Abstract

1350520 Bis-basic esters and amides of xanthene and xanthone RICHARDSONMERRELL Inc 23 June 1972 [14 July 1971] 29594/72 Heading C2C Novel compounds of the Formula I and pharmaceutically acceptable acid addition salts thereof wherein Z is oxygen or H 2 ; each R<SP>1</SP> and R<SP>2</SP> is hydrogen, C 1-6 alkyl, C 3-6 cycloalkyl, C 3-6 alkenyl in which the double bond is other than in the 1-position or each set of R<SP>1</SP> and R<SP>2</SP> together with the attached nitrogen is pyrrolidino, piperidine, an N-(C 1-6 alkyl) piperazino or morpholino; each A is C 2-8 alkylene which separates Y and the amino nitrogen by at least two carbon atoms; Y is oxygen, or N-R, where R is H or C 1-4 alkyl may be prepared by (a) reacting a compound II wherein W is hydroxy, halogen, azolide, C 1-6 alkoxy or O-metal with an amine III wherein R<SP>4</SP> and R<SP>5</SP> are as R<SP>1</SP> and R<SP>2</SP> with the proviso that when R<SP>4</SP> or R<SP>5</SP> is hydrogen, the amino group of the amino alcohol is protected during the reaction and subsequently the protective group is removed; (b) wherein Y is oxygen, reacting a metallic salt of an acid II with a compound III; (c) reacting a compound IV wherein Hal is chlorine, bromine or iodine with an amine R<SP>6</SP>R<SP>7</SP>NH, wherein R<SP>6</SP> and R<SP>7</SP> are C 1-6 alkyl or together with the N-atom to which they are attached form a pyrrolidino, piperidino, an N-(C 1-6 alkyl) piperazino or morpholino; (d) wherein R<SP>1</SP> and R<SP>2</SP> are hydrogen, reducing a compound. (e) wherein Z is oxygen, oxidizing the corresponding xanthene in which Z is H 2 ; (f) wherein Z is H 2 , reducing the corresponding 9-oxoxanthene. Intermediate acid chlorides of the Formula II, wherein W is chlorine may be prepared by reacting the free acid with thionyl chloride. Pharmaceutical compositions of the compounds I show antiviral activity when administered orally, topically, parenterally or buccally with the usual excipients.
