Abstract
Description
本出願は、35 U.S.C.§119(e)に基づき、そのそれらすべての内容をここに参考文献として合体させる、2003年5月20日出願の「腫瘍の治療のためのNDGA誘導体の投与のための方法と組成物」と称する米国仮出願第60/472,188号、2003年5月20日出願の「腫瘍の治療のためのNDGA誘導体の投与のための方法と組成物」と称する米国仮出願第60/472,299号、2003年5月20日出願の「神経変性疾患、障害及び状態の治療のための方法と組成物」と称する米国仮出願第60/472,008号、2003年5月20日出願の「神経変性疾患、障害及び状態の治療のためのNDGA組成物と方法」と称する米国仮出願第60/472,144号、2003年5月20日出願の「高血圧症、糖尿病及び肥満治療のためのNDGA誘導体」と称する米国仮出願第60/472,282号、の優先権を主張する。
本発明は、肥満の治療用などの、過体重対象体などに対する投与のためのカテコールブタンを含有する、キット、方法及び組成物に関する。本発明は、更に、前述の組成物の製造方法にも関する。いくつかの実施例において、カテコールブタンが、疾患組織に対する直接注射以外、及び、皮膚に対する局所的塗布以外の投与経路で対象体に投与される。本発明は、更に、そのような治療に適して調合される単数又は複数のカテコールブタンを含む組成物にも関する。
従って、本発明の対象の一つは、そのような治療を必要とする対象体の肥満の治療などの、過体重対象体の予防又は治療のための方法と組成物とを提供することにある。
図1は、肥満の治療のために、組織に対してNDGA誘導体を投与する種々の態様の具体例の略図である。M4Nは、親水性NDGAを表し、G4Nは親油性NDGAを表す。SCは、皮下投与を表す。IPは腹腔内投与を表す。IMは、筋肉内投与を表す。
本発明者等は、下記の式のカテコールブタンを発見した。
ここでの使用において「活性剤」、「化合物」及び「薬剤」という用語は、NDGA及びNDGA誘導体を含む単数又は複数のカテコールブタンを意味する。
本発明のカテコールブタンは、任意の従来方法によって調製することができる。例えば、そのような化合物は、米国特許5,008,294号に記載されているように調製することができる。
前記NDGA化合物及びその製剤は、任意の従来式の方法によって製造することができる。例えば、前記NDGA化合物は、米国特許5,008,294号(ジョーダン(Jordan)他,1991年4月16日発行)、米国特許6,291,524号(フアン(Huang)他、2001年9月18日発行)、フー,ジェイ・アール(Hwu, J.R.)(1998)又はマクドナルド,アール・ダブリュ(McDonald, R.W.)他 (2001)に記載されているように製造することができる。
本発明は、更に、前記NDGA化合物と、薬剤的に許容可能な担体又は賦形剤を含む前記カテコールブタンを有する、薬用組成物も提供する。これらの組成物は、前記カテコールブタン又はNDGA化合物の所望の利用法に応じて選択可能な緩衝剤を含むことができ、更に、その意図される利用法に適したその他の物質も含むことができる。当業者は、意図される利用法に適した、多種多様なものが知られている緩衝剤を、容易に選択することができる。いくつかの場合、前記組成物は、多様なものが知られている薬剤的に許容可能な賦形剤を含むことができる。ここでの利用法に適した薬学的に許容可能な賦形剤は、例えば、エイ・ジェンナーロ(A. Gennaro)(1990);アンセル,エイチ・シー(Ansel, H.C.)他 (1999);およびキッベ,エイ・エイチ(Kibbe, A.H.)(2000)を含む、種々の刊行物に記載されている。
本発明の前記NDGA化合物組成物を含む前記カテコールブタンは、肥満に苦しむ対象体の治療を提供することが望まれる状況において治療剤として使用される。
上述したように、有効量の前記有効物質が前記ホストに投与され、ここで、「有効量」とは、所望の結果を作り出すのに十分な投与量を意味する。いくつかの実施形態において、前記所望の結果は、少なくとも体重の減少又は体重増加の減少である。
