本発明はα−グルコシダーゼインヒビター及びα−アミラーゼインヒビターを含む栄養サプリメント組成物に関する。更に詳しくは、本発明はリパーゼインヒビターの実質的に不在下に有効量のα−グルコシダーゼインヒビター及びα−アミラーゼインヒビターを含む栄養サプリメント組成物に関する。また、本発明は食品に含まれる炭水化物の吸収の制限方法、及び個人の体重減の促進方法に関する。

【発明が解決しようとする課題】
炭水化物は多数の食品中に存在する栄養化合物である。それらは種々のサイズ及び形状で生じる多種の化合物のグループである。全ての炭水化物はグリコシド結合により一緒に結合された1個以上の糖単位を含む。澱粉の如き複雑な炭水化物は多分岐鎖構造に形成された多くの反復単位からなる比較的大きい分子である。一方、単糖類及び二糖類は夫々単一及び二つの糖単位を含む簡単な炭水化物である。
人体は複雑な炭水化物を比較的簡単な単位、単糖類に分解し、次いでこれらの単位を腸の如き組織中に吸収することによりそれらを消化する。多種の消化酵素が段階的様式で作用して複雑な炭水化物を吸収可能な単位に分解する。初期の工程において、アミラーゼとして知られている唾液及び膵臓の酵素が大きい炭水化物をオリゴ糖及び二糖類の一層小さい単位に分解する。炭水化物消化の最終工程の一つにおいて、小腸の刷子縁中のα−グルコシダーゼ酵素が二糖類マルトース及び蔗糖をそれらの夫々の単糖類単位に分解し、次いでこれらが生体により吸収され得る。

【課題を解決するための手段】
本発明はリパーゼインヒビターの実質的な不在下にα−グルコシダーゼインヒビター及びα−アミラーゼインヒビターを有する栄養サプリメント組成物を含む。本発明者らはリパーゼインヒビターの実質的に不在下のα−グルコシダーゼインヒビターとα−アミラーゼインヒビターの組み合わせが炭水化物吸収の有効なインヒビターを与えることを発見した。本発明の組成物は食品中に含まれる炭水化物の吸収を制限し、体重減を促進するのに特に有益である。
驚くべきことに、本発明者らはα−グルコシダーゼインヒビターであるトウチ(touchi)抽出物と、α−アミラーゼインヒビターであるファセオラミンの組み合わせが、本発明の栄養サプリメント組成物に予想されるよりも有効に炭水化物吸収をブロックさせる相乗効果を生じることを発見した。トウチはペーストの形態のシーズニングとして主として使用される伝統的な中国の食物である。トウチはアスペルギルス種菌類であるコウジにより発酵されたスチーム処理大豆から誘導される。本発明の栄養サプリメント組成物中に使用されるトウチはトウチの抽出物である。トウチ抽出物は水抽出物であることが望ましい。

望ましい実施態様において、本発明の栄養サプリメント組成物は組成物の約7〜35質量%の量のトウチの抽出物を含む。トウチ抽出物は組成物の約13〜27質量%の量で存在することが更に望ましい。トウチ抽出物が組成物の約18〜19質量%の量で存在することが特に望ましい。

本発明の組成物は更に駆風薬を含んでもよい。望ましい実施態様において、有機パセリが駆風薬として使用される。

【発明の実施の形態】

栄養サプリメント組成物はリパーゼインヒビターを実質的に含まない。本明細書に使用される“リパーゼインヒビターを実質的に含まない”という用語は組成物が許容し得る汚染により、又はα−グルコシダーゼ成分及び/又はα−アミラーゼ成分により示されるリパーゼ抑制活性のレベルにより存在するリパーゼインヒビターの量のみを含むことを意味する。許容し得る汚染により存在するリパーゼインヒビター(即ち、α−グルコシダーゼインヒビター及びα−アミラーゼインヒビターを除く)は栄養サプリメント組成物の0.001質量%未満を構成することが望ましい。

TECHNICAL FIELD OF THE INVENTION
The present invention relates to a nutritional supplement composition comprising an α-glucosidase inhibitor and an α-amylase inhibitor. More particularly, the invention relates to nutritional supplement compositions comprising an effective amount of an α-glucosidase inhibitor and an α-amylase inhibitor in the substantial absence of a lipase inhibitor. The invention also relates to a method for limiting the absorption of carbohydrates contained in foods and a method for promoting weight loss of individuals.

