Description
多くの場合、宿主細胞における増殖性アデノウイルスのライフサイクルは短いことが好ましい。増殖性アデノウイルスを製造する時、又は増殖性アデノウイルスをベクターとして用いてタンパク質を細胞で製造する時には、アデノウイルスのライフサイクルが短いと、製造工程の速度が上がる。増殖性アデノウイルスを細胞集団の殺傷に用いる時も、短いライフサイクルはこの工程の効率を高める。短いライフサイクルは、in vivoで増殖性アデノウイルスを使用する際には特に重要である。アデノウイルスは、自らを不活化する強力な免疫応答を動物生体内で誘導する。この現象が、投与した増殖性アデノウイルスのin vivoにおける複製の継続期間を限定する。ライフサイクルが短ければ、増殖性アデノウイルスの投与から不活化までの期間内で娘ウイルスの産生をより多くの回数実施することができる。In vivoにおいて増殖性アデノウイルスの短いライフサイクルが特に重要になる状況は、不適切な細胞寿命が関与する疾患の治療に関連した場合である。このような疾患の典型例は癌である。In vivoの腫瘍に投与した増殖性アデノウイルスの抗癌効果は、(1)ウイルスが、近傍の腫瘍細胞に感染することができる娘ウイルスを産生しながら腫瘍の中を拡散する効率、及び(2)ウイルスが複製と細胞溶解によって腫瘍細胞を殺傷する効率に依存する。従って、短いライフサイクルは、より速い腫瘍退縮、時間当たりの新ウイルス産生周期の増加、一定時間内にウイルスに感染する腫瘍細胞の増加をもたらし、その結果、腫瘍の根絶がより効果的となる。
本発明とその好ましい態様は、添付の請求の範囲から明らかとなる。
ゲートウェイ組み換えデスティネーションカセット(Gateway recombination destination cassette)をアデノウイルスのE4領域と右方向ITRの間に含有するアデノウイルスシャトルベクターを構築するために、pEndK/SpeI構築物(スペイン国、バルセロナ、カタロニア癌研究所(Institut Catala d'Oncologia)のR. Alemany博士による寄贈)を使用した。pEndK/SpeIの作製には、初めにpTG3602(Chartier et al., J. Virol, 70 (1996): 4805-4810)をKpnIで消化し、次にAd5の地図上の0〜7単位と93〜100単位を包含するベクターの断片を再度連結してpEndKを作製した。続いて、部位特異的突然変異でAd5の第35,813番ヌクレオチドをAからにTに変更して、ここにしかないSpeI部位をpEndKに導入し、pEndK/SpeIを作製した。PEndK/SpeIは、Ad5の2つのITRのそれぞれに隣接したPacI制限部位を有する。pEndK/SpeIにゲートウェイシステムとの互換性を与えるために、平滑断片にしたゲートウェイデスティネーションカセットrfa(ゲートウェイベクター変換システム(Gateway Vector Conversion System);カリフォルニア州、カールスバッド、Invitrogen製)を(Klenowポリメラーゼで埋めた)SpeI部位に連結した。アデノウイルスのR鎖又はL鎖上のccdB遺伝子コード配列を有するゲートウェイデスティネーションカセットを含有するプラスミドを選択し、それぞれpEndK/DEST-RとpEndK/DEST-Lと命名した。
プラスミドpSHAG-1(Paddison et al., Genes Dev. 16 (2002) 948-958;ニューヨーク州、コールドスプリングハーバー研究所のG.J. Hannon博士による寄贈)を、ゲートウェイシステム(カリフォルニア州、カールスバッド、Invitrogen製)への導入クローンとして用いる。pSHAG-1は、attL1組み換え部位とattL2組み換え部位に挟まれた、U6プロモーター駆動性の発現カセットを含有し、この発現カセットは、ゲートウェイシステムを用いて、実施例1で使用したpEndK/DEST-R、pEndK/DEST-L、pBHG11-DEST_R及びpAdΔ24-DEST_Rを含む目的プラスミドベクターに導入することができる。shRNAコード配列は、BseRIとBamHIで消化したpSHAG-1を、互換性のある突出したDNA配列を有する2本のアニーリングした合成オリゴヌクレオチドと連結することによって、導入することができる。