本発明は、NP製剤としてNDGA化合物を含むカテコールブタンの製剤を含む。ここでの使用に適した様々なNP製剤を、投与方法に応じて製造することができる。前記NP製剤は、分子量、共重合体比率、薬剤添加(drug loading)、マイクロ粒子寸法、多孔度、製造条件、を制御することによって、薬剤放出プロファイルに基づいて、異なるものとすることができる。前記NP製剤は、又、製造工程に使用される重合体、安定化剤、及び界面活性剤に基づいても、異なるものとすることができる。異なる賦形剤によっても、薬剤吸収、体内での薬剤分布、血漿中での薬剤の残留率、に対して異なる影響を与えることができる。当業者は、所望の性質又は特性を決定することができ、従って、使用される適切なNP製剤を決定することができるであろう。
本発明は、ミセル形成担体に製剤される、前記NDGA化合物を含むカテコールブタンを含み、ここで、前記ミセルは、従来の方法によって作られる。そのようなものの具体例は、例えば、リッギンズ,アール・ティ(Liggins, R.T.)及びバート,エイチ・エム(Burt, H.M.)(2002)、ツァン,エックス(Zhang, X.)他 (1996)及びチャーチル,ジェイ・アール(Churchill, J.R.)及びハッチンソン,エウ・ジー(Hutchinson, F.G.)(1988)に記載されている。そのような方法の1つにおいて、親水性(ポリエーテル)セグメントと疎水性(ポリエステル)セグメントとを有する両親媒性重合体であるポリエーテル−ポリエステルブロック共重合体がミセル形成担体として使用される。
多小胞リポソーム(“MVL”)を、例えば、マントリプルガダ,エス(Mantripragada, S.)(2002)に記載されている二重乳化法などの、任意の公知の方法によって調製することができる。簡単に説明すると、前記二重乳化法において、先ず「油中水」エマルジョンが、少なくとも1つのトリグリセリド等の中性脂肪を含有するリン脂質を、投与単数又は複数の揮発性有機溶剤中に溶解させ、この脂質成分を、非混合性の第1水性成分と、疎水性NDGA化合物などの親水性カテコールブタンに添加することによって作られる。次に、この混合物を、乳化して油水中エマルジョンを形成し、その後、第2の非混合性水性成分と混合し、その後、機械混合して、前記第2水性成分中に懸濁した状態の溶剤小球を形成し、水中油中水エマルジョンを形成する。前記溶剤小球は、その中に溶解した前記NDGA化合物などの、カテコールブタンを備える複数の水性小滴を含んだものとなる。次に、一般的に、蒸発、減圧、又はその懸濁液上又は中にガス流を通過させることによって、前記有機溶剤を前記小球から除去する。溶剤が完全に除去されると、前記小球は、DepoFoam粒子などのMLVとなる。前記中性脂肪がこの工程から省略される場合、前記MLVの代わりに、従来式の多重層小胞又は単層小胞が形成される。
G4N等のような、NDGA化合物などの一部のカテコールブタンは、水溶性親水性化合物である。本発明は、薬学的許容可能な担体又は賦形剤中の親水性化合物の製剤と、その化合物の水溶液形態などの、経口製剤などの投与を含み、或いは、前記化合物は、凍結乾燥して散剤として投与したり、錠剤にしたり、又は前記化合物はカプセル剤化することも可能である。
前記経口製剤中に含まれるNDGA化合物などの前記カテコールブタンの量は、対象体に投与される所望の投与量に応じて調整することができる。そのような調整は、当業者の技量範囲内である。
本発明は、鼻腔内投与用の、NDGA化合物によって例示されるカテコールブタンの製剤と、その鼻腔内投与とを含む。鼻腔内投与は、静脈内投与によって達成可能であるよりも、脳内での活性剤の濃度を有利に高めることができる。又、この投与形態は、その薬剤を受ける対象体の肝臓と腸とにおける初回通過代謝の問題を回避するものである。
前記NDGA化合物によって例示されるここでのカテコールブタンは、前記NDGA化合物を含有する生物分解性重合体の皮下移植などの、外科的移植によって治療のために対象体に投与することができる。この治療は、外科手術以外又はそれに加えて、他の従来の療法と組み合わせることができる。
