Abstract
Description
本発明は、HLAクラスI分子に結合するヘテロクリティックアナログを設計する方法、およびそのようなアナログを含むポリペプチド、またはそのようなアナログからなるポリペプチドを産生する方法に関する。本発明は、ポリペプチド自体、ならびにそのようなポリペプチドをコードするポリヌクレオチドにも関する。ヘテロクリティックアナログによる免疫化は、腫瘍に対するワクチン接種のより有効で効率的な戦略であり、有効なCTLの産生が困難であることがこれまでに判明している場合には特にそうである。
「ヒト白血球抗原」または「HLA」は、ヒトクラスIまたはクラスII主要組織適合複合体(MHC)タンパク質である(例えば、非特許文献59を参照されたい)。
本発明は、対応する類似の野生型エピトープよりも高い免疫応答を起こし得るエピトープを含むペプチドを調製する方法を提供する。得られる「ヘテロクリティックアナログ」は、標的細胞上に表示された抗原によって特徴付けられるウイルス性疾患、癌、および他の病気を治療するための免疫学的組成物に有用である。
上記のように、「アナログポリペプチド」は、アナログを含み、またはアナログからなり、本発明の一部でもある。好ましいアナログは、上記項に示されている。
アナログポリペプチドを改変して、血清半減期の改善など所望の性質を与え、または免疫原性を高めることができる。
ワクチンに使用するポリエピトープ組成物に入れる多数のアナログおよびエピトープを選択するとき、あるいはワクチンに入れる個別のエピトープおよび/またはミニ遺伝子などの核酸によってコードされる個別のエピトープを選択するために、以下の原理を利用することが好ましい。以下の原理の各々は、この選択をするために調和がとられていることが好ましい。所与のワクチン組成物に組み込まれる複数のエピトープは、エピトープが由来するネイティブ抗原の配列中で隣接することができるが、隣接しなくともよい。

ワクチン、および免疫を生じるのに有効な量の本明細書に記載する1個または複数のアナログポリペプチドまたはアナログポリヌクレオチドを含有するワクチンを調製する方法は、本発明のさらなる実施形態である。適切な免疫原性のポリペプチドを定義した後、それらを分類し、本明細書では「ワクチン」組成物と称する様々な手段によって送達する。この開示を通して記述する組成物を、ワクチンとして使用することができる。

本発明の一実施形態においては、本明細書に記載するヘテロクリティックアナログポリペプチドを試薬として使用して免疫応答を評価する。評価される免疫応答は、試薬として使用されるアナログポリペプチドを認識し、これに結合する抗原特異性CTLを誘導することができる任意の薬剤を免疫原として使用して誘導される。ポリペプチドを免疫原として使用する必要はない。このような分析に使用されるアッセイシステムは、細胞内リンホカイン用四量体染色、およびインターフェロン放出アッセイ、またはELISPOTアッセイなどの比較的最近の開発技術を含む。
本発明のポリペプチドおよびポリヌクレオチド組成物を、ワクチン投与の説明書とともにキットの形で提供することができる。一般に、このキットは、容器に入った、好ましくは単位剤形の所望のポリペプチド組成物、および投与説明書を含む。別のキットは、容器に入った、好ましくは単位剤形の本発明の所望の核酸を含むポリヌクレオチド(例えば、ミニ遺伝子)構築体、および投与説明書を含む。IL−2、IL−12などのリンホカインもこのキットに入れることができる。望ましいこともある他のキット成分は、例えば、無菌シリンジ、追加投与製剤などを含む。
過去数年で、HLAクラスI分子の大部分を、各々がほとんど重複するペプチド結合レパートリーと、主要なペプチド結合ポケットのコンセンサス構造とを特徴とする比較的少数のスーパータイプに分類できることを示す証拠が蓄積された。したがって、本発明のペプチドは、いくつかのHLA特異的アミノ酸モチーフ(例えば、表3〜4参照)のいずれか1個によって特定され、またはモチーフの存在がいくつかの対立遺伝子特異的HLA抗原、スーパーモチーフに結合する能力に対応するかどうかで特定される。特定のアミノ酸スーパーモチーフを有するペプチドに結合するHLA分子は、HLA「スーパータイプ」と総称される。
