本発明は、一般的に生殖生物学に関する。より詳細には、本発明は妊娠特異的糖タンパク質(PSG)に関する。
卵母細胞のインビトロでの受精(IVF)は、様々な形態の雌性及び雄性不妊を克服するのに用いられる広く実用的な医療技術であり、その結果不妊カップルに提供される。標準的なIVF処置は、卵母細胞の成熟を誘導する外因性ホルモンを用いた、女性患者の排卵誘発(COH)に基づいている。この処置は、典型的には、ゴナドトロピン放出ホルモン(GnRH)アゴニスト又は患者自身の卵胞刺激ホルモン(FSH)及び黄体形成ホルモンを抑制するアンタゴニストを投与することにより開始される。その後、外因性ゴナドトロピン(FSH及び/又はLH)の注入へと続き、多数の排卵前卵胞への発達を確実にする。排卵の直前に、多数のインビボで成熟させた卵母細胞を卵巣から取り除く。単離した成熟卵母細胞はその後受精させ、発達した胚を4〜8細胞分裂期に子宮に戻す前に、典型的に3〜6日間培養する。
COH処置は、5組のカップルの約1組のカップルにおいて成功せず、多嚢胞卵巣障害の女性などの多くの女性には薦められていない。更に、COH処置において用いられる外因性ホルモン処理は、卵胞の発達及び卵胞の成熟を過剰に刺激し得る。COHを受ける患者の一部は、卵巣過剰刺激症候群(OHSS)に罹患し、これは重度の潜在的な致命的状態である。結果として、COHを受ける女性は、毎日の卵巣の超音波検査及び血中ホルモン測定により厳密に観察されなければならない。
COHを用いる標準的なIVF処置の制限により、様々な代替のプロトコルが提案されてきた。COHプロトコルの危険性、副作用及び経済的不利益を軽減する1つの方法は、インビボでの卵母細胞の成熟が後に続く未熟細胞の回収に関連する。この方法においては、女性は刺激を受けず、又は軽減した刺激を受け、回収した卵母細胞をインビトロでのホルモン処理にかける。卵母細胞のインビトロでの成熟(IVM)は、典型的には投与する外因性ホルモンの量を減少又は無くすことを可能にし、その結果議論された問題を軽減する。

発明の概要

PSGは卵母細胞のインビトロでの成熟に用いることができる。
a.卵母細胞
卵母細胞が、発達の時期、例えば早期腔卵胞及び腔卵胞(これらに限定されない)にある間に、未熟卵母細胞を女性から取り除くことができる。

未熟卵母細胞は、外因性ホルモン治療を受けていない女性から取り除くことができる。或いは、未熟卵母細胞は、外因性ホルモン治療を受けた女性から取り除くことができる。女性には、ホルモン、例えばGnRH、FSH、LH又はhCG(これらに限定されない)を投与しておくことができる。ホルモンは、別のホルモンとの組み合わせにおいて、又は任意の順序で連続的に投与しておくことができる。
未熟卵母細胞は、超音波検査及び吸引を含む(これらに限定されない)方法により、女性から取り除くことができる。未熟卵母細胞は、単離の後に凍結保存し、インビトロでの成熟の時期に解凍することができる。
b.成熟
単離した未熟卵母細胞は、PSGを含んで成る培地中でインキュベートすることができる。培地は、任意の生理学的に許容される培地、例えばTCM 199、αMEM及びHam’s F10(これらに限定されない)であることができる。培地は、任意に1又は複数の因子、例えばFSH、hCG、エストラジオール、システアミン、ピルビン酸ナトリウム、グルタミン、自己加熱不活性化血清及び卵胞液(これらに限定されない)を更に含んで成ることができる。
単離した未熟卵母細胞は、PSGを含んで成る培地中でインキュベートすることができる。培地は、任意の生理学的に許容される培地、例えばTCM 199、αMEM及びHam’s F10(これらに限定されない)であることができる。培地は、任意に1又は複数の因子、例えばFSH、hCG、エストラジオール、システアミン、ピルビン酸ナトリウム、グルタミン、自己加熱不活性化血清及び卵胞液(これらに限定されない)を更に含んで成ることができる。
未熟卵母細胞は、約6、12、18、24、30、36、42、48、54、60、66又は72時間(これらに限定されない)、約37℃〜約39℃(これらに限定されない)で、培地中でインキュベートすることができる。卵母細胞は、顕微鏡下での卵核胞崩壊(GVBD)、卵丘拡張(expansion)、中期IIプレート形成(Mil)、又は極体押し出し(これらに限定されない)の外観検査を含む方法によって、証明したように成熟が起こるまで、卵母細胞をインキュベートすることができる。
