Abstract
Description
The present invention eliminates the dryness and discomfort of the eye part of the contact lens wearer by stabilizing the tear film around the lens (lens front and back) when wearing the contact lens. The present invention relates to a tear film stabilization system that provides a moist feeling and a feeling of wearing.
In recent years, contact lenses have been widely used, and not only hard contact lenses and soft contact lenses but also disposable contact lenses are widely used. These contact lenses are classified into ionic and non-ionic lenses. As materials for ionic contact lenses, hydroxyethyl methacrylate, methyl methacrylate, methacrylic acid, etc. are polymerized (or copolymerized). The resulting polymer (or copolymer) is used.
When wearing contact lenses, the tear film present on the lens surface of the contact lens wearer and the tear film present between the lens back surface and the eyeball surface are extremely thin and unstable. The tear fluid on the surface of the eyeball of the person disappears earlier than that of the person who does not wear the contact lens. For this reason, the contact lens wearer often lacks tear fluid on the surface of the eyeball for a short time before blinking, and the wearing feeling of the contact lens deteriorates (dryness and discomfort in the eye), Furthermore, chronic shortage of tears on the eyeball surface of contact lens wearers can cause serious damage to the external eye such as the conjunctiva and cornea.
An extremely thin tear film exists on the lens surface and the lens back surface during wearing of the contact lens, and these tear films serve as a cushion for the eyeball and eyelid of the contact lens wearer. Therefore, in order to improve the wearing feeling of the contact lens and to prevent the external eye such as the conjunctiva and cornea from being damaged, the tear film on the surface of the eyeball of the contact lens wearer is stably held. A system is needed.
The inventors of the present invention pay attention to ionic contact lenses that are widely used, and the adsorption of various polymer compounds to ionic contact lenses and the moist feeling of the eyes of ionic contact lens wearers. As a result of diligent research on the relationship with wearing feeling, polyvinylpyrrolidone is specifically adsorbed on ionic contact lenses and has the effect of removing dryness and discomfort in the eye part of contact lens wearers, and is applied as an ophthalmic solution I found out that it is excellent in the feeling of comfort. Furthermore, if an ophthalmic composition containing polyvinylpyrrolidone and a thickener is used, the water retention of the ionic contact lens is improved. As a result, the above-described improvement effect of the eye part of the ionic contact lens wearer can be improved for a long time. I found that it was sustainable.
That is, the present invention comprises an ophthalmic composition containing polyvinylpyrrolidone, preferably an ophthalmic composition containing 0.05 to 3.0% by weight of polyvinylpyrrolidone having an average molecular weight of 500,000 or less, and An ophthalmic composition containing polyvinylpyrrolidone, which is a system that stabilizes a tear film around the ionic contact lens (lens surface and lens back surface) by adsorbing polyvinylpyrrolidone to the ionic contact lens. By using the object, not only the moist feeling and wearing feeling of the eye part of the ionic contact lens wearer can be improved, but also damage to the outer eye part such as the conjunctiva and cornea can be prevented.
Further, by using an ophthalmic composition containing polyvinylpyrrolidone and a thickener, the adsorption of polyvinylpyrrolidone on the contact lens surface lasts for a long time, and the water retention of the contact lens is improved. For example, the above ophthalmic composition When applying as an eye drop, the number of eye drops can be reduced.
The present invention relates to a polyvinyl chloride in which a negative charge of a contact lens is adsorbed on the front and back surfaces of the contact lens by causing the ophthalmic composition containing positively charged polyvinylpyrrolidone to act on the negatively charged contact lens. A system that stabilizes the tear film around the contact lens by neutralizing with a pyrrolidone plus charge, and the stabilized tear film is a dry feeling of the eye of the contact lens wearer. It is inferred that when it is applied as an ophthalmic solution to remove unpleasant feeling, it provides an excellent feeling of touch.
If a thickener is added to the ophthalmic composition of the present invention in addition to polyvinylpyrrolidone, which is an essential component, the tear film present around the contact lens is stably maintained for a long time, A feeling of wearing can be maintained.