前記NDGA化合物によって例示される前記カテコールブタンは、吸入による肺への投与によって、全身的および/又は局所的に投与することが出来る。薬剤の吸入投与は、大きな全身毒性を引き起こすことなく肺組織における高薬剤濃度を達成する方法として、又、薬剤の全身循環を達成する方法として、十分に受け入れられている。そのような製剤を製造するための技術は、従来のものである。肺疾患に対する有効性は、このように投与された疎水性NDGA化合物と親水性NDGA化合物とのいずれかで見ることができる。
ここでM4Nと称されるテトラ−O−メチル−NDGAを、NDGAを、例えば、水酸化カリウム等の塩基の存在下で、過剰硫酸ジメチルと反応させることによって合成した。その生成物を、生成物の沈殿を引き起こす水の添加によって分離した。反応生成物を、塩基性アルミナのプラグを通過させて、微量フェノール不純物、主として、種々のジ−O−メチル及びトリ−O−メチル−置換NDGA、を取り除いた。反応混合物の溶液を前記アルミナプラグに通過させた後、溶媒を回転蒸発装置で除去し固体生成物を得た。これを、2−プロパノールで粉砕(triturated)し、真空炉で乾燥し、粗製テトラ−O−メチル−NDGAを得た。2−プロパノールからの結晶化によって、99.66%以上の純度のテトラ−O−メチル−NDGAを得た。
機械式攪拌装置、冷却器、不活性雰囲気用の入口を備えた22Lのフラスコを冷却浴として使用されるタブ内にセットした。このフラスコを、アルゴン雰囲気下に置き、これに484.3グラムのNDGA(Western Engineering & Research Co, El Paso, TX)と4850mLのエタノールとを入れ、攪拌した。この攪拌されたスラリに、1210mLの脱イオン水中の387.5グラムの水酸化カリウムの溶液を添加した。この反応混合物を含むフラスコを、氷浴を使用して冷却し、硫酸ジメチル(1210mL)をゆっくりと(滴下)添加した。添加は、発熱を避けるように制御した。この添加の最後において、温度は約13℃であった。反応のpHをモニタし、50%KOH溶液を、塩基性pHを維持すべくその日の間、一部分、添加した:50%KOH溶液の全1400mLを添加した。過剰塩基の反応混合物は、pH指示片の使用による検出で約12のpHを示した。溶液は塩基性pHでは暗色であったが、中性又は酸性pHでは淡色に変化した。
機械式攪拌装置、冷却器、不活性雰囲気用の入口を備えた3Lのフラスコを加熱マントルにセットした。このフラスコに415.5gの前記生成物を入れた。このフラスコに1245mLの2−プロパノールを入れ、攪拌した混合物を加熱し、穏やかな還流を与え、溶液を得た。熱を止め、混合物を一晩冷却させた。結晶生成物を濾過によって分離し、濾過ケーキを200mLの低温2−プロパノールで洗浄した。この生成物を真空炉内で、50℃にて高真空状態で404.7g(NDGAからの70.5%総収率)まで乾燥させた。
前記NDGA化合物は、任意の従来方法によってナノ粒子製剤として製剤できる。例えば、前記ナノ粒子は、ランプレヒト,エイ(Lamprecht, A.)他(2001a)及びランプレヒト,エイ(Lamprecht, A.)他(2001b)に記載されているように、下記のようにして調製可能である。
PLGAと別のマトリクス剤,d−α−トコフェリルポリエチレングリコール1000コハク酸塩(ビタミンE TPGS又はTPGS)とを含むNPを、ムー,エル(Mu, L.)及びフェン,エス・エス(Feng, S.S.)(2003)に記載されているように、改変の水中油の単一エマルジョン溶媒蒸発/抽出法で製造することができる。この方法において、既知量の重合体質量と、NDGA化合物とを塩化メチレン(ジクロロメタン)に添加する。前記重合体、例えば、ポリ−(DL−ラクチド−co−グリコリド (PLGA;L/G=50/50,MW40,000−75,000;L/G=75/25,MW90,000−120,000及びL/G=85/15,MW90,000−120,000)を、Sigma(USA)から購入することができる。