2本のオリゴヌクレオチドの内の1本は、その5’から3’の方向に以下に記載の配列を有するように設計する: 標的mRNA(即ち、アンチセンス)に相補的な、少なくとも19ヌクレオチドで好ましくは29ヌクレオチド以下の第1伸長鎖、ループ配列、第1伸長鎖と同じ長さで逆相補的な配列である第2伸長鎖、及び少なくとも4つのチミジンからなる伸長鎖。2本目のオリゴヌクレオチドは、1本目のオリゴヌクレオチドに対して逆相補的な配列でなければならない。更に、アニーリングの結果として得られる2本鎖オリゴヌクレオチドは、BseRI制限部位とBamHI制限部位とに互換性のある突出部位を形成しなければならない。19〜29ヌクレオチド長の配列の選択によって、所望の標的に対して有用な、本発明のshRNAを作成することができる。例えば、ホタルルシフェラーゼのターゲティングに有用なオリゴヌクレオチドとしては: オリゴヌクレオチド1: 5'-GATTCCAATTCAGCGGGAGCCACCTGATgaagcttgATCGGGTGGCTCTCGCTGAGTTGGAATCCATTTTTT-3' 及び オリゴヌクレオチド2: 5'-GATCAAAAAATGGATTCCAACTCAGCGAGAGCCACCCGATcaagcttcATCAGGTGGCTCCCGCTGAATTGGAATCCG-3' が挙げられ、上記配列において小文字で示したのはループ配列である。オリゴヌクレオチド1と2のアニーリングに続く、BseRIとBamHIで消化したpSHAG-1への連結によって、pSHAG-shRNAが得られる。ルシフェラーゼ特異的shRNAを例とした場合には、ホタルルシフェラーゼ遺伝子のコード配列の第1,340番〜第1,368番ヌクレオチドに対して相同的なshRNAをコードするpSHAG-Ff1が上記実験によってもたらされる。
E4領域と右方向ITRの間に挿入したshRNA発現カセットを含有するアデノウイルスシャトルベクターを構築するために、ゲートウェイLRクロナーゼエンザイムミックス(GATEWAY LR Clonase enzyme mix)(Invitrogen製)を製造者のプロトコルに従って使用して、in vitroゲートウェイLR組み換え反応(GATEWAY LR recombination reaction)で、shRNA発現カセットを、実施例2のpSHAG-shRNA構築物から実施例1のpEndK/DEST-Rプラスミド又はpEndK/DEST-Lプラスミドに移す。その結果として、pEndK/shRNA-R又はpEndK/shRNA-Lが得られた。例えば、pSHAG-Ff1をpEndK/DEST-R又はpEndK/DEST-Lと組み換えて、それぞれpEndK-Ff1-R又はpEndK-Ff1-Lを得る。
プラスミドpEndK/shRNA-R及びプラスミドpEndK/shRNA-Lは、KpnI及び/又はEcoRVで直鎖化することができる。この結果、Ad5の地図上の0〜7単位と、挿入したshRNA発現カセットを有するAd5の地図上の93〜100単位とが分離される。このような直鎖化した分子は、細菌、例えば大腸菌BJ5183株において、増殖性アデノウイルス全長DNAと組み換えることができる。このような増殖性アデノウイルス全長DNAは、アデノウイルス粒子から単離するか、あるいは、増殖性アデノウイルス全長DNA挿入物を含有するプラスミドの消化によって取り出すこともできる。二重相同組み換え(Double homologous recombination)によって増殖性アデノウイルスゲノム挿入物を含有するプラスミドを作成するが、この時、shRNA発現カセットは、E4領域と右方向ITRの間に挿入する。この方法においては、更に修飾(例えば、腫瘍選択性や腫瘍退縮の増強、指向性の変化、導入遺伝子の挿入)を有する組み換えアデノウイルスを含む、いかなる全長増殖性アデノウイルスもshRNA発現カセットの挿入に用いることが可能であることに注目されたい。しかし、上記全長増殖性アデノウイルスが、PacI制限部位をそのゲノムに有していないことが好ましい。挿入したshRNA発現カセットを有する完全な増殖性アデノウイルスゲノムは、プラスミドのPacI消化に続いて放出される。このDNAを、例えば、リポフェクタミン試薬を用いてヒト細胞にトランスフェクトする。結果として得られる本発明の組み換え増殖性アデノウイルスを単離し、公知の、標準的な細胞培養法とウイルス学的方法とを用いてウイルスを増殖し精製する。