c.胚産生
成熟卵母細胞は、インビトロで精子と共にインキュベートし、Textbook of Assisted Reproductive Techniques Laboratory & Clinical Perspectives(Gardner,et al.,2001 Martin Ldunetz Ltd., London)(この内容は、引用文献により本明細書中に組み込まれる。)に記載されている標準的なインビトロでの受精を用いて哺乳動物の胚を産生することができる。胚は、出産まで胚を保持することができる女性の子宮に移植することができる。
成熟卵母細胞は、インビトロで精子と共にインキュベートし、Textbook of Assisted Reproductive Techniques Laboratory & Clinical Perspectives(Gardner,et al.,2001 Martin Ldunetz Ltd., London)(この内容は、引用文献により本明細書中に組み込まれる。)に記載されている標準的なインビトロでの受精を用いて哺乳動物の胚を産生することができる。胚は、出産まで胚を保持することができる女性の子宮に移植することができる。
2.妊娠特異的糖タンパク質
PSGは、妊娠の成功に必須であり得る妊娠した女性の血清の主な要素を構成する。PSG遺伝子は、11の高度に保存されたメンバーのファミリーを含んで成る。PSGは母体循環系に放出され、妊娠の進行とともに濃度が増大し、出産においては400μg/mlの濃度まで達する。PSGが女性による妊娠の保持以外の機能を有するという事実は、胎児肝臓、唾液腺、睾丸及び骨髄細胞に存在するPSGmRNAにより証明される。
PSGは、妊娠の成功に必須であり得る妊娠した女性の血清の主な要素を構成する。PSG遺伝子は、11の高度に保存されたメンバーのファミリーを含んで成る。PSGは母体循環系に放出され、妊娠の進行とともに濃度が増大し、出産においては400μg/mlの濃度まで達する。PSGが女性による妊娠の保持以外の機能を有するという事実は、胎児肝臓、唾液腺、睾丸及び骨髄細胞に存在するPSGmRNAにより証明される。
成熟を誘導するのに用いられるPSGは、インビトロでの卵母細胞の成熟を誘導する任意のPSG、例えばヒトPSGであることができる。ヒトPSGは、PSG1、PSG2、PSG3、PSG4、PSG5、PSG6、PSG7、PSG8、PSG9、PSG10又はPSG11であることができる。PSGは、PSG1、PSG2、PSG3、PSG4、PSG5、PSG6、PSG7、PSG8、PSG9、PSG10又はPSG11のアナログ、誘導体、フラグメント、ホモログ、又は変異型或いはそれらの組み合わせであることができる。アナログ、誘導体、フラグメント、ホモログ又は変異型は、PSG、例えばPSG5と、少なくとも75%、80%、85%又は90%の配列の同一性を有することができる。PSGフラグメントは、Ig可変様ドメインを含んで成ることができる。PSGフラグメントは、シグナルペプチドを含んで成ることができない。
本明細書中で用いる場合、PSGとの関連で用いる場合には、「アナログ」という用語は、1つ以上の非標準的なアミノ酸又は従来のアミノ酸の組からの他の構造的な変異型を含んでなるペプチド又はポリペプチドを意味する。
本明細書中で用いる場合、PSGとの関連で用いる場合には、「誘導体」という用語は、一次構造(アミノ酸及びアミノ酸アナログ)以外が異なるペプチド又はポリペプチドを意味する。例として、誘導体は、グリコシル化(翻訳後修飾の1つの形態)されることにより異なり得る。例えば、ペプチド又はポリペプチドは、異種系における発現により、グリコシル化パターンが存在し得る。少なくとも1つの生物学的活性が保持されるならば、これらのペプチド又はポリペプチドは誘導体である。他の誘導体としては、共有結合的に修飾されたN末端又はC末端を有する融合ペプチド又は融合ポリペプチド、PEG化ペプチド又はポリペプチド、脂質部分と結合したペプチド又はポリペプチド、アルキル化ペプチド又はポリペプチド、アミノ酸側鎖官能基により他のペプチド、ポリペプチド又は化学物質に結合したペプチド又はポリペプチド、及び当業界で理解される更なる修飾型があげられるが、これらに限定されない。