[Problems to be solved by the invention]
Carbohydrates are nutritional compounds that are present in many foods. They are a group of different compounds that occur in different sizes and shapes. All carbohydrates contain one or more sugar units linked together by glycosidic bonds. Complex carbohydrates, such as starch, are relatively large molecules composed of many repeating units formed in a multibranched structure. Monosaccharides and disaccharides, on the other hand, are simple carbohydrates containing single and two sugar units, respectively.
The human body breaks down complex carbohydrates into relatively simple units, monosaccharides, and then digests them by absorbing these units into tissues such as the gut. A variety of digestive enzymes act in a stepwise manner to break down complex carbohydrates into absorbable units. In an early step, salivary and pancreatic enzymes known as amylases break down large carbohydrates into smaller units of oligosaccharides and disaccharides. In one of the final steps of carbohydrate digestion, the α-glucosidase enzyme in the brush border of the small intestine breaks down the disaccharide maltose and sucrose into their respective monosaccharide units, which can then be absorbed by the organism.

A balanced diet contains several types of nutritional compounds, including carbohydrates. However, recent suggestions indicate that unbalanced levels of carbohydrates in the diet may contribute to obesity. As a result, several types of low-carbohydrate and non-carbohydrate foods and meals have been proposed.
Given the common use of carbohydrates in many diets around the world, it is difficult for individuals to avoid carbohydrates. In addition, individuals interested in losing weight may not want to sacrifice the enjoyment of foods containing carbohydrates. As a result, considerable attention has been focused on finding alternatives to reducing and / or eliminating carbohydrates from the diet.
Enzymes that digest carbohydrates into absorbable units offer the opportunity to prevent the absorption of these compounds. By inhibiting the function of the appropriate enzymes, carbohydrates will not be broken down into their absorbable units and will pass through the organism without being absorbed. This allows individuals to enjoy carbohydrate-containing foods and still avoid carbohydrate absorption.

The art has several examples of compositions that prevent the absorption of carbohydrates. For example, U.S. Patent No. 6,200,958 to Odaka et al. Discloses a composition comprising an [alpha] -glucosidase inhibitor. The composition is used to delay the digestion of carbohydrates such as starch and sucrose. US Patent No. 5,643,974 to Bremer et al. Discloses a nutritional supplement composition comprising a glucosidase inhibitor and / or an amylase inhibitor in the presence of a lipase inhibitor. The composition prevents the absorption of carbohydrates and can be used to treat obesity. US Pat. No. 5,643,974 teaches that the presence of a lipase inhibitor is important to the function of the composition.
Although the art includes these and other examples of compositions that limit the absorption of carbohydrates, there is a need for compositions that have improved ability to prevent such absorption.

[Means for Solving the Problems]
The present invention includes a nutritional supplement composition having an α-glucosidase inhibitor and an α-amylase inhibitor in the substantial absence of a lipase inhibitor. The present inventors have discovered that a combination of an α-glucosidase inhibitor and an α-amylase inhibitor in the substantial absence of a lipase inhibitor provides an effective inhibitor of carbohydrate absorption. The compositions of the present invention are particularly beneficial for limiting the absorption of carbohydrates contained in foods and promoting weight loss.
Surprisingly, we have found that the combination of the α-glucosidase inhibitor touchi extract and the α-amylase inhibitor phaseolamine are more effective than would be expected in the nutritional supplement compositions of the present invention. It has been found that it produces a synergistic effect that blocks carbohydrate absorption. Torch is a traditional Chinese food used primarily as seasoning in the form of a paste. Torch is derived from steam-treated soybeans that have been fermented by koji, an Aspergillus species fungus. The torch used in the nutritional supplement composition of the present invention is an extract of torch. Preferably, the torch extract is an aqueous extract.

