Description
第3の面において、本発明は、以下の工程を含む、イオンチャンネル受容体の負のモジュレーター、インヒビターまたはアンタゴニストを選択または同定する方法に関する:
A)工程b)において上記組換え細胞をATPなどの上記P2X受容体のアゴニストおよびイオンチャンネル受容体のアゴニストの存在下で試験すべき化合物と接触させることを特徴とする、イオンチャンネル受容体の活性を調節しうる化合物を選択または同定するための本発明方法により、イオンチャンネル受容体の活性を調節しうる化合物を選択または同定する工程、および
B)本発明方法の工程d)において行われる比較により、第1の参照対照について得られる結果と比べて、該化合物の存在下で組換え細胞においてカルシウム流入の有意な増加が見られることが示された場合、負のモジュレーターとして該化合物を選択する工程。
A)工程b)において上記組換え細胞をATPなどの上記P2X受容体のアゴニストおよびイオンチャンネル受容体のアゴニストの存在下で試験すべき化合物と接触させることを特徴とする、イオンチャンネル受容体の活性を調節しうる化合物を選択または同定するための本発明方法により、イオンチャンネル受容体の活性を調節しうる化合物を選択または同定する工程、および
B)本発明方法の工程d)において行われる比較により、第1の参照対照について得られる結果と比べて、該化合物の存在下で組換え細胞においてカルシウム流入の有意な増加が見られることが示された場合、負のモジュレーターとして該化合物を選択する工程。
(P2X2およびGABA−A受容体を共発現する卵母細胞での記録)
ATPの適用(100μM)はカルシウムプローブの蛍光の著しい一時的増加を誘導し、これはP2X受容体の活性化とそれに続くカルシウム流入を実証する(図3)。ATPの連続適用は同様の大きさの応答を誘導する。この蛍光における増加が細胞の静止電位で見られることに注意することは重要である。
ATPの適用(100μM)はカルシウムプローブの蛍光の著しい一時的増加を誘導し、これはP2X受容体の活性化とそれに続くカルシウム流入を実証する(図3)。ATPの連続適用は同様の大きさの応答を誘導する。この蛍光における増加が細胞の静止電位で見られることに注意することは重要である。
Claims (29)
- 下記工程を含む、イオンチャンネル受容体の活性を調節しうる化合物を選択または同定するための方法:
a)該イオンチャンネル受容体およびP2X受容体を発現する組換え細胞を用意する工程、
b)該組換え細胞を、該P2X受容体のアゴニストの存在下、または該P2X受容体のアゴニストおよび該イオンチャンネル受容体のアゴニストの存在下で、試験すべき化合物に接触させる工程、
c)P2X受容体を介したカルシウム流入を測定する工程、
d)工程c)で得られた測定値を、工程b)において試験すべき化合物が存在しない第1の参照対照について同じ条件で得られた測定値と比較する工程、および
e)工程d)における比較により、該化合物の存在下で組換え細胞におけるカルシウム流入が有意に増加または減少することが示された場合、該化合物を選択する工程。 - 工程a)において前記組換え細胞が、前記P2X受容体のアゴニスト、または前記イオンチャンネル受容体のアゴニストにより調節されうる受容体を内在的に発現しない、請求項1記載の方法。
- 工程a)における前記組換え細胞が、哺乳動物細胞またはアフリカツメガエル卵母細胞である、請求項1または2記載の方法。
- 前記P2X受容体が天然のP2X受容体であるか、前記イオンチャンネル受容体と相互作用しうる変異型のP2X受容体である、請求項1〜3のいずれかの項記載の方法。
- 前記P2X受容体が、P2X 1、P2X 2、P2X3、P2X4、P2X5、P2X6およびP2X7受容体、およびATP活性化チャンネル−受容体機能を果たしうるP2Xサブユニット受容体の会合もしくは組み合わせにより得られる任意のP2X受容体からなる群より選ばれる、請求項4記載の方法。
- 前記イオンチャンネル受容体またはP2X受容体がヒト、ラットまたはマウス由来である、請求項1〜5のいずれかの項記載の方法。
- 前記P2X受容体のアゴニストが、ATP、α, βmATP(アルファベータ−メチルATP)、ベンゾイルベンゾイックATP(2’および3’混合異性体または2’,3’−O−(4−ベンゾイルベンゾイル)−ATP(BzATP)など)、および2−メチルチオ−ATP(2−MeSATP)からなる群より選ばれ、好ましくはATPである、請求項1〜6のいずれかの項記載の方法。
- 工程c)においてカルシウム流入が、カルシウムプローブにより測定される、請求項1〜7のいずれかの項記載の方法。
- カルシウム流入が、カルシウムグリーン、Fluo−3もしくはFluo−4などの蛍光カルシウムプローブにより測定される、請求項8記載の方法。
- 前記試験すべき化合物が、前記P2X受容体と特異的に相互作用する能力について試験され、好ましくは該特異的相互作用が、第2の参照対照について同じ条件でカルシウム流入の有意な増加または低下が得られるかどうかを測定することにより証明され、ここで工程a)において該第2の参照対照に使用される組換え細胞は前記P2X受容体を発現し、前記イオンチャンネル受容体を発現しないものである、請求項1〜9のいずれかの項記載の方法。
- 前記第2の参照対照について組換え細胞でカルシウム流入の有意な増加または低下が見られる場合、前記試験すべき化合物を選択しない、請求項10記載の方法。
- 下記工程を含む、イオンチャンネル受容体のアゴニストを選択または同定する方法:
A)工程b)において前記組換え細胞を前記P2X受容体のアゴニストの存在下のみで試験すべき化合物と接触させることを特徴とする請求項1〜11のいずれかの項記載の方法により、前記イオンチャンネル受容体の活性を調節しうる化合物を選択または同定する工程、および