ホタルルシフェラーゼに対するshRNA分子を発現する増殖制御可能なアデノウイルス(CRAd)を構築するために、E1AのpRb結合性CR2ドメインに24bpの欠失(Suzuki et al., Clin. Cancer Res. 8 (2002): 3348-3359)を有するAd5誘導CRAdであるAd5-Δ24E3を骨格として用いた。実施例4に記載した一般的な方法によると、大腸菌BJ5183株の中で、全長Ad5-Δ24E3ウイルスDNAとKpnI消化pEndK-Ff1-R又はKpnI消化pEndK-Ff1-L(実施例3参照)との相同組み換えを行って、それぞれプラスミドpAdΔ24E3-Ff1-R及びプラスミドpAdΔ24E3-Ff1-Lを得た。これらプラスミドをPacIで消化して、Ff1 shRNA発現カセット挿入物を有する全長アデノウイルスDNAをプラスミド骨格から取り出し、ヒト293細胞にトランスフェクトした(Graham et al., J. Gen. Virol. 36 (1977): 59-74)。Ad5-Δ24E3.Ff1-R CRAd及びAd5-Δ24E3.Ff1-L CRAdを回収し、A549細胞(ATCCより入手)で更に増幅した。最終産物について、E1Δ24欠失、U6-Ff1挿入及びその方向をPCRで確認し、標準的な方法に従って、293細胞を用いた限界希釈プラーク滴定で機能的なPFU力価を求めた。
サイレンシング効率を正確に定量し、実験間の差異を的確に補正するために、我々は二方向hARプロモーター(Barski et al., Biotechniques 36 (2004): 382-4, 386, 388)によってホタルルシフェラーゼ遺伝子とRenillaルシフェラーゼ遺伝子を発現し、そのために、ホタルルシフェラーゼのサイレンシグをRenillaルシフェラーゼの発現に対して正規化することが可能な、レポータープラスミドphAR-FF-RLを使用した。phAR-FF-RLを構築するために、サブクローンしたゲノムDNAをテンプレートとし、両端にSacI部位(下線で示した部位)を作製する以下のプライマーを用いたPCRでhARプロモーター(転写開始部位に対して−124〜+29ヌクレオチド;Genbankアクセッション番号:AF112482)を得た: 5'-CCAGAAGAGCTCGCAACGTGGCATCTGCTA-3'と5'-GTTTGGAGAGCTCCTGGGCACAATGAGGC-3'。PCR産物を、プロモーターの3’末端がホタルルシフェラーゼ遺伝子に向くようにpGL3-basic(ウイスコンシン州、マディソン、Promega製)のSacI部位に挿入し、phAR-FFを作製した。次に、pRL-TK(Promega製)をテンプレートとし、両端にKpnI部位(下線で示した部位)を作製する以下のプライマーを用いたPCRでRenillaルシフェラーゼcDNAを得た:5'-ACAACGGTACCGAACTTAAGCTGCAG-3'と5'-CCGAAAGGTACCACCTGGATCCTTATC-3'。得られたcDNAは、ホタルルシフェラーゼ遺伝子とは逆向き、即ちhARプロモーターの5’末端がRenillaルシフェラーゼ遺伝子の5’末端側を向くように、phAR-FFのKpnI部位に挿入した。
HPV-18 E6に対する、PolIII H1プロモーター駆動性のshRNAを発現する、Ad5-Δ24E3(Suzuki et al., Clin. Cancer Res. 8 (2002): 3348-3359)の誘導体を以下の方法で作製した。初めに、Ad5-Δ24E3直鎖状dsDNAをウイルス粒子から単離し、BJ5183株の中で直鎖状pEndK/Spe(上記参照)と組み換えて、プラスミドクローンpAdΔ24E3を得、これをPacIで消化することで全長AdΔ24E3 DNAを取り出した。