pUHD15−1neo、pUHD19−1neo、pUHD20−1neoおよびpUHD26−1neoとした。
VP16は、大きな負電荷の周囲に位置する嵩張った疎水性アミノ酸を特徴とする2つの異なる転写活性化ドメインを含有する(Regier,J.L.、Shen,F.およびTriezenberg,S.J.(1993)Proc.Natl.Acad.Sci.U.S.A. 90、883〜887)。それぞれのドメインは、GAL4のDNA結合ドメインなどの異種のDNA結合ドメインに融合させた場合、転写を活性化することが示された(Seipel,K.、Georgiev,O.およびSchaffner,W.(1992)EMBO−J 11、4961〜4968)。436位〜447位(このアミノ酸配列を配列番号1に示す)によって明らかにされる酸性ドメインをコードするオリゴヌクレオチド[F]を合成して、tetR遺伝子の3′末端との読み枠を合わせてプラスミドpUHD141−1/Xに挿入した。多数の組み込みのために、1個、2個、3個または4個の活性化モジュールを含有するトランス活性化因子をコードする配列が得られた。それらの配列を、それぞれ、TetR−F、TetR−FF、TetR−FFF、TetR−FFFFとした。これらの構築物の構造を図1に図示する。反復ユニットによって誘導される構造的な制約の可能性を低下させるために、それぞれのドメインをプロリンによって連結した:最初のドメインもまたプロリンによりTetRに結合する。それぞれのトランス活性化因子構築物を配列分析によって確認した。
本発明の新しいTetRキメラのtetOに対する結合をDNA遅延実験により調べた。様々なタンパク質を、HeLa細胞でのプラスミドpUHD141−1/XおよびpUHD18−1〜pUHD21〜1の一過性発現により産生させた。トランスフェクションの30時間後に、抽出物を調製し、放射能標識したtetO DNAとインキュベーションした。タンパク質−DNA複合体の電気泳動分離により、本発明の新しい融合タンパク質は、TetRの効率と比較できる効率でtetO DNAと結合し(図2を参照のこと)、そしてそれぞれの融合タンパク質の分子量に従って移動する複合体を形成することが明らかにされる。結合アッセイにおけるTcの存在は、複合体の形成を妨げる。さらに、この分析により、本発明の新しい融合タンパク質は、分解産物が検出できないほど安定であることが示唆される。
本発明の新規なTetR融合物の活性化能を評価するために、HeLaX1/6細胞を、それぞれのタンパク質をコードするプラスミドで一過性にトランスフェクションして、ルシフェラーゼ活性を測定した。HeLa細胞株X1/6を35mm培養ディッシュで50%コンフルエンスまで増殖させた:この細胞は、染色体に組み込まれ、tTAに依存するプロモーターPhCMV★-1の転写制御下にあるルシフェラーゼ遺伝子(Gossen,M.およびBujard,H.(1992)Proc.Natl.Acad.Sci.USA 89、5547〜5551)を含有する。細胞を、TetRまたは新しい融合タンパク質の1つのいずれかをコードするDNAで一過性にトランスフェクションした。培養物をテトラサイクリン(1μgのTc/ml)の存在下または非存在下で30時間インキュベーションし、その後、ルシフェラーゼ活性を測定して、(pUHD16−1との同時トランスフェクションによって導入される)β−ガラクトシダーゼ活性に対して標準化した。2つの独立したトランスフェクション実験の測定値を下記の表1に示す。これは、tTA活性(100%)に対する相対値である。
安定的にトランスフェクションされた細胞における新規なトランス活性化因子のいくつかの特性を特徴づけるために、PhCMVにより制御されるtTA、tTA3またはtTA4をコードする遺伝子をHeLaX1/6細胞に入れた。pUHD19−1またはpUHD26−1(表1)、およびハイグロマイシン耐性を有するpHMR272(Bernard,H.U.他(1985)Exp.Cell.Res.158、237〜243)との同時トランスフェクションによって耐性クローンを単離して、耐性クローンをTcの存在下および非存在下でルシフェラーゼ活性について調べた。効率的な調節に関して選択された4クローンの細胞を、10,000細胞/35mm培養ディッシュの密度で播種して、テトラサイクリン(1μg/ml)の存在下または非存在下で5日間増殖させた。結果を下記の表2にまとめる。示す結果は、5つの独立した培養物の算術平均である。
最小のドメインを有するトランス活性化因子が、tTAよりも大きな細胞内濃度で寛容であるかどうかを調べるために、HeLa細胞を、それぞれ、tTA、tTA2、tTA3およびtTA4をコードするプラスミドと同時にトランスフェクションした。対応するプラスミド(表1)は、G418耐性クローンもまたトランス活性化因子遺伝子を確実に発現するようにneo耐性マーカーを備えた(上記の「方法」節を参照のこと)。G418耐性に関する選抜によって、300〜500コロニーの集団が得られた。そのような集団を増殖させ、そしてタンパク質抽出物を、放射能標識したtetオペレーターDNAを用いた電気泳動移動度シフト実験によりトランス活性化因子タンパク質について分析した。図4Aに示すように、TetRおよび最小の活性化ドメインからなるトランス活性化因子はすべて、TetR−VP16融合タンパク質であるtTAよりも高い濃度で細胞内に存在する。興味深いことに、tTAと同じ活性化能を有するtTA2(表1)は、それにもかかわらず、3倍高い濃度で寛容である。しかし、新しいトランス活性化因子において、細胞内濃度は、それぞれの活性化能と反対に増大する。従って、tTA3およびtTA4の濃度は、tTAの濃度よりも、それぞれ、5倍および9倍高い。tTAまたはtTA3のいずれかを産生するそれぞれのクローンをDNA遅延アッセイによりトランス活性化因子の相対量について分析した場合、同じ結果が得られた:tTA3の細胞内濃度は、再度ではあるが、tTAの細胞内濃度の約5倍高かった(図4B)。tTAを発現するHeLa細胞から得られた抽出物により、DNA遅延アッセイにおいて、tTAの分解産物であると考えられる第2のタンパク質−DNA複合体が明らかにされる(図3)。この生成物は、他の細胞株においてもまた一定しない程度で見出されている。この複合体の移動度から、約42アミノ酸が、最もありそうなこととしてC端から切断されていることが推定され得る:なぜなら、N端からのこのような大きさの欠失はトランス活性化因子のオペレーター結合能を損なうからである。従って、この分解産物は、VP16分子の第2の(C端側の)活性化ドメインを失っていることが十分に考えられる。そのような短縮型タンパク質が依然としてトランス活性化因子として作用するかどうかは不明である。
当業者は、日常的な範囲を超えることのない実験を行うことによって、本明細書中に記載されている本発明の特定の実施形態に対する多くの均等物を認識し、あるいはそのような均等物を見出すことができる。そのような均等物は、下記の請求項により包含されるものとする。

