Claims (1)
- 【特許請求の範囲】 1. 発現系として形質転換された繊毛虫細胞を用いることを含む、発現系 で異種DNAを発現させる方法。 2. 異種DNA配列が遺伝子である、請求項1記載の方法。 3. DNA配列がヒト起源である、請求項1または2に記載の方法。 4. DNAが動物起源である、請求項1または2に記載の方法。 5. DNAが野菜起源である、請求項1または2に記載の方法。 6. DNAが細菌起源である、請求項1または2に記載の方法。 7. DNAが真菌起源である、請求項1または2に記載の方法。 8. 以下の段階: a)以下のDNA部分配列 i)繊毛虫内で活性であり、発現させるべき異種DNAの転写を 引き起こすプロモーター; ii)センス方向でプロモーターに連結された、発現させるべき異 種DNA; iii)転写を終結させ、そして繊毛虫内で活性である、終止シグ ナル; iv)所望であれば、宿主細胞内でのベクターの複製をもたらす、 適当なori(複製開始点): を含む発現ベクターの調製、および、 b)段階(a)に記載した発現ベクターでの繊毛虫の形質転換: を特に含む、請求項1から7のいずれか一項またはそれ以上の項に記載の方法。 9. ミクロキャリヤーボンバードメント法を用いて繊毛虫細胞をDNA充 填金粒子で形質転換する、請求項1から8のいずれか一項またはそれ以上の項に 記載の方法。 10. 異種DNAを含むプラスミドpRT103gusを繊毛虫細胞の形質 転換に用いる、請求項8または9に記載の方法。 11. プラスミドが、カリフラワーモザイクウイルス35Sプロモーター、 異種DNAのコード領域、およびポリアデニル化シグナルを含む、請求項10記 載の方法。 12. 繊毛虫細胞がホロトリチア(Holotrichia)、ペリトリチ ア(Peritrichia)、スピロトリチア(Spirotrichia) およびスクトリア(Suctoria)の群から選択される、請求項1から11 のいずれか一項またはそれ以上の項に記載の方法。 13. 繊毛虫細胞が、テトラヒメナ(Tetrahymena)、パラメシ ウム(Paramecium)、コルピディウム(Colpidium)、コル ポダ(Colpoda)、グラウコマ(Glaucoma)、プラチオフリア( Platyophrya)、ボルチセルラ(Vorticella)、ポトマク ス(Potomacus)、シュードコニレムブス(Pseudocohnil embus)、エウプロテス(Euplotes)、エンゲルマニエルラ(En gelmaniella)およびスチロニチア(Stylonychia)の群 から選択される、請求項12記載の方法。 14. 繊毛虫スチロニチア・レムナエ(Stylonychia lemn ae)種の細胞を発現系として用いる、請求項13記載の方法。 15. 異種DNAがインビトロで修飾される、請求項1から14のいずれか 一項またはそれ以上の項に記載の方法。 16. 発現生成物がタンパク質である、請求項1から15のいずれか一項に 記載の方法。 17. 異種DNAで形質転換することによって得ることのできる、繊毛虫。 18. 形質転換が、ミクロキャリヤーボンバードメント法を用いて、DNA を充填した金粒子で行われた、請求項17記載の繊毛虫。 19. 以下の手順段階: a)以下のDNA部分配列: i)繊毛虫内で活性であり、発現させる異種DNAの転写を引き 起こすプロモーター; ii)センス方向でプロモータに連結される、発現させる異種DN A; iii)転写を終結させ、そして繊毛虫内で活性である、終止シグ ナル; iv)所望であれば、宿主細胞内でのベクターの複製をもたらす、 適当なori(複製開始点): を含む発現ベクターの調製、および b)段階(a)に記載の発現ベクターでの繊毛虫細胞の形質転換: によって得ることのできる、請求項17または18に記載の繊毛虫。 20. 異種DNAを含むプラスミドpRT103gusで形質転換された、 請求項17から19のいずれか一項またはそれ以上の項に記載の繊毛虫。 21. プラスミドがカリフラワーモザイクウイルス35Sプロモーター、異 種DNAコード領域、およびポリアデニル化シグナルを含む、請求項20記載の 繊毛虫。 22. 発現ベクターが、細胞から分泌される遺伝子生成物をもたらすシグナ ル配列をさらに含む、請求項8記載の方法。 23. 発現ベクターが、細胞から分泌される遺伝子生成物をもたらすシグナ ル配列をさらに含む、請求項19記載の繊毛虫。
