Abstract
A találmány tárgyát képezi eljárás a thyA-Vibrio cholerae törzselőállítására, amely eljárás irányított mutagenezis lépést tartalmaz aV. cholerae kromoszómáján lévő thyA-gén (1. ábra, a szekvencialistában1. azonosítószámon megadott szekvencia) lokuszán. A találmány tárgyatovábbá működésképtelen thyA-gént tartalmazó Vibrio cholerae DthyAtörzs. E törzs tartalmazhat egy vagy több episzómális eredetű, autonómreplikálódó DNS-elemet, pl. plazmidot, amely hordozhat idegen, példáulE. coli eredetű, működésképes thyA-gént, továbbá homológ vagyheterológ proteint kódoló struktúrgént. Továbbá, a találmány tárgyátképezi a thyA-struktúrgén és szegélyező régióinaknukleotidszekvenciái, továbbá általuk kódolt proteinek (4. és 5. ábrántalálható szekvenciák). Ezenfelül, a találmány tárgyát képezi atalálmány szerinti Vibrio cholerae DthyA törzset vagy thyA- törzsettartalmazó vakcina. A találmány szerinti megoldás előnyösenalkalmazható a a protein-előállítás területén, ahol szelekcióra nyújtlehetőséget, továbbá vakcinázáshoz. Ó