ビタミンE TPGSは、Eastman Chemical, USAから入手可能である。全ての原料が溶解されるように前記混合物を攪拌する。次に、有機相の溶液を、乳剤有り又は無しで、攪拌された水溶液にゆっくり注ぎ、同時に50Wパルスモード(Misonix, USA)で超音波処理する。形成されたo/wエマルジョンを、一晩、磁気攪拌装置で室温(22℃)にて、ゆっくりと攪拌して前記有機相を蒸発させることができる。得られたサンプルは、10,000rpm、10分間,16℃(Eppendorfモデル5810R,Eppendorf,ハンブルグ、ドイツ)等での遠心分離によって収集することができ、一部のサンプルについては脱イオン水で1回又は2回洗浄することができる。調製された懸濁液を、凍結乾燥(Alpha-2,Martin Christ Freeze Dryers,ドイツ)し、ナノ粒子の細かい粉体を得て、これを真空乾燥器に入れ、そこに保存することができる。
前記親油性薬剤等のNDGA化合物は、従来方法によって持効性ポソーム中にカプセル剤化することができる。そのような方法のひとつは、例えば、シャルマ,ユー・エス(Sharma, U.S.)他 (1997)に記載されている。
前記NDGA化合物は、例えば、マーティン,イー(Marttin, E.)他(1997)に記載されるもののような任意の従来方法によって鼻腔内投与用の乾燥散剤又は煙霧剤として製剤することができる。
ここでのNDGA化合物は、移植のために生物分解性重合体に組み入れることができる。そのような生物分解性重合体は、例えば、フレミング,エイ・ビー(Fleming, A.B.)及びソールツマン,ダブリュ・エム(Saltzman, W.M.)(2002)に記載されているもののような、任意の従来方法によって製造することができる。この生物分解性重合体の単数又は複数のウエハを、所望の化合物の投与量に応じて、一度に移植することができる。前記重合体の生物分解性マトリクスは、ポリフェプロサン(polifeprosan)20、1,3−ビス−(p−カルボキシフェノキシ)プロパンとセバシン酸[p(CCP:SA)]の共重合体を20:80のモル比で含むものとすることができる。前記重合体を移植用に形成するために、p(CPP:SA)とここでの化合物を、ジクロロメタン中で同時に溶解し、噴霧乾燥させて、約1μm〜約20μmの寸法範囲の球状粒子を形成することが出来る。その結果得られる「微小球体(microsphere)」を圧縮成形して、任意のサイズ、例えば、直径が約14mmで厚みが約1mmのウエハを形成することができる。これらのウエハ、小さな空隙に取り囲まれた密につめられた微小球体から成る均質構造を有する。NDGA化合物の濃度は、例えば、3.8%活性化合物など、治療される対象体に適切な任意の量にすることができる。
前記NDGA化合物を含有するPLGA−mPEGナノ粒子を、ソン,シー・エックス(Song, C.X.)他(1997)によって記載されている二重エマルジョン法を少し改変した方法を使用して調製することができる。ここで、NDGA化合物の水溶液を、プローブ超音波処理(Bioblock Scientific,モデル75038)を使用して、前記共重合体が溶解されたジクロロメタン中で乳化することができる。この水/油エマルジョンを、コール酸ナトリウムの水溶液に移し、その混合物をプローブ超音波処理することができる。結果形成された水/油/水エマルジョンを、有機相の蒸発が完了するまで、室温でゆっくりと攪拌することができる。このようにして作られたナノ粒子は、遠心分離によって精製し、脱イオン化され蒸留された水で再構成することができる。次に、これらのナノ粒子を、例えば、1.2μmのフィルタ(Millex AP, Millipore)等を通して濾過することができる。
