【従来の技術および発明が解決しようとする課題】
β−ヒドロキシアミノ酸誘導体は、一般的に医薬品の中間体として、重要な化合物であり、多くの合成法法が開発されている。例えば、Synthesis, 34-36, (1990)には、反応工程式1で示される方法が記載されている。

反応工程式1で示される従来法では、β−ヒドロキシアミノ酸として、(2S,3S)−2−アミノ−3−ヒドロキシ−4−メチルペンタン酸の合成方法が知られている。そこで、本発明者らは、出発原料としてホルミルシクロヘキサンを用いて、反応工程式1に示される方法と同様の方法で、(2R,3R)−2−アミノ−3−ヒドロキシ−3−シクロヘキシルプロパン酸が製造されることを確認し、さらに保護反応を行うことによって、(2R,3R)−2−(t−ブトキシカルボニル)アミノ−3−ヒドロキシ−3−シクロヘキシルプロパン酸が製造できることを確認した(反応工程式2)。

反応工程式中、

しかしながら、上記した従来の方法では、以下に示したような問題点があった。
(1)ステップ4において、過酸を用いるために、大量合成においては、爆発の危険性がある。
(2)ステップ4において、低温反応が存在するため、大量合成には、不向きである
(3)ステップ5においての酸化反応は、大量合成においては、爆発の危険性がある。
(4)総ステップ数が7ステップであり、ステップ数が多い。
(5)総収率が22.0%であり、総収率が低い。

以上の点から、従来の方法は、必ずしも満足いくものではなかった。そこで、上記した問題点を解決する方法の開発が望まれていた。

一方、反応工程式3に示したピネン誘導体を用いたβ−ヒドロキシアミノ酸誘導体の合成方法が知られている(Tetrahedron Letters, 39, 2191-2194 (1998)、J. Org. Chem., 59, 3240-3242 (1994) 参照)。

これらの文献には、ホルミルシクロヘキサン誘導体についての反応は、全く記載されていない。

【問題解決を解決するための手段】
本発明者らは、上記の目的を達成すべく鋭意研究を行なった結果、下記反応工程式4で示される方法を見出した。

さらに詳しくは、
ステップ1:一般式(II)で示されるピネン誘導体と式(III)で示されるホルミルシクロヘキサンをジクロロメタン中、チタニウムクロライドトリエトキシド、トリエチルアミン存在下、0℃で不斉アルドール反応に付し、一般式(I)で示されるピネン誘導体およびそれらの塩を製造し、
ステップ2:一般式(I)で示されるピネン誘導体およびそれらの塩をテトラヒドロフラン中、1.2N塩酸存在下、室温で加水分解反応に付し、一般式(IV)で示されるヒドロキシアミノ酸誘導体およびそれらの塩を製造し、
ステップ3:一般式(IV)で示されるヒドロキシアミノ酸誘導体およびそれらの塩をエタノール中、1N水酸化リチウム水溶液存在下、室温で加水分解反応に付し、さらに、ジ−t−ブチル ジカーボネート存在下、保護反応に付し、式(V)で示される(2R,3R)−2−(t−ブトキシカルボニル)アミノ−3−ヒドロキシ−3−シクロヘキシルプロパン酸およびそれらの塩の製造方法を見出した。

本発明の方法によれば、反応工程式2で示された従来技術の問題点がすべて解決した。すなわち、
(1)すべてのステップにおいて、過酸を用いないために、爆発の危険性がない。
(2)すべてのステップにおいて、低温反応が存在しないため、大量合成には、有利である。
(3)すべてのステップにおいて、酸化反応がなく、爆発の危険性がない。
(4)総ステップ数が3ステップであり、従来の方法に比べステップ数が短い。
(5)総収率が64.8%であり、従来の方法に比べ総収率が高い。
(6)ステップ2において、(1R,2R,5R)−(+)−2−ヒドロキシ−3−ピナノンが回収され、この回収された化合物を再度グリシン誘導体と反応させることによって、一般式(II)で示される化合物を再合成することができる。すなわち、光学補助剤である(1R,2R,5R)−(+)−2−ヒドロキシ−3−ピナノンが回収でき、再利用可能である。