HLA−A1スーパーモチーフは、エピトープの2位の小さな(TまたはS)または疎水性(L、I、VまたはM)一次アンカー残基、およびC末端位置の芳香族(Y、FまたはW)一次アンカー残基がペプチドリガンド中に存在することを特徴とする。A1スーパーモチーフ(すなわち、HLA−A1スーパータイプ)に結合するHLA分子の対応するファミリーは、少なくともA*0101、A*2601、A*2602、A*2501およびA*3201を含む(例えば、非特許文献109〜111参照)。A1スーパーファミリーのメンバーであることが予測される別の対立遺伝子特異的HLA分子を表5に示す。
対立遺伝子特異的HLA−A2.1分子の一次アンカー特異性(例えば、非特許文献112〜115参照)、およびHLA−A2および−A28分子中の交差反応性結合が記述された。(関連データの総説は、例えば、非特許文献116〜119参照。)これらの一次アンカー残基は、HLA−A2スーパーモチーフを規定する。ペプチドリガンド中のHLA−A2スーパーモチーフの存在は、いくつかの異なるHLA−A2および−A28分子を結合する能力に対応する。HLA−A2スーパーモチーフは、エピトープの2位の一次アンカー残基としてL、I、V、M、A、TまたはQ、およびC末端位置の一次アンカー残基としてL、I、V、M、AまたはTを含むペプチドリガンドを含む。
HLA−A3スーパーモチーフは、ペプチドリガンド中のエピトープの2位に一次アンカーとしてA、L、I、V、M、SまたはTが存在し、C末端位置、例えば9量体の9位に正に帯電した残基RまたはKが存在することを特徴とする(例えば、非特許文献120参照)。A3スーパーモチーフに結合する対応するHLA分子ファミリーの例示的なメンバー(HLA−A3スーパータイプ)は、少なくともA*0301、A*1101、A*3101、A*3301およびA*6801を含む。A3スーパータイプのメンバーであることが予測される別の対立遺伝子特異的HLA分子を表5に示す。
HLA−A24スーパーモチーフは、エピトープの2位の一次アンカーとして芳香族(F、WまたはY)残基または疎水性脂肪族(L、I、V、MまたはT)残基、およびC末端位置の一次アンカーとしてY、F、W、L、IまたはMがペプチドリガンド中に存在することを特徴とする(例えば、非特許文献121参照)。A24スーパーモチーフに結合する対応するHLA分子ファミリー(すなわち、A24スーパータイプ)としては、少なくともA*2402、A*3001およびA*2301が含まれる。A24スーパータイプのメンバーであることが予測される別の対立遺伝子特異的HLA分子を表5に示す。
HLA−B7スーパーモチーフは、エピトープの2位の一次アンカーとしてプロリンを有するペプチド、およびC末端位置の一次アンカーとして疎水性または脂肪族アミノ酸(L、I、V、M、A、F、WまたはY)を特徴とする。B7スーパーモチーフに結合する対応するHLA分子ファミリー(すなわち、HLA−B7スーパータイプ)は、B*0702、B*0703、B*0704、B*0705、B*1508、B*3501、B*3502、B*3503、B*3504、B*3505、B*3506、B*3507、B*3508、B*5101、B*5102、B*5103、B*5104、B*5105、B*5301、B*5401、B*5501、B*5502、B*5601、B*5602、B*6701およびB*7801を含めた少なくとも26個のHLA−Bタンパク質を含む(関連するデータの総説は、例えば、非特許文献122〜125参照)。B7スーパータイプのメンバーであることが予測される別の対立遺伝子特異的HLA分子を表5に示す。
HLA−B27スーパーモチーフは、エピトープの2位の一次アンカーとして正に帯電した(R、HまたはK)残基、およびC末端位置の一次アンカーとして疎水性(F、Y、L、W、M、I、AまたはV)残基がペプチドリガンド中に存在することを特徴とする(例えば、非特許文献121参照)。B27スーパーモチーフに結合する対応するHLA分子ファミリーの例示的なメンバー(すなわち、the B27スーパータイプ)としては、少なくともB*1401、B*1402、B*1509、B*2702、B*2703、B*2704、B*2705、B*2706、B*3801、B*3901、B*3902およびB*7301が含まれる。B27スーパータイプのメンバーであることが予測される別の対立遺伝子特異的HLA分子を表5に示す。