In a preferred embodiment, the nutritional supplement composition of the present invention comprises an extract of Touchi in an amount of about 7-35% by weight of the composition. More preferably, the torch extract is present in an amount of about 13-27% by weight of the composition. It is particularly desirable that the torch extract be present in an amount of about 18-19% by weight of the composition.
Phaseolamine is a partially purified protein extract of white kidney bean that binds to α-amylase enzymes, which are responsible for the digestive degradation of starch. It has been proposed that phaseolamine binds α-amylase in a non-competitive manner to prevent hydrolysis of α-1,4-glycosidic bonds in the starch molecule, thereby suppressing α-amylase degradation of starch.
In a preferred embodiment, the nutritional supplement composition of the present invention comprises phaseolamine in an amount of about 15-48% by weight of the composition. More desirably, the phaseolamine is present in an amount of about 24-40% by weight of the composition. It is particularly desirable that phaseolamine be present in an amount of about 31-32% by weight of the composition.

A particularly preferred nutritional supplement composition of the present invention comprises a solid dosage unit comprising individually about 100 mg of Touchi extract and about 166.67 mg of phaseolamine.
The compositions of the present invention may further comprise a carminative. In a preferred embodiment, organic parsley is used as a carminative.
The present invention also provides a method for limiting the absorption of carbohydrates present in food. The method comprises administering to the individual a nutritional supplement composition of the present invention. In a preferred manner, the administration occurs at a point prior to consumption of the carbohydrate by the individual. In a more desirable manner, the administration is performed 1 second to 30 minutes prior to carbohydrate consumption. In a more desirable manner, the administration is performed 10-25 minutes prior to carbohydrate consumption. In a particularly desirable manner, the administration occurs about 15 minutes prior to carbohydrate consumption. The present invention also provides a method for promoting weight loss of an individual. The method comprises repeatedly administering the nutritional supplement composition of the present invention to an individual over a period of time. In a preferred method, the composition is administered once daily for a period of about 4 to 52 weeks. In another desirable method, the composition is administered multiple times daily with a carbohydrate-containing food over a period of about one month.

BEST MODE FOR CARRYING OUT THE INVENTION
While the invention is defined by the claims, a further understanding of the invention may be had from the following description of the preferred embodiments. The description is not intended to limit the scope of the invention, but rather can be used to enable one of ordinary skill in the art to make and use the invention.
According to the present invention, the nutritional supplement composition comprises an effective amount of an α-glucosidase inhibitor and an effective amount of an α-amylase inhibitor. The nutritional supplement composition may include a carrier acceptable for the nutritional supplement.
The nutritional supplement composition is substantially free of lipase inhibitors. As used herein, the term "substantially free of lipase inhibitors" refers to the level of lipase inhibitory activity exhibited by the composition's tolerable contamination or by the α-glucosidase component and / or the α-amylase component. It is meant to include only the amount of lipase inhibitor present. It is desirable that lipase inhibitors (i.e., excluding α-glucosidase inhibitors and α-amylase inhibitors) present due to acceptable contamination comprise less than 0.001% by weight of the nutritional supplement composition.

α-Glucosidase is an enzyme that breaks down disaccharides into their respective monosaccharide units. α-Glucosidase inhibitors prevent the enzyme from performing this function. A wide variety of α-glucosidase inhibitors are known, and any suitable inhibitor may be used in the compositions and methods of the present invention. Examples of suitable α-glucosidase inhibitors include voglibose (see US Pat. No. 6,200,958 to Odaka et al.), Acarbose (see US Pat. No. 5,643,874 to Bremer et al.), And torch extract.
In a preferred embodiment, the α-glucosidase inhibitor comprises an extract of Touchi. Torch is a traditional Chinese food derived from soy. Torches are prepared by first steaming soybeans and then fermenting them with Aspergillus species bacteria. Typically, torches are used for food purposes in paste form. A torch useful in the present invention is an extract of torch. As used herein, the terms “touch extract”, “touch extract”, and “touch” all refer to touch extract.

