Abstract
Description
本出願は、その開示の全体が参照により本明細書に組み込まれる、2002年2月1日に出願された米国仮出願60/353,758について、35 U.S.C.§119(e)に基づく利益を主張する。
本発明は、部分的に、国立衛生研究所(NIH)からの認可番号RO1 GM36624に基づく政府支援、および国立糖尿病・消化器病・腎臓病研究所(NIDDK)からの認可番号5P30-DK36836-14に基づく政府支援によってなされた。政府は、本発明についてある種の権利を有し得る。
本発明は、IIβ型ホスホイノシチドリン酸キナーゼ(PIPKIIβ)関連疾患を処置するための、PIPKIIβ活性の調節に関する。さらに本発明は、PIPKIIβ関連疾患の診断、モニタリングおよび処置へのPIPKIIβ核酸分子およびポリペプチドの使用に関する。本発明はまた、PIPKIIβ関連疾患の処置に有用な、PIPKIIβ活性を調節する剤のスクリーニングにも関する。
最近まで、II型ホスホイノシチドリン酸キナーゼは、ホスファチジルイノシトール−4−リン酸(PI4P)の5位をリン酸化することによりホスファチジルイノシトール−4,5−二リン酸(PI4,5P2)を生成すると考えられてきた。しかしながら、1997年に、これらの酵素が、ホスファチジルイノシトール−5−リン酸(PI5P)からのPI4,5P2の生成に関与する、新規な、以前には知られていない経路中にあることが示された(Rameh et al., 1997)。この経路のための酵素は、哺乳類から蠕虫類まで保存されているが、生物機能に対する重要性は知られていない。
本発明は、部分的に、患者におけるインスリン感受性を高める方法に関し、そしてII型糖尿病、肥満、脂肪の過剰蓄積およびインスリンに対する低下した感受性などの疾患を処置する方法を提供する。
本発明のこれらのおよび他の側面を、以下でさらに説明する。
ホスファチジルイノシトール−5−リン酸(PIP5)からのPI4,5P2の生成を伴う経路の生理学的な役割を決定するために、筋肉に高度に発現しているII型PIPK酵素であるPIPKIIβの発現に障害を有するマウスを作製した。驚くべきことに、PIPKIIβ−/−マウスは、野生型同腹子に比べ、インスリンに対し高感受性であった。雄ノックアウトマウスはまた、普通食または高脂肪食を給餌した場合に、予期せぬことに、野生型同腹子に比べ体脂肪の蓄積が少なかった。PIPKIIβノックアウトマウスは、生存性の低下またはその他の全体的な生理学的異常を何ら示さなかった。
断片は、かかる核酸を同定するため、サザンおよびノザンブロットアッセイにプローブとして用いることができ、または、限定されることなく、RT−PCRおよびリアルタイムPCRを包含するPCRを用いるものなどの、増幅アッセイに用いることができる。当業者に知られているように、サザンおよびノザンブロットなどある種の用途のためには、200、250、300ヌクレオチドまたはそれ以上の大きなプローブが好まれる一方、より小さな断片は、PCRなどの用途に好ましいだろう。断片はまた、抗体を作出するため、もしくはポリペプチド断片の結合を決定するための融合タンパク質を作製するのに用いることができ、または、イムノアッセイの構成要素を作出するのに用いることができる。同様に、断片は、例えば、抗体の作製およびイムノアッセイなどに有用な、PIPKIIβポリペプチドの非融合断片を作製するのに用いることができる。好ましい断片は、PIPKIIβポリペプチドに特異的に結合する基質剤により認識される、触媒的に活性な断片である。
本医薬組成物は、都合良く単位用量形態とすることができ、製薬分野でよく知られた任意の方法により製造することができる。すべての方法は、活性な剤を1種または2種以上の補助的な成分を構成する担体と会合させる工程を含む。一般的に、本組成物は、活性な化合物を、液体担体、微細に分割した固体担体、またはその両方と、均一かつ密接に会合させ、次いで、必要に応じて、製品を成形することにより製造する。
本発明のキットの別の例は、本発明のPIPKIIβ核酸分子の発現のレベルを、ハイブリダイゼーション法によって決定するのに必要な構成要素を備えるキットである。
本発明のキットの別の例は、本発明のPIPKIIβポリペプチドの活性レベルを、酵素アッセイ法によって決定するのに必要な構成要素を備えるキットである。
前記キットは、該キットの種々の構成要素を、診断目的のためにどのように使用するかについての使用説明書またはその他の印刷物を含むことができる。
