本発明は、比色により試料中の特定成分を分析する際に使用される分析用具に関する。

【従来の技術】
比色により血糖値を測定する際に利用される分析用具としては、たとえば図9に示したグルコースセンサ9がある。このグルコースセンサ9は、第1および第2透明板材91,92を、一対のスペーサ93を介して接合した形態を有しており、各要素91〜93により、キャピラリ94が規定されたものである。キャピラリ94の内部には、血液が供給されたときに溶解する試薬部95が設けられている。この試薬部95は、発色剤、酸化還元酵素および電子伝達物質などの反応成分を含むものとして構成される。

このようなグルコースセンサ9では、開口96を介して血液を導入した場合、キャピラリ94の内部において生じる毛細管力により、キャピラリ94に導入された血液が開口97に向けて移動する。このとき、試薬部95が溶解し、キャピラリ94の内部には、グルコース、発色剤、酸化還元酵素および電子伝達物質を含む液相反応系が構築される。

【発明が解決しようとする課題】
上述のように、発色剤を発色させるためには、少なくとも、グルコースから電子を取り出す反応と、取り出した電子を発色剤に供給する反応が必要となる。一方、液相反応系において効率よく発色剤を発色させ、測定時間を短くするためには、液相反応系において、試薬部の反応成分を均一に分散させる必要がある。しかしながら、試薬部として、試料の供給により溶解する構成を採用した場合には、局所的に反応成分の濃度が高い状態を経た後、反応成分が経時的に拡散することにより、反応成分の濃度が徐々に均一化されていく。そのため、グルコースセンサ9を用いた濃度測定では、測定時間が反応成分の拡散性に依存する傾向にある。また、グルコースは、血液をキャピラリ94に導入した初期段階では液相反応系において略均一濃度で存在するが、反応の進行にともなってグルコースが消費され、未反応のグルコースの濃度は、反応成分の濃度が高い部分において低くなる。そのため、反応成分ばかりでなく、グルコース濃度の濃度分布ひいてはグルコースの拡散性も測定時間に影響を与えることとなる。

グルコースセンサ9では、第1透明基板と第2透明基板との距離が、小さくても、200〜300μmに設定されており、しかも試薬部95が第1透明基板91にのみ形成されている。そのため、液相反応系において試薬部95に含まれる反応成分の濃度を均一化させるためには、目的成分の拡散距離が大きくなってしまう。もちろん、グルコースの拡散距離も大きくなる。その結果、グルコースセンサ9では、目的とする反応状態(液相反応系の着色)を得るための時間が比較的に長く、測定時間が長いといった問題がある。この場合に、測定時間を短く設定すれば、発色剤が十分に発色していないために、高濃度領域での測定精度が低下し、また測定精度を十分に確保しようとすれば測定レンジが狭くなる。

本発明は、このような事情のもとに考えだされたものであって、分析時間を短くでき、また高濃度領域においても精度よく分析できる比色用の分析用具を提供することを課題としている。

【発明の開示】
本発明では、上記した課題を解決するため、次の技術的手段を講じている。すなわち、本発明により提供される分析用具は、試料中の特定成分と試薬を反応させるための反応空間を備え、かつ比色により上記特定成分を分析する際に利用される比色分析用具であって、上記反応空間を規定する面には、試薬を保持させるための試薬保持領域と、上記試薬保持領域の法線方向において上記試薬保持領域に対面して存在し、かつ試薬が保持されない対面領域と、が設定されており、上記試薬保持領域と上記対面領域との最短対面距離が150μm以下であることを特徴としている。

本発明では、「対面」という用語は、特段の限定がない限りは、平面どうしの状態ばかりでなく、平面と曲面との状態および曲面どうしの状態も含むものとして使用している。また、「対面距離」とは、試薬が試薬保持領域からその法線方向に拡散したときに、対面領域に到達するのに必要な距離の最大値を意味している。

以下、本発明の好ましい実施の形態について、図面を参照しつつ具体的に説明する。

図1ないし図3に示したグルコースセンサXは、使い捨てとして構成されたものであり、比色によりグルコース濃度を測定することができるように構成されたものである。このグルコースセンサXは、長矩形の第1および第2板材1,2を、一対のスペーサ3を介して接合した形態を有しており、各要素1〜3により、第1および第2板材1,2の長手方向に延びるキャピラリ4が規定されている。

(グルコースセンサの作成方法)
本実施例においては、3種類のグルコースセンサについて測定時間を検討した。これらのグルコースセンサは、基本的には図1ないし図3に示したグルコースセンサXと同様な構成であり、両面テープ(スペーサ)の厚み寸法を規定することにより、キャピラリの高さ寸法(対面距離)が異なったものとされている。

各グルコースセンサの作成においては、まず、寸法が10mm×30mm×250μmであるPET製の第1透明板に、一対の両面テープを3mmの間隔を隔てて貼着した。両面テープは、キャピラリの厚み寸法を規定するものであるが、各グルコースセンサに使用した両面テープの厚みは、表1に示した通りである。次いで、上記間隔における3mm×3mmの領域に試薬液を分注した後に、試薬液を送風乾燥(30℃、10%Rh)させて試薬部を形成した。各グルコースセンサを作成するときの試薬液の分注量は、表1に示した通りである。すなわち、試薬液の分注量は、キャピラリの容積に応じて設定されており、3種類のグルコースセンサ1〜3は、キャピラリに血液を導入したときにキャピラリ内での試薬濃度が同一となるように構成されている。続いて、寸法が10mm×30mm×250μmであるPET製の第2透明板を、先の両面テープを介して第1透明板に接合することにより本実施例において使用するグルコースセンサを得た。
