本発明は、3−アルコキシアクリル酸ハライドの製造方法に関するものである。3−アルコキシアクリル酸ハライドは、例えば、殺菌剤として有用なポリオキシン又はニコマイシンの合成原料として用いられている。(非特許文献1参照)

【従来の技術】
3−アルコキシアクリル酸ハライドの製造方法としては、(特許文献1)に、ビニルエーテルをホスゲンと反応させる製法が記載されている、しかしながら、有毒なホスゲンを使用するため、工業的製法として望ましくない。

(非特許文献2)には、3,3−ジアルコキシプロパン酸をチオニルクロライドと反応させる製法が記載されているが、収率は64%と充分ではない。

【発明が解決しようとする課題】
本発明の課題は、殺菌剤の中間体等として有用である3−アルコキシアクリル酸ハライドを、工業的に合成容易であり大量に入手可能な原料を用いて、毒性の問題が無く、且つ収率良く工業的に製造する方法を提供することである。

【課題を解決するための手段】
本発明者らは、前記の課題を解決するために検討した結果、3−アルコキシアクリル酸ハライドを、3−アルコキシアクリル酸とチオニルハライドをN−置換アミド類、N−置換尿素、ピリジン類又はアミノ燐酸類の触媒存在下に反応させることによって、簡便かつ安全で、収率の良い工業的な製造する方法を見出し、本発明を完成した。

【発明の実施の形態】
本発明により、前記式(2)で示されるアルコキシアクリル酸ハライド類(2)は、前記式(1)で示されるアルコキシアクリル酸類(1)をN−置換アミド類、N−置換尿素、ピリジン類又はアミノ燐酸類の触媒存在下、チオニルハライドと反応させて得られる。

これら化合物は工業的に製造されており、購入可能な化合物である。
これら触媒は、単独又は混合して用いる事もできる。

触媒の使用量は、アルコキシアクリル酸類(1)に対して0.01〜1.0当量であるが、好ましくは0.05〜0.5等量である。

チオニルハライドとしては、チオニルクロライド、又はチオニルブロマイドなどが挙げられる。これら化合物は工業的に製造されており、購入可能な化合物である。

以下、実施例により、本発明を具体的に説明する。

実施例1.
アルゴン雰囲気下、25mlの3ッ口フラスコにチオニルクロライド(12.52g, 100mmol)を加え、内温4〜5℃で3−メトキシアクリル酸(10.21g, 100mmol)を10分間かけて加えた。引き続きDMF(0.73g, 10mmol)を内温4〜6℃で加え、15分間内温4℃で撹拌した。単蒸留(流出33mmHg, 83〜84℃)を行い、目的の3−メトキシアクリル酸クロライドを無色液体として10.8272g(収率89.9%)得た。

実施例2
アルゴン雰囲気下、300mlの4ッ口フラスコにチオニルクロライド(125.23g, 1.0mol)を加え、内温1〜2℃で3−メトキシアクリル酸(102.09g, 1.0mol)を30分間かけて加えた。引き続きDMF(7.31g, 0.1mol)を内温2〜6℃で加え、1時間内温1〜3℃で撹拌した。単蒸留(流出20mmHg, 73〜74℃(文献値105−110℃(35−37mmHg)(非特許文献3参照)))を行い、目的の3−メトキシアクリル酸クロライドを無色液体として117.85g(収率97.8%)得た。釜残を定量すると、3−メトキシアクリル酸クロライドが1.81g(15mmol)残存した。従って、3−メトキシアクリル酸クロライドの全反応収量は119.69gであり全収率は99.3%、選択率は99.3%であった。

蒸留釜の残渣をHPLCで定量したところ3−メトキシアクリル酸が360mg含まれており、転化率は96.5%であった。従って、目的の3−メトキシアクリル酸クロライドの選択率は63.1%であった。
