百日咳菌の繁殖のための新規な方法
本発明は百日咳ワクチンの生産に有用な抗原タンパク質の単離のためのボルデ
テラ属に属する細菌を培養する方法に関する。特に本発明は、百日咳菌の細菌の
増殖の間に、培地中に分泌される主要なタンパク質成分 FHA及びPTの収量を改良
する方法に関する。
百日咳は主に子供を冒す、高度に感染性の病気である。呼吸性の合併症を引き
起こすのに加えて、百日咳は特に低社会経済群の子供及び母親の抗−百日咳抗体
を有しない新生児に、神経の損傷及び高い死亡率をもたらす。百日咳の病原体は
グラム陰性球杆菌である百日咳菌である。細菌は気道に侵入し、数日後に細菌が
消滅した後ですら、毒性状態を引き起こすものと信じられている。
長年にわたり、この病気は発酵槽中で百日咳菌生物体を増殖させ、次いで熱処
理及び/または化学試薬の添加により生じた細胞を不活性化することによって調
製された「全細胞」ワクチンを用いる免疫化により抑制してきた。世界保健機構
は現在、百日咳の発生及び蔓延を防ぐために小児の免疫化を勧めているけれど、
種々のワクチン製剤に起因する報告された不利な作用についての心配が生じた。
結果として生じた通常の百日咳菌ワクチンの利用の減少は百日咳感染の発生の増
加をもたらした。全細胞ワクチンからの報告された不利な作用を避けるために、
少数の高度に精製された抗原タンパク質成分を含む効能のある無細胞ワクチンの
開発にかなりの研究努力が向けられた。
多数の抗原タンパク質が、たとえば、ヒスタミン感受性因子、島(islet)促進
タンパク質またはもっと一般的に百日咳毒素(PT)と
しても知られているリンパ球増加促進因子(LPF)、糸状ヘマグルチニン(FHA)、ふ
さアグルチノーゲン及びパータクチン(pertactin)として知られている分子量約6
9,000ダルトンの細菌の外側の膜タンパク質を包含する、ワクチン成分としての
有用性を有するものと同定されてきた。
百日咳菌細菌はその増殖条件で培養しにくい。炭水化物、ピルビン酸塩、乳酸
塩及び一連の解糖酵素触媒反応の中間体を利用することができず、代りに培地の
炭素及びエネルギー源としてアミノ酸、特にグルタミン酸塩をあてにする。適切
なグルタミン酸塩含有培地はステイナー及びショルテにより記載されている(「
J.J.Gen.Microbral.」63,P.211 〜220,1971 年)。
適切な液体培地中での病原体である、百日咳菌の増殖を包含する百日咳に対す
るワクチンに用いるための抗原物質の生産は、培地への増殖促進剤の添加により
促進することができることが立証された。
欧州特許出願公開第0077646 号明細書は、培地、適切にはステイナー−ショル
テ培地に、シクロデキストリン、特にβ−2,6−O−ジメチルシクロデキスト
リンを補足することを含む、無細胞ブロス中に分泌された抗原物質の量を増強す
る方法を記載している。
米国特許第4,551,429 号明細書は、培地を振とうしながら冠水好気性条件下で
の百日咳菌の繁殖における代りの増殖促進剤の使用を記載している。米国特許第
4,551,429 号によると、約1,500 〜16,000の分子量のポリビニルアルコール(PVA
)、適切には10,000の分子量の物質を0.5 〜2.0 g/l培地のレベルで加えるこ
とも実質的にPT及びFHA のレベルを増強する。
欧州特許出願公開第0077646 号及び米国特許第4,551,429 号明細書に記載のよ
うに、振とうしながらの冠水好気性条件下の繁殖は小
規模生産、たとえば約5lまでの容積の振とうフラスコでの生産に適切である。
しかしながら、これらの条件は、機械的撹拌が必要な大容量発酵槽を必要とする
、大規模の企業生産の典型ではない。
百日咳ワクチンの企業規模の生産に適切な大規模発酵槽中で百日咳菌を繁殖さ
せる時に問題が生じる。この問題は百日咳菌生物体の液体培地の表面に泡層とし
