BORDETELLA PERTUSSIS 線毛を含有するワクチン接種結合体および 経口ワクチンにおけるキャリアとしてのBORDETELLA PERTUSSIS抗原
本発明は、Bordetella pertussis抗原を含有するワクチン、結合体ワクチン、
キャリアと免疫成分を結合体化してワクチン結合体を形成する方法、ならびにヒ
トおよび動物にワクチン接種するための結合体ワクチンの使用、また百日咳に対
する経口ワクチンに関する。
重篤な侵襲性疾患を引き起こす多くの病原性細菌は、本質的に毒性成分である
糖質莢膜を有する。糖質莢膜は、潜在的なワクチン成分である。なぜなら、それ
らに対して指向する抗体は、補体に媒介される殺菌性活性によって、通常は防御
性であるからである。糖質に対して産生された抗体は、糖質が得られた特定の血
清型に対して特異的である;Haemophilus influenzaeについての1つの主要な病
原性血清型、Neisseria meningitidisについての4つの主要な血清型、およびSt
reptococcus pneumoniaeについての80を超える血清型が存在する。
莢膜ワクチンの主要な不利な点は、糖質がT細胞非依存性の抗原であり、それ
故、糖質が引き起こす免疫応答が低く(特に小児において)、持続時間が短く、
追加免疫ができず、そして成熟しない親和性を有することである。抗原は、免疫
応答を増強するタンパク質との結合体化によってT細胞依存性(記憶応答の提供
を含む)に変換され得る。
Haemophilus influenzae b型(Hib)の莢膜多糖タンパク質結合体化ワクチン
を用いる免疫は、幼児においてHib疾患に対する防御を付与することが実証され
た。これは、例えば、Neisseria meningitidis、およびStreptococcus pneumoni
aeでの小児の感染の制御のために、同様の小児科のワクチン戦略の導入を支持す
る説得力のある議論である。しかしこれらの新たなワクチンは抗原の複雑な混合
物である。Hibワクチン接種の導入の間に有害な抗原の相互作用および処方技術
の制限に遭遇した。新たなワクチンが、確立された小児科の免疫プログラムにお
いて導入される場合、これらの問題は悪化するにすぎないようである。
多糖に対する小児免疫は、T細胞依存性の機構を介してのみ作動するという事
実から問題が生じ、従ってこのタイプのワクチンはキャリアタンパク質との結合
体化を必要とする。現在のところ、ヒトでの使用のための利用可能な唯一のキャ
リアタンパク質は、破傷風トキソイド(TT)または遺伝的にトキソイド化され
たジフテリア毒素(DT)である。これらのキャリアの増加する使用は、これら
に対する前から存在している免疫(生涯の初期に受動的に移行した母性抗体、ま
たは既存のワクチンに対する免疫学的記憶のいずれかを介して生じ得る)が糖質

もしれない。そのような相互作用は、初期免疫および追加免疫適用のそれぞれの
ための、現在の多糖結合体ワクチンの有効性を明らかに減少させ得る。
従って、既存のトキソイドキャリアの拡大した使用は、ジフテリア/破傷風の
過剰負荷を生じ、そしてトキソイドと結合体化した糖質に対する免疫応答を減少
させるという問題が存在する。さらに、トキソイドはその免疫学的特徴を変化さ
せ得る解毒を必要とする。キャリアとして産生された外膜タンパク質は、上記の
問題を有さないが、それらは特徴付けが難しい複雑な混合物であり、そして混合
物間において組成が変化する。
別の困難さは、小児科の免疫のためのワクチンの増加する数と複雑さの実用性
に関連する。ワクチン製造業者は、1つの注射器から同時に送達され得る小児科
ワクチンの組み合わせの産生(従って免疫プログラムの単純化)において成功し
ている。現在の処方物およびアジュバント技術を使用するワクチンの組み合わせ
中の成分の範囲を拡大することは、今やますます技術的に困難である。ますます
複雑になるワクチン接種プログラムの対応する問題の全てをともなう複数回の注
射の再導入の予測は、適切な別の送達系(例えば、粘膜表面)が導入されなけれ
ば生じるようである。
従って、代わりのキャリアタンパク質が新規のまたは第2世代の結合体ワクチ
ンの導入のために必要とされることが、一般に認識されている。
本発明の目的は、今までに遭遇した問題および潜在的問題の改善または減少を
ともなう、既存のトキソイドベースのワクチンと並行してまたは引き続き使用さ
れ得る結合体中の免疫原性糖質の提示のための結合体ワクチンを提供することで
ある。さらなる目的は、結合体ワクチンの製造のための、既存のトキソイドキャ