プルロニックは、トリブロックPEO−PPO−PEO共重合体であり、PEOはポリ(エチレン酸化物)を表しPPOはポリ(プロピレン酸化物)を表す。疎水性の中央のPPOブロックはミセルコアを形成し、側方のPEOブロックは、ミセルを細網内皮系(“RES”)による認識から保護する、シェル又はコロナを形成する。プルロニック共重合体は、BASF Corp,及びICIから市販されている。前記NDGA化合物を、例えば、ラポポート,エヌ・ワイ(Rapoport, N.Y.)他(1999)に記載されているもののような任意の従来方法でプルロニックミセルに導入することができる。
ここでのNDGA化合物は、乾燥散剤や水溶液を含む、任意の従来の製剤を使用して吸入を介して投与することができる。前者は、安定性、微生物の成長に対する低い感受性、及び、ひと吹き当たりの高い質量、の利点を有する。水溶液は、より良好な再現性を提供し、疑集塊の問題を回避する。
前記NDGA化合物は、吸入の目的ためのその他多数の薬学的に許容可能な担体に製剤することができる。この例において、ここでの化合物のいくつかを、エンク,エイ・エイチ(Enk, A.H.)他(2000)に記載の方法によって投与することができる。そのような化合物は、約5%のグルコースと2%のヒトアルブミンとを含有する溶液に溶解することができる。次に、特別に設計された吸入装置を使用して吸入を行うことができる(Jetair, Fa,ホイヤー,ドイツ)。
NDGA誘導体を肥満の治療のために使用することができる。一例において、NDGA誘導体、G4N、を無菌液、例えば、150mM塩化ナトリウム及び40mMリン酸ナトリウム中の1mg/ml(pH7.4)、として製剤する。静脈内投与のためには、前記薬剤を、更に、10ml/kg(約100ml以下)の通常の生理食塩水、中で希釈する。各治療は、標準投与量のジフェンヒドラミン(ベナドリル:Benadryl)及びアセトアミノフェン(タイレノール:Tylenol)の前投与後の15〜20分として、1時間の吸入、等の、その特定のNDGA誘導体の半減時間に依存した所定時間の連続的吸入から構成される。患者は、そのそれぞれが、10日間の連続治療、又は各週が全部で9の投与の、3日連続の投与を3週間のサイクル、で治療される。患者の特性、疾患の状態に応じて与えられる投与量は、例えば、0.1mg/kg/日、又は0.18mg/kg/日、又は0.32mg/kg/日、以上、とすることができる。
以下の実験は、当初、M4Nの腫瘍に対する作用を判定するために行われたものである。しかしながら、それらの結果は、驚くべきことに、M4Nが腫瘍の治療に有効であるばかりでなく、M4Nは、マウスの体重減少も引き起こしたことを示した。これらの結果は、真に思いがけないものであって、ここでの前記化合物が肥満の治療に有効であるということを示している。
6−8週齢のメスのICRマウスを、Harlan Sprague Dawley (Indianapolis, IN)から購入した。C57bl/6マウスを、Charles River Laboratories (Wilmington, MA)から購入した。胸腺欠損(thy−/thy−)ヌードマウス、5−6週齢のオスとメス、をCharles River Laboratoriesから購入し、国際動物保護及び使用ガイドラインに基づき、制御された温度と湿度の無病原室に収納した。C3−細胞誘発腫瘍を有するC57bl/6マウスを、キム,イー・エイチ(Kim, E.H.)他(2004)に記載されているように準備した。
M4Nを、ロガンツォ(Loganzo)他(2003)に記載されているように、6% Cremaphor EL,6%エタノール、88%生理食塩水中に溶解させた。マウスに、2mgのM4Nを含有する一日当たり100μL.i.p.の注射を3週間与えた。対照マウスには、同量の賦形剤を与えた。腫瘍を、7日間毎に、二つの垂直寸法(LとW)で測定し、腫瘍の体積を、下記の式:V=(LxW/2)3xπ/6、によって算出した。それぞれのマウスからの結果を、時間対の平均腫瘍体積としてプロットした。腫瘍体積の平均差の統計上の有意性を、スチューデントT検定によって評価した。実験の最後に、cdc2とサバイビン(survivin)発現の免疫組織学的な解析のために、腫瘍生検を収集した。