て蓄積する傾向に存する。これは細菌の増殖培地からの除去及び培養過程の早す
ぎる終結を生じる。細菌により分泌された高割合の FHAタンパク質もこの泡層に
蓄積する。泡生成により生じた問題は培地に増殖促進剤を添加することによって
は解決されず、さらにシクロデキストリンの代りにPVA を用いる時、特にPVA が
米国特許第4,551,429 号明細書に開示された濃度範囲の上限で存在する時に、悪
化することが分かった。このように泡生成は、増殖促進剤、特に増殖促進剤とし
てPVA の使用する際の有利な効果を妨害する。
培養発酵に普通に用いられる方法を用いる、百日咳菌に伴う泡問題の経済的解
決はすぐには得られない。化学的消泡剤は通常の培養条件下での実用的な解決を
もたらさないことが分かった。多数の試薬が試験された。それらは百日咳菌細菌
により許容されないか、または、その後で泡がすぐに戻ってくる、短時間のみに
効果があることが分かったかのいずれかであった。
泡立ちを機械的に抑制する破泡機が市販されている。FHAタンパク質の物理的
分解への感受性のため、破泡機により高剪断条件が発生するなら、特に大規模百
日咳菌培養発酵槽のための機械的破泡機の設計に気を配らなくてはならない。
欧州特許出願公開第0121249 号明細書は、シクロデキストリンを培地で増殖促
進剤として用いる時の工業的規模での百日咳菌の防御抗原の生産のための改良方
法を開示している。この方法は、培養温
度及び溶解酸素の制御を伴う脱泡条件下に行なわれる発酵を必要とする。脱泡条
件は化学的でも機械的でもよいが、機械的が好ましい。欧州特許出願公開第0121
249 号明細書は、丸菱理化株式会社(日本)により製造された適切な脱泡条件を
作り出す発酵槽を記載している。機械的脱泡装置のための脱泡翼が、日本特許出
願公開第55-11011号公報(丸菱理化株式会社)に記載されている。
欧州特許出願公開第0121249 号明細書に記載された方法により、シクロデキス
トリンの代りに培地中で増殖促進剤としてPVA を用いて実施した時、百日咳菌の
大規模繁殖は、泡生成レベルの減少及び許容量の抗原タンパク質、特にFHA の単
離に関して、変わりやすく、がっかりさせる結果を示した。
培地においてPVA が用いられるなら、過剰な泡の生成及びそれに伴う、大規模
発酵槽中の百日咳菌からのFHA 分泌レベルの低下の問題は、培地に加えられるPV
A の分子量及び加水分解度の両方を選択することにより克服できる。
したがって、本発明は百日咳菌の企業規模の培養方法を提供し、それは、機械
的撹拌を伴う冠水好気性条件下の水性栄養培地における細菌の繁殖及び前記栄養
培地の、10,000〜50,000の範囲の分子量及び80〜90モル%の加水分解度のPVA の
補足を含むものである。好ましくは加水分解度は85〜90モル%である。
本発明によると、百日咳菌の企業規模の培養は適切には少なくとも10l、たと
えば20〜1000lの範囲の容量の発酵槽で行なう。泡生成を適切には、発酵の間に
発生する泡を消散させるが発酵槽中に高剪断条件を発生しない機械的破泡機、た
とえば日本特許出願公開第55-11011号公報に記載された型の破泡機を用いて制御
する。
本発明において用いるための百日咳菌の適切な菌株は文献中に記載されており
、商業的なコレクション、たとえば米国メリーランド
州ロックビルのアメリカン・タイプ・カルチュア・コレクション(American Type
Culture Collection)で容易に入手できる。液体培地中で抗原タンパク質PT及び
FHA の少なくとも1つ及び好ましくは両方を生産する限りにおいては、本発明の
方法においてはいかなる利用可能な菌株を用いてもよい。
用いることができる菌株の例は、百日咳菌I期(phase I)、百日咳菌II期、
百日咳菌I期CS、百日咳菌トーハマ(Tohama)、百日咳菌株185-30、百日咳菌株18
