実施例1−無脊椎動物神経節に対するレチクリン曝露は、内因性モルヒネレベルを増大する
以下の実験は、組織をオピエートアルカロイド前駆体のレチクリンに対して曝露することにより、内因性モルヒネレベルの増大を生じるかどうかを決定するために行われた。
Long Island Soundの地方の水域から収集されたムラサキイガイ(Mytilus edulis)を、実験に使用する少なくとも14日前に実験条件下で維持した。イガイを、以前に記載されたとおり(Stefano et al., Electro-Magnetobiol., 13:123-36 (1994))、30PSUの塩分にて、かつ18℃の温度にて、人工海水(Instant Ocean, Aquarium Systems, Mentor, Ohio)中で保持した。
モルヒネは、液体および固体抽出物に続くアセトニトリルの勾配を使用する逆相HPLCによる神経節抽出物中に同定され、信頼のおける標準と比較した(図1)。材料は、信頼のおけるモルヒネと同じ滞留時間を示した。モルヒネの電気化学的検出感度は、80ピコグラムであった。モルヒネの濃度は、外部標準について算出したピーク面積から推定される、Chromatogram Manager 3.2市販ソフトウェア(Millemmium32, Waters, Milford, MA)によって決定すると、1.43±0.41ng/mg±SEM神経節湿重量であった。50ngのレチクリンと共に1時間インキュベートした神経節は、これらの内因性モルヒネレベルの統計的増大を示した(6.7±0.7ng/mg 組織湿重量;P<0.01;図1)。
ヒト細胞(NCI-H295R)は、副腎皮質の癌腫に由来するNCI-H295多能性副腎皮質癌腫株化細胞(ATCC CRL-10296)から適応された。原細胞を培地に適応させ、これにより5日から2日に集団倍加時間が減少した。原細胞は、懸濁液中で増殖したが、適応された細胞は、単層中で増殖するように選択した。これらの細胞は、副腎アンドロゲンを産生する能力を保持しており、アンジオテンシンIIおよびカリウムイオンに応答した。これらの細胞を増殖するために、培地は、15mM HEPES、0.00625mg/mL インスリン、0.00625mg/mL トランスフェリン、6.25ng/mL セレン、1.25mg/mL ウシ血清アルブミン、および0.00535mg/mL リノール酸(97.5%);Nu-Serum I、2.5%を含むダルベッコ変法イーグル培地とハムF12培地との1:1混合物であった。たとえば、Rainey et al., Mol. イガイ足神経節を得て、10μg L-DOPA単独、10μgドーパミン単独、または10μg L-DOPAに加えて10μg ドーパミンと共にインキュベートした。対照神経節は、L-DOPAまたはドーパミンと共にインキュベートしておらず、11.9ng/mLのモルヒネを示した。L-DOPA単独またはドーパミン単独と共にインキュベートした神経節は、それぞれ9.31および8.82ng/mLのモルヒネを示した。対照的に、L-DOPAおよびドーパミンの両方と共にインキュベートした神経節は、5.52ng/mLのモルヒネを示した。これらの結果は、L-DOPAおよびドーパミンの両方での処置によりモルヒネ産生を減少させることができることを示す。

本明細書に記述したとおり、インビトロおよびインビボの実験から得て、別々に試験した、切開してプールした神経節を使用して、モルヒネ抽出プロトコルを行った。
モルヒネRIA測定は、125I標識したモルヒネを抗体結合部位に対して試験試料中のモルヒネと一定期間競合するという固相の定量的RIAであった。使用した市販のキットは、Diagnostic Products Corporation(USA)から得た。検出限界は、0.5ng/mLであった。
HPLC解析は、Waters 626ポンプ(Waters, Milford, MA)およびC-18 Unijet小口径カラム(BAS)で行った。小口径カラムのために必要とされる低容積流速を提供するために、フロースプリッター(BAS)を使用した。分割比は、1/9であった。0.5mL/分にてポンプを操作することにより、50μL/分の小口径カラム流速を得た。注入体積は、5μLであった。モルヒネ検出は、アンペロメトリック検出器LC-4C(BAS, West Lafayette, Indiana)で行った。小口径カラムは、デッドボリュームを最小限にするために、検知セルに直接連結した。電気化学的検出システムには、グラッシーカーボン-作用電極(3mm)および0.02Hzのフィルター(500mV;レンジ10nA)を使用した。セル容積は、16μmガスケットによって減少させた。クロマトグラフィー・システムは、Waters Millennium32 Chromatography Manager V3.2ソフトウェアによって制御し、クロマトグラムは、Chromatographソフトウェア(Waters)で統合した。