Harlan ICRマウス又はC3細胞誘発腫瘍C57bl/6マウスに、尾静脈を介して、100μCiのトリチウム標識M4Nと60mMの非放射性のM4Nとを含有する100μLのCremaphor−エタノール系溶剤を腹腔内注射した。特定の注射後時に、これらのマウスを犠牲にし、その器官と血液を収集し、計量し、4Mグアニジンイソチオシネート(GITC)中に一晩溶解させた。その後、不溶性ペレットを更にEtOHで抽出した。GITC抽出物とEtOH抽出物との両方のトリチウム含有率を、パッカード シンチレーション カウンターで測定し、各器官のM4Nの量を、接種原の特異的活性に基づいて算出した。
短期給飼実験のために、300μLのひまし油中に溶解させた30mgのM4Nを、6匹のマウスのそれぞれに経口投与した。投与の2時間後、4時間後、及び8時間後に、2匹のマウスを犠牲にし、器官と血液を収集し、下記のように、M4Nを抽出し、定量化した。長期給飼実験では、マウスに、コーン油に溶解させたM4NとBasal Mixとから成るフードボール(Harlan Teklad;Madison, WI;Cat.#TD02273)を14週間与えた。フードボールの重さは9gで、それぞれが242mgのM4Nを含有していた。2匹のマウス、一匹のオスと一匹のメス、を長期薬剤保持研究のために確保し、14匹のマウス、オスとメスの双方、の長期薬剤毒性研究のために使用した。給飼の終わりに、マウスを犠牲にし、器官と血液を収集し、下記のようにしてM4Nを抽出し、定量化した。
M4N給飼マウスから収集した器官と血液を、一晩−80℃で凍結させた。凍結の前に、胃腸器官(胃と、小腸と結腸)を長手方向に切開し、全ての含有物を除去するためにPBSで徹底的に洗浄した。次の日、器官を剃刀でドライアイス上にて小片に切断し、スピード−バック(Speed-vac)中で乾燥させ、その後、乳鉢と乳棒とを使用して粗い粉体に粉砕した。サンプルを、振とうしながら、100%エタノール中で一晩、抽出した。サンプルを遠心分離し、上清を収集した。ペレットを、振とうしながら、更に2回、100%エタノール中に抽出した。貯蔵されたエタノール抽出物を、ベンチトップ上で数日間蒸発させ、その後、酢酸エチルで再度抽出し、スピード−バック内で完全に乾燥させた。これらの乾燥サンプルを、次に、HPLCによって定量化分析し、M4Nを、標準として純粋M4Nを使用した質量分析法によって同定した。
各時点における一匹のマウスからのサンプルを、HPLC分析のために、KP Pharmaeuticals(Bloomington, IN)に送った。HPLC条件は以下の通りであった。35%:0.1% TFA/H2O、65%=“CAN”。N4N標品を、100mLのCANに10.01mgのM4Nを希釈することによって調製し、その後、5分間超音波処理した(2002ng/注射)。サンプルを、400μL EtOHを添加し、2分間、又は、完全な溶解が達成されるまで、超音波処理することによって調製した。これらのサンプルの注射量は100μLであった。
腹腔内、静脈内及び経口投与後、M4Nは、種々の組織に対して全身的に、かつ、検出可能な毒性無く、分散する。
トリチウム標識M4Nと非放射性M4Nの混合物を、6% Cremaphor EL,6%エタノール、88%生理食塩水溶媒に溶解し、マウスに、腹腔内(i.p.)及び尾静脈を介した静脈内(i.v.)で注射した。注射の3時間後、器官と血液を採取し、計量し、M4Nを抽出した。各器官からの抽出物のトリチウム含有量をパッカード シンチレーション カウンターで測定し、各器官のM4Nの量を、接種原の特異的活性に基づいて算出した。M4Nは、i.p.とi.v.の投与経路の両方による注射の3時間後において、種々の器官に有効に分布していた。興味深いことに、投与経路の違いにも拘わらず、非常によく似た組織分布プロファイルが得られ、このことは、薬剤分散のランダムでは無く、恐らくは規制された機序を示すものである。これについて詳述すると、下記の経口投与を使用して、非常によく似た分布プロファイルが観察された。回収された放射性活性の大半は、組織1g当たり3μg〜20μgM4Nの範囲で、胃腸管器官、胃、小腸、盲腸、大腸、に局在していた。有意な量のM4Nは肝臓と脂肪にも存在しており、組織1g当たり150ng〜400ngの範囲での、より低い濃度が、脳、腎臓、及び脾臓に検出された。但し、注射後3時間で、心臓や血液中にはM4Nは全く、又はほとんど検出されなかった。
