【解決手段】網膜及びそれに付着した硝子体を含み、かつ網膜から硝子体中に伸展している管状新生血管を有する、培養組織、並びに上記培養組織を含む、血管新生、特に増殖性糖尿病網膜症、網膜静脈閉塞症、加齢黄斑変性症、糖尿病黄斑浮腫及び癌における血管新生の評価のモデル系;血管形成誘導作用を有する因子の存在下で、硝子体が付着した摘出網膜を培養することを含む、管状新生血管を有する網膜培養物の製造方法;血管新生を抑制する物質のスクリーニング方法;血管の様式を調節する物質のスクリーニング方法など。

(1)in vitroの系
本発明者らが開発した系は、網膜環境下での血管の動態を評価可能である。従って、この系は、網膜環境下にないin vitroの実験系に比し、病態をより再現できるという点で優れている。
(2)未熟児網膜症モデル
本発明者らが開発した系は、一旦成熟し静止期に入った網膜血管からの血管新生の観察が可能である。従って、この系は、このような血管新生と網膜血管発生とを区別できない未熟児網膜症モデルに比し、成体網膜における血管新生をより純粋に評価可能であるという点で優れている。
(3)糖尿病モデル
本発明者らの開発した系は、網膜を器官培養することで、糖尿病網膜症の初期変化である内皮・周皮連関の破綻を起こすことができ、その上でVEGFを投与することで網膜血管新生を誘導することができる。従って、この系は、網膜血管障害を再現できるが血管新生を起こさない糖尿病モデルに比し、網膜における血管新生の誘導に成功したという点で優れている。
(4)VEGF硝子体投与
本発明者らの開発した系は、網膜を器官培養することで、糖尿病網膜症の初期変化である内皮・周皮連関の破綻を起こすことができ、その上でVEGF等の血管形成誘導因子を投与することで網膜血管新生を誘導することができる。従って、この系は、内皮周皮連関のしっかりした正常網膜へのVEGF硝子体投与に比し、網膜における血管新生の誘導に成功したという点で優れている。
(5)in vivoの系
本発明者らの開発した系は、血液網膜柵による薬物送達の問題が解消されている。従って、この系は、網膜への薬物送達が血液網膜柵のために非常に困難となり得るin vivoの系に比し、網膜への薬物送達、ひいては薬物のスクリーニングが容易であるという点で優れている。
(6)正常な成体網膜からの新生血管
本発明者らの開発した系は、網膜の切断部位からの血管内皮細胞の進展ではなく、網膜の非切断部位(網膜の正常部位)からの血管新生が観察可能である。また、新生血管の硝子体中への伸展が認められる。従って、本発明者らの開発した系は、上述した従来の系に比し、増殖性糖尿病網膜症の病態をより反映し得るという点で優れている。
以上のとおり、本発明者らは、従来の系の問題点を克服した増殖性糖尿病網膜症モデルを開発することに成功し、以って本発明を完成するに至った。
〔1〕網膜及びそれに付着した硝子体を含み、かつ網膜から硝子体中に伸展している管状新生血管を有する、培養組織。
〔2〕房状新生血管を有しない、上記〔1〕の組織。
〔3〕管状新生血管が、一旦正常に成熟した網膜および網膜血管から発生している、上記〔1〕の組織。
〔4〕管状新生血管が網膜の非切断部分から発生している、上記〔1〕の組織。
〔5〕網膜及びそれに付着した硝子体を含み、かつ網膜から硝子体中に伸展している管状新生血管を有する培養組織を含む、血管新生の評価のモデル系。
〔6〕血管新生の評価のモデル系が、増殖性糖尿病網膜症、網膜静脈閉塞症、加齢黄斑変性症、糖尿病黄斑浮腫及び癌における血管新生の評価のモデル系である、上記〔5〕の系。
〔7〕血管形成誘導作用を有する因子の存在下で、硝子体が付着した摘出網膜を培養することを含む、管状新生血管を有する網膜培養物の製造方法。