このようなことから、本発明の製造方法は、従来の方法に比べて、大量合成に有利な方法であるといえる。

各反応の生成物は、工程ごとに単離、洗浄、乾燥、精製を行ない次の反応に供してもよいし、またそれらの操作をまったく行なわないか、あるいは適当な段階で中止し、次の工程に進んでもよい。各反応における反応生成物は、通常の精製手段、例えば常圧下または減圧下における蒸留、シリカゲルまたはケイ酸マグネシウムを用いた高速液体クロマトグラフィ、薄層クロマトグラフィ、カラムクロマトグラフィ、洗浄、再結晶等の方法により精製することができる。

グリシンエチルエステル・塩酸塩(9.21g)のベンゼン(50ml)懸濁液にトリエチルアミン(9.2ml)を加えた。混合物を70℃で10分間攪拌した。混合物に、(1R,2R,5R)−2−ヒドロキシ−3−ピナノン(5.55g、99%e.e.)と三フッ化ホウ素・エーテル錯体(90μl)を加えた。反応混合物をDean-Starkを用いて、2時間還流した。析出した不溶物をろ過し、ろ液を濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン:酢酸エチル=2:1→1:1→1:2→0:1)によって精製し、以下の物性値を有する標題化合物(8.23g、99%)を得た。

参考例1で製造した化合物(253mg)のジクロロメタン(1.5ml)溶液に、アルゴン雰囲気下、0℃でチタニウムクロライドトリエトキシド(218mg)のジクロロメタン(1ml)溶液を加えた。この溶液に、ホルミルシクロヘキサン(218mg)とトリエチルアミン(0.28ml)を加えた。反応混合物を同温度で4時間攪拌した。反応混合物を飽和塩化ナトリウム水溶液に注ぎ、析出した不溶物をろ過し、ろ液を酢酸エチルで抽出した。抽出物を無水硫酸マグネシウムで乾燥し、濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン:酢酸エチル=5:1→3:1)によって精製し、以下の物性値を有する本発明化合物(309mg、84%)を得た。

実施例1で製造した化合物(300mg)のテトラヒドロフラン(2.4ml)溶液に1.2N塩酸(5.5ml)を加えた。反応混合物を室温で3日間攪拌した。反応混合物を濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ジクロロメタン:メタノール=70:1→20:1→10:1)によって精製し、以下の物性値を有する本発明化合物(196mg、95%)を得た。また、ピナノン(65mg、47%)を回収した。

実施例2で製造した化合物(190mg)のエタノール(8ml)溶液に、1N水酸化リチウム水溶液(1.8ml)を加えた。反応混合物を室温で3.5時間攪拌した。反応混合物に、ジ−t−ブチルジカーボネート(0.24ml)を加えた。反応混合物を室温で一晩攪拌した。反応混合物を水に注ぎ、t−ブチルメチルエーテルで洗浄した。水層に5%硫酸水素カリウム水溶液をpH3になるまで加えた。水層を酢酸エチルで抽出した。抽出物を飽和塩化ナトリウム水溶液で洗浄し、無水硫酸マグネシウムで乾燥し、濃縮し、以下の物性値を有する本発明化合物(175mg、81%、97.3%e.e.)を得た。

[Background Art and Problems to be Solved by the Invention]
β-hydroxyamino acid derivatives are generally important compounds as intermediates for pharmaceuticals, and many synthetic methods have been developed. For example, Synthesis, 34-36, (1990) describes the method represented by reaction scheme 1.

In the conventional method represented by the reaction process formula 1, a method for synthesizing (2S, 3S) -2-amino-3-hydroxy-4-methylpentanoic acid is known as a β-hydroxyamino acid. Therefore, the present inventors have used (2R, 3R) -2-amino-3-hydroxy-3-cyclohexylpropanoic acid by a method similar to the method shown in Reaction Scheme 1 using formylcyclohexane as a starting material. As a result of intensive studies to achieve the above object, the present inventors have found a method represented by the following reaction process formula 4.

Step 1: A pinene derivative represented by the general formula (II) and a formylcyclohexane represented by the formula (III) are subjected to an asymmetric aldol reaction at 0 ° C. in the presence of titanium chloride triethoxide and triethylamine in dichloromethane. According to the method of the present invention, all the problems of the prior art shown by the reaction process formula 2 are solved. That is,
(1) Since no peracid is used in all steps, there is no risk of explosion.
(2) Since there is no low-temperature reaction in all steps, it is advantageous for mass synthesis.