The present invention relates to an ophthalmic composition containing 0.05 to 3.0% by weight of polyvinylpyrrolidone having an average molecular weight of 500,000 or less, 0.05 to 0.3% by weight of a thickener and sorbic acid or a salt thereof. Then, an ophthalmic composition for stably adsorbing polyvinyl pyrrolidone on an ionic contact lens and stably maintaining a tear film present around the ionic contact lens for a long time is provided.
In the present invention, the thickener is preferably 0.01 to 0.3% by weight of hydroxypropylmethylcellulose.
The material of the ionic contact lens is preferably a polymer or copolymer obtained by homopolymerization or copolymerization of hydroxyethyl methacrylate.
The kinematic viscosity of the ophthalmic composition of the present invention is preferably a 1mm 2 / s~50mm 2 / s.
A preferred form of the ophthalmic composition according to the present invention is an ophthalmic solution or a mounting solution for contact lenses.
The present invention also provides an ophthalmic composition comprising 0.05 to 3.0% by weight of polyvinylpyrrolidone having an average molecular weight of 500,000 or less, a thickener and 0.05 to 0.3% by weight of sorbic acid or a salt thereof. Instilling an ophthalmic solution for a contact lens made of a product, or dropping the ophthalmic composition onto an ionic contact lens before wearing, or immersing the contact lens in a wearing solution made of the ophthalmic composition Thus, a method is provided in which polyvinyl pyrrolidone is continuously adsorbed on an ionic contact lens to stably maintain a tear film present around the ionic contact lens for a long time.
The present invention further comprises an ophthalmic composition comprising 0.05 to 3.0% by weight of polyvinylpyrrolidone having an average molecular weight of 500,000 or less, 0.05 to 0.3% by weight of a thickener and sorbic acid or a salt thereof. Instilling an ophthalmic solution for a contact lens made of a product, or dropping the ophthalmic composition onto an ionic contact lens before wearing, or immersing the contact lens in a wearing solution made of the ophthalmic composition Provides a method for improving the water retention of ionic contact lenses.
In the tear film retention method and the water retention improvement method, the thickener is preferably 0.01 to 0.3% by weight of hydroxypropylmethylcellulose.
The polyvinyl pyrrolidone of the present invention is not particularly limited, but the average molecular weight is desirably 500,000 or less. This is because when the average molecular weight of polyvinylpyrrolidone exceeds 500,000, the amount of polyvinylpyrrolidone adsorbed on the contact lens decreases. The blending amount of polyvinyl pyrrolidone is desirably 0.05 to 3.0% by weight. If the amount of polyvinylpyrrolidone exceeds 3.0% by weight, the ophthalmic composition may become excessively viscous, and if it is less than 0.05% by weight, the tears cannot be sufficiently adsorbed to the lens. This is because a satisfactory moist feeling and wearing feeling cannot be obtained. A more preferable blending amount of polyvinylpyrrolidone is 0.5 to 2.0% by weight, and most preferably 0.8 to 1.2% by weight.
Examples of the thickener of the present invention include methylcellulose, hydroxypropylmethylcellulose, polyvinyl alcohol, sorbitol, sodium carboxymethylcellulose, hydroxyethylcellulose, triisopropanolamine, and the like, but are not particularly limited as long as the additive has a thickening action. When hydroxypropyl methylcellulose (HPMC) is used as a thickener, it is desirable to blend in the range of 0.01 to 1.0% by weight. This is because if the amount of hydroxypropylmethylcellulose exceeds 1.0% by weight, the ophthalmic composition becomes too viscous. The blending amount of hydroxypropylmethylcellulose is more preferably 0.05 to 0.5% by weight, and most preferably 0.1 to 0.3% by weight.