〔8〕血管形成誘導作用を有する因子がVEGF又はPlGFである、上記〔7〕の方法。
〔9〕硝子体が付着した摘出網膜の培養が、網膜部分を培養膜上に固定した条件下でおこなわれる、上記〔7〕の方法。
〔10〕硝子体が付着した摘出網膜の培養が、該摘出網膜の培養膜への非固定部分を培養ゲルに包んだ条件下でおこなわれる、上記〔9〕の方法。
〔11〕培養膜上に固定され、かつ培養ゲルで包まれた摘出網膜を培地中に浸しつつ、該網膜を培養することを含む、網膜培養物の製造方法。
〔12〕血管形成誘導作用を有する因子の存在下で、該網膜を培養することを含む、管状新生血管を有する網膜培養物の製造方法である、上記〔11〕の方法。
〔13〕血管形成誘導作用を有する因子の不在下で、該網膜を培養することを含む、血管が退縮した網膜培養物の製造方法である、上記〔11〕の方法。
〔14〕以下の工程(a)〜(c)を含む、血管新生を抑制する物質のスクリーニング方法:
(a)硝子体が付着した摘出網膜を、血管形成誘導作用を有する因子及び被験物の存在下で培養する工程;
(b)上記(a)で培養された該網膜における血管新生の程度を測定し、該程度を、該因子の存在下及び該被験物の不在下で培養された該網膜における血管新生の程度と比較する工程;ならびに
(c)上記(b)の比較結果に基づいて、血管新生を抑制する被験物を選択する工程。
〔15〕血管新生を抑制する物質が、増殖性糖尿病網膜症、網膜静脈閉塞症、加齢黄斑変性症、糖尿病黄斑浮腫及び癌を予防・治療し得る物質である、上記〔14〕の方法。
〔16〕以下の工程(a)〜(c)を含む、血管の様式を調節する物質のスクリーニング方法:
(a)培養膜上に固定され、かつ培養ゲルで包まれた摘出網膜を培地中に浸しつつ、該網膜を被験物の存在下で培養する工程;
(b)上記(a)で培養された該網膜における血管の様式を測定し、該様式を、該被験物の不在下で培養された該網膜における血管の様式と比較する工程;ならびに

〔17〕工程(a)において血管形成誘導作用を有する因子の存在下で、該網膜を培養することを含み、かつ測定される血管の様式が血管新生の程度である、血管新生を抑制する物質のスクリーニング方法である、上記〔16〕の方法。
〔18〕工程(a)において血管形成誘導作用を有する因子の不在下で、該網膜を培養することを含み、かつ測定される血管の様式が血管退縮の程度である、血管退縮を抑制する物質のスクリーニング方法である、上記〔16〕の方法。
〔19〕血管退縮を抑制する物質が、前増殖性糖尿病網膜症を予防・治療し得る物質である、上記〔18〕の方法。
本発明の製造方法は、上記の通り有用な網膜培養物の製造に有用である。
本発明のスクリーニング方法は、例えば、網膜及び/又は硝子体における血管新生を伴う疾患、並びに網膜における血管退縮を伴う疾患の予防・治療に有用な薬物の開発に有用である。
(a1)硝子体が付着した摘出網膜を、血管形成誘導作用を有する因子及び被験物の存在下で培養する工程;
(b1)上記(a1)で培養された該網膜における血管新生の程度を測定し、該程度を、該因子の存在下及び該被験物の不在下で培養された該網膜における血管新生の程度と比較する工程;ならびに
(c1)上記(b1)の比較結果に基づいて、血管新生を抑制する被験物を、網膜及び/又は硝子体における血管新生を伴う疾患を予防・治療し得る物質として選択する工程。
(a2)培養膜上に固定され、かつ培養ゲルで包まれた摘出網膜を培地中に浸しつつ、該網膜を被験物の存在下で培養する工程;
(b2)上記(a2)で培養された該網膜における血管の様式を測定し、該様式を、該被験物の不在下で培養された該網膜における血管の様式と比較する工程;ならびに
(c2)上記(b2)の比較結果に基づいて、血管の様式を調節する被験物を選択する工程。
