Abstract
3 - Acetamido - 5 - amino - 2,4,6 - tri - iodobenzoic acid is prepared by reacting 3-acetamido - 5 - amino - 2,6 - di - iodobenzoic acid in an aqueous medium having a pH less than 2, with iodine chloride or an alkali metal iododichloride at a temperature below 70 DEG C. The 3-acetamido - 5 - amino - 2,6 - di - iodobenzoic acid may be produced in situ by the addition of at least 3 equivalents, preferably 3.3 equivalents, of iodine chloride or alkali metal iododichloride to an aqueous solution of 3-acetamido-5-aminobenzoic acid. The di-iodo derivative is first precipitated, and then the di-iodo derivative reacts with the remaining iodine chloride or alkali metal iododichloride to produce the tri-iodo derivative. The 3-acetamido-5-amino-2,6-di-iodobenzoic acid is preferably used in the form of an aqueous suspension containing not more than 6% by weight of the di-iodo derivative and also containing at least a 0.5 molar excess of acid over that required to neutralize the di-iodo derivative. In the one step process, an aqueous solution containing 3-acetamido-5-aminobenzoic acid in a proportion of not more than 3% by weight and one mol. of acid is preferably used.
