【発明の詳細な説明】
イソマルツロースおよび他の生成物の製造方法
本発明は、転化された、および転化されていない糖類および/または非糖類生
成物を同時に得る方法に関する。特に本発明は、イソマルツロース(isomaltulos
e)および/またはトレハルロース(trehalulose)およびベタインまたは転化した
糖類の同時製造に関する。
イソマルツロース(またはパラチノース)はスクロースから製造された還元二
糖類である。食品産業では甘味料としてイソマルツロースを使用することが提案
されており、またこれは水素化によって甘味料イソマルト(isomalt)(パラチニ
ト(pala-tinit))を製造するための原料でもある。イソマルトは本質的に均一な
α−D−グリコピラノシル−(1,6)−ソルビトールおよびα−D−グルコピ
ラノシル−(1,1)−マンニトールの混合物である。
トレハルロースはスクロースから得られるもう一種の還元糖である。トレハル
ロースの水素化によりα−D−グルコピラノシル−(1,1)−ソルビトールお
よびα−D−グルコピラノシル−(1,1)−マンニトールの混合物が得られ、
これもまた甘味を有する。
スクロースからイソマルツロースおよび/またはトレハルロースへの異性化ま
たはトランスグルコシル化に関して種々の方法が報告されている。例えばプロト
アミノバクタールブルム(Protaminobacter rubrum)、セルラチカプリムテイカ(S
erratica plymuthica)、エルウィニアラポンチシ(Erwinia rhapontici)、プセウ
ドモナスメソアシドフィル(Pseudomonas mesoacidophile)等のような微生物中に
存在することが発見されているトランスグルコシラーゼ酵素によって異性化が行
われる。公知の異性化技術には、生きている微生物細胞もしくは死んだ微生物細
胞を使用する異性化、または抽出された状態で酵素を使用する異性化が含まれる
。該酵素は固定化された状態および遊離状態の両方で使用されている。
イソマルツロースおよび/またはトレハルロースの製造に関する従来技術の方
法は、例えば欧州特許EP028900号、米国特許US4386158号、米
国特許US4640894号、独国特許DE1049800号、欧州特許EP1
099号、欧州特許EP77971号、欧州特許EP049801号、欧州特許
EP200069号、欧州特許EP160253号、欧州特許EP483755
号および欧州特許EP625578号のような刊行物に記載されている。スクロ
ースのトランスグルコシル化に関する検討は、刊行物、ムニア,エム.(Munir,M
)等,カルボヒドレート リサーチ(Carbohydrate Res.),164(1987)477-485に記
載されている。酵素によって糖蜜からイソマルツロースを製造する方法の技術背
景については本出願人が1996年5月17日に出願したフィンランド出願96
2095号に述べられている。上記特許、特許出願および刊行物の明細は本文中
に参考として取り入れられている。
ベタインは式(CH3)3N+CH2COO-を有する第四アンモニウム化合物で
ある。ベタインが砂糖大根中に蓄積されることは公知であり、そしてこれは一般
的に砂糖大根糖蜜または蒸留かすからイオン交換、結晶化、抽出のような種々の
技術により、またはクロマトグラフィーにより回収される。ベタインは動物飼料
ならびに薬学的用途および化粧品用途のために使用される。
ベタインの回収に関する従来技術の方法は、例えば欧州特許EP54544号
および欧州特許EP345511号に記載されており、この明細は本文中に参考
として取り入れられている。
サトウキビの液体(juice)にはベタインが含まれないが、その代わりそれらは
グルコースとフルクトースの混合物である転化した糖類を多量に供給する。転化
した糖類をそのまま甘味料として使用することもできるが、精製後に更にグルコ
ースおよびフルクトースに分離することも可能である。
転化した糖類をサトウキビから得るための従来技術の方法は、例えば刊行物、
サスカ,エム.(Saska,M.)等,インターナショナル シュガー ジャーナル(Int
.Sugar Jnl.),1994,96巻,1150号,403-410頁;およびホンギスト,エッチ.(