HLA−B44スーパーモチーフは、エピトープの2位の一次アンカーとして負に帯電した(DまたはE)残基、およびC末端位置の一次アンカーとして疎水性残基(F、W、Y、L、I、M、VまたはA)がペプチドリガンド中に存在することを特徴とする(例えば、非特許文献126参照)。B44スーパーモチーフに結合する対応するHLA分子ファミリーの例示的なメンバー(すなわち、B44スーパータイプ)としては、少なくともB*1801、B*1802、B*3701、B*4001、B*4002、B*4006、B*4402、B*4403およびB*4006が含まれる。
HLA−B58スーパーモチーフは、エピトープの2位の一次アンカー残基として小さな脂肪族残基(A、SまたはT)、およびC末端位置の一次アンカー残基として芳香族または疎水性残基(F、W、Y、L、I、V、MまたはA)がペプチドリガンド中に存在することを特徴とする(関連するデータの総説は、例えば、非特許文献121参照)。B58スーパーモチーフに結合する対応するHLA分子ファミリーの例示的なメンバー(すなわち、B58スーパータイプ)としては、少なくともB*1516、B*1517、B*5701、B*5702およびB*5801が含まれる。B58スーパータイプのメンバーであることが予測される別の対立遺伝子特異的HLA分子を表5に示す。
HLA−B62スーパーモチーフは、エピトープの2位の一次アンカーとして極性脂肪族残基Qまたは疎水性脂肪族残基(L、V、M、IまたはP)、およびC末端位置の一次アンカーとして疎水性残基(F、W、Y、M、I、V、LまたはA)がペプチドリガンド中に存在することを特徴とする(例えば、非特許文献121参照)。B62スーパーモチーフに結合する対応するHLA分子ファミリーの例示的なメンバー(すなわち、B62スーパータイプ)としては、少なくともB*1501、B*1502、B*1513およびB5201が含まれる。B62スーパータイプのメンバーであることが予測される別の対立遺伝子特異的HLA分子を表5に示す。
HLA−A1モチーフは、エピトープの2位の一次アンカー残基としてT、SまたはMがペプチドリガンド中に存在し、C末端位置の一次アンカー残基としてYが存在することを特徴とする。別の対立遺伝子特異的A1モチーフは、2位ではなく3位の一次アンカー残基を特徴とする。このモチーフは、エピトープの3位の一次アンカー残基としてD、E、AまたはSが存在し、C末端位置の一次アンカー残基としてYが存在することを特徴とする(関連するデータの総説は、例えば、非特許文献110、111、127参照)。
HLA−A2*0201モチーフは、9残基ペプチドの2位の一次アンカー残基としてLまたはM、およびC末端位置の一次アンカー残基としてLまたはVがペプチドリガンド中に存在することを特徴とすることが決定され(例えば、非特許文献112参照)、さらに、9アミノ酸ペプチドの2位にI、C末端位置にIまたはAを含むことが見出された(例えば、非特許文献114、128参照)。A*0201対立遺伝子特異的モチーフは、エピトープの2位の一次アンカー残基としてV、A、TまたはQ、およびC末端位置の一次アンカー残基としてMまたはTをさらに含むことも本発明者らによって特定された(例えば、非特許文献119参照)。したがって、HLA−A*0201モチーフは、エピトープの2位の一次アンカー残基としてL、I、V、M、A、TまたはQ、およびC末端位置の一次アンカー残基としてL、I、V、M、AまたはTを有するペプチドリガンドを含む。HLA−A*0201モチーフの一次アンカー位置を特徴づける許容される好ましい残基は、A2スーパーモチーフを記述する残基と同一である。