インビトロにおける種々の濃度のL-DOPAまたはレチクリンとの足神経節のインキュベーションにより、時間および濃度依存的な様式で神経節モルヒネレベルが増大した(図5および6)。ビヒクルまたは5-ヒドロキシトリプトファン(1μg/gm組織)(セロトニン前駆体)に曝露した対照神経節では、それぞれ神経節湿重量あたり2.1±0.44および2.1±0.41ngモルヒネを示したが、L-DOPAに曝露したものは、神経節あたり3.6±0.45ngモルヒネを示し、統計学的に有意な増大を表している(P<0.05)。100ngのレチクリンに対する切除した神経節の曝露では、mg神経節あたり約5.0±0.47ngモルヒネを生じた(図5および6;P<0.001)。L-DOPA曝露後の神経節モルヒネレベルの増大は、曝露の10分後に始まり、60分のインキュベーション期間を通して段階的に生じた(図6)。これらの結果から、約5%のL-DOPAがモルヒネに変換されたと思われる。モルヒネHPLC測定の間の空試験ならびにネガティブ組織対照、すなわち外套を試験しても、RIAでモルヒネ残留物は見えなかった。また、プロトコルに使用した海水および種々の化学薬品の解析でもモルヒネがなかった。

本明細書に記述したとおりに、モルヒネ抽出プロトコルをプールした神経節において行った。次いで、固相抽出の前に、乾燥抽出物を0.05%のトリフルオロ酢酸(TFA)水に溶解した。試料は、事前に100%のアセトニトリルで活性化して0.05%のTFA-水で洗浄したWaters Sep-Pak Plus C-18 カートリッジに充填した。モルヒネ溶出は、10%のアセトニトリル溶液で行った(水/アセトニトリル/TFA、89.5%:10%:0.05%(v/v/v)。溶出した試料をCentrivap Consoleで乾燥して、HPLC解析の前に溶解した。

インビトロにおける種々の濃度のノルラウダノソリン(テトラヒドロパポベリン(THP)とも呼ばれる)との足神経節のインキュベーションにより、濃度および時間依存的な様式で神経節モルヒネレベルが増大した(図8および9;P<0.01)。ノルラウダノソリン曝露後の神経節モルヒネレベルの増大は、60分のインキュベーション期間を通して段階的に生じた(図9)。約20パーセントのノルラウダノソリンがモルヒネに変換されたと思われる。モルヒネHPLC測定の間の空試験ならびにネガティブ組織対照、すなわち外套を試験しても、RIAでモルヒネ残留物は見えなかった。また、プロトコルに使用した海水および種々の化学薬品の解析では、モルヒネがないことを証明した。
ヒト末梢血細胞は、Long Island Blood Services (Melville, New York)から得た。ACK溶解緩衝液(8.29g NH4Cl、0.15M;1g KHCO3、1.0mM、37.2mg Na2EDTA、0.1 mM、800mL H2Oを添加し、1N HClでpHを7.2〜7.4に合わせる);0.2μmフィルターを通して滅菌濾過し、室温にて貯蔵した)を使用して、白血球を含むバフィーコートから全ての赤血球を除去した。細胞を溶解緩衝液中で室温にて5分間インキュベートして、RPMI培地(ATCC)を使用して溶解反応を止め、続いて200gにて10分間遠心分離した。上清をデカントして、ペレットをRPMI培地で洗浄した。白血球をピペット操作によってRPMI培地に再懸濁した。

Lett., 25:323 (2004))、インキュベーション期間後に洗浄してペレットにしたWBCおよびPMNにより行った。固相抽出の前に、乾燥抽出物を0.05%のトリフルオロ酢酸(TFA)水に溶解した。試料は、事前に100%のアセトニトリルで活性化して0.05%のTFA-水で洗浄したWaters Sep-Pak Plus C-18 カートリッジに充填した。モルヒネ溶出は、10%のアセトニトリル溶液で行った(水/アセトニトリル/TFA、89.5%:10%:0.05%(v/v/v)。溶出した試料をCentrivap Consoleで乾燥して、HPLC解析の前に溶解した。
モルヒネRIA測定は、125I標識したモルヒネを抗体結合部位に対して試験試料中のモルヒネと一定期間競合するという固相の定量的RIAであった。使用した市販のキットは、Diagnostic Products Corporation(USA)から得た。検出限界は、0.5ng/mLであった。