Any suitable torch extract may be utilized in the present invention. The extract need only retain the desired inhibitory effect on α-glucosidase activity. Water extracts of touchi are particularly desirable. U.S. Patent No. 6,299,911 to Kawabata et al. Describes suitable torch extracts and methods for preparing the extracts, and herein is incorporated by reference for the purpose of describing applicable torch extracts and methods for their preparation. Included as is for reference. Desirably, the torch extract comprises a powder form of an aqueous extract of torch.
The nutritional supplement compositions of the present invention include an effective amount of an α-glucosidase inhibitor. The effective amount utilized will depend on the desired inhibitory effect of the nutritional supplement composition. An effective amount of an α-glucosidase inhibitor includes an amount capable of blocking the absorption of carbohydrate in an amount obtainable from a typical high carbohydrate diet. As used herein, the term "high carbohydrate food" is defined as food that contains more than about 80 grams of carbohydrate. Therefore, it is desirable that an effective amount of an α-glucosidase inhibitor include an amount capable of blocking the absorption of about 60-100 grams of carbohydrate. It is particularly desirable that the effective amount of the α-glucosidase inhibitor comprises an amount capable of blocking the absorption of about 70-90 grams of carbohydrate. Most preferably, an effective amount of an α-glucosidase inhibitor comprises an amount capable of blocking the absorption of about 80 grams of carbohydrate.

α-Amylase is an enzyme that functions to break down α-1,4-glycosidic bonds present in starch. This breaks down complex starch molecules into smaller units, such as disaccharides, which can be further digested by other enzymes, such as α-glucosidase. The α-amylase inhibitor prevents the enzyme from hydrolyzing α-1,4-glucosidic bonds, and thus prevents starch degradation. A wide variety of α-amylase inhibitors are known, and any suitable inhibitor may be used in the compositions and methods of the present invention. Examples of suitable α-amylase inhibitors include inhibitors extracted from wheat (see US Pat. No. 3,950,319 to Schmidt et al.), Amylostatin-A (US Pat. No. 4,010,410 to Murao et al.). 258), and phaseolamine.
In a preferred embodiment, the α-amylase inhibitor comprises phaseolamine. Phaseolamine is an extract of white kidney beans. The extract is water-soluble and is rich in protein content. Phaseolamine is readily available from many commercial suppliers. Phaseolamine 2250®, available from Pharmachem Laboratories of Kearney, NJ and also known as Phase 2, is a standardized extract that is particularly well suited for incorporation in the compositions of the present invention. This phaseolamine shows a high ability to block α-amylase activity.

The nutritional supplement compositions of the present invention include an effective amount of an α-amylase inhibitor. The effective amount utilized will depend on the desired inhibitory effect of the nutritional supplement composition. An effective amount of an α-amylase inhibitor desirably includes an amount capable of blocking the breakdown of starch in an amount that may be present in a typical high carbohydrate food product. Therefore, it is desirable that the effective amount of the α-amylase inhibitor comprises an amount capable of blocking the degradation of about 60-100 grams of starch. It is particularly desirable that the effective amount of the α-amylase inhibitor comprises an amount capable of blocking the degradation of about 70-90 grams of starch. Most desirably, the effective amount of the α-amylase inhibitor comprises an amount capable of blocking about 80 grams of starch degradation.

