1311286 Migraine treatment H WILD 4 Dec 1970 [5 Dec 1969] 59643/69 Heading A5B Pharmaceutical compositions for the treatment and prevention of migraine comprise (a) a 1, 4-disubstituted piperazine having the general formula: wherein X is H, Cl or Br and R<SP>1</SP> and R<SP>2</SP> each represent H or a C 1-6 alkyl, at least one of R<SP>1</SP> and R<SP>2</SP> being an alkyl group, or a salt thereof e.g. salts with hydrochloric, phosphoric, lactic, maleic, fumaric and tartaric acids, and (b) N-acetyl-p-aminophenol (paracetamol), pethoxyacetanilide (phenacetin), salicylamide and/or acetylsalicylic acid, the weight ratio of (a) to (b) being from 1:20 to 1:200. A preferred composition comprises 1-p-chlorobenzhydryl-4- p-(tertiary butyl) benzyl piperazine (buclizine) and paracetamol. The compositions may be administered orally or rectally in the form of tablets, capsules, suspensions, syrups, and suppositories. Migraine may be prevented by the simultaneous administration of dosage units comprising appropriate quantities of (a) and (b) and the invention also relates to a pharmaceutical package comprising separate dosage unit forms containing (a) and (b). The composition may also be formulated as a dosage form divided into separate portions by an internal division which is easily ruptured or dissolved by body fluid, compounds (a) and (b) being confined to different compartments.