ボランティア供血者から得たヒトのヘパリン処置した全血(Long Island Blood Services; Melville, NY)を1-Step Polymorphs(Accurate Chemical and Scientific Corporation, Westbury, NY)勾配媒質を使用して直ちに分離した。5mLのヘパリン処置した血液を14mLの丸底チューブ内の5mLの多形核球に重層し、次いで、500×gでスインギングバケットローターにおいて18℃にて35分間遠心分離した。遠心分離後、単核細胞(MN)からなる試料/培地界面の上部バンドと、多形核細胞(PMN)からなる下部バンドとを14mLチューブに収集し、次いで400×gにて10分間遠心分離することによってPBS(Invitrogen, Carlsbad, CA)で洗浄した。
MNおよびPMN細胞(5×106)を遠心分離によってペレットにして、総RNAをRNeasy Protect Mini Kit(Qiagen, Stanford, CA)で単離した。ペレットにした細胞を緩衝液RLTに再懸濁して、可溶化液をシリンジに装着した20ゲージ針に5回通すことによってホモジナイズした。次いで、試料を製造業者の説明書に従って処理した。最終工程では、10,000rpmにて1分間遠心分離することによって、50μLのRNaseを含まない水でRNAを溶出した。
第1鎖cDNA合成は、ランダムプライマー(Invitrogen, Carlsbad, CA)を使用して行った。1μgの総RNAを95℃にて変性させ、Superscript III Rnase H-RT(Invitrogen, Carlsbad, CA)を使用して40℃にて1時間逆転写した。10マイクロリットルのRT産物をCYP2D6遺伝子に特異的なプライマーおよびPlatinum Taq DNAポリメラーゼ(Invitrogen, Carlsbad, CA)を含むPCR混合物に添加した。フォワードプライマー配列は、

であった。PCR反応は、94℃にて5分間変性させ、続いて95℃にて1分間、60℃にて1分間、および72℃にて1分間を40サイクル、次いで72℃にて10分間の伸長工程サイクルとした。PCR産物は、臭化エチジウムで染色した2%アガロースゲル(SIGMA, St. Louis, MO)で解析した。PCR産物の予想されるサイズは、他に記述されたとおり(Zhuge and Yu, World J. Gastroenterol., 10:3356 (2004))、700bp、300bp、およびその他であった。
本明細書に記述されたとおりに得られたPMNを、他に記述されたとおり(Schon et al., Adv. Neuroimmunol., 1:252 (1991))、細胞高次構造の画像解析のためにも処理した。PMNの形態学的測定は、画像解析ソフトウェア(Compix, Mars, PA)を使用することにより、細胞面積および周囲長測定に基づいた。形状因子(FF)算出は、他に記述されたとおり行った(Stefano et al., Proc. Natl. Acad. Sci. USA, 89:9316 (1992); Stefano et al., Proc. Natl. Acad. Sci. USA, 90:11099 (1993);およびStefano et al., J. Neuroimmunol., 47:189 (1993))。測定決定およびフレーム取り込みのために使用した観察領域は、直径0.4μmであった。コンピューターを利用した画像解析システム(ノマルスキーおよび位相差オプティクスを装着したZeiss Axiophot)は、他に記述されたものと同じであった(Stefano et al., Proc. Natl. Acad. Sci. USA, 89:9316 (1992); Stefano et al., Proc. Natl. Acad. Sci. USA, 90:11099 (1993);およびStefano et al., J. Neuroinnnunol., 47:189 (1993))。
対照(ビヒクル曝露した)白血球(WBC)では、モルヒネが、12.33±5.64pg/1,000,000細胞±SEMのレベルにて同定された(図10)。これらの細胞を無血清RPMI培地中で広範に洗浄し、細胞に見いだされ得るあらゆる血漿モルヒネを制限した。しかし、細胞は、血漿から非特異的にモルヒネを蓄積したかもしれない。WBCが内因性合成によってモルヒネを含むかどうかを決定するために、細胞を、アミノ酸チラミンを含む特異的モルヒネ前駆体と共にインキュベートした。チラミン、ノルラウダノソリン(THP)、レチクリン、およびL-DOPAは、未処置細胞において見いだされるものを上回ってWBCモルヒネ濃度を有意に増大した(ANOVA検定、P<0.001;図11)。前駆体と共にインキュベートした細胞におけるモルヒネ濃度は、THP、レチクリン、およびL-DOPAで1時間処置後に、それぞれ90.25±10.42pg、136.04±8.71pg、および136.5±12.43pg/1,000,000細胞であった。さらにまた、モルヒネ濃度は、濃度依存的様式で前駆体に対する曝露と共に増大した(図11)。これらの結果は、WBCが、低いものの、モルヒネの生理的に有意な量を含むこと、およびモルヒネ前駆体に対するこれらの細胞の曝露により、モルヒネ合成を増大することができることを示す。
