1388915 3 - Nitro - imidazo[1,2-b] - pyridazines AMERICAN CYANAMID CO 23 Feb 1972 [24 Feb 1971 (2) 8 April 1971] 8417/72 Heading C2C Compounds (I), (II) and (III) and their acid addition salts in which R 1 is hydrogen, C 1-8 alkyl phthalimido- C 1-4 alkyl, phenyl-C 1-4 alkyl, C 1-4 alkylamino- C 1-4 alkyl, C 1-4 alkoxy-C 1-4 alkyl hydroxy-C 1-4 alkyl, C 2-4 alkenyl or halobenzoyl-C 1-4 alkyl, R 2 is hydrogen or C 1-4 alkyl and X is oxygen or sulphur; R is C 1-11 alkyl and R 3 is hydrogen or C 1-4 alkyl; R 4 is hydrogen, C 1-6 alkyl, hydroxy- C 1-4 alkyl, C 1-4 alkoxy-C 1-4 alkyl, C 2-4 alkenyl, phenyl-C 1-4 alkyl pyridyl-C 1-4 alkyl, C 3-6 cycloalkyl or di-(C 1-4 alkyl)amino-C 1-4 alkyl, R 5 is hydrogen C 1-4 alkyl or hydroxy-C 1-4 alkyl or together represent piperidinyl, pyrrolidinyl, C 1-4 alkyl-piperazinyl C 1-4 alkoxyphenyl-piperazinyl, morpholino, imidazolyl (C 1-4 alkoxy)- carbonyl - piperazinyl or di - (C 1-4 alkyl)- amino-C 1-4 alkyl-piperazinyl and R 6 is hydrogen or C 1-4 alkyl are prepared by (1) nitrating a 3- desnitro derivative of a compound I, II or III with a nitric acid-sulphuric acid mixture, (2) reacting a compound IV in which Z is alkali metal with a compound R 1 -Y in which Y is halogen to form a compound I, (3) reacting a compound V with a compound R 1 -X-Z to form a compound I (4) acylating a compound VI with the anhydride or acid chloride of to form a compound II or (5) reacting a compound VII in which X<SP>1</SP> is halogen with a compound to form a compound III. Compounds I, II and III show anti-protozoal activity and form with a carrier or diluent a pharmaceutical composition which may be administered orally, parenterally or rectally.