本発明のいくつかの態様では、1または2以上の対照ペプチドまたはタンパク質分子が、基材に付着している。好ましくは、対照ペプチドまたはタンパク質分子により、ペプチドまたはタンパク質の品質および結合特性、試薬の品質および有効性、ハイブリダイゼーションの成否、および、分析の閾値および成否などの因子を決定することが可能となる。
別の態様では、基材は、プローブの基材への結合を増強する化合物でコートされていてもよい。かかる化合物は、限定されることなく、ポリリシン、アミノシラン、アミノ反応性シラン(Chipping Forecast, 1999)またはクロムを包含する。この態様では、接触プリント方式で、または、インクジェットもしくはピエゾ電気送達などの非接触プリント方式でプローブを基材に適用するコンピュータ制御ロボットを用いることにより、事前に合成したプローブを、正確な、所定の容量およびグリッドパターンで基材に適用する。プローブは、UV照射を包含するが、これに限定されない方法により、基材に共有結合させてもよい。別な態様では、プローブは熱で基材と連結される。
本発明の態様では、1または2以上の対照核酸分子が、基材に付着している。好ましくは、対照核酸分子により、核酸の品質および結合特性、試薬の品質および有効性、ハイブリダイゼーションの成否、および、分析の閾値および成否などの因子を決定することが可能となる。対照核酸は、限定されることなく、ハウスキーピング遺伝子などの遺伝子の発現産物、またはその断片を包含することができる。
いくつかの態様では、1または2以上の対照核酸分子が、基材に付着している。好ましくは、対照核酸分子は、結合特性、試薬の品質および有効性、ハイブリダイゼーションの成否、および、分析の閾値および成否などの因子を決定することを許容する。

方法
PIPKIIβ −/− マウスの作出
PIPKIIβの発現を欠くマウスは、Lexicon Genetics, Inc.(The Woodlands, TX)から得たPIPKIIβ+/−ES細胞から作出した。Lexiconは、レトロウイルス挿入ジーントラップ法を用いて、129Sv/Evの遺伝的背景を有するマウス胚性幹細胞(Zambrowicz et al., 1998)においてランダムに遺伝子を破壊した。PIPKIIβ遺伝子座が破壊されていることが報告されたクローン#39557は、Lexicon Geneticsから得た。これらの細胞は、Lexiconのプロトコルに従って発明者らの実験室で成長および増殖させ、Beth Israel Deaconess Transgenic Facilityにて、胚盤胞に注入した。3匹のキメラマウスが得られ、それぞれ、2匹の野生型C57Bl/6マウスの雌と交配させ、C57Bl/6×129Sv/Evの混合された遺伝的背景のコロニーを確立した。すべての実験は、C57Bl/6×129Sv/Evの遺伝的背景におけるPIPKIIβ+/−マウス間の交雑に由来する野生型、ヘテロ接合およびノックアウト同腹子で行った。
プライマー3種の組を用いて、野生型試料、または、ノックアウト試料のいずれかに存在するゲノムDNAの領域を増幅した。このために、イントロンの塩基924-942に相当する単一のアンチセンスプライマー(pR)を用いた。2つのセンスプライマー、一方は、イントロンの塩基327-350に相当するもの(pwtF)、そして他方は、Lexiconの挿入の3’末端の範囲にあるもの(pkoF)も用いた。挿入物の第2カセットのコード部分の3’末端の配列は、挿入物の境界に及ぶPCRによって得られた4kbの断片を配列決定することにより決定した。用いたプライマーの配列は、pR:5’−ACC ATC CCA AAG CAC CCA GGA CC−3’(配列番号3)、pwtF:5’−CGT GCGT ATG CCG TCG TCG TTT CC−3’(配列番号4)、pkoF:5’−AGA AGC GAG AAG CGA ACT GAT TGG−3’(配列番号5)であった。プライマー対pwtF/pRは597bpの断片を増幅することが予想され、プライマー対pkoF/pRは651bp の断片を増幅することが予想された。
内因性のPIPKIIβ転写物に相補的なcDNAは、PIPKIIβのエクソンVIIIにおける29塩基に相補的なプライマーを用いて作製した(5’−CCT CGT CCT CTG CCC GCT CCT CCA CCT CC−3’、配列番号6)。内因性のコード配列の塩基51-902に相当する断片は、エクソンIからのフォワードプライマー(5’−CGC CAG CAA GAC AAG ACC AAG AAG AAG−3’、配列番号7)と、エクソンVIIIに対する入れ子リバースプライマー(5’−CGC TCC TCC ACC TCC ATC TCC TCC−3’、配列番号8)を用いて増幅した。