HLA−A3モチーフは、エピトープの2位の一次アンカー残基としてL、M、V、I、S、A、T、F、C、GまたはDがペプチドリガンド中に存在し、C末端位置の一次アンカー残基としてK、Y、R、H、FまたはAが存在することを特徴とする(例えば、非特許文献129、130参照)。
HLA−Allモチーフは、エピトープの2位の一次アンカー残基としてV、T、M、L、I、S、A、G、N、C、DまたはF、およびC末端位置の一次アンカー残基としてK、R、YまたはHがペプチドリガンド中に存在することを特徴とする(例えば、非特許文献130、131参照)。
HLA−A24モチーフは、エピトープの2位の一次アンカー残基としてY、F、WまたはM、およびC末端位置の一次アンカー残基としてF、L、IまたはWがペプチドリガンド中に存在することを特徴とする(例えば、非特許文献130、132参照)。
本発明のヘテロクリティックアナログを合成した後、それがT細胞応答を誘発する能力について試験することができる。ヘテロクリティックアナログなどのモチーフを有するペプチドの調製および評価は、(特許文献1、2)に記載されている。手短に述べると、特定の抗原由来のエピトープを含むペプチドを合成し、それが適切なHLAタンパク質に結合できる能力について試験する。これらのアッセイは、精製HLAクラスI分子への本発明のペプチドの結合を、放射ヨウ素標識基準ペプチドの結合に関連して評価することを含む。あるいは、空の(すなわち、その中にペプチドを欠く)クラスI分子を発現する細胞を、免疫蛍光染色法およびフロー微小蛍光定量法(flow microfluorimetry)によってそのペプチド結合を評価することができる。ペプチド結合を評価するために使用することができる他のアッセイとしては、ペプチド依存的クラスIアセンブリーアッセイおよび/またはペプチド競合によるCTL認識の阻害などが含まれる。一般に500nM以下の親和性でクラスI分子に結合するペプチドを、感染個体または免疫個体に由来するCTLに対する標的として役立つかどうか、ならびに選択された疾患関連標的細胞と反応可能なCTL集団を生ずることができるインビトロまたはインビボでの一次CTL応答を誘発できるかどうかさらに評価する。
ペプチド合成およびペプチドアナログの産生
これらの実施例に使用するペプチドを表1に示す。腫瘍関連抗原に由来する野生型ヒトCTLエピトープのすべてが、ヒトおよびトランスジェニックマウス系において免疫原性を示した(非特許文献140〜142)。
単一のアミノ酸置換を選択するスキーム
表2は、任意の所与のアミノ酸対の間で割り当てられた類似度である。これによって、所与のアミノ酸置換を、保存的、半保存的または非保存的置換として特徴づけることができた。
アッセイ用材料
1.APC系
HLA−A2.1に関連したペプチドを提示する細胞系を以下のとおり調製した。
MAGE3.112、MAGE2.170、および癌胎児性抗原(CEA)エピトープCEA.691に対してペプチド特異的CTL系を産生するために、正常者から得たPBMCをインビトロで繰り返し上記ペプチドで刺激した(非特許文献140)。手短に述べると、ペプチドを間欠投与した樹状細胞(GM−CSFおよびIL4中で培養したことによって接着PBMCと区別される)を、48ウェルプレート中で抗体被覆ビーズ(Dynal A.S.、Oslo、NorwayまたはMiltenyi Biotec、Auburn、CA)を用いてポジティブ選択することによって得た自己CD8+T細胞と同時培養した。IL2、IL7およびIL10の存在下で7日間培養した後、各PBMC培養物(ウェル)を、ペプチドを間欠投与した接着PBMCを用いてインビトロで再刺激した。次いで、培養物のCTL活性を、ペプチドの存在下または非存在下で、.221A2.1腫瘍APC(A2エピトープ)またはGM3107腫瘍細胞(B7エピトープ)で刺激した後にIFN−γ産生を測定することによって試験した。CTL系を、ペプチドを間欠投与した接着PBMCを用いてさらにインビトロで刺激することによって、ペプチド特異的IFN−γ応答を示すPBMC培養物から発達させた。
