- 【特許請求の範囲】 1.タンパク質の免疫グロブリン(Ig)スーパーファミリーの哺乳動物DS-CAMメン バーをコードする単離された核酸、またはその断片であって、前記DS-CAMが少 なくとも7つのIg様ドメインを含むものである、上記核酸。 2.前記核酸またはその断片が、 (a)配列番号2もしくは配列番号11に示したアミノ酸配列、または配列番号 7、配列番号8もしくは配列番号9のDS-CAMコード領域をコードするDNA、 (b)中程度のストリンジェント条件下で(a)のDNAとハイブリダイズするD NAであって、生物学的に活性なDS-CAMをコードするもの、または (c)前記の(a)または(b)に対する縮重DNAであって、生物学的に活性なDS- CAMをコードするもの、 から選択される、請求項1に記載の単離された核酸。 3.前記核酸が、高度のストリンジェント条件下で、配列番号1、配列番号7、 配列番号8、配列番号9または配列番号10のヌクレオチドのDS-CAMコード部分 とハイブリダイズする、請求項2に記載の核酸。 4.前記核酸のヌクレオチド配列が配列番号1、配列番号7、配列番号8、配列 番号9または配列番号10に示したものと実質的に同一である、請求項2に記載 の核酸。 5.前記核酸のヌクレオチド配列が配列番号1、配列番号7、配列番号8、配列 番号9または配列番号10に示したものと同一である、請求項2に記載の核酸。 6.前記核酸がcDNAである、請求項2に記載の核酸。 7.請求項2に記載の核酸を含有するベクター。 8.請求項2に記載の核酸を含有する組換え細胞。 9.配列番号1、配列番号7、配列番号8、配列番号9または配列番号10に示し たヌクレオチド配列の核酸の配列と特異的にハイブリダイズすることが可能な 少なくとも15ヌクレオチドを含む、オリゴヌクレオチド。 10.前記オリゴヌクレオチドが検出可能なマーカーで標識されている、請求項9 に記載のオリゴヌクレオチド。 11.請求項2に記載の前記核酸によってコードされるmRNAと特異的に結合す ることが可能なアンチセンス・オリゴヌクレオチド。 12.請求項10に記載の少なくとも1種のオリゴヌクレオチドを含む、DS-CAMのc DNA配列の存在を検出するためのキット。 13.少なくとも7つのIg様ドメインを含む、単離されたDS-CAMタンパク質。 14.肺、肝臓または腎臓よりも脳において有意に高い量で発現されることをさら に特徴とする、請求項13に記載のDS-CAMタンパク質。 15.前記タンパク質のアミノ酸配列が、配列番号2もしくは配列番号11に示した タンパク質配列、または配列番号7、配列番号8もしくは配列番号9のDS-CAM コード領域と実質的に同一のものを含む、請求項13に記載のDS-CAMタンパク質 。 16.配列番号2もしくは配列番号11に示したタンパク質配列、または配列番号7 、配列番号8もしくは配列番号9のDS-CAMコード領域と同一のアミノ酸配列を 含む、請求項15に記載のDS-CAMタンパク質。 17.前記タンパク質が配列番号1、配列番号7、配列番号8、配列番号9または 配列番号10に示したヌクレオチド配列と実質的に同一のものを含むヌクレオチ ド配列によってコードされる、請求項13に記載のDS-CAMタンパク質。 18.前記タンパク質が配列番号1または配列番号10を含むヌクレオチド配列によ ってコードされる、請求項17に記載のDS-CAMタンパク質。 19.前記タンパク質が配列番号1に示したヌクレオチド453〜6185、配列番号10 に示したヌクレオチド453〜5168、配列番号7、配列番号8または配列番号9 と実質的に同一のヌクレオチド配列を含むヌクレオチド配列によってコードさ れる、請求項13に記載のDS-CAMタンパク質。 20.DS-CAM関連タンパク質の発現方法であって、請求項8に記載の細胞を、前記 DS-CAMタンパク質の発現に適した条件下で培養することを含んでなる、上記方 法。 21.請求項13に記載のDS-CAMタンパク質との特異的な反応性を有する、単離され た抗DS-CAM抗体。 22.前記抗体がモノクローナル抗体である、請求項21に記載の抗体。 23.前記抗体がポリクローナル抗体である、請求項21に記載の抗体。 24.DS-CAMタンパク質の発現を抑制するのに有効な量の請求項11に記載のアンチ センス・オリゴヌクレオチドおよび細胞膜を通過することが可能な許容できる 疎水性担体を含有する組成物。 25.DS-CAMタンパク質をコードする外因性核酸を発現するトランスジェニック非 ヒト哺乳動物。 26.前記DS-CAMタンパク質をコードする前記核酸が突然変異を起こしており、そ のため発現されたDS-CAMタンパク質が天然のDS-CAMではない、請求項25に記載 のトランスジェニック非ヒト哺乳動物。 27.トランスジェニック非ヒト哺乳動物がマウスである、請求項25に記載のトラ ンスジェニック非ヒト哺乳動物。 28.哺乳動物DS-CAMタンパク質をコードする核酸の同定方法であって、核酸含有 サンプルを請求項9に記載のオリゴヌクレオチドと高度ストリンジェントハイ ブリダイゼーション条件下で接触させ、そしてそれにハイブリダイズする化合 物を同定することを含んでなる、上記方法。 29.サンプル中の哺乳動物DS-CAMタンパク質の存在を検出する方法であって、被 験サンプルを請求項21に記載の抗体と接触させ、抗体−DS-CAM複合体の存在を 検出し、それによって前記被験サンプル中の哺乳動物DS-CAMの存在を検出する ことを含んでなる、上記方法。 30.配列番号1、配列番号10、配列番号7、配列番号8または配列番号9に示し た核酸配列に由来する核酸配列を含む、DS-CAM核酸増幅用の一本鎖DNAプラ イマー。