PIPKIIβの第1エクソンを、Lexiconのレトロウイルス挿入ベクターの5’カセットにスプライシングすることにより生成されたハイブリッド転写物に相補的なcDNAは、Lexiconベクター内のβgeo配列に相補的なプライマーを用いて作製した。ハイブリッド転写物からの断片は、エクソンIからのフォワードプライマー(上記のもの)と、Lexicon挿入ベクターにおけるβgeo配列からの入れ子リバースプライマー(5’−GCA TCC TTC AGC CCC TTG TTG−3’、配列番号9)とを用いて増幅した。
身体組成分析は、2つの方法によって行った。二重エネルギーX線吸収スキャン(DEXAScan)を、10週齢および26週齢でマウスの体脂肪量を測定するのに用いた。死体分析(carcass analysis)(サポニン化と、それに引き続くグリセロール含量のアッセイ)を、36週齢のマウスの脂肪含量を測定するのに用いた。
PIXIMusデンシトメータ(GE Medical Systems, Waukesha, WI)を用いて、PIPKIIβ−/−マウスの脂肪組織の量を分析した。マウスは、ケタミン/キシラジンで麻酔し、測定のために装置上においた。
マウスを解剖し、胃および腸の内容物を除去し、空の胃および腸を死体に戻した。死体は、重量を測定した後、60℃のオーブン中に置き、3週間までの間乾燥した。死体は、連日重量を測定し、いつ水分が完全に蒸発したかを決定した。乾燥の前と後の重量の変化を、各死体の水分重量とみなした。乾燥後、死体を、30%水酸化カリウム1部と100%エタノール2部との溶液中で、60℃にて1週間までの間サポニン化した。得られた死体に存在するグリセロールの量を、酵素による変換、およびSigmaトリグリセリド試薬A(Sigma #337-40-A)による比色検出、およびトリグリセリド標準物質(Sigma #339-11)(Sigma-Aldrich, St. Louis MO)との比較によって決定した。
マウスを午前10:00に清潔なケージ(餌なし)に置き、午後1:00にノボリン−R(Novo Nordisk Pharmaceuticals Inc, Princeton, NJ)を、体重1kgあたり0.5〜0.75単位の用量で腹腔内注射した。血糖値は、尾部出血により、One Touch Basicグルコメーター用いて、インスリン注射の前、および、インスリン注射の15、30、45、60および90分後に測定した。
身体組成分析実験の結果は、studentのt検定により統計学的有意性について分析した。インスリン耐性試験の結果は、反復測定ANOVAにより分析した。すべての統計分析は、StatView software version 4.1(StatView Software, Cary, NC)で行った。
触媒的に障害されたPIPKIIβD278Aは、CLONTECH部位特異的変異導入キット(CLONTECH Laboratories Inc., Palo Alto, CA)で構築した。細菌により発現させた組み換えPIPKIIβD278Aのキナーゼ活性を、ホスファチジルセリン90%およびEchelon, Inc.からの合成PI5P 10%の基質を用いたキナーゼアッセイを行うことにより、野生型と比較した。PIPKIIβD278Aは、野生型PIPKIIβのキナーゼ活性の約3%を有することが見出された。
CHO−HIR細胞およびCos−7細胞は、DMEM+10%ウシ胎児血清中で増殖させ、DEAE/デキストラントランスフェクションによりトランスフェクトした。細胞は、血清を欠乏させ、10nMのインスリンで10分間刺激した。細胞は、免疫沈降およびウェスタンブロット法による分析のために、NP40ベースの溶解バッファー中で、トランスフェクション約48時間後に溶解した。
CHO−HIR細胞を、対照ベクター、または種々のPIPK遺伝子を発現するベクターでトランスフェクトした。HAタグ付きAktをレポーター遺伝子としてトランスフェクトした。細胞は、血清を欠乏させ、10nMのインスリンで溶解前に10分間刺激し、またはしなかった。HA−Atkを溶解物から免疫沈降し、抗pT308抗体でブロッティングし、その活性状態を決定した。また、免疫沈降物を抗HA−Akt抗体、抗PIPKIIβ抗体および抗ホスホチロシン(pTyr)でもブロッティングした。
