Abstract

Metodologia per l'isolamento di fagi filamentosi capaci di legare la biotina, caratterizzato essenzialmente dalle seguenti operazioni:- biotinilazione di una proteina usata per lo screening di una libreria fagica in cui vengono espressi sulla superficie del capside oligopeptidi codificati da inserti nucleotidici a sequenza casuale, introdotti in un gene codificante per il capside fagico con tecniche di manipolazione genetica;- screening della suddetta libreria con la proteina biotinilata; e - isolamento di fagi capaci di legare la biotina.La figura 1 mostra, in forma di istogramma, il legame di proteine biotinilate a fagi selezionati secondo la metodologia dell'invenzione e immobilizzati su plastica.