網膜の器官培養は、以下のプロトコールに従って行った(図1)。
1)眼球を摘出する。
2)強膜・角膜と脈絡膜を取り外し、網膜を水晶体と一緒に取り出す。
3)、4)網膜を5ヶ所切開入れて、硝子体ごと網膜を平面にする(flat mountにする)。
5)網膜に硝子体をつけたまま、1mlのメディウムを満たしたMillicell-CM(#PICM ORG 50, Millipore)のメンブレン(素材:PTFE)に乗せる。
6)コラーゲンタイプIを用いて、網膜、硝子体をメンブレンに固定する。
7)1mlのメディウムを追加し、コラーゲンゲルで包んだ網膜をメディウムに完全につける。
8)培養の条件は、37℃、5%CO2であり、培地交換は24時間毎とする。
9)96時間の培養後、メンブレンに固定されたまま、4%PFAで網膜サンプルを固定して、免疫染色する。
なお、網膜の器官培養用培地としては、MEM−Hepes 50ml、ハンクス平衡化塩溶液25ml、熱非働化ウマ血清25ml、5.75mg/mlグルコース、25U/mlペニシリン、25μg/mlストレプトマイシン、200μM L−グルタミンを用いた。
マウス(C57BL6J, 雄、7−8週齢)の網膜を、4日間器官培養し、その培養液にVEGF(ヒト組換えVEGF 165 (R & D systems))を添加した。4日後、網膜を固定してPECAM(血管内皮細胞のマーカー。これを利用することにより、正常血管と病的新生血管の両方を染色可能)、コラーゲンタイプ4(正常血管のみ染色可能)で二重染色した。
その結果、VEGFを添加して培養を4日間行ったところ、PECAMのみで染色される網膜新生血管が生じた(図2A)。一方、VEGFを添加せずに培養を4日間行ったところ、成体マウスの正常網膜血管におけるPECAMとコラーゲンタイプ4の発現様式は完全に一致していた(図2B)。また、VEGFを添加せずに培養を4日間行ったところ、硝子体に浸潤する血管内皮細胞は存在しなかった(図2C)。一方、VEGFを添加して培養を4日間行ったところ、硝子体に血管内皮細胞が浸潤した(図2D)。
以上より、VEGFの存在下における網膜器官培養により観察された網膜新生血管が糖尿病網膜症のものと非常に類似することが示された。
次に、VEGFの存在下での培養により生じた網膜新生血管を評価するため、1個の網膜あたりにおける、PECAMで染色されるが、コラーゲンタイプ4で染色されない網膜新生血管を計数した。
その結果、濃度依存性試験では、VEGF濃度が25ng/mlのときに新生血管数が最大であった(図3A)。また、経時変化を観察したところ、新生血管は3日から観察され、その後単調に増加した(図3B)。なお、5日では、各新生血管が融合し計数が困難となったため、以下の実験では、4日の時点で新生血管を計数することとした。
次いで、他の増殖因子についてもVEGFと同様に検討した。増殖因子としては、近年その血管新生作用が注目されているPlGF、並びにin vitroにおいて網膜新生血管を発生させることが知られているbFGF及びIGF−1を用いた。PlGFとしてはヒトPlGF2を用いた。
その結果、PlGFの存在下で培養をおこなったところ、PECAMのみで染色される網膜新生血管が生じたが、その形態はVEGFのそれよりもはるかに細かった(図4A)。また、濃度依存性試験では、PlGF濃度が50ng/mlで新生血管数が最大となり(図4B)、VEGF 25ng/mlのときと同程度の数の新生血管を生じた。一方、bFGF、IGF−1では、血管新生の程度は低かった(図5A、B)。
以上より、VEGF、PlGFが、bFGF、IGF−1に比し網膜血管新生作用が優れることが明らかとなった。また、本発明者らが開発したこの系を用いることにより、網膜血管作用を有する新たな因子のスクリーニング、及びin vitroスクリーニングよりも精度の高いスクリーニングなどが可能になると考えられる。