The presence of both an α-glucosidase inhibitor and an α-amylase inhibitor in a composition of the invention provides an effective blocker of carbohydrate digestion. Indeed, in a preferred embodiment comprising Touchi extract and phaseolamine, the composition is surprisingly more effective at blocking carbohydrate absorption than individually acting ingredients (see Example 2). While not wishing to be limited to a particular mechanism, we hypothesize that the two inhibitors act synergistically to enhance the potency of each individual inhibitor.
The nutritional supplement compositions of the present invention may further include one or more acceptable carriers. A wide variety of acceptable carriers are known in the nutritional supplement industry, and the carrier may be any suitable carrier. The carrier need only be suitable for administration to animals, including humans, and capable of acting as a carrier without substantially affecting the desired activity of the inhibitor. In addition, one or more carriers may be chosen based on the desired route and mode of administration of the composition. For example, the nutritional supplement compositions of the present invention are suitable for use in a variety of dosage forms, eg, liquid and solid forms. In the preferred compositions described below, the nutritional supplement compositions include solid dosage forms, such as tablets or capsules. Examples of carriers suitable for use in tablet compositions and capsule compositions include organic and inorganic inert carrier materials such as gelatin, starch, magnesium stearate, talc, gum, silicon dioxide, stearic acid, cellulose and the like. But not limited thereto. It is desirable that the carrier be substantially inert, but it should be noted that the nutritional supplement compositions of the present invention may include additional active ingredients in addition to the α-glucosidase inhibitor and the α-amylase inhibitor.

In a preferred embodiment, silicified microcrystalline cellulose is utilized as a carrier. Silicified microcrystalline cellulose is a physical mixture of microcrystalline cellulose and colloidal silicon dioxide. A suitable form of silicified microcrystalline cellulose for use in the compositions of the present invention is Prosolve 90, available from Penwest, Patterson, NJ. Additional silicon dioxide is advantageously added to the composition of the present invention as a processing aid in addition to that provided by silicified microcrystalline cellulose. For example, additional silicon dioxide may be included as a glidant to improve powder flow during compression in the manufacture of solid dosage units such as tablets.
Other inert ingredients, such as lubricants and / or glidants, may be utilized in the compositions of the present invention. Lubricants facilitate tablet handling during manufacture, for example, during removal from a die, and glidants improve powder flow during tablet compression. Stearic acid is an acceptable lubricant / glidant for use in the nutritional supplement compositions of the present invention.

Example 1 illustrates a particularly desirable formulation for the composition of the present invention.
Advantageously, the nutritional supplement compositions of the present invention are in solid dosage forms, such as tablets and capsules. This form provides a product that is more easily carried by the individual to a dining location, for example, a restaurant, and can be taken before consuming the food. Also, the composition can be formulated in dosage units containing a suitable amount of inhibitor to allow the individual to determine the appropriate number of units to take based on appropriate parameters, such as weight, food size, and carbohydrate content. The α-glucosidase inhibitor and the α-amylase inhibitor comprise a nutritional supplement composition in a weight ratio between about 5-95% α-glucosidase inhibitor and about 95-5% α-amylase inhibitor (only of these two components). It is desirable to be present in the object. More preferably, these components are present in a weight ratio between about 10-45% α-glucosidase inhibitor and about 55-90% α-amylase inhibitor. The α-glucosidase inhibitor and the α-amylase inhibitor are present in the nutritional supplement composition as a percentage by weight of the total composition between about 7-35% of the α-glucosidase inhibitor and about 15-48% of the α-amylase inhibitor. In a particularly preferred embodiment, the nutritional supplement compositions of the present invention comprise solid tablets individually comprising about 10 mg to about 3,000 mg of Touchi and about 70 mg to about 4,500 mg of phaseolamine. More preferably, the nutritional supplement comprises a solid tablet individually containing about 100-1000 mg of Touchi extract and about 100-1,500 mg of phaseolamine. It is particularly desirable that the nutritional supplement comprises a solid tablet individually containing about 100 mg of Touchi extract and about 166.67 mg of phaseolamine. As indicated above, it is particularly desirable that these amounts correspond to about 18-19% by weight of the nutritional supplement composition and about 31-32% by weight of the composition, respectively. The dosage composition provides a level of inhibitory effect in a single unit that may be effective for certain individuals and / or certain foods, and is effective for other individuals and / or other foods. It allows for relatively simple dose escalation to provide other levels of inhibitory effect that may be required. For example, a three tablet dose of this preferred embodiment would provide about 300 mg touchi and about 500 mg phaseolamine. These amounts may be able to block up to 500 kcal of carbohydrate, or up to 100 grams, as shown in the examples.