In addition to polyvinylpyrrolidone, the ophthalmic composition of the present invention includes the above thickeners; isotonic agents such as sodium chloride, potassium chloride, calcium chloride, glycerin, propylene glycol; boric acid, borax, citric acid, phosphorus Buffering agents such as sodium oxyhydrogen and epsilon-aminocaproic acid; preservatives such as benzalkonium chloride, chlorhexidine gluconate, benzethonium chloride, sorbic acid, potassium sorbate, ethyl paraoxybenzoate, butyl paraoxybenzoate; sodium edetate, etc. Additives that are pharmaceutically acceptable, such as a stabilizer of
As the most preferable ophthalmic composition of the present invention, for example, an ophthalmic solution and a mounting solution containing 0.8 to 1.2% by weight of polyvinylpyrrolidone and 0.1 to 0.3% by weight of hydroxypropylmethylcellulose can be mentioned. When this composition is applied as an ophthalmic solution or a mounting solution, for example, 0.05 to 0.3% by weight of sodium edetate is used as a stabilizer, and 0.05 to 0.3% by weight as a preservative. % Sorbic acid or a salt thereof is preferably added.
When applying this ophthalmic composition as an ophthalmic solution for contact lenses, it is preferable to set the pH to around 7.0 using a general-purpose buffer such as boric acid or borax, It is preferable to set the osmotic pressure ratio to around 1.0 using a general-purpose tonicity agent such as calcium chloride.
If when applying the present ophthalmic composition as eye drops for contact lenses, it is desirable that the kinematic viscosity is 1mm 2 / s~8mm 2 / s, also be applied as a mounting solution for contact lenses, it is desirable to 1mm 2 / s~50mm 2 / s.
When this ophthalmic composition is used as an ophthalmic solution, it is usually sufficient to instill 1 to 3 drops once a day or 2 to 5 times a day. May be dropped onto a contact lens. Further, when the ophthalmic composition is used as a mounting liquid, it is used by immersing it in the mounting liquid before wearing the contact lens. Further, the ophthalmic composition can be applied as a storage solution or cleaning solution for contact lenses.

3. Water retention test of ionic contact lens A water retention test of the lens over time with respect to the ophthalmic solution and physiological saline of Example 3 was performed using an ionic contact lens. That is, the moisture on the surface of an ionic contact lens (trade name: One Day Accuview) was wiped off, put in a petri dish, capped and weighed the lens, and then dried in a dryer at a temperature of 40 ° C. for 30 minutes. . After drying, it was confirmed that the moisture content of the lens calculated by measuring the weight of the lens almost coincided with the moisture content indicated by the lens manufacturer. Next, 3 ml each of the ophthalmic solution and physiological saline solution of Example 3 were measured, put into 6 ml glass bottles, and the dried ionic contact lens was immersed in each solution at a temperature of 35 ° C. for 1 day. The lens surface was wiped off, placed in a petri dish, capped, and the lens weight was measured. The petri dish was stored in a constant temperature and humidity chamber with a temperature of 25 ° C. and a relative humidity of 60% under open conditions, the lens was taken out over time (six times at 10-minute intervals), and the weight of the lens was measured. Table 3 shows the results of calculating the water retention rate (water residual rate) of the ionic contact lens according to the following formula. In addition, the numerical value in a table | surface shows the average value of 3 examples.

As is apparent from Table 1, polyvinylpyrrolidone (PVP) is specifically adsorbed on the ionic contact lens. In particular, PVP having a K value of 15 and 30 is adsorbed well. In addition, from the results of the eye comfort test shown in Table 2, the ophthalmic composition containing PVP stabilizes the tear film present around the ionic contact lens, and feels comfortable ( Improve moisturizing and wearing feeling). Furthermore, if hydroxypropylmethylcellulose (HPMC) is added in combination to an ophthalmic composition containing PVP, the feeling of comfort is further improved and sustained, and the number of eye drops can be reduced. From the results of the water retention test in Table 3, the ophthalmic composition of the present invention significantly improves the water retention of the ionic contact lens as compared with the case of using physiological saline. Therefore, the ophthalmic composition of the present invention has the effect of stabilizing the tear film on the eyeball surface of the lens of the contact lens wearer by adsorbing the PVP to the contact lens and holding the tear fluid for a long time. In addition to improving the moist feeling and wearing feeling of the eye part of the contact lens wearer, it is possible to prevent the outer eye part such as the conjunctiva and cornea from being damaged.