リアに代わるキャリアタンパク質を提供することである。なおさらなる目的は、
単一のワクチン処方物において1つより多い病原体に対するワクチン接種のため
に使用され得るワクチンを提供することである。
従って、本発明の第1の局面は、(i)Bordetella pertussisの線毛、(ii)百日
咳毒素、(iii)百日咳トキソイド、および(iv)百日咳69kDタンパク質から選択さ
れるキャリアと結合体化した抗原を含有する、ワクチンにおける使用のための結
合体を提供する。
抗原は、適切には、病原性細菌またはウイルスの抗原性成分であり、この文脈
において、「抗原」は病原性細菌またはウイルスの抗原性成分の、改変体、誘導
体およびフラグメントを含み、その結果、抗原を用いる免疫はその病原性生物体
に対する防御免疫を生じることが、理解される。
Bordetella pertussisの線毛は、Bordetella pertussisの培養物から精製され
得るか(例えば、EP-A-0231083は、百日咳抗原の精製を記載する)、または組換
え技術によって生成され得る。そして結果的に、Bordetella pertussisの線毛に
対する論及は、天然の線毛の精製によって得られるか、線毛をコードするDNA
の組換え発現によって得られる線毛の論及として理解されるべきであり、そして
また、論及がBordetella pertussisの線毛の改変体、誘導体またはフラグメント
であるとして認識されるにもかかわらず、線毛の改変体、誘導体およびフラグメ
ントを含むことがまた理解される。なぜならそのような改変体、誘導体またはフ
ラグメントでの免疫は、Bordetella pertussisのチャレンジに対する防御的な抗
体の誘導をもたらすからである。10μg〜50μgの範囲の線毛の量が代表的なワク
チン接種の用量である。
B.pertussisより単離された線毛の精製および特徴付けはまた、ZhangらのInf
ection and Immunity,1985年5月、422〜427頁、およびRobinsonらのVaccine、
7巻、1989年8月、312頁(前出)によって記載されている。
組換え線毛の生成は、Mol.Microbiol.、1990年1月、4(1)巻、39〜47頁、およ
びInfect.Immun.、1991年5月、59(5)巻、1739〜1746頁において記載される。
抗原のキャリアとの結合体化は、従来の手段によって達成される。本発明の実
施態様において、キャリアはC6スペーサーを用いて抗原と結合体化され、ここで
線毛はまず誘導体化され、そして次に抗原溶液に添加される。また、必要に応じ
て、抗原はまず誘導体化され、そしてこれは抗原が誘導体化条件(代表的にpHに
おける変化を含む)によって損傷を受けやすい場合に有利で有り得る。代表的に
は、2つをともに連結するように、抗原とタンパク質との結合体化のために二官
能性の基が導入される。
本発明の使用において、動物は、免疫原性結合体を含有するワクチンを用いて
免疫され、そして結合体の抗原性成分が由来する病原性生物体によるチャレンジ
に対して防御される。この意味において、防御は、病原性生物体の致死用量を用
いるチャレンジに対する生存によってか、またはそのような致死用量を用いるチ
ャレンジに対する延長された平均余命によって認められる。防御はまた、百日咳
の致死用量を下回るチャレンジに続く、影響の少ない、より病気でない患者によ
って、認められる。
本発明は、T細胞エピトープと組み合わせた抗原の提示のための免疫原性結合
体の調製のための代替的なキャリア分子を提供するにおいて、有利である。本発
明の免疫原性結合体に対する免疫応答は、単離された抗原に対する免疫応答と比
較して、増強される。従って、抗原単独によるワクチン接種に比べて有効性を改
善する。本発明はまた、既存のトキソイドキャリアの代わりを提供し、従って、
これらトキソイドを含有するワクチンの拡張されたそして反復する使用にともな
い生じ得るトキソイド過剰負荷の問題を克服する。
結合体の成分として百日咳線毛を使用するさらなる利点は、ワクチン中への取
込みの前の解毒を必要としないということである。当該分野で使用されるジフテ
リアおよび破傷風毒素のために必要とされる解毒は、タンパク質の免疫原性特徴
を変化し得る。なおさらなる利点は、Bordetella pertussis由来の線毛はまた、
小児科において関連する病原体であるBordetella pertussisに対する突出した免
疫性を与えるか、または増強し、従って、線毛を含有する結合体が二重の免疫応
答を誘導することである。