Hongisto,H.)等,フィンシュガー クロマトグラフィー分離法によるサトウキビ
糖蜜の脱糖化(Desuarization of cane molasses by the Finnsugar chromato-gr
aphic separation process),フィニッシュ シュガー コーポレイションリミ
テッド(Finnish Sugar Co.Ltd.),カントヴィク(Kantvik),フィンランド,
1977年3月4日;に記載されており、これらの明細は本文中に参考として取
り入れられている。上述された従来技術ではサトウキビ糖蜜のバッチ式または連
続式クロマトグラフィーによる脱糖化が検討されている。この方法によりスクロ
ースおよび転化した糖類の分離回収が可能である。
更に米国特許第3214293号には、イオン交換およびイオン排除を組み合
わせることにより糖水溶液中の糖類から非糖類を分離することが開示されている
。
英国特許第1448524号には、カチオン交換体における液体分配クロマト
グラフィーにより砂糖大根またはサトウキビ糖蜜の非糖類から糖類を分離する方
法が記載されている。転化した糖類およびベタインを含む糖類および非糖類のフ
ラクションが回収され得る。
イソマルツロースの製造に関連するベタインまたは転化した糖類を回収するた
めに供給された従来技術の方法は一つもない。
本発明の目的は、植物から誘導された水溶液から貴重な糖類および非糖類生成
物を得る方法を供給することである。
本発明のもう一つの目的は、糖蜜から糖類および非糖類を製造する新しい方法
を供給することである。
本発明の特別な目的としては、糖類含有水溶液、特に糖蜜からベタインまたは
転化した糖類ならびにイソマルツロースおよび/またはトレハルロースを回収す
ることを可能にする方法を供給することが挙げられる。
本発明の更なる目的としては、イソマルツロースおよび/またはトレハルロー
スの水素化生成物を得る方法を供給することが挙げられる。
今や、砂糖大根またはサトウキビから誘導された水溶液中のスクロースを、溶
液中に存在するベタインまたは転化した糖類のそれぞれに実質的な影響を及ぼさ
ずにイソマルツロースおよび/またはトレハルロースに転化できること、ならび
に得られた液体から個々の目的生成物を良好な収率で回収できることが発見され
た。また、糖蜜中の糖類は直ちにイソマルツロースおよび/またはトレハルロー
スに転化され得ること、ならびに上記の転化された糖蜜からベタインまたは転化
した糖類を回収しながら、生成した二糖類を効果的に回収できることも発見され
た。
本発明は上述の発見を基礎とするものである。従って本発明は、イソマルツロ
ースおよび他の糖類および/または非糖類生成物を同時に得る方法であって、以
下の段階
a)スクロースならびに目的の糖類または非糖類化合物を含む、植物から誘導さ
れた水溶液を供給すること;
b)上記溶液中の上記スクロースをトランスグルコシル化させること;ならびに
c)上記トランスグルコシル化された溶液からイソマルツロースおよび目的の糖
類および/または非糖類生成物を回収すること;
を組み合わせることからなる方法を供給する。
本発明の好ましい態様に従って、回収方法は個々のフラクションとして目的の
生成物を回収するクロマトグラフィーからなる。
植物から誘導された液体に多量のスクロースが含まれている場合、例えば希薄
または濃厚な溶液である場合、本発明による方法には植物から誘導された溶液か
ら糖蜜を供給するためにスクロースを結晶化する段階が含まれ得る。あるいは上
記植物から誘導された溶液中の全スクロースをイソマルツロースおよび/または
トレハルロースへの転化のために使用することもできる。上記溶液中のイソマル
ツロースの比率が大きい場合、他の生成物の回収の前に結晶化によりイソマルツ
ロースの少なくとも一部分を分離することが好ましい。
本発明の好ましい態様において、植物から誘導されたそのような溶液には糖蜜
が含まれる。植物から誘導された溶液が砂糖大根から誘導される場合、イソマル
ッロース、トレハルロースおよびベタインが本方法の段階c)で回収される主要
な目的生成物である。上記溶液がサトウキビから誘導された場合、イソマルツロ
ース、トレハルロースおよび転化された糖類が段階c)で回収される。
