Claims (1)
- 【特許請求の範囲】 1. リンパ球におけるグルタチオンの細胞内機能のレベルの決定及びリンパ球に おける酸化防止剤機能の生化学分析で使用に有用な細胞培養培地であって、以下 のものを含み: 次の成分を含む緩衝化無血清溶液: グルコース及びリンパ球内で生物学的にグルコース生産が可能な化合物が構 成する群から選択された炭水化物生物学的使用可能形式パントテン酸、コリンあ るいはリンパ球内でコリン生産可能な生物学的使用可能形式基質 塩化物、リン酸塩、カルシウム、マグネシウム、カリウム、ナトリウム及び 鉄を含む生物学的利用可能形式の無機イオン L−ブチオニンー[S.R.]−スルホキシマイン脱イオン水、及び 分析されるリンパ球を刺激する効果的な量のマイトジェン; pHは約6.8から7.6の当該緩衝化無血清溶液、 リンパ球におけるグルタチオンの細胞内機能レベルの決定及び、リンパ球の 酸化防止剤機能の生化学分析に効果的なものとして特徴付けされる当該細胞培養 培地。 2. 請求項1の細胞培養培地で、培地は生物学的に利用可能な形式のアミノ酸及 びビタミンで構成される群から選 択された栄養素補充物が補充されていて、栄養素が栄養素補充において制限ある いは阻害濃度の範囲外に存在するか、その範囲内に存在するのかを検査される。 3. 請求項1の細胞培養培地で、ビタミンは、ビオチン、フォリニン酸あるいは 生物学的に利用可能な形式のフォリニン酸、ニコチンアミドあるいはニコチン酸 、リボフラビン、チアミン、ビタミンB6及びビタミンB12及び細胞内でそれら を作り出す能力をもつ化合物で構成される群から選択され;アミノ酸あるいは生 物学的にアミノ酸を細胞内で生産する能力をもつ化合物は、L−アルギニン、L −システイン、L−グルタミン、グリシン、L−ヒスチジン、L−イソロイシン 、L−ロイシン、L−リジン、L−メチオニン、L−フェニルアラニン、L−セ リン、L−トレオニン、L−トリプトフアン、L−チロシン、及びL−バリンを 含み、アミノ酸は群として存在し、それぞれの総量は阻害濃度を越えることはな い培地。 4. 請求項1の細胞培養培地で、L−ブチオニンー[S.R.]−スルフォキシ マインが約50μlから約500μlの濃度で存在する培地。 5. 請求項1の細胞培養培地で、ピルビン酸、アデニン、及びイノシトールある いはそれらをリンパ球内で生産することのできる化合物で構成される群から選択 された1種以上の刺激性栄養素をほぼ最大の反応を起こす濃度以 外の濃度で補充されている細胞培養培地。 6. 個体におけるグルタチオンの細胞内機能のレベルの測定及び細胞性酸化防止 剤機能の生化学分析の方法で以下の段階を含む方法: 請求項1の細胞培養培地への固体由来のリンパ球の植え付け; 植え付け細胞培養培地のインキュベーション;及び リンパ球の応答とコントロール群の個体由来のリンパ球の平均的な応答と比 較。 7. ヒトリンパ球におけるシステインの細胞内機能のレベルの決定及びリンパ球 における酸化防止剤機能の生化学分析での使用に有用な細胞培養培地であって、 以下のものを含み: 次の成分を含む緩衝化無血清溶液: グルコース及びリンパ球内で生物学的にグルコース生産が可能な化合物が構 成する群から選択された炭水化物生物学的使用可能形式パントテン酸、コリンあ るいはリンパ球内でコリン生産可能な生物学的使用可能形式基質 塩化物、リン酸塩、カルシウム、マグネシウム、カリウム、ナトリウム及び 鉄を含む生物学的利用可能形式の無機イオン クメンヒドロペルオキシド、 脱イオン水、 N−アセチル−L−システイン、及び 分析されるリンパ球を刺激する効果的な量のマイトジェン; pHが約6.8から7.6である当該緩衝化無血清溶液、 栄養素欠乏、不適合、及び不均衡の決定及び、リンパ球の酸化防止剤機能の 生化学分析に効果的なものとして特徴付けされる当該細胞培養培地。 8. 請求項7の細胞培養培地で、培地は生物学的に利用可能な形式のアミノ酸及 びビタミンで構成される群から選択された栄養素補充物が補充されていて、栄養 素が栄養素補充において制限あるいは阻害濃度の範囲外に存在するか、その範囲 内に存在するのかを検査されるもの。 9. 請求項7の細胞培養培地で、ビタミンが、ビオチン、フォリニン酸あるいは 生物学的に利用可能な形式のフォリニン酸、ニコチンアミドあるいはニコチン酸 、リボフラビン、チアミン、ビタミンB6及びビタミンB12及び細胞内でそれら を作り出す能力をもつ化合物で構成される群から選択される;アミノ酸あるいは 生物学的にアミノ酸を細胞内で生産する能力をもつ化合物は、L−アルギニン、 L−システイン、L−グルタミン、グリシン、L−ヒスチジン、L−イソロイシ ン、L−ロイシン、L−リジン、L−メチオニン、L−フェニルアラニン、L− セリン、L−トレオニン、L−トリプトファン、L−チロシン、及びL−バリン を含み、アミノ酸は群として 存在し、それぞれの総量は阻害濃度を越えることはない培地。 10. 請求項7の細胞培養培地で、クメンヒドロペルオキシドが約50μMから約5 00μMの濃度で存在する培地。 11. 請求項7の細胞培養培地で、最大の反応を起こす濃度以外の濃度のピルビ ン酸、アデニン、及びイノシトールあるいはそれらをリンパ球内で生産すること のできる化合物で構成される群から選択された1種以上の刺激性栄養素で補充さ れている培地。 12. 個体におけるシステインの細胞内機能のレベルの測定及び細胞性酸化防止 剤機能の生化学分析の方法で以下の段階を含む方法: 請求項7の細胞培養培地への個体由来のリンパ球の植え付け; 植え付け細胞培養培地のインキュベーション;及び リンパ球の応答とコントロール群の個体由来のリンパ球の平均的な応答と比 較。
