Abstract

The invention comprises compounds of the formula <FORM:0994023/C2/1> (wherein A is hydrogen, alkyl or alkenyl; X, Y and Z are hydrogen, halogen, alkoxy, amino, alkylamino, acylamino, C1- 6 aryl, C1- 6 alkyl-mercapto, C1- 6 alkenylmercapto, sulphamoyl, C1- 6 alkylsulphamoyl or di (C1- 6 alkyl) sulphamoyl, with the provisos that (a) not all of X, Y and Z are hydrogen; (b) if A is hydrogen or C1- 5 alkyl and both X and Z are hydrogen, Y is not hydrogen, chlorine, bromine, amino, alkylamino or sulphamoyl; (c) if A is C1- 5 alkyl and both X and Y are hydrogen, Z is not hydrogen, bromine or chlorine; (d) if A is alkyl of 6 or more carbon atoms and both X and Z are hydrogen, Y is not hydrogen, amono or sulphamoyl; W is alkylene; and R1 and R2 are C1- 6 alkyl or together with N form a five or six membered heterocyclic ring) and acid-addition salts thereof; and their preparation by reacting a substituted diamine of the formula H2NWNR1R2 with the appropriately substituted o-alkoxy-, o-hydroxy- or o-alkenyloxy-, benzoyl chloride. Examples describe the preparation of compounds of the invention by this process and also (1) the preparation of N-(2-diethylaminoethyl) - 2 - methoxy - 5 - nitro - benzamide hydrochloride from the appropriate diamine and acid chloride, and its reduction to the 5-amino compound; (2) the chlorination of N - (2 - diethylaminoethyl) - 2 - methoxy - 4-acetamidobenzamide to give N-(2 - diethylaminoethyl) - 2 - methoxy - 4 - acetamido - 5-chloro-benzamide and the hydrolysis of this to the 4-amino compound; and (3) the preparation of N - (2 - diethylaminoethyl) - 2 - methoxy-4 - nicotinoylaminobenzamide by acylation of the 4-amino compound. Substituted benzyl chlorides used as starting materials are prepared from the corresponding substituted benzoic acids and thionyl chloride. The benzamides of the invention, which are stated to be anti-emetics, anti-naupathic agents, tranquillizers, analgesics, anticonvulsants, antifibrillants, antihistamines, antitussives and antipruriginous agents, may be made up into pharmaceutical compositions with suitable carriers. They may take the form of dragees, lozenges, tablets, aerosols, injection solutions, suppositories, granules, pills, syrups or oral drops. Compositions containing N-(2-diethylaminoethyl) - 2 - methoxy - 3 - chloro - 5 - bromobenzamide phosphate and N - (2 - diethylaminoethyl) - 2 - methoxy - 4 - amino - 5 - chlorobenzamide dihydrochloride are described.