既存の小児科の免疫プログラムは、DTPワクチンを用いる免疫を含み、これは
単一のワクチンにおいてジフテリア、破傷風および百日咳に対する防御を与える
。
本発明は第4の成分(この第4の成分は、例えば、Haemophilus influenzaeに対
する免疫性を与える抗原であり得る)を、百日咳および第4の病原性生物体の両
方に対する免疫を与える本発明に従う結合体ワクチンを取り込むことにより、こ
の3成分ワクチン中に取り込む可能性を開く。
百日咳トキソイドは、本発明の結合体のさらなる代替的成分であり、先行技術
のキャリアを用いて経験される破傷風/ジフテリアトキソイドの過剰負荷の危険
性に寄与することなく、抗原キャリアとして作用する潜在能力を提供する。百日
咳トキソイドはまた、抗原および百日咳それ自体の両方に対するワクチン接種を
提供する。
別の代替的キャリアである百日咳毒素は、患者への投与前に非毒性化させるた
めに、必要に応じて変性またはそうでなければ処理される。この工程は、抗原と
の結合化前か、または結合体化後に行われ得る。あるいは、毒素は低い非毒性用
量にて、結合体ワクチンにおいて使用される。
別の代替的キャリアである69kDタンパク質は、必要に応じて培養物からの精製
によってか、または組換え手段によって精製される。そして69kDタンパク質に対
する論及は、インタクトタンパク質の実質的な免疫原性を保持するタンパク質の
改変体、誘導体およびフラグメントを含むと理解される。
本発明の結合体は、記載したように、抗原を含む。抗原の供給源または性質は
、任意の特定のサブグループの抗原に限定されず、そして実際に、単離状態の抗
原は免疫原性ではなく、本発明の結合体に組み込まれた抗原のみが免疫原性にな
ることが可能である。
適切な抗原としては、糖質、多糖類、単糖類、オリゴ糖類、タンパク質、ペプ
チド、糖ペプチド、リポ多糖、ならびに類似の分子および関連分子が挙げられる
。代表的には、抗原は、細菌またはウイルスの外表面上に現れる細菌またはウイ
ルスの成分(例えば、細菌細胞壁の成分、線毛(fimbria)もしくは線毛(cilia)も
しくはべん毛の成分、またはウイルスの外側のエンベロープの成分(この具体的
な例は、B型肝炎ウイルスの表面抗原である))であるか、またはそれに由来する
。例示によって、抗原は、Bordetella bronchiseptica、Clostridium tetani、
サイトメガロウイルス、デングウイルス、エプスタイン-バーウイルス、フラビ
ウ

本発明の1つの実施態様において、抗原性結合体は、2つの異なる抗原と結合
体化した、本発明のキャリア(例えば、Bordetella pertussisに由来する線毛)を
含む。従って、この結合体は、百日咳、および線毛に結合体化した異なる抗原が
得られるかまたは由来する2つの異なる病原性生物の各々に対する防御免疫を付
与するかまたは増強することにおいて有用である。従って、本発明の免疫原性結
合体は、必要に応じて、Meningococcal C多糖類、およびHib夾膜(capsular)糖質
の両方が結合体化されたBordetella pertussisの線毛を含む。それゆえ、本発明
のこの実施態様を使用して、3つの病原性生物に対する防御免疫を付与し得る。
本発明のこの実施態様の利点は、複数の免疫が、単一のワクチン成分を介して付
与され得、それによって個々のワクチンの混合物を調製する必要性を回避し、か
つわずか1つの生物に対する防御免疫を付与するワクチンを使用して反復されそ
して複雑なワクチン接種スケジュールについての必要性を減少させる。本発明の
実施態様は、抗原が都合良くカップリングされ得る複数位置を含む、長い多量体
分子である線毛の物理的構造によって可能になる。
本発明の結合体は、経口送達を含む多くの種々の経路を介する送達についての
微小粒子への組み込みに適切である。このような微小粒子の調製は、EP-A-02661
19、EP-A-0333523およびEP-A-0706792に記載され、それらの内容は,本明細書中
において参考として援用される。
多くの異なる型のBordetella線毛が存在することは公知である。1つの型は、
凝集原2を有し、そして約22,500ダルトンの分子量を有し、そして第2の型は、
凝集原3を有し、約22,000ダルトンの分子量を有する。これらは、異なる血清型
の百日咳において見出される傾向がある:凝集原2は、血清型1.2.0および1.2.3
において見出され、そして凝集原3は、血清型1.0.3および1.2.3において見出さ
れる。本発明のさらなる実施態様は、2つの免疫原性結合体の混合物を含み、各