The nutritional supplement compositions of the present invention may advantageously include one or more carminatives, which act as leveling agents. A wide variety of carminatives are known in the nutritional supplement industry, and any suitable carminative may be utilized in the present invention. The choice of carminative will depend on the desired properties of the nutritional supplement composition. For example, it may be desirable for the composition to have some texture, appearance, or taste. Carminatives can also be used for additional functions, such as providing nutrition. When present, it is desirable that the carminative comprises from about 0.01% to about 3% by weight of the nutritional supplement composition. Suitable organic parsley is readily available from various commercial sources.
Organic parsley is a preferred carminative for use in the compositions of the present invention. Organic parsley gives the composition a desirable appearance and contributes to a desirable taste. In addition, organic parsley is known to help prevent gas production in the gastrointestinal tract. The production of gas in the gastrointestinal tract has been associated with various compositions for inhibiting the absorption of various compounds. Thus, the incorporation of organic parsley in the nutritional supplement compositions of the present invention makes these compositions more desirable. In addition, organic parsley contains trace amounts of major nutrients, such as vitamin B (eg, thiamine, riboflavin, niacin, folate, and vitamin B6), vitamin C, and vitamin E, as well as minerals, such as calcium, chromium. Additional benefits are provided by providing copper, iron, magnesium, manganese, molybdenum, potassium, selenium, zinc, and others. Parsley also contains phytochemicals with antioxidant properties, such as β-carotene, lutein, quercetin, rosmarinic acid, and others.

The present invention also provides a method for limiting the absorption of carbohydrates contained in foods. The method comprises administering to the individual a nutritional supplement composition comprising an effective amount of an α-glucosidase inhibitor and an effective amount of an α-amylase inhibitor in the substantial absence of a lipase inhibitor. Desirably, the method comprises administering a nutritional supplement comprising an effective amount of Touchi extract and an effective amount of phaseolamine in the substantial absence of a lipase inhibitor.
The nutritional supplement compositions of the present invention inhibit the activity of carbohydrate digestive enzymes. For those who are conscious of carbohydrates, it may be desirable to perform this control before consumption of high carbohydrate foods, or prior to consumption of typical foods. This may allow inhibitors to limit the absorption of carbohydrates contained in special foods. Administration may be at a time prior to consumption of the food concerned, concurrent with consumption of the carbohydrate, or after completion of consumption of the carbohydrate. Desirably, the composition is administered from about 1 second to 30 minutes prior to carbohydrate consumption. More desirably, the composition is administered about 10 to 25 minutes prior to carbohydrate consumption. More desirably, the composition is administered about 15 minutes prior to carbohydrate consumption. The actual time point chosen will depend on several factors, including the dosage of the nutritional supplement composition utilized.

The present inventors have surprisingly discovered that the nutritional supplement compositions of the present invention are effective in promoting weight loss. This occurs despite the absence of a lipase inhibitor in the composition. Without limiting the present invention to a particular mechanism of action, the inventors believe that this effect occurs due to the presence of phaseolamine, which can result in a weight loss of more than 5 pounds (2 kg) and a 10% It was shown to promote the above body fat reduction. Thus, the present invention provides nutritional supplement compositions that inhibit the activity of related carbohydrate digestive enzymes and promote weight loss (all in the absence of a lipase inhibitor). This allows the composition to be used as a pre-meal carbohydrate blocker, a weight loss promoter, or both.
Thus, the present invention also provides a method of promoting weight loss in an individual. The method comprises administering to the individual a nutritional supplement composition comprising an effective amount of an α-glucosidase inhibitor and an effective amount of an α-amylase inhibitor in the substantial absence of the lipase inhibitor over a period of several days. Desirably, the method comprises administering a nutritional supplement comprising an effective amount of Touchi extract and an effective amount of phaseolamine in the substantial absence of a lipase inhibitor. In a particularly desirable manner, administration comprises administering the composition in solid dosage units containing about 100 mg each of Touchi and about 166.67 mg phaseolamine. In another particularly desirable manner, administration comprises administering the composition once daily for a period of about 4 weeks to about 52 weeks. In other desirable methods, administration comprises administering the composition once daily for a period of one month. In other desirable methods, the compositions are administered on a diet containing each carbohydrate for a period of days, such as from 2 days to 52 weeks.