エピトープHBV Pol.455およびHIV Pol.476ペプチドに対するCTL系を、HLA−A2.1/Kbxsトランスジェニックマウスにおいて、別に記載されたDNA免疫化(非特許文献141)によって産生させた。HLA−A2.1/KbxsおよびHLA−A2.1/KbxdトランスジェニックマウスはEpimmuneで飼育された。これらの系統は、C57BL/6を背景に産生されたHLA−A2.1/Kbトランスジェニック系統(非特許文献152)と、それぞれSJLまたはBALB/cマウス(Jackson Laboratories、Bar Harbor、ME)とを交雑させたF1世代に相当する。MAGE2.157エピトープに対するCTL系は、8〜12週齢のHLA−A2.1/Kbxsマウスをペプチド50μg、およびIFAに乳化したHBVコア.128ThエピトープTPPAYRPPNAPIL(配列番号34)140μgを尾の付け根に皮下注射して免疫し、初回抗原刺激した脾細胞をペプチドで繰り返しインビトロで再刺激しすることによって産生させた。
アッセイ方法
1.HLA−A2.1またはHLA−B7分子のペプチド結合親和性の測定
HLA−A2.1と放射標識標準ペプチドの結合性に対する、所与の試験ペプチドによって誘導される競合レベルを決定することによって、HLA−A2.1に対する試験ペプチドの結合性を測定した。MHC結合放射能の割合をゲルろ過によって決定し、標識標準ペプチドの結合の50%を阻害する試験ペプチド濃度(IC50)を計算した(非特許文献143、154)。標準ペプチドは、HBVコア.18エピトープ(配列FLPSDFFPSV)(配列番号35)であった。類似のアッセイを実施して、精製HLA−B7(B*0702)分子に対するペプチドの結合親和性を決定した。後者のアッセイでは、放射標識標準ペプチドは、SS 5−13a(L7→Y)ペプチド(配列APRTLVYLL)(配列番号36)であった。
in situ捕獲ELISAを使用してCTLからのIFN−γ放出を測定した(非特許文献155)。手短に述べると、抗マウスIFN−γ(クローンR4−6A2、Pharmingen、San Diego、CA)または抗ヒトIFN−γmAb(クローンNIB42、Pharmingen)をあらかじめ塗布したELISAグレード96ウェル平底ウェル中でAPCおよびペプチドを用いてCTLを刺激した。細胞を培養した後、ウェルを洗浄し、ビオチン化抗マウスIFN−γ(クローンXMG1.2、Pharmingen)または抗ヒトIFN−γ(クローン4S.B3、Pharmingen)mAbを添加し、その後酵素複合ストレプトアビジン(Zymed、South San Francisco、CA)および3,3',5,5'テトラメチルベンジジン基質(ImmunoPure TMB基質キット、Pierce、Rockford、IL)を添加して発達させた。各ウェルの吸光度をLabsystems Multiskan RC ELISAプレートリーダーを用いて450nmで測定した。各ウェル中で産生されたIFN−γレベルを、同じアッセイで作成したマウスまたはヒトIFN−γ標準曲線から外挿して決定した。
ELISPOTアッセイを標準手順に従って実施した(非特許文献156、157)。手短に述べると、96ウェルの平底ニトロセルロースプレート(Immobilon−Pメンブレン、Millipore、Bedford、MA)に抗IFN−γ mAb(10μg/ml、クローンR4−6A2)でコーティングし、終夜4℃でインキュベートした。プレートを、PBSで洗浄後、10%FBSを含むRPMI培地を用いて37℃で1時間ブロックした。磁気ビーズ(Miltenyi、Auburn、CA)によって単離された4×105個の脾臓CD8+細胞、および10μg/mlのペプチドを間欠投与した5×104個のJurkat−A2.1/Kb細胞を各ウェルに添加し、10%FBSを含むRPMI培地中で20時間細胞をインキュベートした。インキュベーション後、このプレートをPBS/0.05%Tweenを用いて徹底的に洗浄し、ビオチン化抗IFN−γ mAb(2μg/ml、クローンXMG1.2)を各ウェルに添加し、プレートを37℃で4時間インキュベートした。次いで、プレートを(0.1%Tween−20を含む)PBSおよびVectastain ABCペルオキシダーゼ(Vectastain Eliteキット;Vector Laboratories、Burlingame、CA)で4回洗浄した。