従来の網膜新生血管モデルである未熟児網膜症モデルは、もともと活動性の高い生理的網膜血管発生の要素と、虚血網膜症による病的新生血管の要素を併有する。一方、本発明者らが開発した系では、糖尿病性網膜症の網膜新生血管の特徴(即ち、正常に成熟し一旦静止期に入った網膜血管からの病的な血管新生)が認められる。in vivoではVEGFを投与しても成体マウスの網膜から新生血管が生じないこと、並びに正常な成体における網膜では内皮・周皮連関がしっかりしており新生血管を生じにくいことは以前から知られている。本発明者らは、網膜を器官培養することで、糖尿病網膜症の初期変化である内皮・周皮連関の破綻を起こしたうえでVEGFを投与することによって網膜血管新生を引き起こした。本発明者らが開発した方法は、従来のVEGF硝子体投与よりも糖尿病網膜症をより模倣するし、糖尿病モデルではできなかった網膜血管新生を再現したという点で、これらの方法をはるかに陵駕している。従って、本発明者らが開発した系は、糖尿病網膜症モデルとして非常に優れることが示された。
次に、本発明者らの開発した系において薬物の評価が実際に可能か検討した。詳細には、増殖因子による刺激下で活性化され、血管新生作用に関わると考えられているチロシンキナーゼ、PKCの阻害剤であるゲニステイン(20μM)、GFX(5μM)、又はDMSO(コントロール)を負荷した状態で、VEGF(25ng/ml)を添加し、網膜の器官培養を行った。
その結果、コントロールにおいて生じた網膜新生血管が、両阻害剤では減少した(図6A)。また、それらの新生血管を計数すると、ゲニステイン、GFXは、血管新生を有意に抑制していた(図6B)。
以上より、本発明者らの開発した系において薬物の評価が実際に可能であることが明らかとなり、また、PKC阻害剤が網膜において抗血管新生作用を有することが示唆された。
次に、本発明者らの系において、網膜及び硝子体における新生血管の発生過程を経時的に詳細に観察するために、VEGF(25ng/ml)を添加し、網膜を3.5日培養した後、timelapse観察(15分ごとに撮影)を行った。
その結果、網膜及び硝子体における新生血管の発生過程が経時的に観察された(図7)。
以上より、本発明者らの系により、新生血管の様々なステージを詳細に検討することが可能になることが示された。
次いで、VEGF(25ng/ml)にbFGF又はIGF−1(各50ng/ml)を加えた培地で、網膜の器官培養を上記と同様に行った。
その結果、VEGF(25ng/ml)単独のものよりも太い網膜新生血管が形成され、また、静脈の拡張も増強されていた。
以上より、本培養系において、bFGF、IGF−1は血管新生を増強する作用を持っていることが示唆された。
次に、増殖因子の不在下において本発明者らの系を用いることにより、培養された網膜血管の状態を検討した。なお、網膜の器官培養は、増殖因子を用いないこと以外は、上記と同様にして行った。
その結果、内皮細胞については、器官培養を開始した時点と比較して、4日間培養すると網膜毛細血管が減少し、網膜細静脈は狭小化していた(内皮細胞に関する知見)。一方、周皮細胞(Desmin陽性細胞、SMA陽性細胞)は、開始時点では内皮細胞と密に接触していたが、4日間の培養後、内皮細胞から離れた周皮細胞が多く見られるようになった。周皮細胞は、元来、内皮細胞の生存を維持し、細胞分裂を抑制すると云われているため、内皮・周皮連関の破綻が内皮細胞の退縮に寄与している可能性は非常に高いと考えられる。
本発明の製造方法は、上記の通り有用な網膜培養物の製造を可能とする。