B)請求項1〜11のいずれかの項記載の方法の工程d)において行われる比較により、第1の参照対照について得られる結果と比べて、該化合物の存在下で組換え細胞においてカルシウム流入の有意な減少が見られることが示された場合、アゴニストとして該化合物を選択する工程。 - 下記工程を含む、イオンチャンネル受容体の正のモジュレーターを選択する方法:
A)工程b)において前記組換え細胞を前記P2X受容体のアゴニストおよび前記イオンチャンネル受容体のアゴニストの存在下で試験すべき化合物と接触させることを特徴とする請求項1〜11のいずれかの項記載の方法により、イオンチャンネル受容体の活性を調節しうる化合物を選択または同定する工程、および
B)請求項1〜11のいずれかの項記載の方法の工程d)において行われる比較により、第1の参照対照について得られる結果と比べて、該化合物の存在下で組換え細胞においてカルシウム流入の有意な減少が見られることが示された場合、正のモジュレーターとして該化合物を選択する工程。 - 請求項1〜11のいずれかの項記載の方法の工程b)において、前記イオンチャンネル受容体のアゴニストを、非飽和濃度で使用する、請求項13記載のイオンチャンネル受容体の正のモジュレーターを選択する方法。
- 下記工程を含む、イオンチャンネル受容体の負のモジュレーター、インヒビターまたはアンタゴニストを選択または同定する方法:
A)工程b)において前記組換え細胞を前記P2X受容体のアゴニストおよび前記イオンチャンネル受容体のアゴニストの存在下で試験すべき化合物と接触させることを特徴とする、請求項1〜11のいずれかの項記載の方法により、前記イオンチャンネル受容体の活性を調節しうる化合物を選択する工程、および
B)請求項1〜11のいずれかの項記載の方法の工程d)において行われる比較により、第1の参照対照について得られる結果と比べて、該化合物の存在下で組換え細胞においてカルシウム流入の有意な増加が見られることが示された場合、負のモジュレーターとして該化合物を選択する工程。 - 第1の化合物を試験するために使用する同じ組換え細胞が、少なくとも第2の試験すべき化合物を試験するために使用できる、請求項1〜15のいずれかの項記載の方法。
- 前記イオンチャンネル受容体が、GABA受容体、グリシン受容体、アセチルコリン受容体およびセロトニン受容体からなる群より選ばれる、請求項1〜16のいずれかの項記載の方法。
- 前記イオンチャンネル受容体がGABA受容体である、請求項17記載の方法。
- 前記イオンチャンネル受容体が、任意のGABA受容体サブユニット間のホモメリックまたはヘテロメリックな会合から生じるイオンチャンネルGABA受容体であり、該サブユニットがα1〜6、β1〜3、γ1〜3、δ、ε、θ、πおよびρ1〜3サブユニットからなるGABA受容体サブユニットの群から選ばれ、該ホモメリックまたはヘテロメリックな会合がGABAにより活性化され得るイオンチャンネル受容体を形成する、請求項17または18記載の方法。
- 前記イオンチャンネル受容体のアゴニストが、GABAまたは、ムシモールやイソガバシンなどの他の周知のGABA受容体アゴニストの任意のものである、請求項18または19記載の方法。
- 以下を含む、イオンチャンネル受容体の活性を調節しうる化合物の選択のためのキット:
−イオンチャンネル受容体およびP2X受容体を共発現しうる組換え細胞、または場合により該組換え細胞を得るための材料;
−ATP、α, βmATP(アルファベータ−メチルATP)、ベンゾイルベンゾイックATP(2’および3’混合異性体または2’,3’−O−(4−ベンゾイルベンゾイル)−ATP(BzATP)など)および2−メチルチオ−ATP(2−MeSATP)からなる群より選ばれるP2X受容体のアゴニスト、好ましくはATP;
−場合により、該イオンチャンネル受容体のアゴニスト、好ましくはGABA;および
−場合により、該組換え細胞においてカルシウム流入を測定するためのマーカーとしてのカルシウムプローブ、好ましくは蛍光カルシウムプローブ。 - 前記イオンチャンネル受容体、P2X受容体、組換え細胞およびカルシウムプローブが、請求項1〜20に記載のものの中から独立に選択される、請求項21記載のキット。
- 請求項1〜22に記載の必須の要素を含む、イオンチャンネル受容体の活性を調節しうる化合物を選択するための装置。
- 請求項1〜20のいずれかの項記載の方法により選択または同定された化合物。
- 前記イオンチャンネル受容体がGABA受容体、特にGABA−A受容体である請求項1〜20のいずれかの項記載の方法により選択または同定された、請求項24記載の化合物。
- P2X受容体と機能的に相互作用しうるイオンチャンネル受容体に関連する、ヒトを含む哺乳動物の疾患または障害を診断、予防または治療するための、請求項24または25記載の化合物の使用。
- P2X受容体と機能的に相互作用しうるイオンチャンネル受容体に関連する、ヒトを含む哺乳動物の疾患または障害の予防または治療への、あるいはそのための医薬の製造への、請求項24または25記載の化合物の使用。
- GABA−A受容体の機能不全に関連する、ヒトを含む哺乳動物の疾患、障害または症状を診断、治療または予防するための、請求項25記載の化合物の使用。
- 喘息、急性心疾患、低血圧、尿閉、骨粗鬆症、高血圧、狭心症、心筋梗塞、潰瘍、アレルギー、良性前立腺肥大、前立腺がん、パーキンソン病、精神病性および神経学的障害、不安、分裂病、躁病、鬱病、運動異常、記憶障害、睡眠障害、痙攣疾患または癲癇の予防または治療への、あるいはそのための医薬の製造への、請求項25記載の化合物の使用、または請求項28記載の使用。