本発明による他の態様において、ベタインはスクロースのトランスグルコシル
化の前に上記溶液から回収される。
イソマルツロースおよび/またはトレハルロースへのスクロースの転化は、微
生物中にあってスクロースをイソマルッロースおよび/またはトレハルロースに
転化することができるトランスグルコシル化酵素(スクロース−6 グリコシル
ムターゼ)によってもたらされる。イソマルツロースの製造に好ましい微生物は
プロトアミノバクタールブルム(Protaminobacter rubrum)(CBS 574.7
7)であるか、他の転化微生物を使用することもできる。固定化された、もしく
は遊離の微生物細胞を使用して、または細胞から単離された酵素を使用して、従
来技術のいずれかの方法を使用することができる。実際には酵素の転化活性を有
することだけが必要であり、この酵素活性が供給される方法は本発明の性能に関
して重大ではない。例えば活性の増加は再結合の技術によってもたらされ得る。
スクロースを含有し、植物から誘導された水溶液であって本方法の原料を形成
するものは、例えば砂糖大根またはサトウキビ粉砕機の希薄または濃厚な液体の
ような植物液体であってよい。一方、糖蜜を供給するために一回もしくは数回の
結晶化段階での結晶化によりスクロースを製造する慣用の方法で濃厚な液体を使
用でき、すなわち、これは上記された植物から誘導された溶液として適当である
。
本発明に関して“植物から誘導された溶液”という表現は、多量のスクロース
および他の植物から誘導された化合物を含む溶液をいずれも包括すると理解され
るべきであり、これは本発明の原料として適当である。
多量に入手し易く、安価であり、そして残渣のスクロース成分がイソマルツロ
ース製造のための良好な出発原料となるため、糖蜜は本発明の好ましい原料であ
る。糖蜜にはスクロースならびに細菌の増殖に消費され得るタンパク質およびミ
ネラルのような他の栄養分が含まれる。これらの栄養分化合物は微生物の増殖を
助けるために微生物によって消費され、転化微生物の増殖と本発明によるスクロ
ースの転化を同時に保持することが可能である。
砂糖大根糖蜜には乾燥固体(DS)を基準として約50ないし70%のスクロ
ース、およびDSを基準として約3ないし8%のベタインおよび他のいくらかの
窒素含有化合物が含まれる。サトウキビ糖蜜にはDSを基準として約35ないし
55%のスクロース、およびDSを基準として約10ないし35%の転化した糖
類が含まれる。従来技術において糖蜜は細胞懸濁液の製造に関して、スクロース
をイソマルツロースに転化させる微生物増殖のための栄養分として述べられてき
た。しかしながら純粋なスクロース溶液で、または希薄もしくは濃厚な植物溶液
でイソマルツロースが実際に製造された。糖蜜は上記細菌の良好な栄養培地とし
て働き、そして糖蜜はイソマルツロースおよび/またはトレハルロースの実際的
な製造に適当なスクロース源を供給することが今や発見された。糖蜜中に存在す
るベタインまたは転化した糖類はそれぞれ転化の工程中に物質的な変化を起こさ
ずに残り、そして得られた反応混合物からイソマルツロースおよび他の糖類およ
び非糖類とは分離されてベタインまたは転化された糖類をそれぞれ回収できるこ
とも発見された。
本発明の利点は、転化のために糖蜜のスクロースを使用できるので、イソマル
ツロースおよびベタインの両方または転化した糖類フラクションを製造するため
に現存する糖蜜のクロマトグラフィー分離粉砕機を使用できることである。
本発明について本文中の添付の図を参考にして更に詳細に記載する。
図1に本発明による方法の構成図を示す。
以下の詳細な記述において、糖蜜を使用することにより誘導される特別な性質
のために本発明の方法の原料として−般的に糖蜜が使用される。しかしながら、
他の植物から誘導された溶液を溶液中に含まれるスクロースのトランスグルコシ
ル化のために使用できることは明白であり、そして糖蜜に関して記載されたもの
と同様の方法で、得られた液体からイソマルツロースおよび他の糖類および/ま
たは非糖類生成物を分離できることも明白である。
糖類含有溶液は−般的に、希薄な液体を供給する慣用の方法で砂糖大根または
サトウキビのような糖類含有植物を加工することにより供給される。希薄な液体
