Abstract
1446574 Aryl-octahydroisoquinolines E R SQUIBB & SONS Inc 28 Sept 1973 [5 Oct 1972 (2)] 45576/73 Heading C2C The invention comprises compounds of formula and their pharmaceutically acceptable salts, wherein A-B is -CH 2 -NR 1 - or Z is Ph (possibly substituted), naphthyl or aromatic heterocyclyl; R 1 and R 2 are each H, up to C 8 alkyl (=R 0 ) or alkenyl (either may be Arsubstituted), or heterocyclyl; X and Y are each CO 2 H, CONH 2 , CO 2 (R 0 ), NO 2 , OH, CN, NH 2 , (R 0 )NH, (R 0 ) 2 N, HOCH 2 , (R 0 )CO 2 CH 2 , (R 0 )OCH 2 , NH 2 CH 2 , ArSO 3 CH 2 or halomethyl (or one only of X and Y may be H), or X-Y is - Q-Z<SP>1</SP>-Q- where Q is CO or CH 2 and Z<SP>1</SP> is NH, NR, O, S, CH : CH, CH 2 or CH 2 CH 2 , R being C 1-8 alkyl (possibly Ar-substituted) or Ar; and Ar is Ph or naphthyl, either of which may be substituted. These compounds are prepared by reacting a dienophile with a styryl-tetrahydropyridine, if desired followed by conversion or removal of functional groups in the products, by substantially standard procedures. Intermediates otherwise prepared are styrylpyridines (and their quaternary salts), phenacylpyridines and the corresponding secondary alcohols. Therapeutic compositions having blood pressure lowering, antiinflammatory, antianginal and antiarrhythmic activity comprise compounds of the above formula, and may be administered orally, parenterally or rectally.