次に、HPV-18 E6の19ヌクレオチド(第385番〜第403番ヌクレオチド、番号付けはCole and Danos, J. Mol. Biol. 193 (1987): 599-608に基づく)、それに続く9ヌクレオチドのループリンカー(小文字で示した)及びHPV-18 E6の上記19ヌクレオチド配列の逆相補鎖からなる合成2本鎖オリゴヌクレオチドを、プラスミドpSUPER(Brummelkamp et al., Science 296 (2002): 550-553)に挿入し、pSUPER-18E6を作製した。このために、オリゴヌクレオチドFP_super18E6(5'-gatccccCTAACACTGGGTTATACAAttcaagagaTTGTATAACCCAGTGTTAGtttttggaaa-3')とオリゴヌクレオチドRP_super18E6(5'-agcttttccaaaaaCTAACACTGGGTTATACAAtctcttgaaTTGTATAACCCAGTGTTAGggg-3')のアニーリングを行って、BglII/HindIIIで消化したpSUPERに連結した。pSUPER-18E6をHindIIIで消化し、突出末端をKlenowポリメラーゼで埋めて、NheI部位を作製するために再度連結した。続いて、SpeIとNheIで消化することでH1_18E6断片を取り出し、SpeIで消化したpEndK/SpeIに挿入することでプラスミドpEndK.H1_18E6を作製した。HPV−18で形質転換したヒト癌細胞において、p53活性阻害の低下をもたらすHPV-18 E6の機能的なサイレンシングは、HeLa子宮頸癌細胞にpEndK.H1_18E6をp53特異的レポータープラスミドPG13-Luc(el-Deiry et al., Cell 75 (1993): 817-825)と共にトランスフェクトし、ルシフェラーゼの発現を測定することで確認した。この実験は、pEndK/SpeIをPG13-lucと共に用いた対照トランスフェクションやPG13-Lucのみのトランスフェクションに続いて測定した値よりも顕著に高い値を示した。従って、pEndK.H1_18E6の発現したshRNAは、p53アンタゴニストを沈黙させた。次に、pEndK.H1_18E6をKpnIとEcoRVで消化することで直鎖状にし、大腸菌BJ5183細胞内で、PacIで直鎖状にしたAd5-Δ24E3 DNAと組み換えた。こうすることによってpAdΔ24.H1_18E6が得られた。PacIで直鎖状にしたpAdΔ24.H1_18E6で911細胞(Fallaux et al., Hum. Gene Ther. 7 (1996): 215-222)をトランスフェクトし、AdΔ24.H1_18E6ウイルスを単離して、A549細胞で更に増殖した。

HPV-18 E6に対する、PolIII H1プロモーター駆動性のshRNAを発現し、更にヒトp53を発現する、Ad5-Δ24E3(Suzuki et al., Clin. Cancer Res. 8 (2002): 3348-3359)の誘導体を以下の方法で作製した。初めにプラスミドpEndK/p53を作製した。このために、pBluescript SK(-)をSmaIとEcoRVで消化し、自己連結することで、EcoRV部位を欠失したpBluescript SK(-)(カリフォルニア州、ラホーヤ、Stratagene製)の誘導体を作製した。得られたベクターをKpnIとSalIで消化し、pABS.4-p53(Van Beusechem et al., Cancer Res. 62 (2002): 6165-6171)のSV40プロモーター駆動性ヒトp53発現カセットを含むKpnI/SalI断片を挿入して、pBSK-p53を得た。続いて、pBSK-p53のKpnI部位をSpeI部位に変更した。これは、pBSK-p53をKpnIで消化し、オリゴ5'-TCAGGACTAGTGGAATGTAC-3'とオリゴ5'-ATTCCACTAGTCCTGAGTAC-3'をアニーリングすることで作製した合成2本鎖オリゴヌクレオチドを挿入することで行った。2.6kbのSV40-p53断片をSpeI消化によって取り出し、SpeIで消化したpEndK/Spe(上記参照)に挿入した。SV40-p53カセットがアデノウイルスのL鎖に配置されるような向きに挿入物を有するクローンを単離し、pEndK/p53と命名した。