- 転写を調節する融合タンパク質をコードする核酸分子であって、

これと機能的に連結された、転写活性化ドメインを含む第2のポリペプチドと、

前記転写活性化ドメインが、
a)配列番号5、6、7又は8のアミノ酸配列を含む転写活性化ドメイン、
b)2コピー以上の配列番号1のアミノ酸配列の最小活性化ドメインからなる転写活性化ドメイン、及び
c)3コピーの配列番号1のアミノ酸配列の最小活性化ドメインからなる転写活性化ドメイン、
から選択されることを特徴とする、転写を調節する融合タンパク質をコードする核酸分子。 - 請求項1に記載の核酸分子を含む宿主細胞における融合タンパク質の発現に適する形態の組換え発現ベクター。
- 請求項1に記載の核酸分子を宿主細胞における融合タンパク質の発現に適する形態で含む宿主細胞。

- 請求項1に記載の核酸分子又は請求項2に記載の組換え発現ベクターを遺伝子治療で投与される薬物組成物を製造するために使用する方法。
- 請求項1の核酸分子又は請求項3〜5のいずれか1項に記載の宿主細胞を含むキット。
- 培養細胞中、目的タンパク質をインビトロで生産する方法であって、
a)目的タンパク質をコードするヌクレオチド配列の転写を刺激する濃度のテトラサイクリン又はテトラサイクリンアナログの存在下の培養基において、請求項1に記載の核酸と、少なくとも1種類のtetオペレーター配列に機能的に連結された目的タンパク質をコードする核酸の両方が導入された宿主細胞を生産規模で増殖させる工程、及び
b)得られた宿主細胞又は培養基から目的タンパク質を単離する工程、
