Abstract

Fusidic acid which is an antibiotic designated ZN-6 and contains a steroid nucleus is prepared by growing a mutant of the fungus Fusidium coccineum Fuck K. Tubuki capable of forming the said antibiotic under aerobic conditions in an aqueous nutrient solution containing sources of carbon and nitrogen and inorganic salts, the fermentation being continued until a substantial antibiotic activity has been imparted to said solution and recovering the antibiotic therefrom. The mutant of the fungus may be obtained by irradiating a spore suspension with ionizing or non-ionizing rays or the spores may be exposed to the action of mutagenic chemicals such as diethyl sulphute or mustard gas. The antibiotic may be isolated from the solution by filtering, and either adding an adsorbing agent to the filtered solution and eluting the antibiotic therefrom, or adding an amine or an amine salt to convert the antibiotic into a form soluble in an organic solvent which is substantially water immiscible. Alternatively an acid may be added to the filtered broth to ensure that the antibiotic is present as the sparingly water-soluble free acid. Specific mutants of the above fungus are designated Leo-U42, Leo-U79 and Fc-U 404 and are deposited at the Commonwealth Mycological Institute. Specification 930,786 is referred to.
