Abstract

E' descritto un metodo per la determinazione diretta della concentrazione della frazione libera degli analiti presenti nei fluidi biologici in forma libera ed in forma legata ad uno o più leganti endogeni. Il metodo prevede l'impiego contemporaneo di almeno due specie di leganti esogeni. Il primo legante è in grado di sequestrare una quantità di analita proporzionale alla concentrazione di analita libero presente nel fluido biologico e di rilasciarla, dopo allontanamento dei leganti specifici endogeni, in seguito all'aggiunta di un opportuno agente dissociante. Il secondo è in grado di legare sia l'analita precedentemente rilasciato, sia una versione marcata dell'analita, anche in presenza dell'elemento dissociante. La misura della quantità di analita marcato che si lega al secondo legante esogeno, viene utilizzata per determinare la concentrazione nel fluido dell'analita libero.
