Claims (1)
- 【特許請求の範囲】 1. ヒトPKD1ポリペプチドをコードしている単離された核酸。 2. 前記核酸が、配列番号:2で示される配列を含むDNAである請求項1 に記載の単離された核酸。 3. 前記核酸がRNAである請求項1に記載の単離された核酸。 4. 前記核酸が、配列番号:4で示される配列を含むcDNAである請求項 1に記載の単離された核酸。 5. 請求項1に記載の核酸に対してストリンジェントな条件下でハイブリッ ド形成する単離された核酸。 6. 配列番号:3で示される配列を含む請求項5に記載の単離された核酸。 7. 請求項1に記載の核酸によってコードされる単離されたポリペプチド。 8. 配列番号:5で示されるアミノ酸配列を含む請求項7に記載の単離され たポリペプチド。 9. 請求項1に記載の単離された核酸を含むベクター。 10.前記核酸に対して機能的に結合した転写調節要素を更に含み、該要素は 、原核細胞または真核細胞およびそれらのウイルスの遺伝子またはそれらの組合 わせの発現を支配する能力を有する請求項9に記載のベクター。 11.請求項9に記載のベクターを含む宿主細胞。 12.PKD1タンパク質を製造する方法であって、 (a)請求項11に記載の宿主細胞を培地中においておよび該タンパク質の発 現に適した条件下で培養し、そして (b)該発現されたタンパク質を単離することを含む上記方法。 13.トランジション、トランスバージョン、欠失および挿入から成る群より 選択される修飾を有する配列番号:2で示されるDNA配列を含む単離されたヒ トPKD1遺伝子。 14.対象者のゲノム中の1個またはそれ以上のコピーでのその存在が、該対 象者における成人発症多嚢胞性腎疾患に関係しているDNA配列を含む請求項1 3に記載の遺伝子。 15.請求項13に記載のDNA配列を含む組換えベクター。 16.前記DNA配列に対して機能的に結合した転写調節要素を更に含み、該 要素は、原核細胞または真核細胞およびそれらのウイルスの遺伝子またはそれら の組合わせの発現を支配する能力を有する請求項15に記載のベクター。 17.請求項15に記載のベクターを含む宿主細胞。 18.突然変異PKD1タンパク質を製造する方法であって、 (a)請求項17に記載の宿主細胞を培地中においておよび該タンパク質の発 現に適した条件下で培養し、そして (b)該発現されたタンパク質を単離することを含む上記方法。 を含む単離された核酸。 を含む単離された核酸。 を含む単離された核酸。 を含む単離された核酸。 23.PKD1保因者を識別するためにヒト対象者をスクリーニングする診断 方法であって、 (1)該対象者から生体材料の試料を得;そして (2)該生体材料中の突然変異PKD1遺伝子またはそれらのタンパク質産物 の存在を検定する工程を含む上記方法。 24.前記生体材料が核酸を含み、そして前記検定が、 (a)該生体材料から真のPKD1遺伝子またはそのフラグメントを選択的に 増幅させ、そして (b)正常および突然変異PKD1遺伝子の存在を、制限酵素消化、直接DN Aシークエンス法、配列特異的オリゴヌクレオチドとのハイブリダイゼーション 、一本鎖立体配座多型解析、変性勾配ゲル電気泳動(DDGE)、二次元ゲル電 気泳動およびそれらの組合わせから成る群より選択される分析方法を用いて検出 することを含む請求項23に記載の方法。 25.前記増幅を、 から成る群より選択される少なくとも1種類のオリゴヌクレオチドの存在下で行 う請求項24に記載の方法。 26.前記検定工程が、前記PKD1遺伝子産物に特異的な抗体を用いる免疫 検定を含む請求項23に記載の方法。 27.配列(C)SRTPLRAKNKVHPSST(配列番号:15)を含むペプチドまたは その免疫原性フラグメントに対して向けられた単離された抗体。 28.配列番号:2で示される遺伝子配列によってコードされるポリペプチド と免疫反応性の請求項27に記載の抗体。 29.配列番号:2で示される配列によってコードされるポリペプチドと免疫 反応性の単離された抗体。 30.配列番号:5で示されるアミノ酸配列を含むポリペプチドと免疫反応性 の単離された抗体。 31.APKDの特徴を有する疾患状態を治療する方法であって、PKD1遺 伝子機能の欠損を有する細胞に対して、正常ヒトPKD1遺伝子またはそのフラ グメントを投与することを含み、該投与が、治療的有効量の正常PKD1タンパ ク質またはそのフラグメントの発現を引き起こす上記方法。 32.前記正常ヒトPKD1遺伝子が、配列番号:2のDNA配列を含む請求 項31に記載の方法。 33.前記正常ヒトPKD1遺伝子が、配列番号:4のcDNA配列を含む請 求項31に記載の方法。 34.APKDの特徴を有する疾患状態を治療する方法であって、PKD1遺 伝子機能の欠損を有する細胞に対して、治療的有効量の正常PKD1タンパク質 またはそのフラグメントを投与することを含む上記方法。 35.前記PKD1タンパク質が、配列番号:2で示されるDNA配列によっ てコードされている請求項34に記載の方法。 36.前記PKD1タンパク質が、配列番号:4で示されるcDNA配列によ ってコードされている請求項34に記載の方法。 37.配列番号:5で示されるアミノ酸配列を有するポリペプチドを投与する ことを更に含む請求項35に記載の方法。 38.APKDの特徴を有する疾患状態を治療するための組成物であって、配 列番号:2のDNA配列を有する単離されたヒトPKD1遺伝子またはそのフラ グメントおよび薬学的に許容しうる担体または希釈剤を含む上記組成物。 39.前記PKD1遺伝子が組み込まれているベクターを含む請求項38に記 載の組成物。 40.配列番号:4のDNA配列を有する単離されたヒトPKD1遺伝子また はそのフラグメントを更に含む請求項38に記載の組成物。 41.APKDの特徴を有する疾患状態を治療するための組成物であって、配 列番号:2のDNA配列によってコードされる正常PKD1タンパク質またはそ のフラグメントおよび薬学的に許容しうる担体または希釈剤を含む上記組成物。 42.配列番号:4のDNA配列によってコードされるポリペプチドまたはそ のフラグメントを更に含む請求項41に記載の組成物。 43.組換えPKD1遺伝子またはそのフラグメントをそのゲノムが含んでい る単細胞生物または多細胞生物。 44.前記PKD1遺伝子が、配列番号:2のDNA配列またはそのフラグメ ントを有する請求項43に記載の生物。 45.前記PKD1遺伝子が、配列番号:4の配列またはそのフラグメントを 有する請求項43に記載の生物。