を含む。
本発明はまた、(i)Bordetella pertussisの線毛、(ii)百日咳毒素、(iii)百日
咳トキソイド、および(iv)百日咳69kDタンパク質、から選択されるキャリアと抗
原との結合体を調製する方法を提供し、この方法は、キャリアの調製物を抗原調
製物と組み合わせて抗原をキャリアに共有結合させる工程、および、その後、こ
の混合物から結合体を取り出す工程を包含する。結合体は、キャリア分子におい
て第1級アミン基に基づくので、抗原とキャリアとの結合体化は、これらのアミ
ン基がキャリアの表面上で利用可能である場合はいつでも可能である。このよう
な基の2つ以上が利用可能である場合、キャリア+2つの抗原の結合体が可能で
ある。
以下の具体的な実施例に記載される方法の1つの実施態様において、凍結乾燥
した線毛は、抗原溶液に溶解され、次いで、この溶液は、抗原と線毛との結合体
化を可能にするように延長した時間にわたって低下した温度にて維持される。好
ましくは、一定量の線毛は、酸性緩衝液に溶解され、安定化され、透析され、次
いで凍結乾燥される。この抗原は、適切な緩衝液中で抗原を溶解することによっ
て調製され、次いで、凍結乾燥した線毛は、その緩衝液に添加され、得られる混
合物は、延長された時間にわたって透析され、次いで、この混合物は、凍結乾燥
され免疫原性結合体が回収される。
本発明はまた、本発明の免疫原性結合体の使用に関し、従って、本発明はまた
、抗原が得られるかまたはそれに由来する病原性生物に対してハムスターまたは
動物のワクチン接種のための医薬品の製造における本発明の結合体の使用を提供
する。本発明はまた、本発明の結合体の免疫有効量をヒトまたは動物に投与する
工程を包含するヒトまたは動物のワクチン接種方法を提供する。
本発明の免疫原性結合体を組み込んでいるワクチンは、当該分野において標準
的な技術に従って処方され得る。そしてこのワクチンは、当業者が精通している
従来の薬学的に受容可能なキャリアおよび賦形剤を含み得る。
本発明は、抗原と線毛とを結合体化する方法を提供している一方で、本発明の
免疫原性結合体は、抗原とキャリア分子とを共有結合させるための任意の従来技
術に従って調製され得、そして本発明は、これまで記載され、そして以下に例示
される結合体化の特定の方法に制限されると解釈されるべきではない。
ジフテリア、破傷風、および百日咳に対する免疫を提供するDTPワクチンの注
射によって乳児にワクチン接種することは公知である。この経路によってワクチ
ン接種することは、乳児およびその親の両方にとって快適ではなく、注射用処方
物は、厳密に無菌であるという必要条件を満たしていなければならないという付
随した問題がある。
Jones,D.H.ら、Infection and Immunity,1996年2月、489-494頁によって、B
.pertussis線毛の経口投与が感染を防御することが記載される。線毛は、投与
前にマイクロカプセル化される。
本発明の別の目的は、百日咳に対して注射されるワクチンの代替を提供するこ
とである。従って、本発明の第2の局面は、薬学的に受容可能なキャリア中にBo
rdetella pertussisの線毛の経口処方物を含む、百日咳に対するワクチンを提供
することである。線毛に対する参照の意義および範囲は、本発明の第1の局面と
同様である。
驚くべきことに、線毛の経口投与は、百日咳によるチャレンジに対して防御的
な抗体の産生を生じ得ることが見出されている。これは、注射ワクチンに付随す
る問題を回避する。本発明はまた、線毛または線毛-抗原結合体を経口的に投与
することによる百日咳に対するワクチン接種方法、および百日咳に対する経口ワ
クチン接種のための医薬品の製造における線毛または線毛-抗原結合体の使用を
提供する。
本発明の実施態様において、線毛または線毛-抗原結合体は、特定のキャリア
とともに、代表的には、粒子の外側上に吸着されるか、またはそれに結合体化さ
れて、処方される。PLGのようなポリマーおよび無機質粒子が使用され得る。好
ましくは、Bordetella pertussis線毛は、直径10ミクロン以下の粒子上に吸着さ
れる。特に、直径10ミクロン以下の無機質粒子の懸濁液が適切である。経口投与
後、線毛または線毛-抗原結合体を吸着させたこれらの粒子の取り込みは、腸に
おいてパイアー斑を介して生じ得る。本発明の特定の実施態様において、以下で
さらに詳細に記載されるように、経口ワクチン接種組成物は、本発明の線毛を吸
着させたミョウバンのコロイド性懸濁液を含む。本発明のこれらの実施態様にお