本発明のスクリーニング方法は、例えば、網膜及び/又は硝子体における血管新生を伴う疾患、並びに網膜における血管退縮を伴う疾患の予防・治療に有用な薬物の開発を可能とする。
Claims (19)
- 網膜及びそれに付着した硝子体を含み、かつ網膜から硝子体中に伸展している管状新生血管を有する、培養組織。
- 房状新生血管を有しない、請求項1記載の組織。
- 管状新生血管が、一旦正常に成熟した網膜および網膜血管から発生している、請求項1記載の組織。
- 管状新生血管が網膜の非切断部分から発生している、請求項1記載の組織。
- 網膜及びそれに付着した硝子体を含み、かつ網膜から硝子体中に伸展している管状新生血管を有する培養組織を含む、血管新生の評価のモデル系。
- 血管新生の評価のモデル系が、増殖性糖尿病網膜症、網膜静脈閉塞症、加齢黄斑変性症、糖尿病黄斑浮腫及び癌における血管新生の評価のモデル系である、請求項5記載の系。
- 血管形成誘導作用を有する因子の存在下で、硝子体が付着した摘出網膜を培養することを含む、管状新生血管を有する網膜培養物の製造方法。
- 血管形成誘導作用を有する因子がVEGF又はPlGFである、請求項7記載の方法。
- 硝子体が付着した摘出網膜の培養が、網膜部分を培養膜上に固定した条件下でおこなわれる、請求項7記載の方法。
- 硝子体が付着した摘出網膜の培養が、該摘出網膜の培養膜への非固定部分を培養ゲルに包んだ条件下でおこなわれる、請求項9記載の方法。
- 培養膜上に固定され、かつ培養ゲルで包まれた摘出網膜を培地中に浸しつつ、該網膜を培養することを含む、網膜培養物の製造方法。
- 血管形成誘導作用を有する因子の存在下で、該網膜を培養することを含む、管状新生血管を有する網膜培養物の製造方法である、請求項11記載の方法。
- 血管形成誘導作用を有する因子の不在下で、該網膜を培養することを含む、血管が退縮した網膜培養物の製造方法である、請求項11記載の方法。
- 以下の工程(a)〜(c)を含む、血管新生を抑制する物質のスクリーニング方法:
(a)硝子体が付着した摘出網膜を、血管形成誘導作用を有する因子及び被験物の存在下で培養する工程;
(b)上記(a)で培養された該網膜における血管新生の程度を測定し、該程度を、該因子の存在下及び該被験物の不在下で培養された該網膜における血管新生の程度と比較する工程;ならびに
(c)上記(b)の比較結果に基づいて、血管新生を抑制する被験物を選択する工程。 - 血管新生を抑制する物質が、増殖性糖尿病網膜症、網膜静脈閉塞症、加齢黄斑変性症、糖尿病黄斑浮腫及び癌を予防・治療し得る物質である、請求項14記載の方法。
- 以下の工程(a)〜(c)を含む、血管の様式を調節する物質のスクリーニング方法:
(a)培養膜上に固定され、かつ培養ゲルで包まれた摘出網膜を培地中に浸しつつ、該網膜を被験物の存在下で培養する工程;
(b)上記(a)で培養された該網膜における血管の様式を測定し、該様式を、該被験物の不在下で培養された該網膜における血管の様式と比較する工程;ならびに
(c)上記(b)の比較結果に基づいて、血管の様式を調節する被験物を選択する工程。 - 工程(a)において血管形成誘導作用を有する因子の存在下で、該網膜を培養することを含み、かつ測定される血管の様式が血管新生の程度である、血管新生を抑制する物質のスクリーニング方法である、請求項16記載の方法。
- 工程(a)において血管形成誘導作用を有する因子の不在下で、該網膜を培養することを含み、かつ測定される血管の様式が血管退縮の程度である、血管退縮を抑制する物質のスクリーニング方法である、請求項16記載の方法。
- 血管退縮を抑制する物質が、前増殖性糖尿病網膜症を予防・治療し得る物質である、請求項18記載の方法。
