Claims (12)
1. Varijantni polipeptid koji sadrži supstituciju G142S na položaju koji odgovara položaju 142 polipeptida iz SEQ ID NO: 2, pri čemu varijantni polipeptid ima aktivnost beta-glukozidaze i najmanje 80% identičnosti sekvence s polipeptidom iz SEQ ID NO: 2 i pri čemu je varijanta polipeptida manje osjetljiva na inhibiciju glukoze u odnosu na matični polipeptid prikazan u SEQ ID NO: 2.
2. Varijantni polipeptid prema patentnom zahtjevu 1, naznačena time što nadalje sadrži supstituciju na položaju koji odgovara položaju 90, 103, 335, 485 i/ili 606 polipeptida iz SEQ ID NO: 2.
3. Polinukleotid naznačen time što kodira varijantni polipeptid prema patentnom zahtjevu 1 ili 2.
4. Konstrukt nukleinske kiseline ili vektor naznačen time što sadrži polinukleotid prema patentnom zahtjevu 3.
5. Stanica domaćina naznačena time što sadrži polinukleotid prema patentnom zahtjevu 3 ili konstrukt nukleinske kiseline ili vektor prema patentnom zahtjevu 4.
6. Stanica domaćina prema patentnom zahtjevu 5, naznačena time što je stanica gljivična stanica.
7. Postupak za proizvodnju varijantnog polipeptida prema patentnom zahtjevu 1 ili 2, naznačen time što postupak obuhvaća korake:
a) kultiviranje stanice domaćina prema patentnom zahtjevu 5 ili 6 pod uvjetima koji pogoduju proizvodnji varijantnog polipeptida, i
b) po izboru, oporavljanje varijantnog polipeptida.
8. Pripravak naznačen time što sadrži:
(i) varijantni polipeptid prema patentnom zahtjevu 1 ili 2, i
(ii) celulazu i/ili hemicelulazu i/ili pektinazu.
9. Pripravak prema patentnom zahtjevu 8, naznačen time što je celulaza odabrana iz skupine koja sadrži litički polisaharid monooksigenazu, celobiohidrolazu I, celobiohidrolazu II, endo-beta-1,4-glukanazu, beta-glukozidazu i beta-(1,3)(1,4)-glukanazu ili bilo koju njihovu kombinaciju i hemicelulaza je odabrana iz skupine koja sadrži endoksilanazu, beta-ksilozidazu, alfa-L-arabinofuranozidazu, alfa-D-glukuronidazu, acetil-ksilan esterazu, feruloil esterazu, kumaroil esterazu, alfa-galaktozidazu, beta-galaktozidazu, beta-mananazu, beta-manozidazu ili bilo koju njihovu kombinaciju.
10. Pripravak prema patentnom zahtjevu 8 ili 9, naznačen time što da je pripravak čitava fermentacijska juha.
11. Postupak za obradu supstrata koji sadrži celulozu i/ili hemicelulozu koji postupak uključuje korak dovođenja u kontakt supstrata s varijantnim polipeptidom prema patentnom zahtjevu 1 ili 2 i/ili s pripravkom prema bilo kojem od patentnih zahtjeva 8 do 10.
12. Postupak proizvodnje fermentacijskog proizvoda, koji postupak uključuje slijedeće korake:
a) tretiranje supstrata korištenjem postupka u skladu s patentnim zahtjevom 11, i
b) fermentiranje dobivenog materijala radi dobivanja fermentacijskog proizvoda.