室温で1時間インキュベートした後、プレートを1×PBS/0.05%Tweenで3回洗浄し、その後1×PBSでさらに3回洗浄した。AEC溶液(Sigma Chemical、St.Louis、MO)100μlを添加してスポットを成長させた。この反応を4〜6分後に流水下で停止させた。コンピュータ支援画像分析(Zeiss KS ELISPOTリーダー、Jena、Germany)を用いてスポットを計数した。正味スポット数/106CD8+細胞を以下のとおり計算した:[(関連するペプチドに対するスポット数)−(無関係な対照ペプチドに対するスポット数)]×2.5。
ヘテロクリティック活性によるペプチドアナログのスクリーニング
ヘテロクリティックを設計する法則を決定するために、いくつかの異なるCTL系を、一群のアナログに対する反応性によってスクリーニングした。T細胞刺激能力の改変を、一次MHCアンカーを変えずに実現した。
ヘテロクリティック活性によるペプチドアナログのスクリーニング
A.IFN−γ放出の増大に関連するCEA.691およびMAGE3.112アナログの特定
アナログをスクリーニングする前に、CTL系からのIFN−γ産生のペプチド用量滴定を、広範な野生型ペプチドの用量にわたって実施した。.221A2.1腫瘍細胞に可変用量のペプチドを間欠的に投与し、次いで105個のペプチドを装荷した細胞を、同一数のネズミまたはヒトCTLとともに培養した。37℃で24時間(ネズミ)または48時間(ヒト)のインキュベーション後に、CTLによって放出されるIFN−γレベルをin situ捕獲ELISAアッセイによって測定した。用量滴定曲線を求めた後、野生型ペプチドに対する活性がほとんど検出不可能な場合、最適以下のペプチド用量を選択して、一群のペプチドアナログの抗原性をスクリーニングした。ネズミおよびヒトCTL系のすべての場合で、この最適以下の用量は0.1〜1μg/mlであった。ネズミCTL系は、第3のドメインにネズミH−2Kb配列を含むHLA分子を発現するHLA−A2.1/Kbxsトランスジェニックマウスにおいて産生されるが、すべてが、ネイティブHLA−A2.1分子を発現するAPC上に提示されるペプチドに応答したことに留意されたい。
さらなるヘテロクリティックアナログの特定
A3スーパーファミリーエピトープCEA.61(HLFGYSWYK、配列番号10)およびA24スーパーファミリーエピトープMAGE2.156(EYLQLVFGI、配列番号20)のヘテロクリティックアナログの作製に類似の戦略を採用した。表6に、CEA.61およびMAGE2.156に対して作製したヘテロクリティックアナログの一覧を示す。始めに、応答細胞系によるIFN−γ産生が野生型ペプチドの2倍を超えるかどうかによってアナログをスクリーニングした。アナログのすべてに対する応答が2倍未満の場合、アナログをケース・バイ・ケースでスクリーニングした。
ヘテロクリティックアナログによって誘導されるリンホカインプロファイル
ヘテロクリティックアナログは、T細胞からのサイトカイン産生を異なって活性化することがこれまでに判明している。一部のアナログは、T細胞を特異的に活性化してTh1サイトカインを産生するのに対して、別のアナログはTh2サイトカインの産生を優先的に活性化する。本発明のヘテロクリティックアナログに関連するリンホカイン放出パターンを検討するために、CTL系からのTh2サイトカインIL−10の産生を、IFN−γ産生と比較した。MAGE2.157エピトープを使用した代表的なデータを図7Aおよび7Bに示す。
ヘテロクリティックアナログのHLA−A2.1結合親和性
観測されたCTL系による認識の向上が、アナログエピトープのMHC結合能が偶然増大したためではないことを確認するために、すべてのヘテロクリティックアナログのMHC結合親和性を、精製HLA−A2.1分子を利用してインビトロで測定し、調製Dに記載したそれらの未改変野生型対応物と比較した。
ヘテロクリティックアナログは、腫瘍細胞を認識可能なヒトCTLをインビトロで誘導する
