次にL−グルタミン酸生合成系遺伝子、prpC遺伝子(国際公開2006/051660号パンフレット)、ppc遺伝子、gdh(欧州出願公開0999282号明細書)遺伝子を増幅したプラスミドRSFPPGを構築した。
RSFCPG(欧州出願公開1233068号明細書)のgltA遺伝子のORF以外の部分を増幅するプライマー1(配列番号13)とプライマー2(配列番号14)を設計した。このプライマーを用いて、RSFCPGを鋳型にPCRを行い、約14.9kbの断片を取得した。一方、prpCに関してはプライマー3(配列番号15)とプライマー4(配列番号16)を用い、E.coli W3110株の染色体DNAを鋳型としてPCRを行い、約1.2kbの断片を取得した。両PCR産物をそれぞれBglII、KpnIで処理し、ライゲーション後、E. coli JM109株を形質転換した。出現したコロニーを全て集菌し、混合物としてプラスミドを抽出した。このプラスミド混合物でCS欠損株であるE. coli ME8330株を形質転換し、50mg/Lウラシル、5mg/Lチアミン-HClを含有するM9最少培地(グルコース5 g、硫酸マグネシウム2mM、リン酸一カリウム3g、塩化ナトリウム0.5g 、塩化アンモニウム1g リン酸2ナトリウム6g を純水1Lに含む培地)に塗布した。出現した株よりプラスミドを抽出し、これをRSFPPGとした。SC17sucArpoS株にGlu生産プラスミドRSFPPGを導入し、rpoSを欠損したGlu生産菌SC17sucArpoS/RSFPPGを構築した。

SC17sucArpoS/RSFPPGの各種pH条件における生育を調べた。SC17sucArpoS/RSFPPG株とrpoS野生株であるSC17sucA/RSFPCPG株を、L培地に最少培地成分を加えた固体培地で終夜培養を行い、滅菌水で2回洗菌した。この菌体を5mLの最少培地(グルコース5g、硫酸マグネシウム2mM、リン酸一カリウム3g、塩化ナトリウム0.5g、塩化アンモニウム1g、リン酸2ナトリウム6g、リジン塩酸塩100mg、L−メチオニン100mg、ジアミノピメリン酸100mg、及びL−グルタミン酸30gを純水1Lに含み、アンモニアにより種々のpHに調整した培地)にOD660nm=0.05となるように植菌し、ADVANTEC社製自動OD測定装置TN1506を用いて、経時的にOD測定を行った。結果を図1、図2に示す。

L培地(バクトトリプトン10 g、イーストエキストラクト5g、NaCl 5g、寒天15gを純水1Lに含む培地 pH7.0)に、最少培地成分(グルコース5g、硫酸マグネシウム2mM、リン酸一カリウム3g、塩化ナトリウム0.5g 、塩化アンモニウム1g、リン酸2ナトリウム6gを純水1Lに含む培地)と12.5mg/Lテトラサイクリンを加えた培地でSC17sucA/RSFPPG株とSC17sucArpoS/RSFPPG株を終夜培養した。これらのプレート1枚分の菌体を300mLの下記成分の培地を張り込んだジャーファーメンターに植菌し、通気1/1vvm、34℃、アンモニアガスにてpH4.7に制御して培養を行った。初糖が切れた時点で糖のフィードを開始し、培養を継続した。尚、SC17sucArpoS/RSFPPG株に関してはL−グルタミン酸の飽和溶解度に達した時点で、ペクチンを1g/Lとなるよう添加した。
