1369525 10(11) - Piperazino - dibenzo[b, f]- thiepins F HOFFMANN-LA ROCHE & CO AG 16 Nov 1972 [16 Nov 1971] 52973/72 Heading C2C Novel compounds I: in which R is hydrogen, halogen, cyano, nitro, C 1-7 alkyl or C 1-7 alkoxy, R 1 is hydrogen, halogen, cyano, carboxy, nitro, C 1-7 alkyl, C 1-7 alkoxy, C 1-7 alkylthio, C 1-7 alkylsulphenyl or amino (C 1-7 alkyl) and R 2 is C 1-7 alkyl phenyl or benzyl are prepared by oxidation (preferably using m-chloroperbenzoic acid) of a compound II: in which R<SP>1</SP> 1 is R 1 excluding C 1-7 alkylsulphinyl (those compounds in which R 1 is C 1-7 alkylsulphinyl being prepared by oxidation of compound in which R 1 is C 1-7 alkyl thio). Compounds II are prepared by reaction of the corresponding 10 - oxo - 10,11 - dihydro - dibenzo[b, f] thiepin with N-(R 2 )-piperazine and those in which neither R nor R<SP>1</SP> 1 are hydrogen are novel. 8 - Chloro - 2 - nitro 10 - oxo - 10,11 - dihydrodibenzo [b,f] thiepin is prepared by reaction of 2 - chloro - 5 - nitro - benzaldehyde with mchlorothiophenol to form 2-[m-chlorophenylthio] 5- nitro benzaldehyde which is reacted with acetylglycine and sodium acetate to form a compound III: which on reaction with hydrochloric and acetic acid forms a pyruvic acid derivative which is reacted with hydrogen peroxide to form the corresponding phenylacetic acid which cyclizes in the presence of polyphosphoric acid to the 10-oxo-dibenzo[b, f]thiepin. Compounds I and II (excluding those in which either R 1 and R 2 is hydrogen) are antidepressants and form with a carrier a pharmaceutical composition which may be administered orally, parenterally or rectally.