MAGE3.112ペプチドまたはこのエピトープの15およびW7アナログに対して、調製Cに記載したように、PBMCから一次CTLを誘導することによって、MAGE3.112のヘテロクリティックアナログの免疫原性も試験した。2ラウンドのインビトロでの刺激後、48ウェル中のPBMC培養物は、ペプチドの存在下または非存在下で.221−A2.1APCで刺激した後の正味のIFN−γ産生が>100pg/ウェルであり産生がバックグラウンドの少なくとも2倍である場合、CTL誘導に対して正の評点が与えられた。
B7スーパーファミリーCTLエピトープMAGE2.170のヘテロクリティックアナログの特定
HLA−A2以外のHLAスーパータイプファミリーに対する本発明の適用をよりよく定義するために、B7スーパーファミリーエピトープMAGE2.170(配列VPISHLYIL)(配列番号43)のアナログを合成し、前述したA2スーパーファミリーエピトープの場合と類似した方法でスクリーニングした。すべての非アンカー位置における保存的/半保存的および非保存的置換からなるMAGE2.170エピトープアナログの一群を、野生型エピトープに対して産生されたヒトCTL系を用いて、2つの最適以下のペプチド用量でスクリーニングした。前述したように、このスクリーニングアッセイは、野生型ペプチドよりも強いCTL応答を誘導するあらゆるヘ潜在的なテロクリティックアナログを特定するのに役立つ。
HLA B7スーパーファミリーメンバー上のHLA−B7スーパーモチーフに由来するヘテロクリティックアナログの合成および分析
ヘテロクリティック置換法則をさらに有効にするために、追加の試験を、HLA−B7スーパーモチーフ内の配列を有するペプチドに由来するヘテロクリティックアナログを用いて実施する。例えば、このアナログは、インビボでの免疫原性について試験されうる。
野生型ペプチドに対する交差反応性CTLが、アナログで免疫されたマウスにおいて産生されることを確認するために、アナログまたは野生型ペプチドで免疫し、インビトロでさらにCTLを増殖させない脾臓からCD8+細胞を単離する。この材料から、野生型またはアナログに対して反応性であるCTLの前駆体頻度をELISPOTアッセイによって決定する。野生型ペプチドに対して反応性であるCTLの前駆体頻度は、一般に、アナログの前駆体頻度よりもかなり低い。
ヘテロクリティックアナログを、一次インビトロ誘導系におけるCTLの誘導に対して分析することができる。正常ドナーからの新しい未処置のヒトPBMCを、前述した48ウェルプレート中で野生型またはアナログを用いてインビトロで繰り返し刺激する。次いで、ペプチド特異的CTL応答を、野生型ペプチドまたはヘテロクリティックアナログで刺激した培養物中で検出する。これらのアナログによって誘導された培養物は、内因的にプロセシングされ野生型配列を提示する標的を認識することができる。これは、ヘテロクリティックアナログが、細胞上に発現され、内因的に産生される野生型ペプチドを認識する生理学的に関連するヒトCTLを誘導することができ、この現象がヒトおよびトランスジェニックマウス系の両方に関連することを示している。
HLA−A3スーパーファミリー上のHLA−A3スーパーモチーフに由来するヘテロクリティックアナログの合成および分析
ヘテロクリティック置換法則をさらに有効にするために、追加の試験を、HLA−A3スーパーモチーフ内の配列を有するペプチドに由来するヘテロクリティックアナログを用いて実施する。例えば、このアナログは、インビボでの免疫原性について試験されうる。
野生型ペプチドに対する交差反応性CTLが、アナログで免疫されたマウスにおいて産生されることを確認するために、アナログまたは野生型ペプチドで免疫し、インビトロでさらにCTLを増殖させない脾臓からCD8+細胞を単離する。この材料から、野生型またはアナログに対して反応性であるCTLの前駆体頻度をELISPOTアッセイによって決定する。野生型ペプチドに対して反応性のCTLの前駆体頻度は、一般に、アナログの前駆体頻度よりもかなり低い。