pEndK/p53からの機能的なp53の発現は、p53欠失SaOs−2骨肉種細胞にpEndK/p53をトランスフェクトするか、あるいは対照構築物であるpEndK/SpeIをp53特異的レポータープラスミドPG13-Luc又は負の対照であるプラスミドMG15-Luc(el-Deiry et al., Cell 75 (1993): 817-825)と共にトランスフェクトし、次の日にルシフェラーゼの発現を測定することで確認した。pEndK/p53のトランスフェクションの結果、p53特異的PG−13/MG−15比は63となったが、空のpEndK/SpeIベクターをトランスフェクトした際のPG−13/MG−15比はたった0.7だった。次に、pEndK/p53をClaIで消化し、Klenowポリメラーゼで埋め、pSUPER-18E6(上記参照)のH1-18E6断片を含むHincII/SmaI断片を挿入して、pEndK/p53.H1_18E6を得た。最後に、KpnIとEcoRVによる消化によってpEndK/p53.H1_18E6を直鎖状にし、大腸菌BJ5183細胞内で、PacIで直鎖状にしたAd5-Δ24E3 DNA(上記参照)と組み換え、pAdΔ24.p53(L).H1_18E6を作製した。このベクターをPacIで直鎖状にし、パッケージング用の911細胞にトランスフェクトした。AdΔ24.p53(L).H1_18E6ウイルスを単離し、A549細胞で更に増殖した。
Δ24変異をE1領域(Fueyo et al., Oncogene 19 (2000): 2-12)に有し、bcl-2に対して特異的なそれぞれ異なるshRNAを発現するいくつかのCRAdを、実施例7でplk-1やparcに対して特異的なshRNAを発現するCRAdを作製するために用いた方法で作製した。bcl-2 mRNA上の標的配列に特異的な、下記のオリゴヌクレオチドのセットを使用したことだけが異なる点である。

HPV-18 E6に対する、PolIII H1プロモーター駆動性のshRNAを発現し、更にアデノウイルスの指向性を拡大して、感染力の増強と種々のヒト癌細胞に対する腫瘍退縮力の向上をもたらす修飾fiber遺伝子を発現する、Ad5-Δ24RGD(Suzuki et al., Clin. Cancer Res. 7 (2001): 120-126)の誘導体を以下の方法で作製した。公知の方法で直鎖状の全長2本鎖Ad5-Δ24RGD DNAをAd5-Δ24RGDウイルスから単離し、大腸菌BJ5183細胞内で、 KpnI/EcoRVで消化したpEndK.H1_18E6(上記参照)と組み換え、pAdΔ24RGD.H1_18E6を作製した。続いて、PacIで直鎖状にしたpAdΔ24RGD.H1_18E6をパッケージング用の911細胞にトランスフェクトし、pAdΔ24RGD.H1_18E6ウイルスを単離し、A549細胞で更に増殖した。
内因性アデノウイルスMLPによって発現される導入遺伝子を有する増殖性アデノウイルスを構築するために、部分的なAd5ゲノムを含有するプラスミドのE3領域の代わりに、スプライス受容配列とそれに続く複数のクローニング部位及びポリアデニル化部位を挿入した。このために、合成オリゴヌクレオチドである5'-GGCAGGCGCAATCTTCGCATTTCTTTTTTCCAGGAATCTAGAGATATCGAGCTCAATAAAG-3'と5'-AATTCTTTATTGAGCTCGATATCTCTAGATTCCTGGAAAAAAGAAATGCGAAGATTGCGCCTGCCTGCA-3'をアニーリングせしめ、EcoRIとPstIで消化したpABS.4(カナダ国、トロント、Microbix Biosystems製)にクローニングした。得られたプラスミドpABS.4-SA-MCSは、Fiber遺伝子の長いスプライス受容部位、XbaI、EcoRVとSacIの制限部位を含む小さなクローニング部位およびポリアデニル化部位を包含する、長さが32ヌクレオチドのアデノウイルス セロタイプ40の配列を含有し、所望の導入遺伝子を挿入できるような柔軟性を持たせて設計してある。簡単且つ定量的な方法で測定することが可能な、内因性アデノウイルスMLPによって発現される導入遺伝子を有する増殖性アデノウイルスを構築するために、ホタルルシフェラーゼ遺伝子をpABS.4-SA-MCSのMCSに挿入した。