Claims (5)

1. In vitro postupak za razvoj monoklonskih antitijela, pri čemu navedeni postupak sadrži korake: (a) obogaćivanja izvornog materijala hvatanjem specifičnih stanica na čijoj su površini antitijela izložena na plastičnim površinama napunjenim antigenom pri čemu to hvatanje uključuje podloge za hvatanje koje sadrži 0,5% tež/ob BSA i 0,1% tež/ob NaN3 (b) omplifikacije cDNK kodiranja za teški (VH) i laki (VL) lanac promjenljive domene; (c) konstrukcija kombinatorne teške (VH) i lake (VL) knjižnice DNK formata plazmida ekspresije sisavaca (d) identifikacije propisnih za antigen specifičnih kombinacija vezivanja teških (VH) - lakih (VL) lanaca promjenljive domene nakon eksprimiranja antitijela u stanicama sisavaca.
2. Postupak sukladno patentnom zahtjevu 1, pri čemu se stanice izvora izabiru od skupine koju čine stanice slezene iz sirovog homogenata slezene, koštane srži i stanica periferne krvi iz imunizirane životinje.
3. Postupak sukladno patentnom zahtjevu 1 ili 2, pri čemu koraku a) prethodi korak predapsorpcije gdje su stanice izvora kratko pre-adsorbirane s prekomjernom količinom antigena koji nisu ciljani.
4. Postupak za proizvodnju monoklonskih antitijela, pri čemu se antitijela razvijaju sukladno bilo kojem od prethodnih patentnih zahtjeva, i stanice sisavaca identificirane u koraku d) dalje kultiviraju u velikoj količini za proizvodnju antitijela.
5. Postupak sukladno patentnom zahtjevu 3, pri čemu su ta antitijela bez antigena koji nisu ciljani.