Long Island Soundの地方の水域から収集されたムラサキイガイを本明細書に記述したとおりに維持した。ドーパミン(DA)またはモルヒネの生化学的インビトロ解析のために、20匹の動物群を種々の時間帯にて切除して、これらの足神経節を1〜100ng/mLの範囲の種々の濃度のチロシンまたはチラミンと共にインキュベートした。神経節は、煮沸した無細胞人工海水とInstant Ocean(Boston, MA)との50:50混合物中で維持した。足神経節をキニジン、CYP2D6阻害剤、およびチロシン水酸化酵素を阻害するαメチルパラチロシン(AMPT)の存在下において、チロシンまたはチラミンの存在下または非存在下においてインキュベートした。
ドーパミンは、神経節(処置あたり20匹の足神経節;4回繰り返す)および血リンパ(処置あたり10mL;4回繰り返す)の両方から抽出した。神経節の切開後、神経節を1つのエッペンドルフチューブにプールし、1mlの1N HClを添加して、組織をソニック・ディスメンバレーター(sonic dismemberator:Fisher scientific, USA)によって超音波処理した。次いで、ホモジナイズした組織を15mLのポリプロピレン遠心管(Fisher Scientific, PA, USA)へ移した。5mLのクロロホルム/イソプロパノール(9:1、v/v)を添加して、チューブの内容物を室温で5分間勢いよくボルテックスした。チューブを4000rpmにて15分間4℃にて遠心分離した。上清(水溶性層)を事前にシリコン処理した1.5チューブ(Midwest Scientific)へと手早く処理して、HPLC測定のために即座に使用するために4℃にて保持するか、またはさらなる解析のために-80℃に貯蔵した。
足神経節(100)を切開後直ちに処理した。神経節を1.5mLのチューブ内に置き、次いでPBS(Invitrogen, Carlsbad, CA)で洗浄した。総RNAをRNeasyミニキット(Qiagen、Valencia, Ca)を使用して単離した。神経節を600μL緩衝液RLT中でホモジナイズした。試料を製造業者の説明書に従って処理した。最終工程では、10,000rpmにて1分間遠心分離することにより、50μLのRNaseを含まない水でRNAを溶出した。

PCR産物をゲルから切除した後、Qiaquickゲル抽出キット(Qiagen)でDNA精製を行った。PCR産物を35μL H2Oに溶解して、直接シーケンシングのためにLark Technologies, Inc.(Houston, TX)に送付した。
イガイ足神経節をチロシンまたはチラミンと共にインビトロでインキュベートしたところ、これらの両方共が、濃度および時間依存的様式で神経節モルヒネレベルの増大を生じた(図16;P<0.001、チラミンについて1.08±0.27ng/g神経節湿重量〜2.51±0.36ng/g、およびチロシンについて0.96±0.31ng/g〜2.39±0.64ng/g)。チロシンおよびチラミン曝露後の神経節モルヒネレベルの増大は、60分のインキュベーション期間を通して徐々に生じた(図16B)。約7パーセントのチロシンまたはチラミンは、これらのインビトロ条件下でモルヒネに変換されたと思われる。モルヒネHPLC測定の間の空試験ならびにネガティブ組織対照、たとえば外套膜を試験しても、HPLC共役RIAでモルヒネ残留物は見えなかった。また、プロトコルに使用した海水および種々の化学薬品の解析では、モルヒネがないことを証明した。
以下の実験は、低用量のモルヒネが生物学的効果を及ぼす能力を評価するために行った。全てのイガイ動物には、注射によって塩類溶液またはモルヒネ(10-6〜10-10M)を処置した。5分のインキュベーション後、血リンパを収集して、LPS(1μg/mL)と共にインキュベートした。10-7〜10-10Mのモルヒネで前処置した動物からの細胞では、塩類溶液で処置した動物から得られたLPSで処置した細胞で観察される細胞活性化のレベルの減少を示さなかった(図23)。モルヒネ(10-6 M)で前処置した動物からの細胞は、19.3±3.8パーセントの活性化を示した。したがって、より低い用量のモルヒネでは、ほぼ同時様式で投与したときに、免疫細胞に対するLPS刺激作用をあまり変化させなかった。