は精製後そのまま液体に含まれるスクロースをイソマルツロースおよび/または
トレハルロースに転化するために使用でき、また濃厚な液体にも加工され得る。
続いて濃厚な液体を結晶化させ、スクロースのほとんどを回収することができる
。結晶化は慣用のように三段階の結晶化であってよく、または溶液中の糖類をよ
り多く残すために一回もしくは二回だけスクロースを結晶化させることが好まし
い。
本発明に従って、スクロースをイソマルツロースおよび/またはトレハルロー
スに転化できるトランスグルコシル化酵素活性に糖類含有溶液を接触させる。好
ましい方法においてピー.ルブルム(P.rubrum)(CBS574.77)の生成物
のような転化微生物の細胞懸濁液のための増殖培地は、糖蜜または糖蜜、コーン
スティープ(corn steep)溶液、および無機栄養分、例えば(NH4)2HPO4の
混合物である。約30℃およびpH7において良好な通気を伴う醗酵により、
109/mlもしくはそれより多くの微生物細胞の所望のカウントがもたらされ
る。バッチ法により醗酵肉汁を糖類含有溶液に添加し、そして適当な転化条件下
で転化を行う。
当該分野で公知であるように、生きている状態または死んだ状態で担体中また
は担体上に微生物細胞を固定化することも可能である。あるいは酵素を微生物細
胞から抽出し、それをそのまま、または固定化した状態で使用して転化を行うこ
とも可能である。例えば本明細書の緒言部分で言及された参考文献を参考とし、
これらは全て本文中に参考として取り入れられる。
スクロースをトランスグルコシル化する方法はバッチ法または連続法で、固定
化された、または固定化されていない状態の酵素系を含むタンクまたはカラム中
で実行され得る。バッチ法は転化の間に付加的な炭素源を溶液に供給して微生物
の増殖を助ける供給バッチ法として実行され得る。25ないし35℃の最適温度
で、好ましくは約30℃で適当に通気をしながら好ましく転化が行われる。転化
に糖蜜が使用される場合、好ましくは過度の泡形成を防止するために液体中に消
泡剤を添加する。
固定化された酵素を有する担体を詰めたカラム中で連続法として転化工程を行
う場合、例えばその他のものが操作中でありながら、あるカラムに再生物を受け
られるように、連続して、そして/または並列して連結され得るいくつかのカラ
ムが存在する。転化収率を増加させるため、一つまたはいくつかのカラムを通し
て溶液を再循環することもできる。
一般的に好ましくは、十分な時間トランスグルコシル化を続けてスクロースの
80%またはそれより多くを転化する。転化後の溶液には主反応生成物としてイ
ソマルツロースが含まれ、そして−般的には多少のトレハルロース、ベタインに
加えてフルクトースおよびグルコースならびにいくらかの未反応スクロースも含
まれるか、あるいは最初に供給された溶液の転化した糖類および他の転化してい
ない化合物が含まれる。
所望の転化が達成されたら生きている微生物細胞を殺す必要がある。これは例
えば30分間もしくはそれより長い間、温度を約80℃に上げることにより実行
される。溶液をクロマトグラフィーによって分離する前に、死んだ細胞および他
の固体残渣を遠心分離および/またはろ過により除去する必要がある。
スクロースが結晶化されていない溶液で転化を行う場合、イソマルツロースの
量が多いのでクロマトグラフィーによる分離を行う前に慣用の方法で溶液からイ
ソマルツロースの主要部分を結晶化することが好ましい。
イソマルツロース/トレハルロースおよびベタインまたは転化した糖類のクロ
マトグラフィーによる分離は、スクロースおよびベタインまたは転化した糖類を
それぞれ分離するための従来技術で使用されたものと同じ方法および装置を使用
して行われ得る。
転化された溶液から例えばろ過により不溶性不純物をいずれも除去し、その後
イオン交換により軟化して二価のイオンを除去する。その後、例えばNa,Kの
ような一価型のカチオン交換樹脂を含むクロマトグラフィー分離カラム中に溶液
を供給する。
本発明による代表的な方法ではトランスグルコシル化された溶液にパスツール
氏殺菌法を行い、次いで遠心分離および/またはろ過を行い、微生物細胞をいず
れも除去する。サトウキビ糖蜜の場合、透明溶液を得るためにリン酸塩沈澱によ
