図2A−Eは、AD脳および対照脳のT414G mtDNA突然変異のためのPNA固定化PCRアッセイの結果を示す。図2Aは対照のアガロース・ゲル電気泳動の結果を示す。図2BはAD患者のアガロース・ゲル電気泳動の結果を示す。aおよびbの個々の標本は対象の年齢によって同定される。2つのPCRを各対象について示し、野生型mtDNAの増幅を抑える414野生型塩基を包含するPNAが、一方では存在せず(−)、他方では存在する(+)。図2Cは、AD脳からのT414G突然変異の存在を確認するAD脳および対照脳からのT414G突然変異の存在を確認するPNA固定化PCR産物のFokI消化を示す。図2Dは、比較のために同じゲルに入れられた、PNA固定化PCR産物のFokI消化を示す。cのレーンはAD患者の年齢で標識し;−cのレーンは野生型プラスミドからのPCR産物から得られたFok消化の結果;+cのレーンはT414G突然変異プラスミドからの結果。矢印はT414G FokI産物を示す。図2Eは、74歳の対象からのCRフラグメントの配列解析を示すグラフ。414領域はPNA固定化PCR増幅され、得られたフラグメントをPNAなしで再増幅し、最終フラグメントをクローニングおよび配列決定した。突然変異ヌクレオチドG(矢印で示す)が5つのクローンのうちの3つに見られる。
(ダウン症候群関連認知症の診断)
ダウン症候群と確定診断された25歳の対象は、現時点で認知症の症状を呈していない。上記実施例2のように、細胞(例えば、血液細胞、皮膚線維芽細胞、尿管上皮細胞、および/または脳脊髄液細胞)の標本を414および477ヌクレオチド変異の有無について試験し、次いでその突然変異分子をクローニングおよび配列決定した。T4141G、T414CおよびT477C突然変異の総数が対照よりも有意に多い場合、その対象はダウン症候群関連老年性認知症を後年発現しやすいと結論付けてよい。他方、T4141G、T414CおよびT477C突然変異の総数が正常な対照よりも有意に多くない場合、その対象はダウン症候群関連老年性認知症を後年発現しにくいと結論付けてよい。
ダウン症候群と確定診断された25歳の対象は、現時点で認知症の症状を呈していない。上記実施例2のように、細胞(例えば、血液細胞、皮膚線維芽細胞、尿管上皮細胞、および/または脳脊髄液細胞)の標本を414および477ヌクレオチド変異の有無について試験し、次いでその突然変異分子をクローニングおよび配列決定した。T4141G、T414CおよびT477C突然変異の総数が対照よりも有意に多い場合、その対象はダウン症候群関連老年性認知症を後年発現しやすいと結論付けてよい。他方、T4141G、T414CおよびT477C突然変異の総数が正常な対照よりも有意に多くない場合、その対象はダウン症候群関連老年性認知症を後年発現しにくいと結論付けてよい。
Claims (19)
- ヒトまたは動物対象においてβアミロイド沈着またはβアミロイド線維の発達を伴う障害を診断またはそのような障害のために対象に提供される治療の効果を評価する方法であって、該方法は
A)前記対象から得られた組織、細胞または体液の試料を分析することによりmtDNA CR突然変異の存在を判定する工程を含む方法。 - 工程AはmtDNS CR突然変異が存在するか否かを定性的に判定することを含む請求項1に記載の方法。
- 工程AはmtDNS CR突然変異を定量することを含む請求項1に記載の方法。
- B)工程Aで行われた定量によって得られたmtDNS CR値を対照のmtDNS CR値と比較して、前記対象が対照よりも有意に多いmtDNS CR突然変異を有しているかどうかを判定する工程をさらに含む請求項3に記載の方法。
- B)工程Aで行われた定量によって得られたmtDNS CR値を、アミロイド沈着またはβアミロイド線維の発達を伴う障害に罹患した対象を表すmtDNS CR値と比較する工程をさらに含む請求項3に記載の方法。
- 工程AはT4141G突然変異、T414C突然変異、T477C突然変異、T146C突然変異、T152C突然変異、A189G突然変異、またはT195C突然変異の試験を含む請求項1に記載の方法。
- 工程Aの少なくとも一部はPNA固定化PCRで実施される請求項1に記載の方法。
- 工程Aの少なくとも一部はオリゴヌクレオチド・ハイブリダイゼーションによって実施される請求項1に記載の方法。
- 工程Aの少なくとも一部はプライマー伸長法によって実施される請求項1に記載の方法

xi.上皮;
xii.上皮細胞;
xiii.線維芽細胞;
xiv.脳脊髄液;および
xv.脳脊髄液に含まれる細胞
から選択される組織、細胞または体液の試料において行われる請求項1に記載の方法。 - 前記方法は、その障害の症状を呈し始めた対象の該障害の発症後診断のために実施される請求項1に記載の方法。
- 前記方法は、その障害の症状を呈し始めていない対象の該障害の発症前診断のために実施される請求項1に記載の方法。
- 前記障害は神経変性疾患、アルツハイマー病、パーキンソン病、ダウン症候群関連認知症、海綿状脳症、II型糖尿病、クロイツフェルトヤコブ病、クロイツフェルトヤコブ病、ハンチントン病、黄斑疾患、またはプリオン病である請求項1に記載の方法。
- 工程Aは、
対象から標本細胞を採取する工程と、
該標本細胞からDNAを抽出する工程と、
該抽出したDNAをミトコンドリアDNA制御領域増幅に供する工程と、
ヘテロプラスミック414および477ヌクレオチド突然変異について直接配列決定することによって、ホモプラスミック414および477ヌクレオチド変異が存在するかどうか判定する工程と、
414および477ヌクレオチド変異が検出された場合に、該突然変異分子をクローニングし、該クローンを配列決定する工程と、を含む請求項1に記載の方法。 - 請求項1から15のいずれかに記載の方法を実施するのに使用可能な試薬および/または物質を含むテスト・システム。
