[0027] In this manner, in the digest of chicken cop which has undergone the proteolytic treatment step, the original protein is decomposed into amino acids and peptides, and it is possible to almost completely separate hyaluronic acid from the tissue. The efficiency of the treatment for dehydrating hyaluronic acid in the cockscomb can be improved.
Such a protein treatment step is an essential step in order to efficiently extract the hyaluronic acid of the chicken wool and reduce the molecular weight.

In the production method of the present invention, it is necessary to carry out the step of reducing the molecular weight of hyaluronic acid, as described above, following the step of treating the protease. Even when the above protease treatment step is completed, the weight-average molecular weight of hyaluronic acid in the digested product of the cockscomb is 100%.
As described previously (JP-B-60-9042 and JP-B-61-21241), at this point, the molecular weight of hyaluronic acid has not been reduced.

As means for reducing the molecular weight of hyaluronic acid, acid treatment, alkali treatment, treatment with a specific enzyme such as hyaluronidase and the like have already been known. However, in these low-molecular-weight treatment steps, there are problems such as mixing of a large amount of salt into the product, coloring, and difficulty in controlling the molecular weight of hyaluronic acid to a constant molecular weight.

In the production method of the present invention, a high-temperature / high-pressure treatment is selected as a means for lowering the molecular weight of hyaluronic acid, so that the molecular weight of hyaluronic acid can be easily adjusted. It is possible to avoid inconvenience in the step of dehydrating hyaluronic acid.
In addition, sterilization can be performed simultaneously by performing high-temperature and high-pressure treatment (if a method other than the above-described high-temperature and high-pressure treatment is selected, sterilization must be performed separately).

The conditions for the high-temperature and high-pressure treatment are to be appropriately selected according to the amount and concentration of digests of the chicken crown to be treated, and are not particularly limited. It is preferable that the temperature is 130 ° C., the processing pressure is 1-2 kg / cm 2 , and the processing time is about 10-600 minutes. When the treatment conditions are more severe than these conditions, hyaluronic acid is excessively decomposed, and the tendency to lower the molecular weight and the tendency to color more than the intended weight average molecular weight often becomes unfavorable, and conversely If the conditions are less relaxed than these conditions, the tendency of hyaluronic acid to become lower in molecular weight tends to be insufficient, and is often unfavorable.

The specific means for applying the high-temperature and high-pressure treatment to the digest of the chicken crown is not particularly limited as long as it can provide desired high-temperature and high-pressure processing conditions. For example, autoclave treatment, pressure kiln Although the processing and the like can be selected, it is preferable to select the autoclave processing in view of the fact that the condition setting is easy, the handling of the apparatus is relatively simple, and the mass processing is easy. In this way, the process of degrading the molecular weight of hyaluronic acid in the digested chicken crown obtained by the proteolytic enzyme treatment can be completed.

When the amount is less than 0.5% by weight of the whole composition in terms of hyaluronic acid, the expected skin effect, physical condition adjusting function, etc. can be substantially achieved in consideration of the amount of food normally consumed by humans. In contrast, if the amount is more than 30.0% by weight, hyaluronic acid becomes more hygroscopic and the processing tends to be difficult. However, the addition of hyaluronic acid in an amount exceeding 30.0% by weight of the total composition in terms of hyaluronic acid does not have any adverse effect on the physical condition adjusting function and the beautiful skin effect of the food composition of the present invention. Since these functions and actions may be enhanced in some cases, the present invention does not exclude the inclusion of hyaluronic acid exceeding 30.0% by weight.

Three strains of hyaluronic acid-degrading bacteria were isolated from the contents of rabbit ileum and cecum and identified as strains belonging to the genus Bacterroides. All three strains were found to completely degrade hyaluronic acid in the culture medium in 24 hours and to be degraded in 6 hours, but still confirmed as hyaluronic acid. This was performed 6 hours after administration of the labeled hyaluronic acid. The measurement of radioactivity in serum was performed by attenuating chemiluminescence by mixing 0.5 mL of serum with 20 mL of Hionic Flow (Packard Japan, Tokyo) and incubating at 37 ° C for 24 hours. . Next, radioactivity was measured with a liquid scintillation counter (Tricarb 4640, Packard Japan KK). Table 1 shows the results.