このために、pSP-Luc+ベクター(Promega製)をテンプレートDNAとして用い、突出したXbaI部位とSacI部位を有するオリゴヌクレオチドである5'-GGGTCTAGAGCCACCATGGAAGACGCCAAAAAC-3'と5'-CCCGAGCTCCTTACACGGCGATCTTTCCGC-3'をプライマーとして用いたPCRによって、ホタルルシフェラーゼのcDNAを得た。PCR産物をXbaIとSacIで消化し、同じ酵素で消化したpABS.4-SA-MCSに連結して、pABS.4-SA-Lucを得た。このプラスミドをPacIで消化し、SA-Lucとカナマイシン耐性遺伝子を含む断片をPacIで消化したpBHG11(Microbix biosystems製)に挿入した。アデノウイルスのR鎖にSA-Lucが配置されるような向きに挿入物を有するクローンを単離し、次にカナマイシン耐性遺伝子を取り除くために、SwaIで消化し、自己連結させて、pBHG11-SA-Lucを得た。
アデノウイルスの複製に関連した導入遺伝子の発現を研究するために、96穴プレートに植えたA549細胞を、細胞あたり20PFUの、ルシフェラーゼを発現する組み換えアデノウイルスに37℃で2時間感染させ、続いて感染培地を、細胞周期阻害剤であるアピゲニン(apigenin)を含有するか含有しない新鮮培地と交換した。アデノウイルスの複製には細胞周期の進行がS期を経ることが必要なため、アピゲニン処理はアデノウイルスの複製を阻害する。これは、Ad5-Δ24E3(上記参考)による感染から32時間後に、75μMのアペニゲンの存在下で培養した細胞の含有するAd5-Δ24E3ウイルスゲノムを定量的PCRで測定したところ、その量が1,000分の1〜10,000分の1にまで減っていたことによって確認された。ルシフェラーゼ活性は、種々の濃度のアペゲニンの存在下又は非存在下で培養したアデノウイルス感染細胞について、感染から32時間後にルシフェラーゼ化学発光アッセイシステム(luciferase chemiluminescence assay system)(Promega製)を用いて行った。図2は、Ad5Δ24-SA-LucとAd5Δ24-CMV-Luc(上記参照)を用いて得られた結果と、非増殖性の対照ウイルスであるAd5-ΔE1-CMV-Lucを用いて得られた結果を示す。Ad5-ΔE1-CMV-Lucは、E1領域とE3領域に欠失を有し、pCEP4(Invitrogen製)由来のCMVプロモーターとpGL3-Basic(Promega製)由来のルシフェラーゼ遺伝子(Yamamoto et al., Mol. Ther. 3 (2001): 385-394;アラバマ州、バーミンガム、アラバマ大学のM. Yamamoto博士による寄贈)からなる発現カセットをE1領域に有する。非増殖性アデノウイルスベクターAd5-ΔEl-CMV-Lucによるルシフェラーゼの発現は、アピゲニン処理による影響を受けず、予想したとおり、導入遺伝子の発現はアデノウイルスの複製に依存していなかった。増殖性アデノウイルスベクターAd5Δ24-CMV-Lucによるルシフェラーゼの発現は、アピゲニン処理によって約35分の1に減少し、導入遺伝子の発現は、アデノウイルスの複製に部分的に依存することを示した。増殖性アデノウイルスベクターAd5Δ24-SA-Lucによるルシフェラーゼの発現は、アピゲニン処理によって約4,600分の1に減少し、このウイルスにおけるMLP駆動性の導入遺伝子発現は、アデノウイルスの複製に強く依存していることを示した。従って、Ad5Δ24-SA-Lucは、第1の種類の細胞におけるアデノウイルスの複製を第2の種類の細胞におけるアデノウイルスの複製と比較する工程を含む本発明の方法に有用である。Ad5Δ24-SA-Lucに感染した細胞におけるルシフェラーゼ活性は、この細胞におけるAd5Δ24-SA-Lucの複製と直接関連し、簡単なアッセイ(下記参照)によって測定することができる。