いて、経口ワクチンは、線毛または線毛-抗原結合体以外の抗原性成分または免
疫成分を実質的に含まなくともよい。
ミョウバンは、周知のワクチンアジュバントであり、現在のところ、もっぱら
注射によって使用される。本発明者らは、ミョウバン+線毛が、経口によって与
えられた場合、良好な免疫応答が得られることを見出した。生じた抗体応答は、
防御を与えるために十分であり、この防御は、血清応答および粘膜応答の両方を
含む。
ワクチンの全身投与は、良好な血清応答、特にIgGを与えることが当該分野で
公知である。本発明の第2の局面による経口ワクチン接種は、IgG応答および同
様にIgA応答の両方を与えることが見出された。このことは意義深いことである
。なぜなら、IgAは、大部分の病原体についての侵入点である粘膜表面に出現す
るからである。
好ましくは、線毛を含む処方物の投与前またはそれと同時に、患者の胃は、予
め中和される。その結果、酸によって活性化されるプロテアーゼは、ワクチン成
分を破壊しない。これは、従来の酸を減少させて中和する医薬品を使用して達成
され得る。必要に応じて、本発明のワクチンは、胃酸を中和するための有効量の
化合物を含むように処方される。本発明の特定の実施態様において、以下に記載
されるように、ワクチン接種組成物は、重炭酸緩衝液(特に8.2〜8.5の範囲のpH
を有する、強度0.1Mの緩衝液)を使用して処方されるが、他の酸を中和する溶液
もまた、本発明の組成物に適切であることが予測される。ワクチン処方物を投与
する前に酸を中和する医薬品を投与することは、さらなる選択肢である。
本発明のこの局面の使用において、動物は、本発明の処方物を経口的に投与さ
れ、必要に応じて、追加免疫量の、本発明の処方物を引き続いて投与される。そ
してこのことによって、百日咳に対して防御される。
連続的腸内容物試験プロセスの一部として、腸におけるM細胞が粒子状物質を
取り込み、そしてその内容物はリンパ節を通過し、最終的に免疫応答の基礎を形
成するマクロファージに取り込まれると考えられる。ミョウバン粒子は、M細胞
中へと通過するので、免疫応答を導く連鎖に参加すると考えられる;しかし、本
出願人らは、この理論に拘束されることは望まない。
本発明の実施態様の経口ワクチンは、抗原提示細胞および線毛への通過または
他の輸送について適合されるキャリアの懸濁液を含む。ミョウバンがキャリアで
ある場合、注射用に当該分野で現在使用されている処方物は適切である。小児へ
の投与のために、矯味・矯臭剤または甘味剤が、必要に応じて添加される。本発
明の第2の局面のワクチンはまた、保存剤、または簡便な、室温保存のためにワ
クチンを凍結乾燥させることを補助する賦形剤を含む。
以下は、図面によって例示された本発明の特定の実施態様の説明である。
図1は、リン酸緩衝化生理食塩水(PBS)、線毛(Fim)、または線毛-多糖類結合
体(Fim Conj)を用いて0日目、14日目、および28日目に、免疫したマウスからの
血清抗多糖類抗体応答を示し、そして42日目に特異的な抗多糖類抗体について試

Neisseria meningitidis血清型C多糖類の精製。
本方法は、本質的にGotschlich,E.(1975)によって記載されるようなもの
である。群特異的な多糖類の精製、Monogr,Allergy 9,245-258。多糖類はL91
543系統から精製される(C2a P1.2R,Manchester Public Health Laboratoryよ
り入手する)。
細菌を、血液寒天プレート上で一晩増殖させ、そして250mlのコニカルフラス
コ中の100mlのフランツ培地に接種し、そして7時間振とうしてインキュベート
する。次いで、10mlの種培養物を、750mlフランツ培地を含むコニカルフラスコ
に接種し、37℃で一晩振とうしてインキュベートする。
多糖類の精製。
1.100mlの10%(w/v)ヘキサデシルトリメチルアンモニウムブロミド(CTB)
を、各々1L遠心ポットに加える。
2.遠心ポットを、1Lまで培養物で満たし、室温で1時間静置させる。
3.次いで、細菌および沈殿させた多糖類を、遠心分離(RC3B遠心分離機、50
00rpm、30分間)によって収集し、そして上清を捨てる。
4.ペレットを約200mlの水で再懸濁し、ホモジナイズして滑面懸濁物を調製
し、そして等容量の2M CaCl2を添加する。これを、1時間攪拌してCTB複合体か
ら多糖類を放出させる。
5.無水エタノールを25%(v/v)まで添加してDNAを沈殿させ、そして90分間
攪拌する。
6.これを、25,000gで20分間遠心分離し、そして上清を保持する。
