分光蛍光計
蛍光信号により作成されるピークは典型的には幅広くて、励起および/または発光の波長を正確に選択する能力は、使用する機器および蛍光検出システム、または読取り装置の検定に大きく依存している。本明細書において説明される実験のためには、単一のウエル読取り装置を持つSpex FluoroMax分光蛍光計(認識番号2093、Spex Industries, New Jersey)が0.1nmの通過帯域をもって使用された。或る場合には、Molecular Devices FLEXstationプレート読取り装置(認識番号FX 01090、California)を使用したが、それは10nmの通過帯域を有していて、それ故にSPEX Fluoromax走査装置のように正確には蛍光信号を検出できない。
以下の試薬が、本明細書において記載された実験で使用された。
ADP(A−2754 Sigma Ultra、ロット番号073k7007、アデノシン5'−二リン酸ナトリウム塩);
ATP(A−7699 Sigma Ultra、ロット番号053k7042、アデノシン5'−三リン酸二ナトリウム塩);
EGTA(E0396 Sigma Ultra、エチレングリコール−ビス(2−アミノエチルエーテル)−N,N,N',N'−四酢酸);
EDTA Sigma−Aldrich(E2−628−2、エチレンジアミン四酢酸);
HEPES(J848、AMRESCO、4−(2−ヒドロキシエチル)ピペラジン−1−エタンスルホン酸);
CaCl2(C−4901、Sigma、脱水塩化カルシウム);および
MgCl2(M8266、Sigma、無水塩化マグネシウム)

10mMのHEPES pH8.0緩衝液中の5mMのADPまたはATPのいずれかを含有する溶液を、SPEX Fluoromax蛍光走査装置を用いて400〜600nmの範囲内で蛍光発光について分析した。この実験のために使用された励起波長は405nmであった。この実験によれば、約500nmにおいて測定されたADPおよびATPの間には弱い蛍光発光の相違があることが示されている。
50mMのEDTAの存在下もしくは不在下において、10mMのHEPES pH8.0緩衝液中の10mMのADPまたは10mMのATPを含有する溶液について、SPEX Fluoromaxを用いて蛍光走査を実施した。図2に示されるように、ADPを含有する溶液の蛍光スペクトルは、EDTAの存在下において大きく強化されている。これとは対照的に、ATPはEDTAの存在下において蛍光の増加を示していない。同様に、図3に示されるように、EGTAの存在下においてADP蛍光の増加があるが、これはATP蛍光には見られない。475nmで見られるピークは、バックグラウンドスペクトルの部分である。
50mMのEDTAの溶液を、10mMのHEPES pH8.0緩衝液中、ADPで滴定した。Molecular Devices FlexStationプレート読取り装置で500nmにおける蛍光を読み取った。図4に示されるように、EDTA溶液のADPでの滴定の結果得られるデータの点は、二ログ単位にわたり直線となる。100μMの濃度以下のADPは、使用された手法の検出限界を超えていた。
この実験では、10mMのHEPES pH8.0緩衝液中に1mMのADPおよび30mMのEDTAを含有する溶液へのCa2+およびMg2+の添加の効果を検査した。図5に示されるように、EDTAの存在下においてADPにより作成される蛍光発光信号は、30mMのMg2+またはCa2+の添加により逆転することが判明し、このことはMg2+およびCa2+がADPのEDTAとの相互作用と競合することを示している。蛍光は500nmにおいて監視された。
ATPはEDTAの存在下において蛍光発光するようには見えないが、ADPは蛍光発光するので、EDTA−ADPで誘発される蛍光に対するATPの存在の効果を検査した。10mMのHEPES pH8.0緩衝液200μl中に5mMのADPおよび10mMのEDTAを含有する溶液に、増加する濃度のATPを加えた。410nmの励起波長を用いて500nmにおいて、蛍光を読み取った。試料をFlexStation(Molecular Devices)プレート読取り装置の上で読み取った。図6に示されるように、10mMのATPは、ADPおよびEDTAの間の相互作用を完全に壊している。この観察により、ATPがEDTAと直接に相互作用することができることを示唆している;しかしながら、ATPは、これらの条件下では蛍光の増加を引き起こさない。
この実験では、10mMのHEPES pH8.0緩衝液中、種々の濃度におけるADPおよびATPの一連の溶液を50mMのEDTAで処理した。HEPES−EDTAを対照として使用した。図7は、対照の蛍光読取りをそれぞれのデータ点から差し引いたデータを示している。
ATP、ADP、グアニジン二リン酸(GDP)、およびグアニジン三リン酸(GTP)の各5mM溶液の蛍光スペクトルを、50mMのEDTAの不在下および存在下において測定した。蛍光発光信号を450〜600nmの間で解析した。図8に示されるように、EDTAの存在下におけるADPのみが、強化された蛍光を示しており、490〜500nmの間で蛍光のピークを有する。
ランタニド金属、キレート剤、およびヌクレオチドの間で形成される三次複合体の使用を確立して、キレート剤の存在下においてヌクレオチドにより作成される蛍光信号を変更するために、以下の実験を行う。
Claims (23)
- (a)溶液にキレート剤を加える工程、および(b)キレート剤の添加時に作成されるか、または変更される信号を分光検出システムを用いて検出する工程を含んで成る、溶液中のヌクレオチドを検出する方法。
- ヌクレオチドがアデノシン二リン酸である、請求項1に記載の方法。
- 分光検出システムが蛍光に基づくシステムである、請求項1または2に記載の方法。
- キレート剤がポリカルボキシレート分子である、請求項1、2又は3のいずれか1項に記載の方法。
- キレート剤がEDTA、EGTA、またはそれらの組合せである、請求項4に記載の方法。
- (a)溶液にキレート剤を加える工程、および(b)キレート剤の添加時に作成されるか、または変更される信号を分光検出システムを用いて検出する工程を含んで成る、少なくとも二つのヌクレオチドの混合物の溶液中における一つのヌクレオチドを示差的に検出する方法。
- ヌクレオチドがアデノシン二リン酸である、請求項6に記載の方法。
- アデノシン三リン酸が溶液中にある、請求項6または7に記載の方法。
- キレート剤がポリカルボキシレート分子である、請求項6、7または8のいずれか1項に記載の方法。
- キレート剤がEDTA、EGTA、またはそれらの組合せである、請求項9に記載の方法。

- 蛍光に基づく信号が、450および550nmの間で発生する、請求項13に記載の方法。
- 1またはそれ以上のキレート剤を溶液に加える前のアデノシン二リン酸の蛍光に基づく信号と比較して、信号の強度が増加する、請求項7〜14のいずれか1項に記載の方法。
- キレート剤の添加後に信号の強度が増加する、請求項1または15のいずれか1項に記載の方法。
- キレート剤の添加後に信号のピーク強度が移動する、請求項1または15のいずれか1項に記載の方法。
- キレート剤の添加後に信号の強度が減少する、請求項1または15のいずれか1項に記載の方法。
- (a)適当な量のキレート剤、および(b)請求項1〜18のいずれか1項に記載の方法のいずれかを実施するための指示書を含むキット。
- 工程(b)に先立って、溶液へのランタニドの添加をさらに含む、請求項1〜18のいずれか1項に記載の方法。