The nutritional supplement compositions of the present invention were formed into single dose tablets containing the ingredients in the amounts and percentages indicated below.

In this form, the composition provides the desired inhibitory effect and allows for relatively easy dose escalation based on various parameters, such as food size, carbohydrate content, and body weight. For example, a three tablet dose would contain about 300 mg touchi and about 500 mg phaseolamine.
Example 2
The combination of an α-glucosidase inhibitor and an α-amylase inhibitor provides a composition that is beneficial in blocking carbohydrate absorption. In one embodiment, an extract of the α-glucosidase inhibitor torch, combined with the α-amylase inhibitor phaseolamine, has a synergistic effect in which the composition has a greater blocking potential than the separate components acting alone. Is generated.
How to measure enzyme inhibition
Three substances (Touchi (soybean extract), phaseolamine (white kidney bean extract) and the composition of Example 1) were tested for their inhibitory effect on α-glucosidase (AGH) activity and α-amylase activity. Known assays (Bernfeld, P. "α-amylase and β-amylase in enzymological methods", Vol. 1 (Cowwick, SP and Kaplan, NO., Edited by p. 149), Academic Press, New York. Using an improved assay from (1955), we evaluated the blocking potency (kcal) of the Touchi extract alone, phaseolamine alone, and the composition of Example 1.

Sample calculations for units of inhibited enzyme and carbohydrate inhibition potential as kcal
Briefly, the production of a product (ie glucose for α-glucosidase activity or maltose for α-amylase activity) from a suitable substrate using standard α-glucosidase or α-amylase. The measurement resulted in a standard calibration of the enzyme activity. The maximum amount of enzyme from the calibration curve was used to determine the enzyme inhibition conferred by Touchi extract or Phaseolamine by measuring the resulting activity reduced due to the presence of Touchi or Phaseolamine. A similar method was used to measure α-glucosidase inhibition and α-amylase inhibition in the composition. The synergistic effect on the inhibition of both enzymes was determined from the composition, where unexpectedly high inhibitions of both α-glucosidase activity and α-amylase activity were respectively inhibited based on their mass% in the composition. Observations were made in comparison to theoretical values derived from calculations of units of enzyme inhibited by the compound.

Units of enzyme per gram are calculated by conversion from units of enzyme inhibited by reaction per amount of compound present. Enzyme units are defined as μmoles of product (glucose or maltose) released under optimal conditions per minute. Convert μmoles of glucose (molecular weight 180) or maltose (molecular weight 360) based on the units of the suppressed enzyme to grams of glucose or maltose, which are easily converted to Kcal as 4 kcal / g.
For α-glucosidase inhibition (AGH) contributed by Touchi extract, the results are reported as grams of blocked glucose release per gram of inhibitor per minute.
For α-amylase suppression contributed by the phaseolamine extract, the results are reported as grams of maltose release blocked per gram of inhibitor per minute.

If the torch extract is found to have 900 units of AGH inhibited per gram, the available inhibition per minute per gram of torch is represented by the following equation:

If the phaseolamine extract is found to have 6000 units of α-amylase inhibited per gram, the available inhibition per minute per gram of phaseolamine is expressed by the following equation:

If the composition is found to have 292 units of AGH inhibited per gram and 3167 units of α-amylase inhibited per gram, the inhibition per minute per gram of composition is represented by the following formula:

If 1 gram of the composition contains 19% by weight torch and 32% by weight phaseolamine, the theoretical calculation for AGH suppression from the torch based on the weight% of the formulation should be as follows.

Theoretical calculations for α-amylase inhibition from phaseolamine based on the weight percent of the formulation should be as follows.
