Claims (9)
1. Postupak za proizvodnju šećera ili šećera od lignoceluloznog materijala, koji obuhvaća:
a) predobradu lignoceluloznog materijala prije enzimatske obrade izlaganjem (vrućoj) vodi, pari (eksplozija pare), kiselini, bazi, otapalu, toplini, peroksidu, ozonu, mehaničkom usitnjavanju, brušenju, mljevenju ili brzom smanjenju tlaka, kombinaciji bilo koje dvije ili više njih;
b) predsaharifikaciju lignoceluloznog materijala inkubacijom s enzimom ili smjesom enzima;
c) dovođenje u kontakt lignoceluloznog materijala s (i) polipeptidom koji sadrži aminokiselinsku sekvencu navedenu u SEQ ID NO:2 ili njegovom varijantom polipeptida, pri čemu varijanta polipeptida ima najmanje 98% istovjetnosti sekvence sa sekvencom navedenom u SEQ ID NO:2, pri čemu polipeptid ima aktivnost beta-glukozidaze i pri čemu varijanta polipeptida ima katalitički ostatak Asp277 - Glu505 (položaji kao u SEQ ID NO: 2), ili (ii) pripravkom koji sadrži polipeptid iz (i).
2. Postupak u skladu s patentnim zahtjevom 1, naznačen time što se temperatura koraka predsaharifikacije kreće u rasponu od 20°C do 85°C.
3. Postupak u skladu s patentnim zahtjevom 1 ili 2, naznačen time što se pH smjese predsaharifikacije kreće u rasponu od 3,0 do 7,0.
4. Postupak u skladu s bilo kojim od patentnih zahtjeva 1 do 3, naznačen time što se predsaharifikacija odvija u vremenu od 2 sata do 48 sati.
5. Postupak prema bilo kojem od patentnih zahtjeva 1 do 4, naznačen time što pripravak koji sadrži polipeptid iz (i) nadalje sadrži celulazu i/ili hemicelulazu i/ili pektinazu.
6. Postupak u skladu s patentnim zahtjevom 5, naznačen time što je celulaza celobiohidrolaza, endo-beta-1,4-glukanaza, beta-glukozidaza ili beta-(1,3)(1,4)-glukanaza.
7. Postupak u skladu s patentnim zahtjevom 5 ili 6, naznačen time što je hemicelulaza endoksilanaza, β- ksilozidaza, α-L-arabionofuranozidaza, α-D-glukuronidaza, acetil-ksilan esteraza, feruloil esteraza, kumaroil esteraza, α-galaktozidaza, β-galaktozidaza, β-mananaza ili β- manozidaza.
8. Postupak prema bilo kojem od patentnih zahtjeva 5 do 7, naznačen time što je pektinaza endo poligalakturonaza, pektin metil esteraza, endo-galaktanaza, beta-galaktozidaza, pektin acetil esteraza, endo-pektin liaza, pektat liaza, alfa ramnozidaza, egzo-galakturonaza, egzo-poligalakturonat liaza, ramnogalakturonan hidrolaza, ramnogalakturonan liaza, ramnogalakturonan acetil esteraza, ramnogalakturonan galakturonohidrolaza ili ksilogalakturonaza.
9. Postupak prema bilo kojem od patentnih zahtjeva 1 do 8, naznačen time što pripravak nadalje sadrži enzim iz obitelji GH61 ili drugi enzim koji ima aktivnost pojačavanja celulaze.