323、百日咳菌株134、百日咳菌株509、百日咳菌株ウェルカム(Wellcome)28及
び生物学局(Office of Biologics)百日咳菌株165 を包含する。本発明で用いる
のに好ましい菌株は百日咳菌I期であるトーハマであって、それは受理番号IFO-
14073 の下に日本国大阪市の発酵研究所から入手できる。
選択された百日咳菌株は当業者に公知の多くの方法で増殖できる。量及び起原
または種培養の維持方法に依存して、種々の培養段階、及び液体または固体培地
を用いる培養方法が公知である。しかしながら、大規模生産に慣用の許容し得る
植え込み数をもたらすいかなる方法も本発明で用いるのに十分であろう。
百日咳菌接種原の増殖のための適切な培地はいかなる当業者によっても選択で
きる。適切な培地は、限定するものではないが、ジェンゴウ(Gengou)培地(欧州
特許出願公開0077646 号明細書)、エヌ.アンドーン(N.Andorm)他の「Appl.M
icrobiol.Biotechnol.」28、第356 〜360 頁(1988年)及びその中に引用され
た参考文献に記載された培地、ステイナー−ショルテ培地(「J.Gen.Microbio
l.」63、第211 〜220 頁、1971年)、エー.今泉他の「Infect.Immun.」41,3,
第1138〜1143頁(1983年)及び「J.Microbiol.Methods」2、第339 〜347 頁
(1984年)に記載された変性ステイナー−ショルテ培地、バーウェイ(Verway)
培地(米国特許第4,784,58
9 号明細書)、合成培地B2(P.Van Hemert「Prog.Indust.Microbiol.(Bul
l,M.J.編)」Vol.13、第151 頁、Elsevier Sci,Amsterdan,1997 年)また
はそれらの記載された培地を包含する。
本発明による百日咳菌の増殖のために適切な液体培地に接種原を加え、発酵を
前記のように行なった。結果は発酵槽の設計PVA を補足された選択された発酵栄
養培地、方法及び発酵パラメータに依存して変化し得ることが当業者には分かる
だろう。本発明で用いるための好ましい栄養培地はステイナー−ショルテ培地で
ある。
0.5 〜2.5 g/lの範囲、適切には1または2g/lの濃度のPVA がPT及びFH
A の繁殖を増強するのに最も有効であることが分かった。
必須の分子量及び加水分解度を有するPVA は多数の製造者から市販されている
。たとえばアルドリッチは分子量範囲31,000〜50,000及び87〜89モル%の範囲の
加水分解度のPVA を供給し、シグマは平均分子量30,000〜70,000で87〜89モル%
範囲の加水分解度のPVA、フルカ(Fluka)は分子量15,000で86〜89モル%の範囲の
加水分解度のPVA を供給し、ジャンセン(Janssen)は、特定された分子量22,000
及び特定された加水分解度88モル%のPVA を提供する。
PT及びFHA を培養上清から単離し、公知方法により精製できる。次に精製され
た抗原を任意にアジュバント、典型的には水酸化アルミニウムと一緒にして、ワ
クチンとして製剤化できる。ワクチンは有利にパータクチン〔69KDa の外膜タン
パク質(OmP)〕を含むことができ、ワクチンはたとえば本発明の方法に従って増
殖された百日咳菌細胞から調製できる。
次の例は、本発明を説明するものであるが本発明を限定するものではない。
例1
マスター種(Master Seeds)/作業用種(Working Seeds)の調製
a)マスター種
百日咳菌トーハマ菌株(テイジン株式会社、日本)を含有するアンプルの内容
物を脱イオン水(0.5ml)で再構成した。生じた懸濁液を、2%の脱繊維素羊血で
補足されたB.G.寒天(Bordet Gengou、パスツール研究所、パリ)を含有する5枚