それはしばしば多くのガンの治療において主要な限定要因であるので、予期される全身化学療法剤の毒性は、極めて重要である。M4Nでの長期腹腔内全身投与の毒性を評価するとともに、下記の異種移植片実験のための安全な投与量を決定するために、下記の毒性試験を行った。12匹のマウスを、それぞれ4匹のマウスから成る3つのグループに分け、以下の治療を2週間受けさせた。即ち、グループ1は、一日一回、i.p.で、100マイクロリットルの6% Cremaphor EL,6%エタノール、及び88%生理食塩水に溶解させた2mgのM4Nの注射(20mg/mLが、前記溶剤中に溶解することのできるM4Nの最大量である)を受け、グループ2は、i.p.で毎日賦形剤のみの注射を受け、グループ3は、治療を受けなかった。活性の毎日の評価と、治療中における総体重の変化による測定で、これらのマウスのいずれにおいても毒性は観察されなかった。
この実験において、オス及びメスのビーグル犬に対するM4Nの二種類の製剤[Cremaphor−エタノール(CET)又はジメチルスルホキシド(DMSO)]の製剤の最大耐容量(MTD)を測定した。この例は、M4Nが、犬に対して、四時間にわたる静脈内注入で、CET賦形剤の場合は10mg/kgまで、DMSO賦形剤の場合は100mg/kgまで、安全に投与されたことを示している。これらの製剤で、最小の毒性で、14,000ng/ml M4N以下の血中レベルが達成された。
VAPをビーグル犬に対して、注入カテコールの先端が、上大静脈の準位に位置するように移植した。犬は、外科手術の日に、鎮痛剤と抗生物質とによって、又、手術後は、抗生物質および/又は鎮痛剤によって予防的に処理された(適切な場合には、Gene Logic Inc. SOP Nos. 324.0.2、325.0.1及び326.0.2によって)。その他の処置が、スタッフの獣医による推奨により、提供された。カテーテルラインを、研究指導者によって適切であると見なされた頻度で、術後の回復時期中に、生理食塩水によって洗浄した。
前記CETグループ動物からの血液サンプルを、下記の時点、即ち、投与前、約4時間の注入の完了後の、0.25,0.5.1,2,4,8及び16時間後、研究日(SD)1,SD3,SD6及びSD8に頚静脈を介して収集した。
前記血漿及び血清サンプルを、TK分析のために本提供者が指定した研究所である、MedTox Laboratoriesに送った。M4N血漿及び血清濃度−時間データのTK分析を、MedTox Laboratoriesによる確証方法(M200406)を使用して行い、非コンパートメント法によって分析し、毒物動態学的パラメータ(データによって可能である場合)、但し、Cmax,Tmax及びAUCに必ずしも限定されるものではない、の推定値を得た。
a)M4N−CETグループ
オス犬:CET注入に対して、最初の一時間半で、紅斑、蕁麻疹、そう痒、嘔吐、下痢、及び一般的な傾眠で反応した。そのあと、回復を始め、歩き回り始め、水を飲み始めた。注入終了後直ぐに正常に挙動した。
メス犬:嘔吐と下痢が無かったこと以外はオス犬と類似の反応であった。そのアレルギー反応も、オス犬よりも軽かった。注入終了後直ぐに正常に挙動した。
オス犬:DMSO注入に対して、軽度の紅斑、軽いそう痒、で反応した以外は、正常であった。最後の注入終了後直ぐに正常に挙動した。メス犬:オス犬と類似の反応。最後の注入終了後直ぐに振る舞いは正常になった。4匹の犬全部がそのそれぞれの賦形剤処理の注入に生き残った。それらはすべて治療後、健康で正常に挙動した。
a)M4N−CETグループ
オス犬:M4N− CET(1mg/kg)注入に対して、軽い紅斑、蕁麻疹、そう痒、で反応した。この日のこれらの反応は、SD1のものよりも軽度であった。具体的には、動物には、嘔吐、下痢及び傾眠は無く、SD1においてよりもより機敏であった。注入終了後直ぐに正常に挙動した。メス犬:M4N−CET(1mg/kg)当日の注入に対するその反応は、SD1に観察されたものよりも更に軽度であった。そのアレルギー反応は、軽い紅斑とそう痒を含んでいたが、その4時間の注入期間全体を通じて一般的に極めて機敏であった。注入終了後直ぐに正常に挙動した。