本明細書中で用いる場合、PSGとの関連で用いる場合には、「フラグメント」という用語は、約8〜50アミノ酸長のペプチドを意味する。フラグメントは、8、9、10、11、12、13、14、15、16、17、18、19、20、21、22、23、24、25、26、27、28、29、30、31、32、33、34、35、36、37、38、39、40、41、42、43、44、45、46、47、48、49又は50アミノ酸長であることができる。
本明細書中で用いる場合、PSGとの関連で用いる場合には、「ホモログ」という用語は、共通の進化的先祖を共有するペプチド又はポリペプチドを意味する。
本明細書中で用いる場合、PSGとの関連で用いる場合には、「変異型」という用語は、アミノ酸の挿入、欠損、又は保存的置換によりアミノ酸配列が異なるが、少なくとも1つの生物学的活性を保持するペプチド又はポリペプチドを意味する。本発明の目的に関して、「生物学的活性」としては、特異的抗体により結合される能力が挙げられるが、これらに限定されない。
本発明は、以下の非限定的な実施例により説明される複数の側面を有する。
実施例1
マウス卵丘−卵母細胞複合体の単離
PMSG(5IU/雌性、Calbiochem 367222)は、7〜8週齢のCD−1雌性マウス(35トータル;Charles River)を準備するのに用いた。マウスは、進行性の低酸素血症により48時間後に屠殺した。アルコール(70%)を、動物の腹部領域に適用してその部分を消毒し、毛による試料の汚染も減少させた。腹部切開を行い、腹腔を露出させた。卵管に接続した卵巣を、子宮角及び内臓脂肪症から取り除いた。卵巣/卵管試料を、3mlのL−15培地(Gibco 11415−064)と10%のウシ胎仔血清(FCS;Invitrogen 16000−044)を含む15mlチューブ(10/チューブ、Corning 430052)に置いた。卵巣/卵管試料を37℃で保持した。
マウス卵丘−卵母細胞複合体の単離

その後、卵巣/卵管試料をペトリ皿(Falcon 353004、60×15mm)に移した。1mlのツベルクリン注射器に取り付けた一組のハサミと針(27ゲージ)を用いて、実体顕微鏡(サーモプレート加熱ステージを有するNikon SM2−800)下で、卵巣及び卵管を脂肪パッドから取り除き、2〜3mlの新鮮な培地(L−15+10% FCS)で満たした新しいペトリ皿に置いた。COCを針による各卵巣の機械的破裂により回収し、2〜3mlの新鮮な培地(L−15+10% FCS)で満たした新しいペトリ皿に置いた。
実施例2
マウス卵丘−卵母細胞複合体のインビトロでの卵丘拡張におけるPSG5の効果
同種細胞質を有する卵丘−無傷卵母細胞を、低倍率(20〜30×)の実体顕微鏡を用いて、実施例1に記載のとおりに調製したCOCから選択し、マウス(mouth)ガラスピペットを用いて、ウエルミネラルオイルあたりに90μlの培地(10% FCSを含むαMEM(Gibco 32571−036)及びPenStrep−Antibiotics(Invitrogen 15140−122))を含む96−ウエルプレート(2/ウエル)に移した。COCを96−ウエルプレートに添加する前に、プレート中の培地を、空気中に5%のCO2を含む加湿したインキュベーター中で37℃において1時間事前に平衡化した。
マウス卵丘−卵母細胞複合体のインビトロでの卵丘拡張におけるPSG5の効果
同種細胞質を有する卵丘−無傷卵母細胞を、低倍率(20〜30×)の実体顕微鏡を用いて、実施例1に記載のとおりに調製したCOCから選択し、マウス(mouth)ガラスピペットを用いて、ウエルミネラルオイルあたりに90μlの培地(10% FCSを含むαMEM(Gibco 32571−036)及びPenStrep−Antibiotics(Invitrogen 15140−122))を含む96−ウエルプレート(2/ウエル)に移した。COCを96−ウエルプレートに添加する前に、プレート中の培地を、空気中に5%のCO2を含む加湿したインキュベーター中で37℃において1時間事前に平衡化した。