るろ過段階と遠心分離段階を組み合わせる必要がある。例えばイオン交換により
ろ液を軟化し、そして蒸発させて体積を減少させる。ろ液を(好ましくはpH5
.5またはそれより高いpHで)分離カラム中に供給し、そして水で展開して糖
類および非糖類生成物の種々のフラクションを供給する。フラクションの実際の
組成は使用された初期供給溶液に依存する。−般的に個々のフラクションを再び
蒸発させ、次の加工段階のために濃度を増加させる。
本発明の方法により、フラクションにはイソマルツロース、多少のトレハルロ
ースおよびベタインまたは転化した糖類がそれぞれ含まれる。フラクションの一
部分は更に純粋な生成物を供給するために再生されてよい。分離で得られたイソ
マルツロースを結晶化し、イソマルツロース結晶を供給する。イソマルトを供給
するためにイソマルツロースを使用することができ、これは甘味料として市販さ
れ、使用されている。イソマルトはイソマルツロースから、イソマルツロースの
結晶化の後またはクロマトグラフィーカラムからのイソマルツロースフラクショ
ンから直接、水素化することにより製造される。トレハルロースの水素化も甘味
料混合物を供給するので、イソマルツロースとトレハルロースを組み合わせて水
素化してもよい。水素化の前にイオン交換をして精製することができる。同様に
、例えば欧州特許EP54544号または欧州特許EP345511号に記載さ
れているように溶液からベタインを回収することもでき、これらの文献は本文中
に参考として取り入れられている。転化した糖類のフラクションは液体の状態で
保持されるか、または蒸発させてシロップ状とされる。転化した糖類を蒸発の前
に更に精製するために、例えばイオン交換を使用できる。
以下の実施例で本発明について説明するが、いずれかの方法に限定するもので
はない。
実施例1 イソマルツロース/トレハルロース製造のための供給材料としての糖蜜および スクロース
プロトアミノバクタールブルム(Protaminobacter rubrum)(CBS 574.
77)醗酵肉汁の製造:
ピー.ルブルム(P.rubrum)の材料培養を+4℃の栄養寒天培地斜面上に保持し
た。培養からの細胞を2mlの塩水で希釈した。得られた懸濁液の0.4ないし
0.6mlのアリコートを使用し、滅菌済の遮蔽された500ml振とうフラス
コに増殖培地の9×300mlのアリコートを接種した。接種されたフラスコを
230rpm、30℃で1日間振とうした。増殖培地には、0.01M Na2
HPO4緩衝液,pH7中に5%スクロース、1%ペプトン、0.5%酵母エキ
ス、0.3%ビーフエキスが含まれた。濃度が10×109細胞/mlに達した
ら、一つは砂糖大根糖蜜から、そしてもう一つは純粋なスクロース溶液からの、
二つの別々の実験におけるトランスグルコシル化に醗酵肉汁を使用した。
トランスグルコシル化:
DSを基準として60.7%のスクロースを含む砂糖大根糖蜜(クルター リ
ミテッド(Cultor Ltd.))を糖蜜試験に使用した。上述されたピー.ルブルム増
殖培地2600mlを7lの殺菌済の糖蜜−水溶液と一緒に転化醗酵機(バイオ
スタット(Biostat),10l)に移した。転化開始にはスクロース濃度は200
g/lであり、前乾燥固体濃度は325g/lであり、そしてpHはpH7に調
節した。転化は13時間、殺菌された条件下、30℃で、10l/分の通気をし
ながら、そして350rpmで攪拌しながら、ほとんどのスクロースが転化され
るまで続けられた。
糖蜜が転化に使用される場合、泡形成が非常に問題であった。泡形成の連続的
な検出および消泡剤の添加が必要であった。転化が完了してスクロースが検出さ
れなくなったら、30分間溶液の温度を80℃まで上げて微生物を殺すことによ
り転化を停止した。ボディーフィード(body feed)として0.1%のケナイト(Ke
nite)300を使用してサイズ圧ろ過により細胞および他の固体残渣をろ過した
。
もう一方の実験では、砂糖大根糖蜜のものと同様の条件下で純粋なスクロース
溶液を転化したが、ただしスクロースの濃度は325g/lであった。
両方の転化された溶液の組成をHPLC(イオン交換樹脂はPb2+型)により
分析し、そして結果を以下の表に示す。 実施例2 ピー.ルブルム細胞による糖蜜スクロースのトランスグルコシル化
