Claims (1)
- 【特許請求の範囲】 1.植物細胞の細胞増殖を再誘導および刺激することによりインビトロ培養植 物細胞の増殖速度および細胞濃度を高める方法であって、 a)植物細胞培養物を栄養培地中で細胞分裂の開始に適した増殖条件下に増殖 させ;そして b)細胞培養物に、初期増殖段階と細胞培養物の死の前との間のいずれかの時 点で、細胞培養物にとって有毒でなく細胞分裂を再誘導するのに十分である量の 追加のアンモニウムイオンを補充し; これにより、培養物の植物細胞濃度を高めるために細胞分裂を再誘導、刺激、維 持および増大させる 工程を含む方法。 2.工程a)の増殖を、培養物の酸素取込み速度を監視しながら行わせる、請 求項1記載の方法。 3.工程b)のアンモニウムイオンを、酸素取込み速度が実質的にプラトーに 達したときに添加する、請求項2記載の方法。 4.工程b)のアンモニウムイオンが、(NH4)2SO4、NH4NO3、(N H4)2PO4、酢酸アンモニウムおよびグルタミンよりなる群から選択される、 請求項1記載の方法。 5.工程b)がさらに、炭水化物、溶存酸素、ホスフェートおよびカリウムよ りなる群から選択される栄養素を添加することを含む、請求項1記載の方法。 6.炭水化物が、ショ糖、グルコースおよびフルクトースよりなる群から選択 される、請求項5記載の方法。 7.工程a)の増殖を、フラスコ、いずれか適切な培養器、またはバイオリア クター内で行わせる、請求項5記載の方法。 8.工程a)の増殖を、培養器またはバイオリアクター内において、バッチ式 、供給バッチ式または連続式で行わせる、請求項7記載の方法。 9.適切な増殖条件が、無菌性、混合速度、温度、光、酸素供給および栄養培 地である、請求項7記載の方法。 10.さらに工程b)の後に実施する2工程: c)ファイトケミカル産生のために植物細胞培養物を改変すること;そして d)ファイトケミカルを産生させ、産生したファイトケミカルを増殖した植物 細胞バイオマスおよび培地から単離すること; を含む、請求項9記載の方法。 11.ファイトケミカルが、アルカロイド類、タキサン類、タキシン類、テル ペン類、ステロイド類、キノン類、フラボノイド類、タンニン類、サポニン類、 クマリン類、カロテノイド類、およびその任意の生合成中間体よりなる群から選 択される、請求項10記載の方法。 12.さらに工程b)の後に実施する2工程: c)組換えタンパク質および抗体の産生のために植物細胞培養物を改変するこ と;そして d)組換えタンパク質および抗体を産生させ、産生した組換えタンパク質およ び抗体を増殖した植物細胞バイオマスおよび培地から単離すること; を含む、請求項9記載の方法。 13.植物ベースの培養法を工業的規模にまで拡大する方法であって、 a)植物細胞培養物を請求項1記載の方法に従って増殖させて、高い細胞濃度 の第1容量の培養物を得て;そして b)植物細胞培養を工業的規模にまで拡大するために、濃厚な第1容量の少な くとも一部を第2容量の培養物に接種し、その際第2容量は第1容量より大きく ; これにより、植物細胞培養物の接種容量比が従来のバッチ式培養法を用いた場合 より低くなる; 工程を含む方法。 14.高濃度が細胞数約3×106〜60×106個/mlである、請求項13 記載の方法。 15.接種容量比が約1:20〜1:60である、請求項14記載の方法。