RNAiを用いて標的細胞の細胞遺伝子を沈黙させ、これら細胞を増殖性ウイルスに感染させ、そして複製及び/又は細胞溶解に対するサイレンシングの効果を測定することで、アデノウイルスの複製又は細胞溶解に対する阻害因子のみならず、このような阻害因子の発現を低下させうるサイレンシング因子を同定することができる。従来技術の項で説明したように、合成siRNA又はshRNAをコードするプラスミドの形態で、大規模なサイレンシング因子ライブラリーが既に入手可能である。ライブラリーの個々のメンバーについて大規模のトランスフェクションを行うための方法も既に存在し(Berns et al., Nature 428 (2004): 431-437; Paddison et al., Nature 428 (2004): 427-431)、このような方法は、公知の方法に基づいた増殖性ウイルスによる感染と組み合わせることも容易である。このようなライブラリーを用いた阻害因子とサイレンシング因子の同定を成功させるためには、ウイルスの複製及び/又は細胞溶解の測定に簡便且つ定量的なアッセイを用いることが好ましく、スクリーニングの工程を自動化できるような機械化プラットフォーム(robotic platform)と互換であることが好ましい。アデノウイルスの複製を測定するために、我々は、マーカー遺伝子であるルシフェラーゼを主要後期プロモーターの制御下で発現する、増殖可能なAd5Δ24-SA-Lucウイルスを開発した(実施例12を参照)。このウイルスのゲノムからのルシフェラーゼ発現はウイルスの複製に依存するので、このウイルスに感染した細胞によるルシフェラーゼの発現は、ウイルスの複製に対して感受性の高いマーカーとして用いることができる。ウイルスに感染した細胞の溶解を測定するための方法としては、損傷した細胞の細胞質から培養上清に放出された乳酸デヒドロゲナーゼの活性の測定に基づく、細胞死と細胞溶解を定量するための比色定量的、蛍光定量的又は化学発光的なアッセイが既に市販されている(Roche Applied Science; Cambrex; Promega)。
CRAdであるAdΔ24とAdΔ24-p53(Van Beusechem et al, Cancer Res. 62 (2002): 6165-6171)及びAdΔ24.p53(L).H1_18E6(実施例8を参照)を、HPV−18陽性のHeLa子宮頸癌細胞(ATCCより入手)を感染させるのに用いた。感染は、リポフェクタミンプラス(LipofectAMINE Plus)(Invitrogen製)を製造者のプロトコルに従って使用して、200ngのPG13-Lucプラスミド(el-Deiry et al, Cell 75 (1993): 817-825)をHeLa細胞にトランスフェクトした24時間後に、10PFU/細胞のCRAdを用いて行った。PG13-Lucはp53依存性プロモーターによって駆動されるホタルルシフェラーゼ遺伝子を発現する。感染から72時間後には細胞をレポーター溶解バッファー(Reporter Lysis Buffer)(Promega製)の中で溶解し、ルシフェラーゼ化学発光アッセイシステム(Promega製)及びLumat LB 9507発光計測器(Lumat LB 9507 luminometer)を用いて化学発光を測定した。測定した相対光学単位は擬似感染対照に基づいて正規化した。図3に示したように、機能的なp53の発現は、AdΔ24感染細胞と比べてAdΔ24-p53感染細胞でほんのわずかばかり上昇したにすぎず、これはHeLa細胞において、p53はHPV-18 E6タンパク質によって効果的に阻害されていることを示した。AdΔ24.p53(L).H1_18E6感染細胞においては、相当に高いレベルのp53活性が測定され、AdΔ24.p53(L).H1_18E6内のHPV-18 E6特異的shRNAは、HeLa細胞におけるp53阻害を抑制したことを示した。

Claims (25)
- 標的細胞内で複製可能であり且つ細胞溶解能を示す増殖性ウイルスであって、該ウイルスは、標的細胞内の標的遺伝子の発現を抑制するサイレンシング因子をコードする少なくとも1種のDNA配列をそのゲノム内に包含し、該DNA配列は、該標的細胞内で機能する少なくとも1種の発現制御配列に発現可能な状態で連結していることを特徴とする増殖性ウイルス。
- サイレンシング因子が2本鎖RNA分子を含むことを特徴とする、請求項1に記載の増殖性ウイルス。
- 2本鎖RNA分子の各RNA鎖の長さが、19ヌクレオチド以上で30ヌクレオチド未満であることを特徴とする、請求項2に記載の増殖性ウイルス。
- 該RNA分子がヘアピンRNAを含むことを特徴とする、請求項1〜3のいずれかに記載の増殖性ウイルス。