醗酵
プロトアミノバクタールブルム菌株(CBS574.77)の寒天斜面からの
細胞を塩水2mlで希釈した。得られた懸濁液の0.4ないし0.6mlのアリ
コートを使用して、滅菌済の遮蔽された500ml振とうフラスコに増殖培地の
300mlを接種した。接種されたフラスコを230rpm、30℃で1日間振
とうした。増殖培地には0.01M KH2PO4緩衝液,pH7中に60g/
lスクロース、10g/lペプトン、6g/lビーフエキスが含まれた。一日古
い増殖培地1200mlを8.5lの栄養培地(スクロース70g/l,コーン
スティープ溶液40g/l(NH4)2HPO4 0.9g/l,pH7)と共に
醗酵機(バイオスタット,10l)に移した。
温度30℃、pH6ないし7、350rpmで混合しながら、そして12l/
分の通気速度で、細胞密度が10×109カウント/mlまで増加するまで醗酵
を続けた。1500l醗酵機中で、上述の組成の殺菌された栄養培地1100l
に接種するために全醗酵肉汁を使用した。30℃、1100l/分の通気速度お
よび攪拌速度350rpmで醗酵が行われた。
濃度が10ないし20×109細胞/mlに達したとき、糖蜜中のスクロース
をイソマルツロースおよび他の糖生成物へトランスグルコシル化するために全醗
酵肉汁を使用した。
トランスグルコシル化
DSを基準として60%のスクロースを含む砂糖大根糖蜜(クルター リミテ
ッド(Cultor Ltd.))(1800kgDS)を反応容器(10m3)中で370g
DS/lの濃度まで希釈し、20分間120℃で殺菌し、30℃まで冷却した。
pHをH2SO4によりpH6に調節する。1100lのピー.ルブルム醗酵肉汁
(10ないし20×109細胞/ml)を砂糖大根糖蜜溶液(4800kg)に
導入し、30℃で、1500l/分で通気し、そして200rpmで混合しなが
らスクロースに作用させる。消泡剤マズ(Mazu)100(ピーピージー インダス
トリーズ インコーポレイテッド(PPG Industries Inc.),英国)を使用して、
転化の間の重大な泡形成を回避する。25時間以内に実際的に糖蜜中のスクロー
スの全量を転化し、転化された溶液の糖類組成を以下に示した。
イソマルツロース 糖類を基準として82% DSを基準として41%
トレハルロース 8.8% 5.3%
フルクトース 3.3% 2%
グルコース 2% 1%
オリゴ糖 3.9% 2%
転化の後、ピー.ルブルム細胞物質をセパレーターを介して溶液から分離し、
そして細菌ミルク(bacteria milk)を新しい反応に再利用した。セパレーターか
らの溶液に80℃で30分間パスツール氏殺菌法を行い、残りの細菌を殺した。
溶液を冷却し、ろ過した。
実施例3 イソマルツロース、トレハルロースおよびベタインのクロマトグラフィー分離
実施例2のトランスグルコシル化された溶液を、クロマトグラフィー分離カラ
ム中でクロマトグラフィーにより分離する。
分離はバッチ法であるパイロットクロマトグラフィー分離カラムにおいて行わ
れる。全装置は、供給タンク、供給ポンプ、熱交換機、カラム、排出ポンプ、生
成物コンテナ、供給溶液および希釈水の導入および排出のためのパイプラインな
らびに排出される液体を制御する装置およびバルブから構成された。
直径1.0mのカラムにカチオン交換樹脂(フィネックス オイ(Finex Oy),
フィンランド製)を詰め、樹脂ベッドの高さは約5.3mであった。樹脂の架橋
結合度は5.5%DVBで、そして平均粒径は約0.35mmであった。ナトリ
ウム(Na+)型に樹脂を再生し、次いで供給装置を樹脂ベッドの上部に組み立
てた。カラムならびに供給溶液および展開水の温度は約80℃であった。カラム
の流速を630l/hに調節した。
供給溶液の最初の前処理段階はろ過であり、これはろ過助剤としてケイソウ土
を使用した圧ろ過により行われた(溶液の温度は約80℃,濃度は100g溶液
中に約30gの乾燥固体)。この後、カルシウムイオンを除去するためにイオン
交換により溶液を軟化した(温度は約55℃,濃度はろ過と同じ)。次いで分離
の前に溶液を再びろ過した。
クロマトグラフィーによる分離は以下のように行われた。
段階1:供給溶液の乾燥固体含有量を決定し、そして溶液の屈折率(RI)に