in vivo標識は、細胞を、32P−ATPの存在下で、インスリン刺激(10nMインスリン中で10分)および溶解の前4時間増殖させることによって行った。脂質は、クロロホルム−メタノールで抽出し、脱アシル化し、分離およびPIP3レベルの検出のために、高速液体クロマトグラフィー(HPLC)に供した。
PIPKIIβ −/− マウス
PIPKIIβの発現を欠くマウスは、Lexicon Genetics, Inc.から得たPIPKIIβ+/−ES細胞から作出した。マウスはPCRにより遺伝子タイピングし、PIPKIIβ遺伝子発現の破壊は、複数の組織のRT−PCR分析により確認した。これらのノックアウトマウスに関する発明者らの初期の研究により、PIPKIIβがインスリン応答性および体脂肪蓄積に関与していることが決定された。PIPKIIβ−/−マウスは生育可能であり、繁殖力があり、概して健康である。PIPKIIβ+/−間の交雑により、261匹の子孫を得、このうち60匹(23%)はPIPKIIβ+/+であり、145匹(55.5%)はPIPKIIβ+/−であり、56匹(21.5%)はPIPKIIβ−/−であった。この数は、常染色体遺伝子のメンデルの期待値の範囲内であり、PIPKIIβ遺伝子発現の破壊は、致死をもたらさないことを示唆するものである。PIPKIIβマウスは、その後、明確な病理組織学的異常なしに、2年間にわたって生存することが観察された。
PIPKIIβの欠損がマウスにおいてインスリン感受性を増大させたため、この酵素の過剰発現が細胞株におけるインスリンシグナル伝達に影響を与え得るかどうかを調査した。CHO−HIR細胞は、ヒトインスリン受容体を発現しており、インスリン受容体活性化の下流の細胞内シグナル伝達事象を研究するのに頻繁に用いられる。CHO−HIR細胞を、野生型PIPKIIβ、PIPKIIβD278A(触媒活性の減少した突然変異体)、または、I型PIPキナーゼであるPIPKIαもしくはPIPKIβ、または、ベクターのみでトランスフェクトした。トランスフェクトした細胞は、インスリン(10nMインスリンで10分間)で刺激し、溶解し、HA−Aktを溶解物から免疫沈降し、抗pT308抗体でブロッティングし、その活性化状態を決定した。Aktは、この酵素の活性化形態に特異的な抗体でのブロッティングにより判断したところ、活性化していた(図6)。しかしながら、PIPKIIβを過剰発現している細胞では、Aktのインスリン依存性の活性化の有意な減少があった。この効果は、触媒的に障害されたPIPKIIβ変異体(PIPKIIβD278A)を用いた場合は、それほど明確ではなかった。I型PIPキナーゼであるPIPKIαまたはPIPKIβの過剰発現は、Aktの活性化に効果がなかった。これらの結果は、高レベルのPIPKIIβ活性は、Aktのインスリン依存性活性化の阻止をもたらすことを示唆する。
別な可能性は、PIPKIIβが、Aktを活性化するのに必要なPI3Kの産物であるPIP3の分解を促進するというものである。PIP3はインスリンに応答してPI3Kにより生成され、SHIP2などのホスファターゼにより分解される。構成的に活性なPI3Kと、PIPKIIβおよび/またはSHIP2とでトランスフェクトしたCos細胞におけるPIP3のレベルを調査した。細胞は、血清を欠乏させ、[3H]−イノシトールで24時間標識した。脂肪を抽出し、脱アシル化し、PtdIns−3,4,5−P3レベルをHPLCで分析した。PIPKIIβまたはSHIP2のいずれかの発現は、PIP3の減少をもたらし、両方のタンパク質の発現は、PIP3のほぼ完全な枯渇をもたらした(図7)。したがって、減少したAktのインスリンに応答した活性は、PIPKIIβを過剰発現した細胞におけるPIP3の増強された分解により説明できる。PIPKIIβがPIP3生成に影響を与える機構をさらに調べるために、脂質ホスファターゼSHIP2の活性および局在化に対するその効果を決定した。

Claims (95)
- II型糖尿病を有するか、または有する疑いがある対象を処置する方法であって、かかる処置が必要な対象に、対象におけるPIPKIIβの活性を減少させる剤の有効量を、II型糖尿病の処置として投与することを含む、前記方法。
- 組織のインスリンに対する感受性を増大させる薬剤を対象に投与することをさらに含む、請求項1に記載の方法。
- 薬剤が、メトホルミン、ピオグリタゾンおよびロシグリタゾンからなる群から選択される、請求項2に記載の方法。
- インスリン放出を増大させる薬剤をさらに投与することを含む、請求項1に記載の方法。
- 薬剤が、スルホニル尿素、ナテグリニドおよびレパグリニドからなる群から選択される、請求項4に記載の方法。