ヒト血液細胞(単核細胞および多形核細胞)を10-7Mのモルヒネ-6β-グルクロニド(M6G)と共に30分間インキュベートして、リアルタイムRT-PCR(Applied Biosystems 5700 SDS)を使用して、μ3オピエート受容体配列の相対的遺伝子発現レベルから評価した。加えて、M6Gを添加する前に、細胞を10-6 M CTOPの存在下または非存在下のいずれかにおいて10分間インキュベートした。
脳における細胞内および細胞外β-アミロイドペプチド(Aβ)の沈着は、よく見られる神経変性障害であるアルツハイマー病(AD)の病的特色である。以下の実験は、ADの症候を生じさせる際のAβの役割をより理解するために行った。
SH-SY5Yヒト神経芽腫細胞(ATCC, USA)は、ダルベッコ変法イーグル培地/ハム栄養剤混合物(DMEM-F12)(Invitrogen, USA)中で培養し、HTB-11ヒト神経芽腫細胞(ATCC, USA)は、最小必須培地アルファ培地(MEM-α)(Invitrogen, USA)中で培養した。細胞は、5% CO2/95% 空気ガスを供給した37℃のインキュベーター(Napco)内に保持した。全ての処置は、無菌フード下で行った。

予想される産物長は、両方のプライマーセットについて556bpであった。次いで、PCR産物および100 bp DNAマーカーを臭化エチジウムで染色した2%アガロースゲルに充填した。ゲル電気泳動は、一定アンペア数で110Vに設定した電源(E-C Apparatus Corp.)を使用して1.5時間行った。次いで、Gel Documentation System(UVP)を使用してゲルを撮影し、P4 Windowsマシン上でGel-Pro Analyzer(MediaCybernetics)を使用してバンドを解析した。発現レベルは、以下のプライマーを使用して、参照遺伝子(サイクロフィリン)に対して標準化した:

未処置のHTB-11細胞は、BACE-1およびBACE-2 mRNAを構成的に発現する。これらの細胞に対するモルヒネ曝露は、24時間後に、濃度依存的様式でBACE-1の発現をダウンレギュレートする(1μM投薬量では、5μMよりも優れた効果を有し、18%と比較して44%である;図32)。同時に、モルヒネは、HTB-11細胞におけるBACE-2発現の発現をアップレギュレートし、Aβ1-42の存在下において効果が増強した(図33)。BACE-1は、Aβ1-42の産生を促進し、BACE-2は、これを阻害するので、BACE酵素のモルヒネ調整がAβ1-42の産生を減少させるであろうから、モルヒネは神経保護的であり得る。BACE-1およびBACE-2発現に対するモルヒネの効果は、ナロキソンによって遮断され(図34)、モルヒネの神経保護作用は、μ3オピエート受容体に対するその結合に直接関連があることを立証している。
以下の実験は、モルヒネおよびNOが、タンパク質代謝を介して細胞ストレスの予防において役割を担っているかどうかを決定するために行った。特に、以下の実験は、モルヒネが、NOの産生の刺激を介して、細胞ストレスの誘導およびタンパク質代謝の不均衡を減らすことによって神経細胞を保護するかどうかを決定するために行った。
ヒトSK-N-SH神経芽細胞腫株化細胞(ATCC #HTB-11)を細胞モデルとして使用した。細胞は、10% ウシ胎児血清(FBS)、2% ペニシリン/ストレプトマイシン、1.5g/L NaHCO3、および1.0mMピルビン酸を含む最小必須培地アルファ(MEMα)中で増殖させた。温度およびpHの維持のために、37℃の5% CO2インキュベータ(NAPCO)を使用した。トリプシン-EDTA溶液(0.25% トリプシン、0.03% EDTA)を使用して接着細胞を吸引し、ペレットにした(室温にて5分間400G)。必要なときに、血球計数器を使用して細胞を6または12ウェルプレートにプレートにまいた(6ウェルプレートに2×105細胞/ウェルまたは12ウェルプレートに1×105細胞/ウェル)。実験操作は、細胞がプレートの底面に付着した後、薄層気流フード下にて無菌条件下で行った。
モルヒネで刺激された、cNOSに由来するNO放出が、神経芽腫細胞において生じた(図43AおよびB;22.3nM±0.85のピーク値、ベースライン2nM NOと比較してp<0.001、ベースラインは、ネガティブ対照と比較したときに有意ではなかった)。ナロキソン(10-6 M)およびL-NAME(10-4M)は、10-6 Mモルヒネによって誘導されるNO放出を遮断した(それぞれ、3.2nM NOおよび0.3nM NO)。