ヘテロクリティックアナログを、一次インビトロ誘導系におけるCTLの誘導に対して分析することができる。正常ドナーからの新しい未処置のヒトPBMCを、前述した48ウェルプレート中で野生型またはアナログを用いてインビトロで繰り返し刺激する。次いで、ペプチド特異的CTL応答を、野生型ペプチドまたはヘテロクリティックアナログで刺激した培養物中で検出する。これらのアナログによって誘導された培養物は、内因的にプロセシングされ野生型配列を提示する標的を認識する。これは、ヘテロクリティックアナログが、細胞上に発現され、内因的に産生される野生型ペプチドを認識する生理学的に関連するヒトCTLを誘導し、この現象がヒトおよびトランスジェニックマウス系の両方に関連することを示している。
追加のヘテロクリティックアナログの特定
3個の追加のA2.1制限エピトープ、MAGE2.157 YLQLVFGIEV、配列番号7腫瘍エピトープ、およびウイルス抗原からの2個のエピトープHBV Pol.455、GLSRYVARL(配列番号55)およびHIV Pol.476 ILKEPVHGV(配列番号57)を分析した。これらのエピトープのすべてが、CTLに対して免疫原性であることがこれまでに示されている。
ネズミp53.261エピトープに対するアナログの予測および免疫原性
免疫原性をインビボで試験するために、HLA−A2.1制限ネズミp53.261エピトープを使用した。というのは、このエピトープに対するCTL応答が、HLA−A2.1/Kbトランスジェニックマウスにおいて部分的に寛容化されていることが示されたからである。これによって、予測されたヘテロクリティックアナログがT細胞寛容性をインビボで打破できるかどうかの分析が可能になる。これまでヘテロクリティックアナログは、野生型エピトープに対して産生されたCTL系を用いたインビトロでのスクリーニングによって検出されたが、本発明者らは、置換法則によって特定され、野生型エピトープに対してヘテロクリティックであるアナログがインビボでCTLを潜在的に誘導できると論理的に考えたが、これは寛容な腫瘍関連エピトープに対するワクチンを設計するための興味深い応用である。
野生型との交差反応性
D3およびH3アナログによって誘導されたCTLの交差反応性も、HLA−A2.1/Kbを形質移入したp53を発現するMeth A腫瘍細胞クローンによって自然にプロセシングされた野生型エピトープに対して試験した。野生型エピトープのヘテロクリティックであるp53.261アナログによって産生されたCTLは、Meth A/A2.1Kb腫瘍細胞によって提示される内因的にプロセシングされたp53.261エピトープに応答することが見出された。
Elispotアッセイによる前駆体頻度分析
野生型ペプチドに対する交差反応性CTLが、アナログで免疫されたマウスにおいて産生されることを確認するために、アナログまたは野生型ペプチドで免疫し、インビトロでさらにCTLを発達させないマウスの脾臓細胞からCD8+細胞を単離し、野生型またはアナログに対して反応性であるCTLの前駆体頻度をElispotアッセイを用いて決定した。
HLA A2スーパーファミリーメンバー上のHLA−A2.1スーパーモチーフに由来するヘテロクリティックアナログの合成および分析
A2スーパーファミリー内の他のHLA分子に対してヘテロクリティック置換法則をさらに有効にするために、3、5および7位における3つの保存的/半保存的置換からなる上述した9個のp53.261エピトープアナログの一群の免疫原性を、以下のヒトHLA分子の1つを発現するトランスジェニックマウスにおいてインビボで試験する:A*0202、A*0203、A*0204、A*0205、A*0206、A*0207、A*0209、A*0214、A*6802およびA*6901。

- 請求項40から52のいずれかに記載の2つ以上のポリペプチドを含む組成物。
- 請求項53に記載の2つ以上のポリヌクレオチドを含む組成物。
- 請求項55または56に記載の組成物であって、および賦形剤、およびアジュバント、および脂質からなる群から選択される少なくとも1個の成分を含むことを特徴とする組成物。
- HLA重鎖、β2−ミクログロブリンおよびストレプトアビジンを含むことを特徴とする、請求項54から57のいずれかに記載の組成物。