PSG5を、各ウエルの最終体積が100μlとなるように、各ウエルに10μlの体積で添加した。各プレートは、4つのウエルの「ネガティブコントロール」(αMEMと10% FCS)及び4つのウエルの「ポジティブコントロール」(αMEMと10% FCSとEGF(5 ng/ml、Sigma E−9644))を含んでいた。1つの試験あたり2つのプレート(重複)を用いて、1つの試験タンパク質あたり2つのウエルを提供した。試験における1:50の最終希釈のために試験プレートにタンパク質を添加する前に、IVM培地(αMEMと10% FCS)にタンパク質を1:5で希釈した。
処理したCOCを含むプレートを、空気中に5%のCO2を含む加湿したインキュベーター中で37℃において18時間インキュベートした。その後、Nikon倒立顕微鏡を用いて各COCを視覚的に検査し、卵丘細胞によるムコイド細胞外マトリクス(卵丘拡張の指標)の形成を同定した。卵丘拡張の割合を、各処理群において用いた全部のCOCと比較した、拡張した卵丘の数として規定した。
PSG5は、最初の試験において、60%のCOCの拡張を誘導した。その後、COC拡張におけるPSG5の効果を、再確認試験において再試験した。それはPSG5が試験した100%のCOCにおいて拡張を誘導することを示した。
最初の試験及び再確認試験の両方において、ネガティブコントロール(EGFを含まない)又はポジティブコントロール(EGFを含む)のプレートは、それぞれ常に0%又は100%の拡張であった(データは示さない)。
実施例3
PSG5の用量反応試験
実施例2に記載した最初及び再確認試験からの結果に基づいて、用量反応試験をPSG5に関して実施した。1つのウエルあたりに4〜5個のCOCを含む3つのウエルが各タンパク質濃度に対して指定されたことを除いて、実施例2に記載の方法と類似の方法において用量反応試験を実施した。特定のタンパク質(試験に添加する前に希釈しないものがあった)の濃度に依存して、試験タンパク質の希釈を行い、1:10の最終濃度とした。
PSG5の用量反応試験
実施例2に記載した最初及び再確認試験からの結果に基づいて、用量反応試験をPSG5に関して実施した。1つのウエルあたりに4〜5個のCOCを含む3つのウエルが各タンパク質濃度に対して指定されたことを除いて、実施例2に記載の方法と類似の方法において用量反応試験を実施した。特定のタンパク質(試験に添加する前に希釈しないものがあった)の濃度に依存して、試験タンパク質の希釈を行い、1:10の最終濃度とした。
PSG5の用量反応試験の結果を、図1に示す。Origin 7 SR3 v7.0475 (B475)を用いた試験は、PSG5による卵丘拡張のEC50が0.64ng/mlであったことを示す。
この記載は、上述の代表的な実施態様に限定されない。
Claims (10)
- (a)未熟卵母細胞を調製すること;及び
(b)卵母細胞を妊娠特異的糖タンパク質(pregnancy specific glycoprotein)と接触させること、
を含んで成る、インビトロで卵母細胞を成熟させる方法。 - 妊娠特異的糖タンパク質が、PSG1、PSG2、PSG3、PSG4、PSG5、PSG6、PSG7、PSG8、PSG9、PSG10及びPSG11から成る群から選択される、請求項1に記載の方法。
- 妊娠特異的糖タンパク質が、PSG5又はそれと少なくとも80%の同一性を有するポリペプチドである、請求項2に記載の方法。
- 未熟卵母細胞が、外部ホルモン治療を受けていない女性に由来する、請求項1に記載の方法。
- 未熟卵母細胞が、外部ホルモン治療を受けた女性に由来する、請求項1に記載の方法。
- 女性が、GnRH、FSH、LH、hCG及びそれらの組み合わせから成る群から選択されるホルモンを投与された、請求項5に記載の方法。
- FSH、hCG、エストラジオール、システアミン、ピルビン酸ナトリウム、グルタミン、自己加熱不活性化血清及び卵胞液から成る群から選択される因子を含んで成る培地中で未熟卵母細胞が調製される、請求項1に記載の方法。
- 請求項1に記載の方法により産生された成熟卵母細胞。
- 卵母細胞が請求項1に記載の方法により産生される、成熟卵母細胞を精子を用いて処理することを含んで成る、インビトロで胚を産生する方法。
- 成熟卵母細胞を精子で処理することを含んで成る方法により胚が産生され、卵母細胞が請求項1に記載の方法により産生される、インビトロでの受精を必要とする胚を移植することを含んで成る、インビトロでの受精の方法。