のペトリ皿(0.1ml/皿)に画線した。35℃で24〜48時間のインキュベーション後
、各ペトリ皿からの表面増殖を3〜5mlの10%(v/v)グリセロールで補足し
た変性ステイナー−ショルテ(ステイナー−ショルテ)培地(「J. Clin.Micro
biol.」17(5)、第781 〜786 頁、1983年)を用いて再懸濁し、その懸濁液をプ
ールした。プールされた懸濁液のアリコート(0.3ml)をプラスチック製凍結管中
に入れ、直ちに凍結し、−70℃で貯蔵した。
b)作業用種
マスター種を含有するプラスチック製凍結管を室温まで融解させ、その内容物
を寒天を補足された変性ステイナー−ショルテ培地を含有する、各2本のガラス
管に画線した。35℃で24時間のインキュベーション後、各管からの表面増殖を、
寒天を補足された変性ステイナー−ショルテ培地(35ml)を各々含有する、2本
のより大きいガラス管中に画線した。さらに35℃で24時間のインキュベーション
後、各管からの表面増殖を変性ステイナー−ショルテ培地(3ml)で懸濁し、変
性ステイナー−ショルテ培地(150ml)を含有するエルレンマイヤーフラスコ(1
l)に加えた。各フラスコをオービタル振とう機上に置き、35℃で24時間インキ
ュベートした。4つのフラスコの内容物をプールし、懸濁液を8000rpm で、4℃
で15分間遠心
した。上清を捨て、ペレットを10%(w/v)のグリセロールで補足された変性
ステイナー−ショルテ培地(150ml)中に再懸濁した。その懸濁液のアリコート(0.
4ml)をプラスチック製の凍結管中に入れ、直ちに凍結し、−70℃で貯蔵した。
例2
接種原液体プレ培養の調製
例1で調製した作業用種(0.08ml)を寒天を補足した変性ステイナー−ショル
テ培地の形の固体培地(10ml)上で35℃で24時間インキュベートした。表面増殖
を同じ培地(35ml)上に画線し、35℃で24時間インキュベートした。表面増殖を
変性ステイナー−ショルテ培地(3ml)に懸濁し、寒天で補足した変性ステイナ
ー−ショルテ培地(100ml)を含有するルー瓶(1l)に加えた。ルー瓶を35℃で2
4時間インキュベートした。
各ルー瓶からの表面増殖を変性ステイナー−ショルテ培地(6ml)中に再懸濁
し、変性ステイナー−ショルテ培地を含有するエルレンマイヤーフラスコ(5l
)に加えた。本発明による分子量と加水分解度を有するPVA は最終濃度の2g/
lで、β−2,6−O ジメチルシクロデキストリンの代りに存在した。そのフ
ラスコを35℃で24時間インキュベートした。フラスコの内容物を20lの発酵槽に
接種原を提供するためにプールした。
例3
20 l発酵槽中の百日咳菌の増殖
例2で調製された接種原を2g/lの最終濃度で適当なPVA を含有する13lの
ろ過殺菌された変性ステイナー−ショルテ培地を含有する発酵槽(Biolofitte、
フランス国 ポアッシ)に加えた。pHを7.2 に調整し、酢酸(50%)の断続的な
添加により7.2 に維持し、温度を35℃に調整し、35℃に維持した。撹拌速度と通
気速度の調整
により溶解酸素を飽和の20〜30%に維持した。発酵の間発生する泡を分散させる
ために機械的破泡機を用いた。培養の試料(10〜20ml)を培養の期間中(30〜40
時間)発酵槽から断続的に取り出し、濁度(650nmでの光学密度)を測定し、増殖
速度論を追求した。
培養の予備ろ過試料中のPT及びFHA の両方の濃度をエリザ技術により2回測定
した。試験の正確性を確かめるために、試験を公知の濃度のPT及びFHA の2つの
参考試料の存在において行なった。試験の精度は平均で正確な値の+/−20%で
あった。
培養の予備ろ過試料におけるPT及びFHA の同一性及び品質をウェスタンブロッ
トで決定した。PTのブロットは、明白な29KDa,25KDa,24KDa,14KDa及び9KDa