オス犬:この犬によるなんら有害な臨床反応はなかった。予想されたように、頚静脈の沿った注射部位にいくらかの炎症があった。
メス犬:この犬によるなんら有害な臨床反応はなかった。予想されたように、頚静脈の沿った注射部位にいくらかの炎症があった。
4匹の犬全部がそのそれぞれの試験物治療に生き残った。それらはすべて治療後、健康で正常に挙動した。
a)M4N−CETグループ
オス犬:以前の二つの投与日に類似し、この動物は、注入に対して軽い紅斑、蕁麻疹及び、そう痒で反応した。これらの反応の強度は、SD3での反応よりも確実に小さかった。動物は、嘔吐や下痢をせず、注入期間全体を通して一般的に機敏であった。注入終了後直ぐに正常に挙動した。
メス犬:以前の投与への反応と一致して、オス犬よりも当日の注入によく耐容性を示した。動物は軽度の紅斑、及び、そう痒を示したが、ほとんど機敏であった。注入終了後直ぐに正常に挙動した。
オス犬:この犬は、最初の3回の投与間隔(投与の間に1/2時間)において非−VAP頚静脈を介してM4N−DMSOをうまく静脈内注射された。前の投与日と同様、この犬も各注射後において、なんら有害な臨床的徴候又は症候群を示さなかった。4回目の注射に先立って、技術者は、注射部位の周りに「卵のサイズ」の腫れに気づいた。皮下投与ミスの可能性は、それは、3回目の注射中に容易に検出されたであろうため、除外することができた。それは、注射部位を通して血液がゆっくりと血管外放出した結果の血腫である可能性が最も高かった。この動物は、三回目の注射後、それ以上の投与を受けなかったが、但し、血液サンプルは収集され、三回目の注射投与後の血液収集の正確な時点は明確に記録された。血腫は、2時間以内に無くなり、注射部位領域をゆっくりマッサージしても動物に炎症を起こさせることはなかった。
メス犬:この犬は、4時間にわたる8回反復投与全体においてM4N−DMSOをうまく注射された。前の投与日と同様、この犬もなんら有害な臨床的徴候又は症候群を示さなかった。

M4N−DMSOを100mg/Kg投与された動物はなんら有害な臨床的徴候又は症候群を示さなかったので、二匹の予備の犬(オス1匹とメス1匹)に、M4N−DMSOを200mg/Kg投与した。200mg/Kgでは、メス犬は、8回の投与の最初のあとに既に呼吸困難(鼻からの泡立ち)を経験し、直ぐに虚脱したが、二回目の投与時では回復することができ。二回目の投与後、その反応は、類似していたが、更に深刻であった。従って、スタッフ獣医はこのメス犬を安楽死させることを提案した。オス犬は、それよりも僅かに耐性を示したが、類似の呼吸困難と虚脱症候群を示した。この動物は全部で三回の投与を受けたが、スタッフ獣医はそれ以上の投与を停止することを提案した。

- 前記組成物が、食用混合物として存在する請求項100に記載の方法。
- 前記組成物が、所定期間毎日投与される請求項100に記載の方法。
- 前記組成物が、毎日、5日間以上、1週間投与される請求項104に記載の方法。
- 前記組成物が、毎日、5日間以上、2週間投与される請求項104に記載の方法。
- 前記組成物が、毎日、5日間以上、3週間投与される請求項104に記載の方法。
- 投与される前記テトラ−O−メチルNDGAの量が、毎投与当たり少なくとも30mgである請求項100に記載の方法。
- 投与される前記テトラ−O−メチルNDGAの量が、毎投与当たり少なくとも90mgである請求項100に記載の方法。
- 前記テトラ−O−メチルNDGAが、前記組成物中において20mg/mLの濃度で含まれる請求項100に記載の方法。
- 前記薬学的に許容可能な担体又は賦形剤が、Cremaphor EL、エタノール及び生理食塩水を含む請求項99に記載の方法。
- 前記Cremaphor EL濃度が、6%である請求項111に記載の方法。
- 前記エタノール濃度が、6%である請求項111に記載の方法。
